Red cell uridine monophosphate kinase polymorphism in Japanese.

Samples from 635 unrelated Japanese blood doners in Tokyo were examined for their red cell uridine monophosphate kinase phenotypes using starch gel electrophoresis. Three phenotypes were found: UMPK1 (570, 89.76%), UMPK 2--1 (63, 9.92%) and UMPK 2 (2, 0.32%). The corresponding gene frequency for UMPK1 was 0.9472 and for UMPK2 was 0.0528. The UMPK2 frequency in the present study was slightly higher than the previously reported value for Caucasians in the United States, but the difference is not statistically significant. A study of 15 twin pairs and their parents was in agreement with the hypothesis of autosomal codominant inheritance for UMPK.

Isoelectrofocusing in acetate membrane: the method and application.

Acetate membrane as a new medium for isoelectrofocusing was used in this study. The acetate membrane was specially produced for isoelectrofocusing, and contains a surface active agent. The ampholine solution system and isoelectric condition for this purpose are described. Polymorphic traits as alpha1-antitrypsin in human serum and 6-phosphogluconate dehydrogenase (6-PGD) in human erythrocyte could be identified by this method. The isoelectric patterns and the isoelectric points of these proteins were demonstrated on the acetate membrane.

The distribution of polymorphic traits in atopic asthmatic patients.

The phenotypic distribution and gene frequencies of haptoglobin (Hp), transferrin (Tf), group specific component (Gc), cholinesterase (Cho E2), and alpha1-antitrypsin (Pi) in plasma proteins, and phosphoglucomutase (PGM), 6-phosphogluconate dehydrogenase ((6-PGD), esterase D (Es D), phosphohexose isomerase (PHI), adenosine deaminase (ADA) and acid phosphatase (AcP) in red cells were studied in 127 atopic, asthmatic patients. The gene frequencies were compared with normal groups. The phenotypic distribution of the Pi system in atopic patients was somewhat different from the normal. No significant differences were found between the two groups in protein systems or in enzyme systems, except Pi systems. In conclusion, except for the Pi system, no definite association between polymorphic traits and atopic asthma was found in this study.

Polymorphism of esterase D in some population groups in Japan.

Electrophoretic types of red cell esterase D (Es D) have been determined in more than 1,000 blood samples from Tokyo. The allele frequency for Es D2 was found to be 0.342. Typing was also carried out in samples of the Ainu of Hokkaido and of the Ryukyuans of the Okinawa Island: the Es D2 frequencies were found to be 0.273 and 0.369, respectively. These values are considerably higher than those described in Europeans, Negroes and even in Asiatic Indians. The results indicate that Es D with a marked degree of heterogeneity is a valuable marker for genetic, anthropological and forensic application in Mongoloid groups.

Studies on lankacidin-group (T-2636) ANTIBIOTICS. IX Preparation of C-labeled lankacidin C 14-propionate.

To investigate the metabolic fate of lankacidin C 14-propionate in experimental animals, the 14-C-labeled antibiotic was prepared by the fermentation of Streptomyces rochei var, volubilis in the presence of various 14-C-labeled organic carboxylic acids, amino acids and carbohydrates. Significant incorporation (20 similar to 40%) was observed with L-methionine-methyl-14-C. Lankacidin C 14-propionate-14-C (specific activity 49.6 muCi/mg) was obtained from lankacidin C-14-C and ethyl propionate by the action of an acylase of the streptomyces.

Studies on lankacidin-group (T-2636) antibiotics. X. Microbial conversion of lankacidin-group antibiotics.

Lankacidin C, a component of lankacidin-group (T-2636) antibiotics, was esterified to lankacidin C 8-butyrate in the presence of methyl butyrate by culture broth and by cell-free extract of Bacillus megaterium IFO 12108. In addition, methyl isobutyrate, methyl valerate and methyl isovalerate served as acyl donors for the esterification, and lankacidin C 8-isobutyrate, lankacidin C 8-valerate and lankacidin C 8-isovalerate were formed respectively. Lankacidin C 8, 14-dibutyrate was hydrolyzed to lankacidin C 14-butyrate by the same organism.

Serologic differences in strains of Sporothrix schenckii.

To obtain evidence that Sporothrix scheneckii enters the body by contact with contaminated materials, the antigenic property of strains from different sources was investigated. The reciprocal absorption test of the antisera against a soil isolate and a human isolate (KO 4606) showed that the absorbed antisera against KO 4606 possessed unique antigen(s) in addition to the common antigen of both strains. Twenty-three clinical isolates were tested with absorbed antisera. Not all of them possessed the unique antigen(s), but there were serologic varieties among S. schenckii strains, regardless of their sources, clinical type of the disease and the morphology of the yeast phase cells.
