Effects of major histocompatibility complex on autoimmune disease of H-2-congenic New Zealand mice.

Autoimmune-prone NZB mice mainly produce IgM-class anti-DNA antibodies and mild SLE develops later in life. The F1 hybrid of NZB and non-autoimmune NZW mice (NZB/W F1 mice) develop a more fulminant SLE, associated with decreases in IgM class, and, in turn, increases in IgG class anti-DNA antibodies. To elucidate the role of the H-2 complex in this mode of anti-DNA antibody production, we established and studied H-2-congenic New Zealand mice, i.e. NZB, NZW, and NZB/W F1 mice with either the homozygous H-2z/H-2z or H-2d/H-2d haplotype or the heterozygous H-2d/H-2z haplotype. The data showed that: (i) although the non-H-2-linked NZB gene(s) seems to determine the IgM anti-DNA antibody production in NZB mice, the effect of this gene is fully expressed only in the case of the H-2d/H-2d homozygous state. (ii) The production of IgG anti-DNA antibodies observed in NZB/W F1 hybrid mice is restricted to the H-2d/H-2z heterozygosity. (iii) Because both NZB and NZW mice with the H-2d/H-2z haplotype produce a lower titer of IgG anti-DNA antibodies than do the NZB/W F1 mice, other complementary non-H-2-linked genetic elements from both NZB and NZW parents are required. The development of lupus nephritis correlated well with that of anti-DNA antibodies. Thus, H-2d/H-2z heterozygosity is a necessary but not sufficient condition for the development of autoimmunity in NZB/W F1 mice.(ABSTRACT TRUNCATED AT 250 WORDS)

Monoclonal antibodies to epitope of CD45R (B220) inhibit interleukin 4-mediated B cell proliferation and differentiation.

A mAb, ALP-2, produced by immunizing rats with proliferating lymph node cells from MRL/Mp-lpr/lpr mice, had an inhibitory effect on recombinant interleukin 4 (rIL-4)-mediated proliferation and differentiation of murine B cells. Kinetic analysis revealed that the ALP-2 exerted these inhibitory effects at an early phase of B cell proliferation and differentiation. Nevertheless, the proliferative response of thymocytes to rIL-4 was not inhibited by ALP-2. In addition, ALP-2 inhibited neither the B cell proliferation induced by interleukin 2 nor the B cell differentiation by interleukin 5. Cross-inhibition experiments, together with immunoblotting analysis, revealed that the LP-2 recognized by ALP-2 appears to be an epitope of CD45R (B220) molecule(s), the isoform(s) of the Ly-5 antigen system. Epitope mapping analysis using CD45 gene transfectants showed that Lp-2 epitope is dependent upon the expression of the first alternatively spliced exon of CD45 gene. Functional studies showed that the mAb to an epitope of CD45R, RA3-2C2, but not RA3-6B2, had similar inhibitory effects on the IL-4-mediated proliferative response of B cells. These findings suggest that the CD45R molecules associated with Lp-2 and RA3-2C2 epitopes are probably related to a signal transduction provided by IL-4 in B cells. The possibility that different pathways are operative for IL-4-mediated signaling between B and T cells has to be considered.

Changes of DNA methylation level during pre- and postnatal periods in mice.

DNA methylation in an adult mammalian body shows tissue-specificity. But when and how the specificity is established in the process of development has not yet been elucidated. Here we have investigated age-dependent changes in the amount of 5-methyldeoxycytidine (5mdC) that DNA of various mouse tissues contains during the late-fetal and postnatal periods, using high-performance liquid chromatography. The tissue-specificity in the 5mdC level was observed in the late-fetal stage, and the level continued to change during the subsequent periods. The most pronounced alterations were observed in brain and liver, where similar biphasic changes were seen, but at different ages. At maturation, the 5mdC levels were high in thymus, spleen and brain, intermediate in lung, and low in liver and sperm. The data demonstrate the importance of the peri- and postnatal periods in establishment of tissue-specificity in 5mdC content.

Effects of interferon gamma on cultured synovial cells from patients with rheumatoid arthritis: inhibition of cell growth, prostaglandin E2, and collagenase release.

The effects of recombinant interferon gamma (rIFN gamma) on the in vitro growth of adherent synovial fibroblast-like cells from patients with rheumatoid arthritis (RA) and also on the release of prostaglandin E2 and collagenase from these cells stimulated with recombinant interleukin-1 beta (rIL-1 beta) were investigated. The growth of adherent synovial cells from six of nine samples, determined by [3H]thymidine incorporation, was inhibited by rIFN gamma in a manner dependent on dose. The release of prostaglandin E2 and collagenase from adherent synovial cells stimulated with rIL-1 beta was also suppressed by rIFN gamma in all samples tested, though the basal release of these inflammatory mediators was little influenced. No apparent correlation between inhibition of proliferation by rIFN gamma and either inhibition by rIFN gamma of rIL-1 beta stimulated prostaglandin E2 release or the endogenous synthesis of prostaglandins was found.

Stability of urokinase in solutions containing sodium bisulfite.

The stabilities of urokinase (UK) in aqueous solution were investigated at pH 5.0-8.0 in the presence (1.0-3.0 x 10(-3) M) and absence of sodium bisulfite (SBS) both under scattered light (1000 lux) and in the dark using the fluorogenic substrate method. Increasing concentrations of SBS tended to increase the inactivation of UK. In the presence of SBS, with the increase in the pH value, UK gained in stability in the pH range of 5.0-8.0. The stability of UK in the presence of SBS in the dark was larger than that under scattered light, especially at pH 5.0. Therefore, it was suggested that the difference in the residual activities of UK between under light and in the dark was due to free radicals formed during the autooxidation of bisulfite under scattered light. UK was stabilized by glucose in the presence of SBS both under scattered light and in the dark. One reason for this phenomenon was postulated to be the formation of inactive bisulfite-glucose addition compound. The degradation products of UK during storage in a solution containing SBS were investigated by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis. UK was revealed to be split into M.W. 36000 form and M.W. 20000 form by SBS.

Stability prediction of nafamostat mesilate in an intravenous admixture containing sodium bisulfite.

The hydrolysis of nafamostat mesilate (NM) in aqueous solution was found to be accelerated by sodium bisulfite (SBS) using high-performance liquid chromatographic assay. The hydrolysis of NM catalyzed by SBS was pseudo-first-order and was considered to be a reaction between nafamostat cation (N+) and sulfite ion. The effects of SBS concentration and temperature on the hydrolysis of NM in buffer solution were examined. From the findings obtained, we estimated the compatible pH range of the intravenous admixture (mixed infusion) of NM after a constant storage time at a constant SBS concentration and temperature employing a simulated pH-profile for the mixed infusion. In order to evaluate the compatibility of the prescribed mixed infusion, the method of pH estimation for the mixed infusion was also investigated using the pH titration curve of each preparation.

The effect of increased intracranial pressure during the appearance of pressure waves on the brainstem.

The effects of increased intracranial pressure (ICP) during the appearance of plateau waves or B-waves to the brainstem functions were investigated by evaluation of evoked potentials. The ICP and systemic blood pressure were continuously recorded in nine cases of intracranial hypertension. In the four cases demonstrating plateau waves in ICP recording, the latencies of the auditory brainstem evoked potentials (ABEP) measured during the appearance of plateau waves showed no significant differences compared to those measured during the interval phase between two plateau waves. In the five cases demonstrating B-waves in ICP recording, four cases showed significant (p less than 0.05) prolongation of the V wave of ABEP and three showed significant (p less than 0.05) prolongation of the P15 of somatosensory evoked potentials during the phase of increased ICP level with B-waves compared to those measured during the phase of decreased ICP level without B-waves, produced by external drainage of the cerebrospinal fluid or mannitol administration. The results indicated that during the appearance of plateau waves there were no significant changes of electrical activity in the brainstem, even though there was a significant reduction in cerebral perfusion pressure. It is suggested that in cases with the appearance of B-waves, an increase in ICP frequently affects the electrical activity in the brainstem.

Distinct surface phenotypes of B cells responsible for spontaneous production of IgM and IgG anti-DNA antibodies in autoimmune-prone NZB x NZW F1 mice.

Autoimmune-prone NZB x NZW F1 (B/W F1) mice produce a high titer of anti-DNA antibodies, In vivo and in vitro studies showed that in the early life of these mice, the immunoglobulin isotype of these antibodies almost exclusively belongs to IgM class, however, IgG anti-DNA antibodies begin to develop when the mice are about 5-6 months old and the titer exceeds that of IgM antibodies from age 7 months on. We asked whether or not the B cell population responsible for IgM and IgG antibody production belongs to the same lineage. The surface phenotypes of B cell populations responsible for the spontaneous production of either IgM or IgG anti-DNA antibodies were examined using panning and sorting methods with several monoclonal antibodies to B cells, including CD5 (Ly-1) and Lp-3; the latter defines a unique B cell differentiation antigen. We obtained evidence that surface phenotypes of B cells secreting IgM anti-DNA antibodies belong to CD5+ Lp-3- and those of B cells secreting IgG anti-DNA antibodies which occur only in old B/W F1 mice belong to CD5- Lp-3+ subpopulations. The majority of peritoneal B cells were CD5+ Lp-3+ throughout the life span of the mice and anti-DNA antibody production was never evidenced. These findings were discussed in relation to age-associated changes of B cell populations in the spleen of this strain of mice.

HLA-DP-positive T cells in patients with systemic lupus erythematosus.

HLA-DP+ T cells in peripheral blood from 23 patients with systemic lupus erythematosus (SLE) were examined using two-colour flow cytometry analysis. A marked increase of HLA-DP+ T cells was observed in patients with SLE (20.5-98.7%; 59.8 +/- 20.8%) in comparison to normal subjects (1.3-20.6%; 11.1 +/- 7.2%), and the ratio of these cells greatly exceeded that of the HLA-DR+ T cells (6.5-49.1%; 21.5 +/- 12.7%). This high frequency of HLA-DP+ T cells in patients with active SLE decreased with prednisolone therapy. When the lymphocytes from normal subjects were stimulated with PHA in vitro, HLA-DP+ T cells increased from 1.8 to 59.2%. Therefore, it appears that the HLA-DP antigen expression on T cells is a practical marker for monitoring changes in the proportion of activated T cells in patients with SLE during the course of therapy.

Lymphocyte responses to peptide M and to retinal S-antigen in uveitis patients.

A synthetic amino acid, peptide M18 (18 amino acids in length which correspond to the amino acid positions 303-322 of bovine S-antigen: DTNLASSTIIKEGIDKTV), is capable of inducing experimental autoimmune uveitis in monkeys and Hartley guinea pigs as well as Lewis rats. Recently, the amino acid sequence of human S-antigen corresponding to peptide M was found to be virtually identical to that of bovine S-antigen. To investigate the role of the epitope corresponding to peptide M in human uveitis, 21 patients with uveitis were tested for proliferation of peripheral mononuclear cells against peptide M18 and bovine S-antigen. Six of the 21 patients responded to S-antigen and 4 of these 6 patients responded to peptide M18. These findings show that peptide M18 is immunogenic in man and provide additional support to the hypothesis that the amino acid sequence which corresponds to peptide M might be involved in uveitic conditions in man.

[Idiopathic thrombocytopenic purpura complicated with circulating lupus anticoagulant].

A 37 year-old female was admitted to our hospital because of hypermenorrhea, prolonged bleeding time, thrombocytopenia and the diagnosis of idiopathic thrombocytopenic purpura (ITP) was made. Though activated partial thromboplastin time (APTT) was markedly prolonged, her coagulation factors were within normal ranges. Activities of the circulating lupus anticoagulant (LAC) was suggested. Kaolin clotting time of the platelet poor plasma was used as a sensitive screening test using the mixture of normal and patient's plasma for the detect of LAC. As a result, LAC positive pattern was observed. The treatment with high-dose gammaglobulin brought out a transient increase of the platelet count, but the prolongation of APTT was not corrected. Both the platelet count and the prolongation of APTT were significantly improved after the treatment with betamethasone.

[Genetic control of experimental autoimmune uveitis in rats].

Differences in susceptibility to experimental autoimmune uveitis (EAU) among rats of various strains have been reported. The present study was aimed at separating the effects of the major histocompatibility antigen complex (MHC) and non-MHC genes on the development of EAU. EAU-susceptible LEW, EAU-resistant WKAH, WKAH, 1L MHC congenic strain rats representing MHC of LEW on WKAH genetic background, and other nine inbred strains of rats were examined for their ability to develop EAU by immunization with bovine S-Ag or peptide M, an synthetic polypeptide corresponding to position 303 to 320 in bovine S-Ag. We found that only LEW rats developed S-Ag induced-EAU, where WKAH, 1L congenic and other rats did not. However, when an additional injection of Bordetella pertussis (BP) was given, all rat strains developed S-Ag induced-EAU. In contrast, only LEW, WKAH, 1L,F344, and NIG-III rats, which have haplotype 1 at RT1 class II subregions developed EAU by immunization with peptide M and BP. The present findings showed that the susceptibility to EAU in rats was controlled by both MHC and non-MHC genes. Eventual development of EAU in rats was governed by non-MHC gene (s). However, this effect of non-MHC gene (s) could no longer be observed when an additional injection of BP was administered. MHC class II (RT1(1] restricted the susceptibility to peptide M induced-EAU. However, this MHC restriction was not observed when multideterminant S-Ag was used as an immunogen.

[Busulfan lung exacerbated during steroid therapy: a review of Japanese literature].

A 61-year-old male was diagnosed as chronic myelocytic leukemia (CML) in 1985 and had been treated with busulfan for 4 years and 5 months (total 3,572 mg). Since he had complained dyspnea with abnormal lung shadow on a chest x-ray film in May 31, 1989, he was treated with 3 mg/day of dexamethasone under a diagnosis of busulfan lung. Dyspnea and lung shadow was remarkably improved after the treatment. On July 11, he was admitted our hospital with high fever, hypoxemia and diffuse interstitial shadow on a chest x-p. Although intensive treatment with 10 mg/day of dexamethasone and various antibiotics were done, he died in Aug 6. An autopsy revealed the intra-alveolar fibrosis, dense hyaline deposits in alveoli associated with atypical type II epithelial cells in the lung. Seven cases of busulfan lung have been reported in the Japanese literature. The clinical and pathological aspects of these cases, including our case, were discussed.

Clinical significance of IgG anti-Sm antibodies in patients with systemic lupus erythematosus.

Free Sm was purified by gel filtration and anti-Sm affinity chromatography. Using this purified antigen, an ELISA for anti-Sm was performed. Three hundred and fifty patients with various rheumatic diseases were studied with respect to immunoglobulin classes of anti-Sm by ELISA. A high frequency of IgG anti-Sm was specifically detected in patients with systemic lupus erythematosus (SLE), but IgA and IgM anti-Sm showed a low frequency and also was detected in other diseases. In patients with SLE, anti-Sm significantly correlated with lung fibrosis and pericarditis. In our longitudinal study, there were increases in titer of IgG anti-Sm preceding central nervous system exacerbation and serositis. IgG anti-Sm was found to be not only a diagnostic marker but also a reliable measure of disease activity in SLE.
