Cloning of CTP:phosphocholine cytidylyltransferase cDNA from Arabidopsis thaliana.

As one of the first steps to elucidate the relationship between the structure and function of CTP:phosphocholine cytidylytransferase (EC 2.7.7.15) in plants, the cytidylyltransferase cDNA of Arabidopsis thaliana was cloned and characterized. The A. thaliana cytidylyltransferase cDNA is 1447 bp long and contains an open reading frame of 993 bp coding for a protein of 331 amino acids. The deduced structure of the enzyme was composed of three main regions; the catalytic domain in the N-terminal half, the hydrophilic C-terminal region and the amphipathic domain in the middle. The catalytic domain region was relatively well conserved among different organisms, showing 76 and 72% homology with the rat and yeast protein sequences, respectively. The hydropathy profile revealed that the C-terminal non-catalytic portion of the protein was very hydrophilic, highly enriched in negatively charged aspartic acid and glutamic acid residues. In the region between the catalytic domain and the C-terminal region, there was an amphipathic alpha-helical domain, which was believed to bind the membrane surface in the active formation. Unlike animal counterparts, there was only one potential site of phosphorylation by protein kinase C and none by Ca2+/calmodulin protein kinase II in the C-terminal region. The identity of cytidylyltransferase cDNA was verified by successful transformation of a yeast mutant defective in the enzyme activity, using an expression vector inserted with the A. thaliana cytidylyltransferase cDNA. This was further confirmed by in vivo analysis of the enzyme reaction product after labeling the yeast transformants with radioactive phosphocholine. Southern analysis indicated the presence of a single copy of the citidylyltransferase gene in A. thaliana.

Validity of the interval method for the determination of the anisotropy factor of high dose rate 192Ir sources.

PURPOSE: This study investigates the validity of the interval method to determine the anisotropy factors of high dose rate (HDR) 192Ir sources. METHODS AND MATERIALS: A calculation method, which is one of the various numerical implementations of the Sievert integral and known as the interval method, is applied to determine the anisotropy factors of a HDR 192Ir source. Calculated results from r = 1 cm to 10 cm are compared to published data. RESULTS: The comparison shows that the error resulting from the interval method increases from 1.3 to 28.1% as the radial distance increases from r = 1 cm to 10 cm and is significantly smaller than that previously reported. CONCLUSIONS: The interval method might be considered as a method that provides a very rough estimate of the anisotropy of HDR 192Ir sources whenever no conclusive anisotropy data is available.

Assessment of the relationship between ischemic damage and brain swelling in frozen brain slices.

The purpose of the study was to verify a method for the measurement of both cerebral infarction and brain swelling in frozen brain slices for histology. The animals were divided into two groups sham-operated control (n = 10) and focal cerebral ischemia group (n = 10). Focal cerebral ischemia was produced by permanent occlusion of the left middle cerebral artery. The rats were sacrificed 24 hours postocclusion. The brain was divided into two through the corpus callosum. Each hemisphere was weighted and frozen. Cerebral infarction and brain swelling were each assessed at 8 predetermined coronal planes. The volume of brain swelling was obtained by subtracting the total volume of nonischemic hemisphere from the total volume of the ischemic one. In the MCA occlusion group, brain infarction and the differences of hemispheric volume and weight between the right and left hemispheres were consistently observed, whereas sham-operated rats demonstrated no brain infarction or significant differences between two hemispheres. There were good corelationships not only between the volumes of brain edema and infarction (p < 0.05) but between the volume of brain edema and the difference in weight (p < 0.01) also. The results indicate that the measurement of the volume of ischemic brain edema in frozen brain slices may be useful in elusidating relationship with ischemic brain damage.

Prevalence of childhood asthma based on questionnaires and methacholine bronchial provocation test in Korea.

BACKGROUND: In most epidemiological survey studies, only subjective symptoms and past medical history of asthma have been used as diagnostic criteria. Even though a questionnaire survey can be performed in a large population study at low cost, limitations such as lack of objectivity and poor predictability in non-specific bronchial hyperresponsiveness cannot be avoided. OBJECTIVES: The purpose of this study was to elucidate the prevalence of current asthma based on questionnaires and methacholine bronchial provocation test, and the prevalence of atopy in Korea. METHODS: We performed modified ATS respiratory questionnaires and allergen skin-prick test with 10 common inhalant allergens among 3219 subjects aged 7-19 years in Seoul and a rural part of a small city, Chungju in Korea. Methacholine bronchial provocation tests were also performed among those who had asthma symptoms according to the questionnaire. The criteria of asthma was presence of both asthma symptoms and non-specific bronchial hyperresponsiveness. Atopy was defined as when an allergen induced weal size is same or larger than that caused by histamine. RESULTS: The prevalence of asthma based on questionnaires and methacholine bronchial provocation tests was 4.6%, while the prevalence of wheeze was 8.2% and 19.3% of total population complained of one or more respiratory symptoms related to asthma on the questionnaires. There was no significant difference according to age, sex and living area. The mean prevalence of atopy was 35.0% and the most common allergens were Dermatophagoides farinae (30.9%), Dermatophagoides pteronyssinus (27.5%), cat fur (20.4%) and cockroach (11.8%). The atopy prevalence in Chungju area was higher than that in Seoul and males showed a higher prevalence than females. The asthma prevalence was higher among atopics (6.8%) than among non-atopics (2.7%). None of questionnaire items were enough to predict the presence of bronchial hyperresponsiveness in terms of sensitivity, specificity and positive predictive value. CONCLUSION: The prevalence rate of current asthma in Korea was 4.6% and the prevalence rate of atopy in Korea was 35.0%. Questionnaire-based surveys are not enough to predict the actual prevalence of asthma.

Complete sequence of the UPP gene encoding uracil phosphoribosyltransferase from Mycobacterium bovis BCG.

The nucleotide sequence of a 799 bp fragment of Mycobacterium bovis BCG containing the putative upp gene that encodes a uracil phosphoribosyltransferase (UPRTase [EC 2.4.2.9]) has been determined. The upp gene of BCG has an open reading frame (ORF) of 621 bp (207 amino acids) starting with GTG (position 112) and ending with TGA (position 733), and its molecular mass was calculated to be 21,864 Da. Comparative analyses of the deduced amino acid sequence of BCG UPRTase with the UPRTase of six bacterial genera revealed that 24% (50/211) of the residues are perfectly conserved and 32% (67/211) of the residues are well conserved.

Calculation of the dose distribution in water from 71Ge K-shell x-rays.

The dose distribution in water from 71Ge K-shell x-rays (Eave = 9.44 keV) was calculated for various source configurations using both analytic and EGS4 Monte Carlo calculations. The point source kernel and the buildup factor are presented. The buildup factor for a point source in water has been found to increase up to about 1.1 as radial distance approaches 1 cm. Comparison between 71Ge and 90Sr/Y shows a similarity between their relative dose distribution in water. The dose distribution from a disc source was calculated using the EGS4 code and compared with the results from analytic calculation. Excellent agreement was observed, confirming the validity of analytic calculations. The dose rate at 0.01 cm from a 71Ge disc source was calculated to be about 1.3 x 10(-5) Gy MBq-1 s-1. Based on the results from this study, 71Ge activity of the order of 3.7 x 10(10) Bq (approximately 1 Ci) might be necessary to obtain dose rates typical of 90Sr/Y ophthalmic applicators. The possibility of using 71Ge as a source of radioactive stents was also investigated. A 71Ge stent was modelled as a cylindrical shell source and the dose rates were determined by Monte Carlo calculations. Some calculated results are compared with published values for a 32P-coated stent. The dose rate at 0.01 cm from a 71Ge stent has been calculated to be about 6.5 x 10(-3) Gy MBq-1 h-1, which is much lower than the reported dose rate at the same distance from a 32P-coated stent. However, an initial source activity of the order of 3.7 x 10(7) Bq (approximately 1 mCi) would easily result in a typical target dose (approximately 24 Gy) needed for intravascular stent applications. In conclusion, 71Ge sources could be used as alternatives to beta sources and, unlike high-energy (approximately MeV) beta sources, may provide easily predictable dose distributions in heterogeneous media and low dose rates, which might be beneficial for some clinical applications.

Milia en plaque associated with pseudoxanthoma elasticum.

We report a woman with numerous milia occurring in a plaque-type distribution in the postauricular area, who has suffered from cutaneous pseudoxanthoma elasticum on both sides of the neck. Milia may arise primarily or may follow a number of dermatoses. We believe this is a unique case because milia en plaque coexisting with pseudoxanthoma elasticum does not appear to have been reported previously.

Evaluation of a commercial three-dimensional electron pencil beam algorithm.

Results from beta testing of a commercially available three-dimensional electron pencil beam algorithm (CMS FOCUS, Computerized Medical Systems, Inc. St. Louis, MO) are reported. Straight on beams were evaluated at normal and extended distances, and obliquely incident beams at angles up to 40 degrees. Shaped electron fields with small circular cutouts, and narrow elongated, centered and offcentered, rectangular field shapes were investigated. Slab inhomogeneities were studied for lung and bone equivalent material, and isodose distributions for small inhomogeneities of these materials were compared with film and TLD measurements. All tests reported here were performed with electrons 6, 12, and 20 MeV from a Cl-1800 accelerator.

Supracondylar osteotomy with Ilizarov fixation for elbow deformities in adults.

Stable fixation after a corrective supracondylar osteotomy in adults is difficult because of the irregularity of the area of bony contact, displacement of the fragments, the predominance of cortical bone, and the need for early mobilisation. We have used the Ilizarov apparatus for fixation in 15 patients who were treated by complex osteotomies with displacement of fragments for cubitus varus or valgus. Most patients with cubitus varus required medial displacement with rotation of the distal fragment. Those with cubitus valgus required lateral shift of the distal fragment to reduce the medial prominence of the elbow that would otherwise result. All osteotomies united within the expected time without loss of correction, despite early mobilisation. Complications related to the fixation were few and had resolved at the long-term follow-up.

Inhibition of neointimal proliferation of rat carotid artery by sulodexide.

Sulodexide, a glycosaminoglycan-containing compound, is known to have an antiproliferative effect on vascular smooth-muscle cells, in vitro, as well as antithrombotic and fibrinolytic effects. But there are few reports about the effect of neointimal proliferation in vivo. In this study, we examined whether Sulodexide was effective in the inhibition of neointimal proliferation after vascular injury. Ten-week-old Sprague-Dawley rats were subjected to vascular injury by endothelial denudation of the common carotid artery by using a balloon catheter. They were then allocated randomly into a control group (saline 2 ml for 3 days, and then 1 ml for 18 days, IM) and a treated group (Sulodexide 10 mg/kg/day for 3 days, and then 4 mg/kg/day for 18 days, IM). Three weeks after vascular injury, we analyzed the neointimal proliferation using morphometry. The neointimal proliferation was significantly reduced in the treated group compared to the control group (Ratio of neointimal area to medial area; 118.39 +/- 6.80% in the treated group, 177.25 +/- 17.25% in the control group). This result showed that Sulodexide might be effective in reducing the rate of restenosis after balloon angioplasty.

Nitrous oxide synthase expression in placenta of preeclampsia.

To compare the expression of endothelial nitrous oxide synthase (e-NOS), which plays a major role in vasoconstriction, in preeclampsia with that in the normotensive placenta and to identify whether or not e-NOS actually influences the pathogenesis of preeclampsia, we have studied the distribution and intensity of the endothelial isoform of NOS expression in preeclampsia (n = 121) and normal (n = 14) pregnancies. Archival paraffin-embedded tissues were immunostained with a polyclonal e-NOS antibody. E-NOS was stained predominantly in the endothelial cells of the umbilical cord and along the surface of the villi, especially within the cytoplasms of the syncytiotrophoblasts with a granular and punctuated pattern, but not in the trophoblastic island or in the intravillous capillaries. The degree of e-NOS expression in pregnancy-induced hypertension (PIH) was significantly lower than in normotensive placenta, and moreover in the patients lacking abruptio. The expression of e-NOS in preeclampsia has some correlation with patient age. In conclusion, the degree of e-NOS expression in cotyledon from PIH is far less than from control, it means a kind of hemodynamic alteration of the fetoplacental circulation in preeclampsia.

Pulmonary asbestosis: radiologic-pathologic brief report.

Pulmonary asbestosis is defined as bilateral diffuse interstitial fibrosis of the lungs caused by exposure to asbestos. Many occupations are at risk for asbestos exposure, particularly in the mining, milling, manufacturing, construction, shipbuilding, and automotive industries. Therefore, the prevalence of asbestosis should be fairly widespread. The diagnosis of asbestosis can be made on either clinical or pathological grounds. We recently encountered one case of asbestosis which was confirmed histologically. On HRCT, there was ground-glass opacity with irregular linear shadows, subpleural curvilinear lines and parenchymal bands. Neither plaque nor calcification were noted. The histologic findings observed on open-lung biopsy specimen were well in accord with those in HRCT. Many asbestos-coated bodies were present along with black dust.

Antibiotic-impregnated cement beads in the treatment of chronic osteomyelitis.

Fifty-four patients with chronic osteomyelitis of the long bones were treated at Gyeong-Sang National University Hospital between 1985 and 1993 using the cement bead technique. We studied the results of thirty-one patients who were followed up for 3 years or more. The average duration of follow-up was 4 years and 2 months (range: 3 to 7 years). All of the patients were treated by a two-stage operation; primary saucerization with implantation of antibiotic-impregnated polymethylmethacrylate beads and secondary bone grafts. The most recent follow-up examinations and analyses revealed that 17 patients (55%) were completely free of infection. Ten patients (32%) required repeated procedures of curettage and/or bone grafting. Amputations were performed on 4 patients.

Efficacy of closantel against Fasciola hepatica in Korean native goats.

Closantel (Flukiver), a salicylanilide antiparasitic compound, was tested in Korean native goats infected with Fasciola hepatica. The goats were administered closantel once orally at a dose of 10 mg/kg body weight. Efficacy was monitored weekly by fecal examination of all infected animals starting the second week post-treatment and continuing for 3 weeks. Closantel elicited 80.3, 97.8 and 92.7% efficacy in goats with naturally-acquired fasciolosis at the second, third and fourth week post-treatment, respectively. It elicited a 100% efficacy in goats experimentally infected with F. hepatica metacercariae and treated at 18 weeks post-infection.

Pathological changes according to the severity of asthma.

BACKGROUND: There have been many studies concerning pathological changes in bronchial mucosa from asthmatics; however, few studies has been carried out to evaluate pathological changes according to the severity of asthma. OBJECTIVE: This study was designed to evaluate the cellular components in bronchoalveolar lavage fluid (BALF) and histological abnormalities in asthmatics according to the severity of asthma. METHODS: Bronchoalveolar lavages, bronchoscopic biopsies and ultrastructural examinations were performed in 13 asthmatics and 11 (BAL) or four (biopsies) non-asthmatic controls. The proportions of epithelial cells and correlations with PC20Meth which reflects bronchial hyperresponsiveness. Light microscopic examination revealed loss of epithelium, inflammatory cell infiltrations and thickening of the basement membrane which also showed significant correlation with PC20Meth. Hypertrophy of airway smooth muscles and hyperplasia of mucous glands were prominent in asthmatics but there was no difference according to the severity of asthma. Ultrastructural examination revealed that basement membrane thickening on light microscopic examination is due to the increased subepithelial collagen deposition with normal thickness of basal lamina. CONCLUSION: These data suggest that loss of epithelial cells, infiltration of inflammatory cells, especially eosinophils, and increased deposition of subepithelial collagen play major roles in determining the severity of asthma and non-specific bronchial hyperresponsiveness.

The role of omentopexy in tracheal transplantation in dogs.

The major step toward successful tracheal transplantation is revascularization of the grafted trachea. There are many reports that although omentopexy is an effective method to facilitate neo-vascularization in tracheal transplantations, the procedure has not been accepted universally in the transplantation field. It remains unclear whether an omentopexy can successfully revascularize tracheal graft regardless of the length of graft. This study was undertaken to assess the usefulness of omentopexy for long-segment(more than 4 cm) tracheal allotransplantation. We have performed six tracheal transplantations with omentopexy (group A) and four tracheal transplantations without omentopexy (group B) in mongrel dogs from July 1993 to February 1995. Five mid-portion tracheal rings were removed from ten donor dogs and ten corresponding tracheal rings were removed from the ten recipient dogs. The excised tracheal rings from the donors were transplanted to the recipient tracheal-excised sites. All the recipients were given cyclosporine, azathioprine, and prednisolone for immunosuppression in the post-operative period. The histologic results of all the surviving members of group B were better than those of the group A. These findings indicate that omentopexy has a limitation, it is not a major method for graft revascularization. Therefore the length of the tracheal graft was greater than 4.0 cm, for its viability, a longer tracheal graft requires some other blood supply aside from the omentopexy.

Metacercarial production of Lymnaea viridis experimentally infected with Fasciola hepatica.

Three groups of Lymnaea viridis snails were infected with one, three or five Fasciola hepatica miracidia, and their metacercarial production was observed. In all groups cercarial shedding commenced in some snails as early as day 27 post-infection. The duration and pattern of cercarial shedding varied from snail to snail in each group. Snails infected with three or five miracidia produced more metacercariae than those infected with a single miracidium. The overall metacercarial production was influenced by the temperature at which the snails were kept during infection.
