Resolution of parentage in dogs by examination of microsatellites after death of putative sire: case report.

A case of disputed paternity in dogs is reported. DNA examinations were carried out from hair samples of the individuals several months after the death of the putative sire. Ten short tandem repeat (STR) loci were analysed by fluorescence-labelled multiplex PCR using ABI PRISM 310 Genetic Analyser. Based on the results the candidate sire was included in the pedigree records as the biological sire. In spite of the genetic homogeneity of pedigree dogs due to inbreeding, canine microsatellites can provide an adequate basis for assigning paternity in pure breeds.

Alteration of body composition in rats: effect of organic chromium and L-carnitine.

A growth trial was carried out to test the effect of organic, trivalent chromium and L-carnitine on the body composition of growing rats. At the same time, an evaluation of different measurement methods (weight of epididymal fat pad, adipocyte morphometry, total body electrical conductivity) was performed. Outbred Wistar rats of 30 days of age were fed diets of different (0, 10 and 20%) protein level. The diets were supplemented with 4 mg/kg Cr as chromium nicotinate, and 100 mg/kg L-carnitine. The experimental feeding lasted 15 days, after a 5-day-long adjustment period. It was found that Cr addition increased feed intake. Both treatments caused changes in body composition, increasing fat and protein deposition. Organic chromium had no effect at either protein level, while L-carnitine improved the protein retention only at an optimum (20%) protein supply. No statistically significant correlation was found between total body electrical conductivity (TOBEC) and body composition, which could be attributed to the great individual differences. A close correlation was found among total body fat percentage, weight of epididymal fat pad and the adipocyte surface. The data suggest that there is an interaction between dietary protein supply and the effect of repartitioning agents.

[Diagnostic and preventive factors in the background of attempted suicide--review of two-years results of the Pécs Center in the WHO/EURO Multicenter Study of Attempted Suicide].

Since parasuicides are not being systematically recorded in Hungary, a monitoring project within the framework of the WHO/Euro Multicentre Study on Parasuicide was started in Pecs catchment area. The study is aimed both to monitor the medically treated parasuicides and to predicts future acts (repetition prediction study) through a follow-up investigation. Pecs centre has been participated for two years in this study. The recent results of this survey are presented and analysed: 553 monitored suicidal events has been collected; 101 structured interview and 37 follow-up cases has been conducted in this period. The most important epidemiologic sociodemographic and psychosocial factors are discussed. Authors paid special attention to the presuicidal psychological characteristics especially the cry for help of the patients, emphasizing the specific diagnostic and preventive possibilities.

[Detection of minimal residual diseases in B-cell tumors using PCR specific for the immunoglobulin heavy chain gene].

In B-cell non-Hodgkin's lymphomas (NHL), clonal rearrangement of the immunoglobulin heavy chain (IgH) gene provides a useful marker for the detection of minimal residual disease (MRD) after treatment. To explore clinical usefulness of polymerase chain reaction (PCR) analysis of clonal IgH gene rearrangement in the detection of MRD a follow up study of 10 patients with B-cell NHL have been performed. At the time of diagnosis, tumor DNAs were PCR-amplified using sense primer specific for the heavy chain variable region (VH) and antisense primer specific for the heavy chain joining region (JH) of the IgH gene. The clonal rearrangement of IgH gene detected by PCR was used as clonal marker to determine MRD after treatment. In three cases, where clinical remission was not achieved, clonal IgH gene rearrangement was detected after the treatment. In seven cases, clinical remission was achieved after induction therapy but the PCR analysis revealed clonal IgH gene rearrangement in three of the cases. In all of the three cases, where MRD was detected by PCR, clinical relapse developed after 7-28 months of the therapy. In all cases that have relapsed, the IgH gene rearrangement was identical at the time of initial diagnosis and at the relapse. This study demonstrates that PCR analysis of clonal IgH gene rearrangement is a useful method to monitor and detect MRD before clinical relapse.

[Comparative study of the attitudes of students and nurses to physician-assisted suicide--first results].

While the popular media and the professional literature deal with the topic of euthanasia intensively, the problems of physician-assisted suicide received insufficient attention in Hungary. The authors review the most important details of the physician-assisted suicide. A twelve-item scale to measure attitude toward PAS (constructed and validated by G. Domino) was administered to the samples of Hungarian social science, medical students and nurses. The distributions of responses for the 12 items for the social and medical samples are compared and discussed. Also a cross-cultural comparison was made with an American student sample examined by Domino. The social science students who have the fewest personal experiences with serious, or terminally ill patients are the most liberal group, characterised by the most permissive attitudes toward PAS. Nurses who have everyday contact and experience with these patients are the most conservative; they show more or less conclusively the lowest acceptability rate of PAS. The attitudes of medical student's group, of the would-be physicians are between them, and they are the most controversial and ambivalent. Future research is necessary to get more information about attitudes toward physician-assisted suicide.

[A case of datura stramonium poisoning--general problems of differential diagnosis].

In past year drug abuse becomes more and more general in Hungary. In addition to consume traditional drugs, other substances are used frequently too. One of them is the Datura stramonium, which contains alkaloids (mostly atropine), and can result in hallucinations. Therefore Datura stramonium is seemingly becoming popular as a hallucinogenic drug. The consumption of any part of the plant causes atropine intoxication, thus anticholinergic delirium. Differential diagnosis of Datura intoxication can be difficult in the everyday medical practise. In our paper the symptomatology, diagnosis, differential diagnosis, and therapy of Datura intoxication are discussed and we report one of our cases.

Ovarian consequences of low dose peroral Fusarium (T-2) toxin in a ewe and heifer model.

The effect of low dose peroral Fusarium produced T-2 toxin intake upon the ovarian function was evaluated in ewes (n = 30; Trial 1) and heifers (n = 7; Trial 2). Half of the ewes and all of the heifers were fed rich, acidosis-inducing concentrate. The 30 ewes were divided into 6 groups of 5 animals each. They were given 0, 0.3 or 0.9 mg/day (0, 5 or 15 ug/kg) purified T-2 toxin per os for 21 days (3x2 factorial design). Four of the 7 heifers were fed 9 mg/day (25 ug/kg) of the same purified T-2 toxin for 20 days while 3 remained untreated. The estrus cycles in all animals were synchronized prior to the trials and the T-2 exposure was started in the mid-luteal phase. The acidic condition in the rumen was estimated by the determination of urinary net acid-base excretion. The ovarian activity was followed with blood sampling for progesterone on alternate days (Trial 1) or with ultrasonography and sampling for progesterone daily (Trial 2). All of the heifers and concentrate-fed ewes showed a compensated acidosis, during first two thirds of T-2 exposure. In Trial 1, ovarian malfunction manifested as lower P4 peak concentration in the midluteal phase, shortening of the CL lifespan and prolonged follicular phases. These malfunctions were detected in 3 and 3 ewes fed concentrate and 0.3 mg and 0.9 mg T-2 toxin. Lower P4 peak concentration was observed in 1 ewe fed regular diet and 0.9 mg T-2 toxin. None of the control and acidotic groups (0 mg T-2), or ewes fed regular diet with 0.3 mg T-2 showed any ovarian malfunction. In Trial 2, after PGF2, administration the ovulation occured later and the plasma progesterone level remained low (< 3 nmol/l) for a longer period in T-2 treated heifers, than their untreated control mates (5.0+/-0.7 vs 3.7+/-0.5 d, P<0.05 and 8.3+/-0.4 vs 6.3+/-0.9 d, P<0.01, respectively). These results show that the peroral T-2 intake can significantly retard the folliculus maturation and ovulation and perhaps the subsequent luteinisation also in ruminants kept on concentrate-rich diet.

Dog feeding test for assessing the nutritional adequacy of practical diets.

The nutritive value of dog foods declared by the manufacturer as nutritionally complete and balanced can be best assessed by feeding trials with dogs. A protocol of a feeding trial has been developed and tested with working dogs fed two different commercial complete and balanced diets for 8 weeks. The parameters used for evaluating the effect of diets were general health status, body and hair coat condition, change of body weight, haematological parameters (white blood cell (WBC) count, red blood cell (RBC) count, haemoglobin, packed cell volume), and biochemical parameters in blood serum (alanine aminotransferase, urea, albumin). The trial protocol proved to be appropriate to monitor the dogs' nutritional status and to reveal differences between diets. This method of evaluation is recommended for use in supporting the nutritional claims (labelling) of dog foods.

[Positron emission tomography ia an effective tool in modern oncology].

A total of 399 positron emission tomography (PET) examinations were carried out with a GE 4096 Plus PET scanner during the past 5 years on patients referred to the National Institute of Oncology in Budapest. The majority (n = 316) of these investigations were performed with the use of [18F]-fluorodezoxyglucose (FDG) to map the glucose metabolism; [11C]-methionine PET was indicated in 79 cases to detect protein transport and metabolism. The perfusion tracer [15O]-butanol was applied in only 4 cases to answer certain oncology-related, differential diagnostic questions. The oncological examinations were related to primary diagnostics, staging/restaging and therapy monitoring. In the staging/restaging and therapy monitoring of known tumours, conclusive results were achieved in 81-82% of the cases by using either FDG or [11C]-methionine as tracer. The concordant numerical data indicated that the PET investigation provides a definite answer to the question of the presence or absence of viable tumour tissue, with similar effectivity in any of the above indications, no matter whether FDG or [11C]-methionine is used. The search for occult primary tumours was the most frequent indication within the primary diagnostics: 10 (37%) primaries were localized by using FDG PET in the 27 investigated cases. This is a remarkably high value, especially in view of the failure of all the conventional diagnostic procedures carried out prior to the PET investigations. Application of PET may be indicated in all cases when the ultimate question is a non-invasive estimation of viable tumorous tissue.

A case of bilateral testicular lymphoma.

Authors report on a 75-year-old man with bilateral testicular lymphoma. He complained of painless right testicular enlargement. Orchidectomy was indicated by ultrasound examination and the diagnosis (large cell, non-Hodgkin lymphoma B-cell origin) was established by histology and immunohistochemistry. Two months later, the left testis enlarged, orchidectomy was performed, and a lymphoma with identical histology was found. PET revealed retroperitoneal spread of the tumor. Irradiation (18 Gy) was applied. Three months later, because of gastric metastases of the lymphoma the patient underwent CVP and CAVP (Cyclophosphamide, Adriablastin, Vincristin, Prednisolone) chemotherapy. Despite of the repeated courses, eleven months after the primary diagnosis the patient died due to of multiple metastases.

Studies on the energy content of pigeon feeds I. Determination of digestibility and metabolizable energy content.

The digestibility coefficient and metabolizable energy (ME) content of the most important pigeon feeds (corn, wheat, barley, red and white millet, sorghum, canary seed, peas, lentils, sunflower, and hemp) were determined. The experiment was carried out using 10 adult male homing pigeons. All feeds were fed alone, in a whole-grain form, ad libitum. Drinking water and grit were offered to the birds on a continuous basis. Each feedstuff was fed to five pigeons in 1-wk cycles. There was no significant difference between the values determined in pigeons and those reported in the literature for chickens among the digestibilities of the CP of the various feeds. For pigeons, the digestibility of carbohydrates (N-free extracts, NFE) was lower (e.g., 62.37 vs 83.00% for barley and 63.45 vs 77.00% for peas), whereas the ether extract (EE) was higher (e.g., 75.58 vs 61.00% for barley and 82.59 vs 80.00% for peas) in pigeons compared with chickens. As a result, the AMEn values determined in pigeons did not differ significantly from those reported for chickens but tended to be slightly higher. For feeds of high-oil content, that difference may be somewhat larger. The correlation between the CP, EE, crude fiber (CF), and NFE contents of the feeds and the ME values determined in this experiment were calculated by multivariate linear regression. It was concluded that it was more accurate to determine and tabulate the ME contents of other potential pigeon feeds directly by experimental methods rather than using an equation.

Studies on the energy content of pigeon feeds II. Determination of the incorporated energy.

The effect of breed and sex on adult body composition of four pigeon breeds: Texan (TEX), Mondain (MON), Szeged Tumbler (SZT), and homing (HOM) and on the digestibility coefficients (DC) and metabolizable energy (ME) content of their feeds was studied. A total of eight groups, each comprising five males and five females of each breed, were used. All birds were fed the same pelleted pigeon feed (17.27% CP) ad libitum. After the metabolic study, the whole body was analyzed for dry matter (DM), ash, CP, and ether extract (EE) contents. The DC of DM, TEX, MON, and HOM pigeons did not differ significantly, whereas DC for the SZT breed were consistently lower. In the majority of cases, the DC values obtained for males were higher, irrespective of the breed. The body composition of the two sexes was first compared within a given breed, and no major differences were detected. Interbreed differences were greater. The DM content of the body of HOM pigeons exceeded that of the other three breeds for both sexes. For ash and CP content of the body, the reverse was found, i.e., the values of both parameters were lower in HOM pigeons. The HOM pigeons had significantly (P < 0.05) higher body fat (EE) content as compared with the other three breeds except TEX females. The NFE value for HOM also tend to be higher than in the other three breeds.

Study of the soil-plant (carrot)-animal cycle of nutritive and hazardous minerals in a rabbit model.

Carrots were grown on soils polluted by heavy metal salts. Each particular microelement reached a high concentration [molybdenum (Mo) 39.00, cadmium (Cd) 2.30, lead (Pb) 4.01, mercury (Hg) 30.00, and selenium (Se) 36.20 mg/kg dry matter] in the carrot. In a metabolic balance trial conducted with 15 male and 15 female New Zealand White rabbits, the control animals (n = 5) were fed ad libitum with concentrate as basal diet, while the other rabbits received the basal diet and carrots containing the particular microelement. Blood samples were taken to determine the activity of serum enzymes. To investigate the metabolism of Mo, Cd, Pb, Hg and Se, samples were taken from the heart, liver, lungs, kidneys, spleen, ovaries/testicles, entire digestive tract, adipose tissue, femur, hair, faeces and urine. Carrot had significantly higher digestibility for all nutrients than the rabbit concentrate. Carrot samples of high Pb content had the lowest digestibility of crude protein. The microelements differed in their rate of accumulation in the organs examined: Mo and Cd accumulated in the kidneys, Pb in the kidneys, liver, bones and lungs, Hg in the kidneys and liver, while Se in the liver, kidneys and heart. The proportions of microelements eliminated from the body either via the faeces and urine (Mo 80.18% and Se 47.41%) or via the faeces (Cd 37.86%, Pb 66.39%, Hg 64.65%) were determined. Pathohistological examination revealed that the rate of spermatogenesis was reduced in the Mo, Cd, Pb and Hg groups compared to the control. Lead, Cd and Hg intake resulted in a considerable decrease in gamma-glutamyltransferase (GGT) and in an increase of alkaline phosphatase (ALP) activity because of damages to the kidneys and bones. All experimental treatments decreased the activity of cholinesterase (CHE) because of lesions in the liver.

[A new radiation-free conditioning in bone marrow transplantation and dibromo-mannitol therapy in chronic myeloid leukemia].

A new, radiation-free, conditioning protocol, containing the original Hungarian mitobronitol (DBM) (DBM/ cytosine arabinoside/cyclosphosphamide) has been applied to 36 chronic myeloid leukemia (CML) patients followed by bone marrow transplantation (BMT) from HLA identical sibling donors between 1990-1997. In spite of some prognostically disadvantageous factors (half of them were above 40 years, 10 out of 36 patients were in accelerated phase, the disease history was longer than 2 years in average) the overall survival (30/36) and the leukemia free survival rate (26/36) were in accordance with the best international results. Transplantation-related toxicity was remarkably reduced in comparison to bone marrow transplantation performed by total body irradiation/cyclophosphamide (TBI/Cy) or busulphan/cyclophosphamide (Bu/Cy) conditioning protocols. Acute graft versus host disease was present in lower percentage (9/36) and the number of serious cases was only 2/36. Chronic GVH disease, generally known to be associated with antileukemic effect (GVL), occurred in 25 of cases. Early haematological relapse among the 34 patients with functioning graft occurred in 6 patients which rate is slightly higher than reported after TBI/Cy or Bu/Cy conditioning treatment. There was no relapse among patients transplanted within one year post-diagnosis and patients having CML with accelerated phase. The leukemia free post-transplant period was in association with the chronic GVH disease and full chimeric state.

Evaluation of the nutritional value of the amaranth plant. I. Raw and heat-treated grain tested in experiments on growing rats.

Two experiments were carried out on rats to determine the nutritional value of raw and autoclaved amaranth (Amaranthus retroflexus) seeds. The test groups of the first experiment were fed only ground amaranth seed. The control group received pure maize meal. Both test groups showed higher growth rate than the control group, due to the higher protein content of amaranth. The final body weight of rats fed autoclaved amaranth seed significantly exceeded that of rats fed raw amaranth seed. The Net Protein Utilisation (NPU) index of raw and autoclaved amaranth was determined in the second experiment. The diets had 10% crude protein concentration and the only protein source was the meal of raw and autoclaved amaranth seed. It was found that the heat treatment of amaranth increased the body weight gain of rats even when the animals consumed diets containing only 10% crude protein. The feed consumption was also higher with the diet containing autoclaved seeds. Despite the finding that there was no significant difference in the NPU index, data suggest the presence of a heat-labile antinutritive compound.

Assessment of nutritional adequacy of the protein in dog foods by trials on growing rats.

In order to offer methods for assessing the protein quality of dog foods, eight commercial dry-type complete dog foods were analysed by in vitro chemical methods (crude protein, amino acid composition, in vitro pepsin digestibility) and rat growth trial (weight gain = WG, feed efficiency = FE, protein efficiency ratio = PER, net protein ratio = NPR, and net protein utilisation = NPU). Three individual samples were taken from each dog food and their homogenous mixture was used as an ingredient for the test diets fed to six rats per diet. All dog food samples were used as a single protein source in isonitrogenous (10% crude protein) and isoenergetic (ME = 14.9 MJ/kg) complete diets. Rat weight gains showed significant differences among most of the test diets as a consequence of the differences in protein quality of the dog food samples. The protein quality indices of the dog food products showed significant variations. The measured ranges of protein quality indices of the samples were as follows: WG = -5.4-21.4 g; FE = 0-0.25 g/g; PER = 0-2.54 g/g; NPR = 1.06-3.52 g/g; NPU = 19.93-65.93%. The limiting amino acid, the sum of essential amino acids and the chemical scores were calculated from gross amino acid composition. Tryptophan was the first limiting amino acids in all samples but one. Methionine was found to be the second limiting amino acid. The crude protein content of the samples showed no correlation with indices of protein quality, indicating that the declaration of crude protein is not a sufficient information to judge the protein quality of dog foods. The declaration of NPR, or NPU indices may be recommended for labelling the protein quality of premium-type dog foods.

Relationship of dietary fat and lysine level with body composition in broiler chickens.

The effect of dietary fat and/or lysine supplementation on the body composition of broiler chickens was studied. Body composition was measured by computed tomography (CT) and direct chemical analysis. Cockerel chicks from a previous experiment (Fekete et al., 1992) were used in this study. Ten chickens from each treatment group were euthanised, deep frozen and subjected to CT. Subsequently the bodies were dissected and ground to obtain homogeneous samples for chemical analysis. Supplementation of the diet with 6 g/kg lysine did not change total body composition but positively influenced the final body weight. In the treatment group receiving 40 g/kg added fat and 3 g/kg feed lysine (F-LYS-I) the higher body weight ran parallel with a higher fat content. The difference between the frozen body weight and final live body weight shows that lysine supplementation increased the water-holding capacity of muscle, which was the best in groups receiving a diet of normal energy content plus high lysine supplementation.

Effect of methionine and its derivatives on the weight gain and protein utilisation of growing rats.

Methionine and its derivatives are used as feed supplements. The objective of this study was to compare the effectiveness of DL-methionine (MET), DL-methionine-hydroxy-analogue (2-hydroxy-4-methylthio-butyric acid, MHA), and DL-S methyl-methionine-sulphonium-chloride (SMM) in rats using low- (10 g/100g, BD10), medium-(15g/100g, BD15) and high-protein (20 g/100 g, BD20) diets. In a 10-day growth assay with weaning male rats (Wistar, SPF, LATI, Gödöllo, Hungary, 6 rats/group) methionine-deficient basic diets (BD10, BD15, BD20) were supplemented with 0, 1.5 ad 3.0 g/kg MET, MHA OR SMM. Extracted soybean meal was used as sole protein source. A protein-free diet was used for determining net protein ratio (NPR) indices. The weight gain of rats fed test diets fortified with 1.5 and 3.0 g/kg MET, MHA, or SMM was significantly higher (p < 0.05) than that obtained with the unfortified diets. The NPR index of the diets was increased by MET, MHA or SMM supplementation, which increase was significant (p < 0.05) at 3.0 g/kg fortification level. However, there were no significant differences between the efficiencies of methionine derivatives on the same dietary protein levels. The weight gains of the rats were higher and the NPR indices of the diets were lower at a higher dietary protein level.
