Excess wild-type p53 blocks initiation and maintenance of simian virus 40 transformation.

Wild-type (wt) murine p53 has been tested for its ability to block and reverse the transforming effects of simian virus 40 (SV40) large T antigen. Established and precrisis mouse cells overexpressing exogenously introduced wt p53 became resistant to SV40 transformation. The introduction of excess wt p53 into SV40-transformed precrisis cells reverted their transformed phenotype. However, the phenotype of SV40-transformed established cells was not reverted by excess wt p53. We conclude that an antioncogenic action of wt p53 is exerted during SV40 transformation and that in precrisis cells, the antitransforming action of wt p53 can be exerted both at initiation and during the maintenance of transformation.

Effects of three sodium-potassium adenosine triphosphatase inhibitors.

Reports from several laboratories suggest the presence of an ouabainlike compound in plasma and various animal tissues, particularly during acute volume expansion and in low-renin hypertension. It has been hypothesized that this compound, through inhibition of the Na(+)-K+ pump, can constrict blood vessels, enhance vasoconstriction in response to agonists, increase cardiac contractility, raise blood pressure, and cause natriuresis/diuresis and therefore is implicated in the pathophysiology of the low-renin, volume-expanded type of hypertension. However, so far, only two steroid Na(+)-K+ pump inhibitors (namely, a bufodienolide derivative [resibufogenin], obtained from toad skin and plasma and a factor with the same carbon, oxygen, and hydrogen content as ouabain obtained from the plasma of volume-expanded humans) have been purified and structurally characterized. To determine whether such endogenous Na(+)-K+ pump inhibitors can in fact produce the above effects on the cardiovascular and renal systems, we infused commercially available bufalin (aglycone, identical to resibufogenin except for one H+), ouabain, and ouabagenin (aglycone) at equimolar doses in normotensive rats. Relative to ouabain, bufalin produced significantly greater dose-dependent increases in blood pressure, left ventricular rate of pressure change, heart rate, and excretion of urinary volume and sodium. Ouabagenin was without effect on any of these parameters. These data indicate that a Na(+)-K+ pump inhibitor can cause an increase in blood pressure despite potent diuretic and natriuretic effects and that, in rats, bufalin is much more potent in this respect than ouabain or ouabagenin.

Behaviour disorder in pre-school children in Hong Kong. A two-stage epidemiological study.

A representative sample of 855 Hong-Kong Chinese children aged 36-48 months were assessed using the BSQ and the PBCL. Good reliability for both instruments were found. For the BSQ and PBCL, 12.75% and 27.5% were above the cut-off points of 10+ and 12+ respectively and 5.9% were above both cut-off points. In the second stage, 234 subjects were recruited by stratified random sampling according to the results of the screening state. A clinician interviewed the parent, child and teacher before making a diagnosis. The prevalence of behaviour disorder was: nil, 53.7%; dubious, 23.1%; mild, 18.0%; moderate, 4.5%; and severe, 0.7%. There were significantly more boys in the categories mild, moderate and severe.

[Effect of captopril on hypoxia-induced pulmonary hypertension in pigs].

The role of angiotensin system in the development of hypoxic pulmonary hypertension, in nine pigs (50 +/- 8 kg) were studied. A Swan-Ganz Catheter and an arterial catheter were inserted into the pulmonary artery and aorta, pulmonary arterial pressure (PAP), pulmonary capillary wedge pressure (PCWP), Cardiac output (CO) and arterial blood gases were monitored before and after hypoxia and captopril injection (7.3 mg/kg.iv). Plasma renin activity (PRA) and angiotensin II (AT-II) were measured by RIA. Angiotensin converting enzyme (ACE) was measured by fluorometry. Results showed that during hypoxemia (PaO2 6.3 +/- 0.2 kPa, PaCO2 5.4 +/- 0.2 kPa), PAP increased from 2.43 +/- 0.17 to 3.76 +/- 0.2 kPa, (P less than 0.05) and right ventricle stroke work index (BVSWI) from 55.7 +/- 7.2 to 91.3 +/- 9.3 mJ/m2 (P less than 0.05). PRA increased from 0.56 +/- 0.19 to 1.28 +/- 6.30 mol. L-1/h (P less than 0.02) and AT-II from 62.4 +/- 17.4 to 133.3 +/- 31.8 ng/L (P less than 0.01); but ACE decreased from 77.6 +/- 5.6 to 58.4 +/- 4.2 mumol.min-1/L (P less than 0.02). After Captopril injection ACE was reduced to 26.7 +/- 3.4 mumol.min-1/L (P less than 0.001) and AT-II dropped to 61.9 +/- 15.5 ng/L (P less than 0.01), as compared with those during hypoxemia. There was significant correlation between PAP and PRA (r = 0.5643 P less than 0.01). We surmise that angiotensin system may play a part in acute hypoxia-induced pulmonary hypertension, and captopril Inhibits AT-II, leading to the drop of pulmonary arterial pressure.

[Molecular study of Ph1 chromosome negative chronic myelocytic leukemia].

Six cases of Ph1 chromosome negative chronic myelocytic leukemia (CML) were studied. In five cases, the BCR gene configuration was analyzed with a probe covering the major breakpoint cluster region (M-bcr). No M-bcr rearrangement was detected in two cases. On the contrary, the BCR gene was found rearranged in three other patients. The breakpoints of the chromosome 22 were localized within the M-bcr in two cases and in a region upstream the M-bcr in the remaining case. These data suggest that Ph1 negative CML can be divided into two groups according to the BCR gene stattus. The clinical characteristics and pathogenesis of these two groups may be different.

[Efficiency evaluation of combined intervention measures with improving drinking water first to prevent infantile acute diarrhea].

The paper analysed factors of acute diarrhea in children under five by Mantel-Haenszel stratification and Logistic regression. The total person-years of observation were 1332. The results showed that the combined risk factors which caused diarrhea were drinking the pond water and failure to wash hands before eating and so on. The combined intervention Measures, especially improvement of drinking water had effectively decreased the incidence rate of infantile acute diarrhea through the observation in the past two years.

[Determination and comparison of beta-glucuronidase activity among strains of B. fragilis and E. coli].

We determined and compared the beta-glucuronidase (beta-G) activity among strains of B. fragilis and E. coli under the optimum condition. Results showed that the mean beta-G activity of strain B. fragilis ATCC25285 was 94.7u. B. fragilis ATCC25285 strain was selected as the model strain to establish the animal model of bilirubin gallstones because its beta-G activity was obviously higher than that of B. fragilis CDC14462 and E. coli 3362 (O157K88) strain.

The role of oxygen free radicals in myocardial ischemia/reperfusion injury.

A series of experiments have been done to investigate the role of oxygen free radicals in ischemia/reperfusion injury. The following results were found: Myocardial MDA content increased significantly after post-ischemic reperfusion in vivo and in vitro. A blockade of the xanthine oxidase pathway for free radical generation could provide effective protection against ischemia/reperfusion injury. Exogenous reactive oxygen intermediates H2O2, .OH and O2- could induce changes in the contractility and electrophysiological properties of myocardial cells similar to those seen in ischemia/reperfusion. An outburst of free radical generation was detected by ESR spectroscopy at low temperature (-173 degrees C) and with the spin trapping technique during the very early phase of reperfusion. The authors emphasize the important role of free radicals in the pathogenesis of myocardial ischemia/reperfusion injury.

[Improvement of memory in mice by extracts from leaves of Ginkgo biloba L].

The study has shown that the extracts from leaves of Ginkgo biloba can significantly improve the NaNO2 and scopolamine induced impaired memory in mice. The potency of the ethanolic extract is greater than that of the aqueous extract. The ethanolic extract acts favorably on the memory function of normal animals. Both extracts help to prolong the survival time of mice receiving 200 mg/kg(ip)NaNO2.

Five cases of neurocysticercosis diagnosed in Sydney.

Cysticercosis, once rare in Australia, is now more frequently diagnosed. This change reflects the countries of origin of new immigrants and the destinations of Australians travelling. Five cases of neurocysticercosis diagnosed at Westmead Hospital in Sydney are described. Two involved Australians, a father and son who had visited eastern and southeastern Asia 10 years before presentation. The other three included immigrants from Chile and India and a visitor from Timor. Ages ranged from 5 to 57 years. Three individuals presented after focal seizures involving the upper limb, one had a long standing history of neurological dysfunction and one suffered from persistent headaches. In all cases computed tomographic scanning (CT) or magnetic resonance imaging (MRI) revealed cystic brain lesions and three of the five were seropositive as well. Four were treated with praziquantel and in one the lesions regressed significantly following treatment. However, the lesion in one case had decreased in size prior to treatment and that in the untreated individual also became smaller.

High expression of era gene.

For high production of Era protein, lambda Ea8.5 gene has been chosen from Gene Bank because the Ea8.5 protein contains a sequence of several amino acids at its N-terminus which is identical to that of Era and also because it can be highly expressed in E. coli. The 5'-end sequence of era gene was substituted by a synthetic oligonucleotide which was identical to that of the 5'-end of Ea8.5 gene, so that the transcripts of era gene was provided with a strong translational initiation signal without changing the amino acid sequence of its translational product. Plasmid pCE31, which harbored the recombined era gene under the control of PL promoter, could express very high levels of Era protein, while synthesis of other cellular proteins was nearly shut off during the period of induction. As a result, Era constituted over 80% of the total cellular protein. Electrophoretic pure Era protein with specific guanine nucleotide-binding activity was obtained by a simple procedure including lyzing the cells and washing the pellet of the lysate.

Role of amygdaloid nucleus in the correlation between the heart and the acupoint neiguan in rabbits.

Amygdaloid nucleus (AMYG) could converge and integrate the signals from electroacupuncture (EA) at Neiguan (P 6) and acute myocardial ischemia (AMI) in rabbits. And EA could regulate the changes in the discharge of single units in AMYG caused by AMI, that is, EA inhibited the increased frequency, and reversed the decreased frequency. This suggested that AMYG is an important link in correlation between the heart and the acupoint Neiguan, providing an experimental basis for the Theory of Channel--Viscera Correlation.
