A-6G variant of the angiotensinogen gene and essential hypertension in Han, Tibetan, and Yi populations.

To investigate the relationship between the A-6G variant in the promoter of the angiotensinogen gene and essential hypertension in Han, Tibetan, and Yi populations. All patients with essential hypertension were selected by WHO criteria. And the polymorphism of the A-6G variant was determined by PCR/RFLP. The data were analyzed by t test and chi2 test. There was no significant difference in the genotype or allele frequencies between normotensives and hypertensives in the Han, Tibetan, and Yi populations, respectively. However, when the subjects were divided into male and female subgroups, the genotype distributions among hypertensives and normotensives of the Tibetan female group were as follows: AA, 37% vs. 48%; AG, 52% vs. 48%; GG, 11% vs. 4%, respectively and the frequency of the G allele was significantly higher in hypertensives than in normotensives in the Tibetan female group (0.37 vs. 0.28, chi2=4.25, p<0.05). In addition, we observed that there was a significant difference between the Han and Tibetan normotensive groups in the distributions of the allele and genotype frequencies of the A-6G variant. The frequency of the G allele was 0.29 and 0.17 in the Tibetan normotensive and Han groups, respectively (p<0.001). The G allele of the A-6G variant was associated with hypertension in the Tibetan females, but not in the Yi or Han females. And we confirmed that there was a significant difference in the prevalence of the allele frequencies of the A-6G variant between the Han and Tibetan normotensive groups.

[Effect of lower androgen levels on arteriosclerosis].

OBJECTIVE: To analyse the effect of lower androgen levels on risk factors for arteriosclerosis of men. METHODS: In 30 cases of primary prostate carcinoma (stage A), serum levels of testosterone (T), free testosterone (FT), dehydroepiandrosterone (DHEA), sex hormone-binding globulin (SHBG), prostatic specific antigen(PSA), triglyceride (TG), total cholesterol (TC), high density lipoprotein-cholesterol (HDL-C), low density lipoprotein-cholesterol (LDL-C), apoprotein alpha 1 (APO alpha 1) and apoprotein beta(APO beta), insulin, plasma fibrinopeptide A (FPA), plasminogen activator inhibitor-1(PAI-1), fibrinogen were determined before and 1 week, 1,4,8 months after castration. RESULTS: T, FT, PSA decreased significantly 1 week after castration, and kept on decreasing afterwards. DHEA and SHBG were not changed. TG, fasting insulin and glucose, 2-hour insulin and glucose elevated significantly 1 month after castration. TG, LDL-C, FPA and PAI-1 elevated 4 months after castration. Insulin sensitive index(ISI) decreased significantly 1 month after operation (-4.4 +/- 0.4 vs. -3.5 +/- 0.4, P < 0.01). HDL-C, APO alpha 1, APO beta and fibrinogen were not changed. There was a negative linear correlation between FT and TG, TC, LDL-C, PAI-1, FPA, fasting insulin and glucose, 2 h insulin and glucose, so did T. CONCLUSION: Lower androgen levels have adverse effect on lipid metabolism, coagulative function and insulin sensitivity, which are related to arteriosclerosis of men.

The relationship between cartilage end-plate calcification and disc degeneration: an experimental study.

OBJECTIVE: To study the relationship between cartilage end-plate calcification and intervertebral disc degeneration. METHODS: An experimental model of cervical disc degeneration in rabbits was established by resection of the cervical supraspinous and interspinous ligaments and detachment of the posterior paravertebral muscles from the cervical vertebrae. Mechanical instability in the cervical spine elicited by this surgical intervention accelerated the process of intervertebral disc degeneration. The extent of intervertebral disc degeneration was graded in morphologically, and the thicknesses of the calcified layer and the uncalcified layer of the cartilage end-plate were measured in each degenerated cervical disc. RESULTS: In less severely degenerative cervical discs, the morphology of the cartilage end-plate showed nearly normal construction, and the tidemark was clear. In severely degenerative discs, the matrix and cells showed fibrosis, the tidemark advanced, and the calcified cartilage thickened. There exists a positive correlation between the thickness of the calcified layer of the cartilage end-plate and the degree of cervical disc degeneration. CONCLUSION: The calcification of the cartilage end-plate is the key factor that initiates and promotes cervical disc degeneration.

[Preparation of cucurbitacinBE polylactic acid nano-particles for targeting cervical lymph nodes].

OBJECTIVE: Anti-cancer nano-particles for targeting cervical lymph nodes were designed and prepared on the basis of the physiologic and anatomic properties of the peri-oral cancer lymphatic capillaries. METHODS: The biodegradable polylactic acid (PLA) was selected as the carrier material, and cucurbitacinBE (CuBE) with very high anticancer activity against oral squamous cell carcinoma was used as the model drug. According to the criteria of the diameter of the nano-particles, drug loading (DL) and embedding ratio (ER), the preparing technology was optimized by uniformly designing experiment on the basis of single-factor experiment. CucurbitacinBE poly-lactic acid nano-particles (CuBE-PLA-NP) was prepared with the method of emulsion solvent evaporation, and then CuBE-PLA-NP lyophilization injection was prepared by using 10% manninositose as the supporting agent. RESULTS: The diameter distribution for optimized CuBE-PLA-NP lyophilization injection ranged from 47 nm to 120 nm, and the average diameter was 85 nm. DL and ER were 23.03% +/- 0.47% and 93.00% +/- 0.10% respectively. The release study of the thermostatics also demonstrated that Higuchi equation was adequate to fit the release curve of CuBE-PLA-NP. CONCLUSION: CuBE-PLA-NP can be used as the release agent, as CuBE-PLA-NP can enter peri-oral cancer lymphatic capillaries for targeting cervical lymph nodes by the peri-cancer submucosal injection. Therefore, it can also used for animal and clinical experiments.

Expression and fast-flow purification of a polyhistidine-tagged myoglobin-like aerotaxis transducer.

A Co(2+)-affinity, fast-flow perfusion chromatography method to purify a polyhistidine-tagged myoglobin-like aerotaxis transducer HemAT-Hs has been developed. The method relies upon a six-histidine affinity tag fused to the C-terminus and N-terminus of HemAT-Hs for expression in the native host, an extremely halophilic Archaeon Halobacterium salinarum, and in the heterologous host Escherichia coli, respectively. The His-tagged HemAT-Hs can be purified rapidly using either low or high ionic strength buffers. Purified His-tagged HemAT-Hs in high or low salt buffers demonstrated no difference in spectral characteristics and retained reversible oxygen binding capacity. This fast-flow Co(2+)-affinity perfusion chromatography provides a simple method for preparation of halophilic heme containing soluble proteins for biophysical and structural studies.

Genome sequence of Halobacterium species NRC-1.

We report the complete sequence of an extreme halophile, Halobacterium sp. NRC-1, harboring a dynamic 2,571,010-bp genome containing 91 insertion sequences representing 12 families and organized into a large chromosome and 2 related minichromosomes. The Halobacterium NRC-1 genome codes for 2,630 predicted proteins, 36% of which are unrelated to any previously reported. Analysis of the genome sequence shows the presence of pathways for uptake and utilization of amino acids, active sodium-proton antiporter and potassium uptake systems, sophisticated photosensory and signal transduction pathways, and DNA replication, transcription, and translation systems resembling more complex eukaryotic organisms. Whole proteome comparisons show the definite archaeal nature of this halophile with additional similarities to the Gram-positive Bacillus subtilis and other bacteria. The ease of culturing Halobacterium and the availability of methods for its genetic manipulation in the laboratory, including construction of gene knockouts and replacements, indicate this halophile can serve as an excellent model system among the archaea.

Association of histone deacetylase with COUP-TF in tumorigenic Ad12-transformed cells and its potential role in shut-off of MHC class I transcription.

Chicken ovalbumin upstream promoter-transcription factor (COUP-TF) is an orphan nuclear receptor that represses transcription of many genes. In adenovirus type 12 (Ad12) transformed cells, a high level of binding activity of COUP-TF to the major histocompatibility complex (MHC) class I enhancer correlates with the down-regulation of class I transcription, which, in turn, contributes to tumorigenesis. The mechanism by which COUP-TF represses transcription has yet to be elucidated. Here we show that COUP-TF represses transcription through its association with histone deacetylase. This was demonstrated using reciprocal binding assays that determined that the interaction between COUP-TF and histone deacetylase requires the COUP-TF C-terminal repression domain. Moreover, a histone deacetylase enzymatic activity was found to be associated with COUP-TF in Ad12-transformed cells. Transfection experiments further revealed that exogenous histone deacetylase facilitates transcriptional repression by COUP-TF. Also, supershift assays suggest that the transcriptional corepressor N-CoR, which is known to associate with histone deacetylases, is a part of the COUP-TF complex bound to the MHC class I enhancer R2 site. Finally, we provide evidence that inhibition of histone deacetylases relieves the repression of MHC class I expression in Ad12-transformed cells. Taken together these results support the notion that deacetylation of histones, mediated through COUP-TF, serves to down-regulate MHC class I transcription in Ad12-transformed cells.

Myoglobin-like aerotaxis transducers in Archaea and Bacteria.

Haem-containing proteins such as haemoglobin and myoglobin play an essential role in oxygen transport and storage. Comparison of the amino-acid sequences of globins from Bacteria and Eukarya suggests that they share an early common ancestor, even though the proteins perform different functions in these two kingdoms. Until now, no members of the globin family have been found in the third kingdom, Archaea. Recent studies of biological signalling in the Bacteria and Eukarya have revealed a new class of haem-containing proteins that serve as sensors. Until now, no haem-based sensor has been described in the Archaea. Here we report the first myoglobin-like, haem-containing protein in the Archaea, and the first haem-based aerotactic transducer in the Bacteria (termed HemAT-Hs for the archaeon Halobacterium salinarum, and HemAT-Bs for Bacillus subtilis). These proteins exhibit spectral properties similar to those of myoglobin and trigger aerotactic responses.

[Measure of vibration protection effect of driver's corset and analysis of its biomechanical effect].

The purpose of this study was to evaluate the vibration protection and biomechanical effect of driver's corset. The frequencies of vertfical and horizontal vibrations were measured at low back of driver. The vehicle driven was ISUZU truck (loading capacity 8 tons). Vibration of the driver's lumbar back was measured real time with wear corset and without wear corset when the truck loaded with 6 tons was driven at the spead of ten, thirty and sixty kilometers an hour on the asphalt road. The results showed: 1. Vibration frequencies at driver's low back was under 10 Hz. It is a low frequency vibration. 2. The value of vertical vibration was higher than the value of horizontal (back and forth) vibration. 3. The vibration value of wear corset was higher than un-wear corset. These indicate the driver's corset is effective for protecting lumbar spine by means of change in the biomechanical characteristics and the resonace requencies of lumbar spine. So the driver's corset is one of the good methods for preventing the back pain of drivers.

[Treatment of ureteric lithioasis with ureteroscope: report of 234 patients].

SUBJECT: To sum up the experience in the treatment of ureteric lithioasis with ureteroscope. METHODS: From January. 1991 to November 1998 234 patients with ureteric lithiasis were treated with lithotripsy of ureteroscopy. RESULTS: 185 of 234 the patients (79%) were successfully treated with lithotripsy of ureteroscopy. Three of the patients (1.3%) had operative complications (ureterperforation 2 patients and ureteral mucosa avulsion in 1). CONCLUSIONS: Lithotripsy of ureteroscope is one of the effective methods in the treatment of ureteric lithiasis, especially in the lower part of ureters.

[Influence of androgen on Bcl-2 mRNA expression in BPH tissue].

OBJECTIVE: To study the expression of Bcl-2 mRNA in BPH tissue and the influence of androgen. METHODS: The circulating testosterone level of BPH patients and rats was decreased by administration of medroxyprogesterone and stilbestrol as well as castration. Using RNA dot blot hybridization technic, we detected the Bcl-2mRNA expression in human BPH tissue and rat prostate under different level of androgen. RESULTS: The average dot IOD (integrated optical density, IOD) value ratio of the untreated patient group to the treated group was 1.71 (P < 0.01). There was an decrease of intensity of Bcl-2 mRNA expression in the latter group compared with the former group. The ratio of the normal rat prostates to the castrated rat prostates was 1.28 (P < 0.05). The Bcl-2 mRNA expression was decreased after castration. CONCLUSIONS: The expression of Bcl-2 plays a role in the development of BPH. The decrease of prostatic cell death is important in the BPH pathogenesis. Apart from promoting the proliferation of prostatic epithelia and stroma cells, androgen might prohibit apoptosis by increasing the expression of Bcl-2 in BPH development.

[Chemical-induced polycyst with renal tumor and expression of 8-OHdG in kidney tissue].

OBJECTIVE: To induce the rat model of polycyst with renal tumor and investigate the expression of 8-OHdG in the kidney tissues. METHODS: The rat model of polycyst with renal tumor, similar to human acquired cystic disease of the kidney (ACDK), was induced by oral administration of 2-amino-4, 5-diphenylthiazole (DPT) and N-nitrosomorpholine (NNM). Immunohistochemical method (LSAB) was used to assay the expression of 8-hydroxydeoxyguanosine (8-OHdG) in the rat kidney tissue. RESULTS: Three of 10 rats in the NNM group had renal solid adenomatous lesions. Bilateral polycysts were observed in all 9 rats of the DPT/NNM group. Seven of the 9 rats had cystic multistage renal tumor. In the DPT/NNM group, 4 rats had cystic adenomatous lesions, but none in the other groups showed this lesion. In the model, adenomatous lesions derived from polycysts in the rats were closely consistent to human ACDK in morphology. The significant expression of 8-OHdG was found on renal tubular cell, cystic epithelial cell, stromal cell and tumor cell in all rats of the NNM and DPT/NNM group. CONCLUSION: A rat model of polycysts with renal tumor, similar to human ACDK, induced by DPT and NNM provides evidences for further study on pathogenic mechanism in human ACDK with renal cell carcinoma. The expression of 8-OHdG, a DNA damage marker, in the renal tissues of rat model might help to explain the mechanism of cysticogenesis and carcinogenesis in human ACDK.

[Middle-aged and elderly patients with hydronephrosis induced by ureteric obstruction: etiology and diagnosis].

OBJECTIVE: To improve the etiologic diagnosis of middle-aged and elderly patients with ureteric obstruction. METHODS: The etiology and diagnosis were retrospectively studied in 141 middle-aged and elderly patients with hydronephrosis in recent 10 years. RESULT: 59 patients had ureterolithiasis, 51 ureteric cancer, 11 ureteric tuberculosis, 9 UPJ stricture, 6 uretero-cyst and 5 ureteric polyp. CONCLUSIONS: Ureterolithiasis and ureteric cancer contribute greatly to ureteric obstruction in middle-aged and elderly patients. Etiologically, B ultrasonography is suggested to be used as the first choice in diagnosis and follow-up of the middle-aged and elderly patients with hydronephrosis, especially those accompanied with hematuria.

[The invasive ability of bladder cancer].

OBJECTIVE: To find a direct method for testing the invasive ability of bladder cancer. METHOD: The invasive ability of 49 specimens of primarily cultured bladder cancer, transfected BIU-87 cells with pcDNA3-E1AF and untransfected BIU-87 cells was evaluated by Matrigel invasion assay. RESULTS: Sixteen of the 49 specimens were found to penetrate the matrigel barrier, and the result of the Matrigel invasion was related to the grade of the tumor cells. The number of positive cells of transfected BIU-87 was remarkably larger than that of untransfected BIU-87. CONCLUSIONS: Matrigel invasion assay is a simple, direct and dependable method, and could be used clinically to test the invasive ability of bladder cancer.

[Chemical constituents of grub and grub eye drops].

The chemical constituents of grub (Holotrichia diomphalia Bates.) were systematically analyzed. The results showed that grab contained amino acids, polypeptide or protein, carbohydrate, alkaloid, organic acid, steroid et al. The analysis of amino acids in the hydrolysate in grub eye drops showed the content of Glu and Gly was high, which occupied 40.33% of total amino acids in the hydrolysate. We inferred that Glu and Gly might be the active principle on catatract and nebula.

[Determination of chromium in geological samples with slurry sampling and probe atomization in GFAAS].

A method of loading a slurry prepared from a solid sample for analysis of chromium in geological samples using graphite furnace atomic absorption spectrometry had been approached by combining slurry sampling with probe atomization. The factors that influence the performances of chromium including ashing and atomization temperature, atomization surfaces were investigated. The results showed that the determination of chromium in geological samples could be performed by direct slurry sampling with the use of a calibration established with aqueous standards. The relative standard deviations(RSD) of the method were 3.0%-6.5%. The detection limit was 5.1 x 10(-12) g Cr. The Analytical results of the geological reference samples were consistent with the certified values.
