New synthetic inhibitor to the alternative complement pathway.

The inhibitory effects of 6-amidino-2-naphthyl 4-guanidinobenzoate (FUT-175) on the activities of factor B, factor D and cobra venom factor (CVF) X Bb were examined. FUT-175 bound specifically to the Bb fragment of factor B or CVF X Bb. FUT-175 was a non-competitive inhibitor of the esterolysis of L-leucyl-L-alanyl-L-arginine naphthylester by factor B and CVF X Bb. FUT-175 also inhibited the haemolytic activity of factor B, the C3 convertase activity of CVF X Bb and the factor B-cleaving activity of factor D. The concentration of FUT-175 causing 50% inhibition of these activities was 10(-5) - 10(-4)M.

Effect of the synthetic protease inhibitor [N,N-dimethylcarbamoyl-methyl 4-(4-guanidinobenzoyloxy)-phenylacetate] methanesulfate on carcinogenesis by 3-methylcholanthrene in mouse skin.

The effect of the potent synthetic protease inhibitor [N,N-dimethylcarbamoylmethyl 4-(4-guanidinobenzoyloxy)-phenylacetate] methanesulfate (FOY-305) on skin carcinogenesis in ddY mice was examined over a total observation period of 105 days. Administration of 0.1% FOY-305 in the diet suppressed the incidence of carcinomas induced by repeated local application of the carcinogen 3-methylcholanthrene (MCA) (P less than .05) in mouse skin and delayed the time of appearance of the skin tumors (P less than .001). There was no significant difference in the number of tumors per tumor-bearing mouse and the size of the tumors between mice treated with MCA and mice treated with MCA plus FOY-305.

Theoretical and experimental approach to recognition of amino acid by tRNA.

The C4N model concludes that the pocket on the complex of the anticodon bases and the discriminator base on a tRNA can accept the corresponding protein amino acid. A theoretical molecular computation by using the energy minimum principle is carried out to find the energy-refined C4N conformation. The results for Gln and Val suggest that not only the lock and key relationship but the induced fitting effect occurs. Some experiments to detect the interaction between the polynucleotides and the amino acid are being attempted.

Flexibility and rigidity of left-handed Z-DNA.

The local variation of torsional angles and helical parameters in Z-DNA was analyzed. The sugar phosphate backbone is fairly rigid but the angles at GpC linkage are more changeable than those at CpG linkage in order to form a variety of structures. The water channel at minor groove is important to stabilize and retain the novel Z-DNA helix.

[Evaluation of left ventricular asynergy by two-dimensional echocardiography: comparison between fixed external axis system and floating axis system].

Detection and quantitative estimation of segmental asynergy in the infarcted area has been impeded by the rotation and anterior movement of the heart during systole. In the present communication, we compared the two measurements determined by the floating and fixed external axis systems. The former takes into account of the effect of cardiac movement. Furthermore, we assessed which system is superior by the left ventriculographic and coronary arteriographic analyses. The materials consisted of 24 cases of myocardial infarction. There was no significant difference between the two systems in the estimation of the number of asynergic segments or between echocardiographic and ventriculographic findings. The floating axis system had a tendency to underestimate asynergy, so that the number of hypokinetic segment estimated by the floating system was greater than that by the fixed external axis system. Therefore, the fixed external axis system was superior to the floating axis system in determining the causative coronary artery.

Molecular structure of (m5 dC-dG)3: the role of the methyl group on 5-methyl cytosine in stabilizing Z-DNA.

The hexamer (m5 dC-dG)3 has been synthesized and its three-dimensional structure determined by a single crystal X-ray diffraction analysis. The structure has been refined to a final R value of 15.6% at 1.3 A resolution. The molecule forms a left-handed Z-DNA helix which is similar to the unmethylated Z-DNA structure. The presence of the methyl group has resulted in slight changes in the twist angle between successive base pairs and modification of some of the interatomic contacts. Methylation of cytosine in the C5 position is associated with a relative destabilization of the B-DNA structure and a stabilization through hydrophobic bonding of the Z-DNA structure.

Molecular structure of r(GCG)d(TATACGC): a DNA--RNA hybrid helix joined to double helical DNA.

The molecule r(GCG)d(TATACGC) is self-complementary and forms two DNA--RNA hybrid segments surrounding a central region of double helical DNA; its molecular structure has been solved by X-ray analysis. All three parts of the molecule adopt a conformation which is close to that seen in the 11-fold RNA double helix. The conformation of the ribonucleotides is partly determined by water molecules bridging between the ribose O2' hydroxyl group and cytosine O2. The hybrid-DNA duplex junction contains no structural discontinuities. However, the central DNA TATA sequence has some structural irregularities.

2-amino-2-deoxy-D-erythrose in Agaricus bisporus.

2-Amino-2-deoxy-D-erythrose was isolated from the cell wall of the fruit body of Agaricus bisporus. The structure of the amino sugar was determined by mass spectrography and 1H-NMR spectrography of its acetylated derivative and by paper chromatographic comparisons with authentic 2-amino-2-deoxy-D-erythrose. This amino sugar is a component of the glycoprotein fraction from the cell wall. Its content in the glycoprotein increased markedly, especially during the ripening stage of the fruit body.

A sensitive colorimetric assay for thrombin, prothrombin and antithrombin III in human plasma using a new synthetic substrate.

Benzoyl-L-leucyl-L-alanyl-L-arginine-alpha-naphthylester (Bz-Leu-Ala-Arg-NE) was synthesized as a new substrate for use in the assay of thrombin. In the assay alpha-naphthol released by the enzyme reaction was measured colorimetrically. With Bz-Leu-Ala-Arg-NE as substrate, the minimum detectable concentration of human thrombin was 0.0025 U. This assay using Bz-Leu-Ala-Arg-NE is a highly sensitive method for detecting prothrombin, thrombin and antithrombin III in human plasma. Prothrombin could be determined with 0.2 microliter of human plasma using Echis carinatus venom (ECV) as activator. Antithrombin III activity could be determined with 2 microliter of human plasma using human thrombin and heparin as cofactor. A zymogram of human prothrombin was prepared with Bz-Leu-Ala-Arg-NE as substrate. The preparation gave one band (pI 4.9) on polyacrylamide disc gel isoelectrophoresis.

Effects of oral and intravenous fat administration on the levels of apoproteins A-I, A-II and C-III in human.

Changes of plasma levels of apoproteins A-I, A-II and C-III were determined after oral and intravenous fat administration. The A-I levels increased in three out of four subjects after fat ingestion but no changes or even a slight decrease in the levels of A-I were observed after intravenous fat infusion. The A-II levels also increased after fat ingestion in two subjects but the levels either did not change or decreased slightly after fat infusion. The levels of C-III increased concomitantly with the increase of triglyceride levels after fat ingestion as well as fat infusion . After intravenous fat infusion, part of the C-III in the d greater 1.006 fraction shifted to the lighter fraction (d less than 1.006). These observations suggest that the increase in the levels of A-I and A-II after fat ingestion are a consequence of an increase in apoprotein synthesis in the intestine during fat absorption. The increase in the levels of C-III after fat ingestion as well as fat infusion seemed to be related to the capture of C-III inthe triglyceride rich particles, i.e. C-III accumulated in the circulation with triglyceride-rich particles. However, it appeared also to be possible that the rate of C-III synthesis increases during hyperlipidemia induced by fat infusion.
