Current contributions of peptide synthesis to studies on brain-gut-skin triangle peptides.

The usefulness of a strong acid, such as MSA or TFMSA/TFA, as a deprotecting reagent in peptide synthesis was examined. By synthesizing several structurally related brain-gut-skin triangle peptides, a number of advantageous features of the thioanisole-mediated deprotecting procedure were demonstrated. New amino acid derivatives, Arg(Mts), Trp(Mts) and Asp(OChp), were introduced to improve the synthetic methodology of complex peptides and the superior properties of Cys(Ad) were evaluated.

The granulopoietic effect of human urinary colony stimulating factor on normal and cyclophosphamide treated mice.

A practically endotoxin-free colony stimulating factor from human urine (CSFHU) was prepared and its granulopoietic effect on normal and cyclophosphamide treated mice was examined. When normal C57BL/6N mice were injected intraperitoneally with 2.5 X 10(6) units/kg of the CSFHU daily for a 5-day period, the numbers of progenitor cells (CFUC) in the femur and spleen were significantly increased. The CFUC in the femur and spleen reached a maximum at day 3 (270%) and day 5 (250%) after the initial injection, respectively. The increase in number of CFUC in the femur exhibited a dose-dependency with respect to the CSFHU and a significant increase was observed even at 4 X 10(5) U/kg (P less than 0.05). However, neither granulocytosis nor monocytosis occurred in normal C57BL/6N mice injected with the CSFHU. In cyclophosphamide induced leukopenic C3H/HeN mice, daily injections of the CSFHU at 2.5 X 10(6) U/kg for 5 days stimulated the restorative granulocyte production (P less than 0.05) as well as the CFUC recovery in both the femur and spleen. These findings suggested that the CSFHU might be involved in granulocyte production in vivo.

Antimicrobial effect of human serum IgA.

Serum IgA, IgG and colostrum secretory IgA prepared from specimens pooled from a large number of human beings were shown to have measurable levels of antibodies against Escherichia coli, Pseudomonas aeruginosa, Klebsiella Pneumoniae, poliovirus, Coxsackie B virus, echovirus and influenza virus. Serum IgA exerted a bacteriostatic effect in vitro on E. coli and P. aeruginosa, which increased in the presence of the iron-binding proteins lactoferrin and transferrin. This bacteriostasis was reduced when the iron-binding proteins were saturated with iron. Similar results were obtained with IgG and secretory IgA. The bacteriostatic effect of serum IgA was also shown in vivo, in the peritoneal cavity of mice. The effect was suppressed by iron. Iron-chelating substances, siderophores, excreted by E. coli diminished the co-operative bacteriostatic effect of serum IgA and transferrin. Siderophore production by E. coli was inhibited in the presence of serum IgA, but not when serum IgA was deprived of specific antibody by absorption with E. coli. These results indicate that serum IgA has a potent bacteriostatic effect in co-operation with transferrin or lactoferrin because of the inhibitory effect of the specific antibody on siderophore production by E. coli.

Clinical and virological features of herpes genitalis in Japanese women.

We report herein clinical and virological findings in 35 Japanese women with a confirmed herpes genitalis. The frequency of herpes genitalis was 0.24% in our obstetrical and gynecological outpatient clinic, while that in cancer screening clinics was 0.01%. Eighty percent of all patients were between 20 and 49 years of age (median age of 38.5 years). Genital herpes was detected more frequently in summer and autumn. Virus isolations were carried out in 20 patients and 13 were positive (65.0%). Of 13 genital isolates with positive cultures, 7 (53.8%) were identified as herpes simplex virus type 1 (HSV-1) and 6 (46.2%) as herpes simplex virus type 2 (HSV-2). Two virgins were diagnosed virologically and cytologically as having progenital herpes, and in these 2 women, HSV-1 was isolated by the virus isolations. Lack of antibodies to either virus was higher in HSV-1 type infection. These studies together with previous findings show that both clinical and virological features of genital herpes differ between Japanese and Caucasian or black women. These differences are attributed to some extent to sexual patterns of behavior in the different races.

Structural studies of asparagine-linked sugar chains of human ceruloplasmin. Structural characteristics of the triantennary complex type sugar chains of human plasma glycoproteins.

The structures of the oligosaccharides released from human ceruloplasmin by hydrazinolysis were studied by sequential exoglycosidase digestion in combination with methylation analysis and periodate oxidation. A molecule of ceruloplasmin was shown to have two biantennary oligosaccharides A-1 and one triantennary oligosaccharide A-2, the structures of which are shown below. Studies on peptides. LXXXI. Application of a new arginine derivative, NG-mesitylene-2-sulfonylarginine, to the synthesis of substance P and neurotensin.

A new devised arginine derivative, NG-mesitylene-2-sulfonylarginine, Arg(Mts), was employed for the synthesis of hypothalamic substance P and neurotensin. The former was obtained in 74% yield by treatment of the protected undecapeptide amide, Z - Arg(Mts) - Pro - Lys(Z) - Pro - Gln - Gln - Phe - Phe - Gly - Leu - Met(O)-NH2, with methanesulfonic acid in the presence of anisole followed by reduction of the sulfoxide with 2-mercaptoethanol. The latter was obtained in 54% yield by the similar treatment of the protected tridecapeptide ester, Z - Pyr - Leu - Tyr - Glu(OBzl) - Asn - Lys(Z) - Pro - Arg(Mts) - Arg(Mts) - Pro - Tyr - Ile - Leu - OBzl, with methanesulfonic acid. As scavenger, a mixture of anisole-thioanisole-o-cresol (1:1:1, by vol.) was employed to suppress the side reaction, O-mesitylene-2-sulfonation of the Tyr residue.

Polygraphic study on the clinical seizures induced during nocturnal sleep.

The subjects of this study consisted of 17 epileptic patients with clinical seizures during all-night sleep recording (the seizure group) and another 17 epileptic patients without clinical seizures (the control group). The results obtained were as follows: 1) The 50-90% out of total clinical seizures were induced in non-REM sleep, while a few clinical seizures were induced in REM sleep. The number of clinical seizures in sleep stage of higher activity level increased as the incidence of clinical seizure during all-night grew. 2) In the seizure group REM sleep could not be detectable during all-night in five cases out of 17 cases, while we could not find cases lacking REM sleep in the control group. This difference between two subject groups was statistically significant (P less than 0.025). 3) As to focal spike group, the spike discharge incidence of total sleeping time and of each sleep stage was higher respectively in the seizure group than that in the control group, and particularly in St.1, St.2 and REM sleep the figure of the discharge incidence was found significantly higher in the seizure group than that in the control group (P less than 0.05). We discussed on above-mentioned results, and we emphasized particularly that REM-sleep suppresses clinical seizures, although non-REM sleep induces clinical seizures.

Characterization of one neutral and two acidic glycoasparagines isolated from the urine of patients with aspartylglycosylaminuria (AGU).

One neutral and two acidic glycoasparagines were isolated from the urine of patients with aspartylglycosylaminuria (AGU). The neutral one was identified as beta-Gal-(1 leads to 4)-beta-GlcNAc-Asn. The acidic ones were composed of 1 mole of sialic acid and 2 moles each of galactose and N-acetylglucosamine, attached to asparagine, and were isomeric with respect to the position of sialic acid attachment since they produced the same glycoasparagine on incubation with the neuraminidase [EC 3.2.1.18] from Clostridium perfringens. The structure of the resulting sialic acid-free glycoasparagine was determined to be beta-Gal-beta-GlcNAc-beta-Gal-(1 leads to 4)-beta-GlcNAc-Asn based mainly on the results of sequential enzymatic degradations.
