[Determination of beryllium in geological samples with slurry sampling and probe atomization in graphite furnace atomic absorption spectrometry].

A method of loading a slurry prepared from a solid sample for analysis of beryllium in geological samples using graphite furnace atomic absorption spectrometry was approached by combining slurry sampling with probe atomization. The factors that influence the performances of beryllium including ashing and atomization temperature, atomization surfaces (probe and tube), the materials of the tube and probe, and the use of matrix modifiers were investigated. The results show that the determination of beryllium in geological samples could be performed by direct slurry sampling with the use of calcium as the matrix modifier for a calibration established with aqueous standards. The relative standard deviations (RSD) of the method were 3.1-5.2%. The analytical results of the geological reference samples were consistent with the certified values. The activation energy during beryllium atomization was calculated using the Arrhenius graph method which was in good with the bonding energy of Be-O.

[Immunohistochemical study on the distribution and coexistence of SP, VIP and NPY in the rat submandibular gland].

The distribution and coexistence of SP, VIP and NPY in the submandibur gland of rat have been studied with immunohistochemical ABC methods. The results indicated that SP, VIP and NPY-IR nerve fibers were presented as varicose and linear profiles, which mainly travelled around acini, duct and blood vessels. The ganglion cells in submandibular gland showed SP, VIP and NPY-IR all positive immunocytochemically. The findings indicated that the neuropeptides were coexisted in the same neuron body and might play the role of regulation of the secretary activity and effective blood supply.

[Extraction, purification and ascertainment of anti-hepatoma active principles for targeted preparation of radix Aconiti Kusnezoffii].

The extraction, purification, pharmacological experiments and chemical analysis of anti-hepatoma active principles of Radix Aconiti Kusnezoffii have been studied. The results show that the medicinal material Radix Aconiti Kusnezoffii is efficacious in inhibiting hepatoma, and the active principles are mainly made up of poisonous ester alkaloids. The liver targeting delivery system will be the first choice for its anti-hepatoma preparation, and the effective component AY3a is fit for the preparation of targeted microspheres.

Evaluation of transforming growth factor beta and bone morphogenetic protein composite on healing of bone defects.

OBJECTIVE: To evaluate the effect of transforming growth factor-beta/bone morphogenetic protein (TGF-beta/BMP) composite on the healing of large segmental bone defects and to discuss the interaction between TGF-beta and BMP during bone repair. METHODS: A 1.5 cm segmental defect was made in the mid-upper part of the radial shaft of 48 adult rabbits. The defects were filled with implants of TGF-beta/carrier, BMP/carrier or TGF-beta/BMP/carrier separately. 120 micrograms of purified bovine TGF-beta and 12 mg of BMP were used in the composite. The defects were examined radiographically and histologically at 4, 8, 12 and 16 weeks post-operation. RESULTS: In the group filled with TGF-beta/BMP composite, the defect areas were bridged at 4 weeks, with callus of uniform radiodensity. Cortices of the cut ends were obscured by new bone. By 16 weeks post-operation, the defects were bridged by uniform new bone and the cut ends of the cortex could not be seen in all groups. In group of BMP/carrier, the defects were filled with more irregular woven bone callus than in other two groups. TGF-beta/BMP implanted defects in animals killed at 16 weeks showed histologically new lamella and woven bone that was formed in continuity with the cut ends of the cortex. Medullar canal was recanalized and contained marrow elements with normal appearance. CONCLUSIONS: These data demonstrate not only the synergistic action between TGF-beta and BMP in the process of bone healing, but also a better effect of TGF-beta/BMP composite than single TGF-beta or BMP on bone repair, especially in the early stage of bone repair process.

Renal disease in pregnancy.

The incidence of acute renal failure in South Africa has been reduced and is a reflection of improved obstetric care. A report of eclampsia in the UK suggests that eclampsia with its attendant risk of acute renal failure may never be eliminated completely. There has been progress in understanding the changes in endothelial cell metabolism and pre-eclampsia but there has been limited success in improving treatment. The development of hypertension or pre-eclampsia during pregnancy may predict the development of hypertension, but it rarely indicates underlying renal disease. It appears that pregnancy does not adversely affect the course of renal disease if renal function is normal at conception, but it may hasten the decline in renal function in women who have impaired renal function at conception. The frequency of complication both for mother and for child remain high in such pregnancies.

Management of pregnancy in women undergoing continuous ambulatory peritoneal dialysis.

A survey in which there were responses from dialysis units caring for approximately 38% of women aged 14-44 receiving continuous ambulatory peritoneal dialysis in the United States revealed a conception rate of 0.2% per year. A total of 44 pregnancies were identified in women treated with peritoneal dialysis at some time during their pregnancies. Two thirds of pregnancies were complicated by hypertension and one third by severe hypertension. There was little difficulty with catheter placement during pregnancy, and peritonitis was an unusual complication, which did not result in fetal loss. Forty-eight percent of pregnancies resulted in surviving infants. All but one were premature and two thirds were small for gestational age.

[Mesh-like tubular stent treatment for chronic urinary retention caused by benign prostatic hypertrophy].

42 patients with chronic urinary retention caused by BPH were treated by means of Mesh-like tubular stent placing from August 1993 to August 1994. The mesh-like tubular stent was made of nickel titanium alloy in sizes of 2.0 x 1.5 cm, 2.5 x 1.5 cm, 3.0 x 1.5 cm respectively. Under direct vision the stent was placed with the deployment tool. The desirable position of the stent is 0.3-0.5 cm proximal to the internal urethral orifice, 0.5 cm distal to the verumontanum. In 38 patients followed up for 1-6 months, 34 were succesful with an effective rate of 80%. IPSS, PUFR, RUV after 1, 3, 6 months of placement were 8.5 +/- 4.4, 6.8 +/- 3.5, 7.1 +/- 5.2; 12.5 +/- 6.0 ml/s, 14.0 +/- 5.2 ml/s, 14.4 +/- 8.1 ml/s; 43 +/- 11.5 ml, 47.4 +/- 13.3 ml, 44.5 +/- 15.7 ml respectively. Four stents were taken away transurethrally in the unsuccessful cases. The results indicat that there are no serious complications except for shorttime hematuria, fever, and bladder irritation. The success depends chiefly upon exactly meassuring the length of prostatic urethra, selecting the rational size of the stent, and correctly placing the stent.

[The relationship between p53 and PCNA in germ cell carcinoma].

To analyse the relationship between cellular proliferating status, p53 expressions and tumor biological characteristics, immunohistochemistry was used in 13 cases of testicular germ cell carcinoma paraffin section. The results show that PCNA-LI is higher significantly in invasive tumor group than in noninvasive group (P < 0.05). Although there were no significant differences of PCNA-LI, p53 between seminoma and nonseminoma, two parameters in the latter group were somewhat higher. There was a correlation of PCNA-LI with p53 in nonseminoma group (r = 0.64). It is concluded that PCNA-LI and p53 are both correlated positively with tumor differentiation.

[Reasonability of extracting procedures for huiyangfanben decoction].

The reasonability of extracting procedures for traditional Chinese prescriptions was studied with Huiyangfanben Decoction as sample. The comparative results of the ingredients in extractions prepared by different procedures revealed. For the prescriptions with complicated ingredients, more attention should be paid to the ingredients and their characteristics when designing the extracting procedurs, and in extraction the drugs should be classified according to the interaction of the ingredients. More extracting solvents could be used according to the characteristics of the active ingredients.

Subverting lymph node trafficking by treatment with the Mel-14 monoclonal antibody to L-selectin does not prevent an effective host response to Sendai virus.

A single 250-micrograms dose of the Mel-14 mAb to L-selectin greatly diminished the extent of L-selectin expression on lymphocytes and decreased (60 to 90%) the massive cellular recruitment to the cervical and mediastinal lymph nodes that follows intranasal infection of naive C57BL/6 mice with Sendai virus. The numbers of CD8+ CTL precursors in the mediastinal lymph nodes were considerably reduced on day 7, when compared with virus-infected mice given a control rat IgG2a, but potent CTL effectors were present in the lungs of both groups by day 10 after infection, and the overall magnitude of CTL precursor generation was not obviously compromised. The early dominance of Sendai virus-specific IgM Ab-forming cells was prolonged in the Mel-14-treated mice, whereas plasma cells producing virus-specific IgA were abnormally prominent in the lymph nodes but not in the spleen. The kinetics of virus-specific Ab-forming cells generation and the serum Ab response for the various IgG isotypes were also delayed. Thus, though L-selectin is clearly important for the localization of naive lymphocytes to regional lymph nodes, the Mel-14-treated mouse can still deal effectively with a virus that causes productive infection only in the respiratory tract. The spleen, where L-selectin does not determine lymphocyte trafficking, is a major site for the compensatory T cell and B cell responses.

Temporal loss of the activated L-selectin-low phenotype for virus-specific CD8+ memory T cells.

Whether the L-selectin-low (L-sel-lo) phenotype of acutely stimulated CD8+ T cells is a permanent characteristic of long-term memory CTL precursors (p) is addressed for mice primed with an influenza A virus or the murine parainfluenza type 1 virus, Sendai virus. In both cases, many of the splenic CD8+ CTLp gradually lose the predominantly L-sel-lo profile associated with recently generated CTLp populations. The influenza-specific CTLp also tend to revert from the activated alpha 4-integrin-high to the resting alpha 4-integrin-low form. The kinetics of the switch back to the "naive" L-sel-hi phenotype differs for the influenza and Sendai virus models, perhaps reflecting events occurring during the acute phases of these responses. The return to being L-sel-hi is not due to irreversible lymphocyte senescence, because restimulation of this set with the inducing virus in vitro causes most of the cells to become L-sel-lo. Also, despite the time-related drift of these particular memory CTLp to the L-sel-hi state, the size of the total pool of L-sel-lo CD8+ T cells increases with age.

Recruitment and proliferation of CD8+ T cells in respiratory virus infections.

The dramatic increase in the cellularity of the mediastinal lymph nodes (MLN) of mice infected intranasally (i.n.) with influenza viruses is a consequence of both recruitment and proliferation. As many as 20% of the CD8+ subset in the MLN can be shown to be in S or G2 + M phase at 6 days after i.n. challenge with the HKx31 influenza A virus, the percentage of of cycling cells being approximately five times greater for the activated/memory substantial evidence of apoptosis was found for CD8+ T cells recovered from the MLN and lung, particularly at 5 and 7 days after infection. Less than 1/100 of the proliferating T cells could be shown, by limiting dilution analysis (LDA), to be influenza virus-specific CD8+ cytotoxic T lymphocyte precursors (CTLp). A single, low dose (20 mg/kg) of the DNA-targeted drug cyclophosphamide (Cy) caused a massive decrease in frequency for the responding CD8+ CTLp, though the mice survived infection with the HKx31 virus and there was no long-term exhaustion of the CTLp pool in the MLN, spleen, or lung. The Cy treatment was also followed by a smaller reduction in the prevalence of memory CTLp (specific for Sendai virus) that were present concurrently in the regional lymph node, indicating that a measure of bystander activation is occurring. The experiments show that respiratory virus infections have no negative impact on established T cell memory, and that there is no phase of transient exhaustion in the acute virus-specific CTLp response in this localized infection.

Clearance of Sendai virus by CD8+ T cells requires direct targeting to virus-infected epithelium.

Minimal numbers of CD8+ T cells are found in bronchoalveolar lavage (BAL) populations recovered from Sendai virus-infected mice that are homozygous (-/-) for a beta 2-microglobulin (beta 2-m) gene disruption. The prevalence of the CD8+ set was substantially increased in the pneumonic lungs of 8-12-week radiation chimeras made using substantially class I major histocompatibility complex (MHC) glycoprotein-negative beta 2-m (-/-) recipients and normal beta 2-m (+/+) bone marrow. Even so, the CD8+ (but not the CD4+) lymphocyte counts were still much lower than in the (+/+)-->(+/+) controls. The (+/+)-->(+/+) and (+/+)-->(-/-) chimeras cleared Sendai virus and potent virus-immune CD8+ cytotoxic T lymphocytes (CTL) specific for H-2Kb+viral nucleoprotein peptide were found in the BAL from both groups. However, following in vivo depletion of the CD4+ population, only the (+/+)-->(+/+) mice were able to deal with the infection. Similarly, adoptively transferred, H-2Kb-restricted CD8+ T cells from previously-primed (+/+) mice also failed to clear virus from the lungs of (+/+)-->(-/-) chimeras infected within 2 weeks of reconstitution with bone marrow, though they were effective in the (+/+)-->(+/+) controls. Sendai virus-immune CD8+ T cells are thus unable to eliminate virus-infected beta 2-m (-/-) lung epithelial cells that might be thought to be expressing very small amounts of either isolated class I heavy chain, or class I MHC glycoprotein that has bound beta 2-m derived from beta 2-m (+/+) T cells or macrophages present in the pneumonic lung. Furthermore, the CD8+ CTL that are being exposed to beta 2-m (+/+) stimulators in the BAL population cannot operate in some bystander mode to clear virus from respiratory epithelium.

Host response to Sendai virus in mice lacking class II major histocompatibility complex glycoproteins.

The development of Sendai virus-specific cytotoxic T-lymphocyte (CTL) effectors and precursors (CTLp) has been compared for mice that are homozygous (-/-) for a disruption of the H-2I-Ab class II major histocompatibility complex glycoprotein and for normal (+/+) controls. The generation of CD8+ CTLp was not diminished in the -/- mice, though they failed to make virus-specific immunoglobulin G class antibodies. While the cellularity of the regional lymph nodes was decreased, the inflammatory process assayed by bronchoalveolar lavage (BAL) of the pneumonic lung was not modified, and potent CTL effectors were present in BAL populations recovered from both groups at day 10 after infection. There was little effect on virus clearance. Production of interleukin-2 by both freshly isolated BAL inflammatory cells and cultured lymph node cells was greatly diminished, though the -/- mice still made substantial levels of gamma interferon. However, treating the mice with a single dose of a monoclonal antibody to this cytokine, at least some of which is made by CD8+ T cells, did not decrease CTLp frequencies. As found previously with CD4-depleted H-2b mice, the development of Sendai virus-specific CD8+ T-cell-mediated immunity is not compromised by the absence of a concurrent class II major histocompatibility complex-restricted response.

[Transrectal hyperthermia in the treatment of benign prostatic hyperplasia].

A group of 53 patients with benign prostatic hyperplasia underwent transrectal hyperthermia using Prostathermer (Biodan) from September 1992 to March 1993. 41 of the 53 patients were followed up for 6 months. The subjective symptoms improved in 75.6% (31/41) of the treated patients. The mean maximum urine flow rate increased by 3.1 +/- 2.6 ml/s. The mean residual urine reduced 38.4 +/- 25.2 ml. The mean prostate volume remained almost unchanged. The results showed that transrectal hyperthermia of the prostate gland is as effective as transurethral approach for the treatment of benign prostatic hyperplasia.

[Excision of mid-lower rectal carcinoma through abdomen and anogenital space].

28 men with male mid and lower rectal carcinoma were treated by dissecting sigmoid colon and rectum in the abdomen through the anogenital space, which refers to the potential coronoid space located between the deep transverse perineal muscles and anal sphincter muscles. The rectal segment was pulled out through the anogenital space to accomplish resection and anastomosis outside the pelvic cavity. Follow-up for 9-57 months showed that 26 cases (92.9%) survived without recurrence of carcinoma and 1 had local recurrence. The advantages of the procedure include clear exploration, minimal trauma, super-lower anastomoses under direct vision, good recovery of defecating function, and less contamination of pelviccavity.
