Expression of deoxyuridine triphosphatase during liver regeneration in rat.

Deoxyuridine triphosphatase (dUTPase) activity increases concomitantly with DNA replication in the course of liver regeneration in rat. We confirmed in this report using Western blot analysis and Northern blot hybridization that the increase of dUTPase activity was derived from expression of the gene. The content of dUTPase protein quantitatively coincided with the activity, they reached to the maximum at about 24 h after partial hepatectomy. We also detected a transcript (1.0 kb) presumed to be a rat spleen dUTPase mRNA by the human dUTPase cDNA probe. This transcript appeared in the liver after a lag of about 16 h, reached to the maximum at 24 h, and could be detected until 48 h after partial hepatectomy. The increase of this transcript nearly coincided with the changes of dUTPase activity and the quantity of enzyme protein. These results indicated that the expression of dUTPase gene is related to the DNA replication.

Deoxyuridine triphosphatase in human hepatoma.

1. Deoxyuridine triphosphatase (dUTPase) in human hepatoma was investigated. The apparent activity in a gram weight tissue was about 6 times that of the activity in rat livers after partial hepatectomy. 2. Most catalytic properties of the hepatoma dUTPase were similar to the enzymes from other sources. 3. The hepatoma dUTPase reacted with the antibodies against the rat spleen dUTPase, although the molecular size of the enzyme (46-48 kDa) was different to the rat enzyme. 4. The immunochemical studies indicated that the enzyme was composed of two identical subunits, whose molecular mass was about 22 kDa.

[A study on wounds caused by cats as basic materials of cat scratch disease].

Between October, 1992 and March, 1993, the incidence of bite or scratch by the cat, was surveyed by questionnaire in 1,619 peoples who were veterinarians, veterinary technicians, students and the general population (male 652, female 967). The wound rate by cat: Veterinarians, veterinary technicians, students of veterinary college, students of veterinary technician school were significantly higher than the general population. Incidence of bite or scratch in the cat owner were significantly higher than the non cat owner, and the incidence of wounds in cat owners was more than 90%. Most injuries for veterinarians were of the occupational nature. Their injuries clearly differed from those of the other groups. In our survey 33 cases were suspected to have CSD; 19 of 102 veterinarians, 4 of 45 veterinary technicians, 2 of 517 students of veterinary college, 1 of 400 students of veterinary technicians, 7 of 555 in the general population. Of the seven cases suspected for CSD among the general population, four were proven to have cat-scratches, suggesting that injuries can be prevented by clipping the cat's nails at home. However, most of the injuries suffered by veterinarians are caused by bites, prevention poses a more serious problem. It is therefore important to educate people in all fields to fully understand sanitation and zoonosis including CSD, and teach them to handle their pets appropriately.

[A case of cat scratch disease: from the clinical pathological point of view].

We described a forty-four-year-old female who was scratched by her cat, and developed lymphadenopathy. Cat scratch disease skin-test was positive. Cat scratch disease is well known, but there are not many reports in Japan. Reports of cat scratch disease will increase as the pathogen was recently detected. Attention must be drawn to zoonosis containing cat scratch disease.

Purification and characterization of cytoplasmic 5'(3')-nucleotidase from rabbit spleen: characteristic differences of the enzyme from the rat spleen nucleotidase.

1. A 5'(3')-nucleotidase, which preferably hydrolyzed 3'-dTMP and 3'-dUMP, was highly purified from rabbit spleen cytosol. 2. The enzyme also hydrolyzes 3'-UMP, 5'-dUMP and guanine nucleotides, but does not hydrolyze any adenine nucleotides or cytosine nucleotides. 3. The activity is dependent upon Mg2+, Co2+ or Mn2+; the addition of deoxyinosine stimulates the activity, and the pH optimum for the hydrolysis of 3'-dTMP is 7.0. 4. Although the catalytic properties of the enzyme are similar to the 5'(3')-nucleotidase from rat spleen, these nucleotidases differ in their molecular disposition. 5. The charge state of the rabbit enzyme is slightly basic, and the subunit M(r) is about 27 kDa, while the value of the rat enzyme is 26 kDa. 6. Immunochemical experiments with the use of antibodies against the purified nucleotidase indicate that enzymes from rabbit spleen and from rat spleen are composed of different polypeptides.

[Studies on the giardiasis as the zoonosis. III. Prevalence of Giardia among the dogs and the owners in Japan].

To obtain the basic data on the route of Giardia infection as zoonosis, of many regions in Japan, the feces from 2218 dogs were examined for detection of Giardia cysts. Giardia cysts were detected in 239 of the 2218 dogs (10.9%), which was the same as previous reports from America. None were found from the owners of 51 dogs in which Giardia cysts were detected. The detection rates of each facilities were, 68 of 366 (18.6%) from the breeder's kennels, 169 of 1811 (9.3%) in individual houses, 2 of 42 (4.9%) from research institutes. The detection rate of the breeder's kennels was higher than the other two facilities (p less than 0.05, p less than 0.001). The detection rates of Kanagawa and Shizuoka prefectures among 17 regions in Japan were higher than the others (p less than 0.001, p less than 0.05). Especially in Shizuoka, the rate of the individual houses was higher than from breeder's kennels. In kanagawa the rates of the individual houses and the breeders kennels were higher than the mean in Japan (p less than 0.001). Therefore one must instruct the breeders when teaching health education to included zoonosis, and that the detection rate of the age groups of less than 3 years old was high-221 of 1276 (17.3%). Since the detection rates of Giardia cysts in the dogs were low, the possibility that human infection acquired from dogs was low. However, some of patients with giardiasis we encountered had never been abroad, and it is not yet clear whether Giardia is strictly host specific or not, so attention should be paid to the possibility of cross-infection between man and animals.

[Detection of intestinal parasites from dogs in Tochigi-City in 1979 and 1991].

To clarify whether sanitary and hygienic conditions surrounding humans and pet animals have improved or not, we examined feces from dogs for intestinal parasites in Tochigi in 1979 and 1991. Intestinal parasites were detected from 109 of the 262 dogs (41.9%) in 1979 and 86 of the 260 dogs (32.8%) in 1991. Among the parasites. Ancylostoma caninum, Tirchuris vulpis and Toxocara canis were most commonly detected, and their detection rates were higher in 1979. On the contrary, Dipylidium caninum, Strongyloides stercoralis, Isospora spp., which are important causes of human diseases, were detected at higher rates in 1991. In the dogs of outdoor feeding, the detection rates of parasites were markedly decreased, but slightly increased in the dogs of indoor feeding in 1991. These results suggest that sanitary conditions surrounding pet animals have not been sufficiently improved, in spite of the fact that humans are in more contact with their pet animals.

[Clinicobacteriological study of Pasteurella multocida as a zoonosis (1). Condition of dog and cat carriers of Pasteurella, and the influence for human carrier rate by kiss with the pets].

Pasteurella multocida is a gram-negative short rod-shaped bacteria, which is a part of the indigenous flora of the oral cavity of many animals other than man. The number of reports on cases of infections with this bacterium due to animal bites and/or scratches, bacterial infections of the respiratory tract, sepsis due to this organism and death caused by the bacteria have been increasing in recent years. We investigated P. multocida in the hair and oral cavity of 3 dogs and 29 cats according to the classification of Mutters et al.. We also studied the relationship between the carrier rate for Pasteurella in the oral cavity and kissing of pets in 24 pet owners (3 dogs and 11 cats). No P. multocida was isolated from the hair of neither dogs nor cats. One strain of P. multocida subsp. multocida and two strains of P. stomatis, were isolated from the oral cavity of dogs, and 35 strains of Pasteurella were isolated from the oral cavity of cats. Two strains of P. multocida subsp. multocida, whose biochemical properties were different, were detected in the oral cavity of one cat. In three cats, Pasteurella other than P. multocida subsp. multocida was isolated from the same oral cavity. No Pasteurella was detected in the oral cavity of 19 pet owners who had not kissed their cats, whereas P. stomatis was isolated from the oral cavity of one of 2 pet owners who had kissed their cats and in 2 of 3 pet owners who had kissed their dogs (the same bacteria was isolated from a dog that was being kept by some of these positive pet owners).(ABSTRACT TRUNCATED AT 250 WORDS)

[Studies on the giardiasis as the zoonosis. II. Giardiasis in dogs and cats].

To obtain the basic data on the route of Giardia infection as zoonosis, we examined feces from 354 dogs and 34 cats for Giardia cysts and trophozoites and evaluated the detection rates. In addition, the feces of owners of the dogs infected with Giardia were examined. Giardia cysts and trophozoites were detected in 58 of the 354 dogs (16.4%) and 3 of the 34 cats (8.8%). Of the 61 animals from which cysts and trophozoites were isolated, 59 were purchased from the breeders and the other two animals had been kept for examinations in the research institute. None of the owners of the dogs had this infection. This result suggests that the possibility of human infection from contact with infected dogs or cats seems to be very low. However, attention should be paid to the possibility of cross-infection between men and animals, because it is not yet clear whether Giardia is strictly host-specific or not.

A 3'-deoxyribonucleotidase isolated from rabbit spleen cytosol differs from the rat spleen nucleotidase.

A novel 3'-nucleotidase, which preferably hydrolyzed 3'-dTMP and 3'-dUMP, was isolated and purified from rabbit spleen cytosol. Since the main catalytic properties of the enzyme were similar to the rat liver nucleotidase reported by Fritzson and Smith, we also purified the latter enzyme from rat spleen. These two enzymes were different in several points: the substrate restriction, the molecular mass and the apparent charge. Further analyses with the antiserum raised in rat against the 3'-deoxyribonucleotidase evidenced that these enzymes were immunochemically different as well.

Granularity of dental X-ray film. Fundamental study of Wiener spectrum measurement.

We developed a 16k dots x 100 lines Wiener spectrum measurement system using a microscope-television camera combination. Heretofore, the granularity of film was evaluated from Wiener spectrum using Wiener spectral value in the spatial-frequency independent region obtained by microphotometer as the input device, but this factor is determined by the measurement system and the film emulsion density and there were problems in the evaluation of granularity. Therefore we established a new method for evaluating the granularity from Wiener spectral values in the spatial-frequency independent region through a Wiener spectrum measurement system that utilized a microscope-television camera setup as the input device.

Calf thymus deoxyuridine triphosphatase differs from rat spleen enzyme in molecular disposition.

Antiserum against calf thymus dUTPase was raised in rats by injection of the partially purified enzyme. The antiserum did not react with dUTPase purified from rat spleen, while antibody against rat spleen dUTPase partially reacted with calf thymus enzyme. Native molecular weight of the calf thymus dUTPase was estimated at 46,000 daltons by gel filtration, and the denatured form of the enzyme was about 22,000, as judged by immunoblot analyses using the antibodies. These results indicate that the calf thymus dUTPase is composed of two identical subunits.

Rat spleen cytoplasmic nucleotidase: characterization and its physiological significance.

1. A cytoplasmic nucleotidase, which preferably hydrolyzed 5'-dUMP, was investigated in rat spleen. 2. Total activity of the nucleotidase increased about 3-fold in the spleen of anemic rat was caused by phenylhydrazine administration. This increase was repressed by the injection of methotrexate, an inhibitor of DNA synthesis. 3. Activities of heme oxygenase or acid phosphatase did not correlate to the change of the nucleotidase activity. 4. The nucleotidase catalyzed dephosphorylation of 3'(5')-dUMP, 3'(5')-dTMP and 3'-UMP more readily, in the presence of Mg2+. Its optimum pH was around 6.0-6.5. It was stimulated by the addition of deoxyinosine. 5. These catalytic properties and tissue distribution of the enzyme, abundant in the thymus, spleen and intestine, were similar to that of 5'(3')-nucleotidase in rat liver [Fritzson P. (1978) Adv. Enzym. Regul. 16, 43-61]. 6. A possible physiological significance of the nucleotidase is in reutilization of preformed pyrimidine nucleotides.

Deoxyuridine triphosphate nucleotidohydrolase: distribution of the enzyme in various rat tissues.

The distribution of deoxyuridine triphosphate nucleotidohydrolase (dUTPase) [EC 3.6.1.23] in the cytosol of various rat tissues was investigated by measuring the enzyme activity and by immunochemical analyses. Among nine rat tissues, thymus, and spleen showed the highest activities of the enzyme per gram of tissue, while intestine, stomach, lung and liver showed very low levels. Rabbit antibodies directed against purified dUTPase of anemic rat spleen showed reactivity with partially purified dUTPases from other rat tissues such as thymus, testis, or regenerating liver. Immunotitration and immunoblot experiments also indicated that the dUTPases in various rat tissues had very similar antigenicity and apparently the same subunit molecular size (Mr = 19,500), suggesting that the enzyme lacks organ-specificity. Immunoblot analysis of dUTPase protein with crude extracts from various rat tissues showed a similar distribution to that of the enzyme activity. No immuno-reactive band corresponding to the dUTPase was detected in intestine, although intestinal mucosa has been recognized as an actively proliferating tissue.

Purification and characterization of deoxyuridine triphosphate nucleotidohydrolase from anemic rat spleen: a trimer composition of the enzyme protein.

Deoxyuridine triphosphate nucleotidohydrolase (dUTPase) was purified to near homogeneity from the spleens of rats made anemic by phenylhydrazine injection; the enzyme activity in these spleens was about 30 times higher than that in spleens of untreated rats. The purified enzyme preparation showed an apparent molecular weight of 58,500 and appeared to consist of three identical subunits each with a molecular weight of about 19,500. The purified enzyme catalyzed specifically the hydrolysis of dUTP, and no other naturally occurring nucleoside triphosphates could be hydrolyzed by this enzyme. The Km value for dUTP was 12 microM. Enzyme activity was inhibited by the addition of EDTA, whereas the enzyme preparation exhibited activity in the absence of added divalent cations. Activity was not affected by the addition of fluoride ion.

The effect of copper on intact swine erythrocytes.

The effect of copper on intact swine erythrocytes was investigated in vitro. When treated with copper, a decrease in the GSH content, accumulation of copper in the cell, loss of potassium and gain of sodium, cross-linking of membrane proteins and echinocytic transformation were observed. All of these phenomena seem to be caused by a potent oxidant action of copper. These cytotoxic effects of copper were markedly inhibited by the addition of bovine serum albumin in the incubation medium. These results may help to understand the mechanism of hemolysis associated with copper poisoning in vivo.
