Role of parathyroid hormone-related protein in the proliferation of vascular smooth muscle cells.

Human parathyroid hormone-related protein(1-34) [hPTHrP(1-34)] inhibited [3H]thymidine incorporation into DNA of cultured vascular smooth muscle cells (VSMC) and increased intracellular cAMP accumulation, both in a concentration-dependent manner. The potency of hPTHrP(1-34) was similar to that of human parathyroid hormone(1-34) [hPTH(1-34)], but their antagonists, hPTHrP(7-34) and hPTH(3-34), showed no effect on VSMC DNA synthesis. The inhibitory effect of PTHrP on VSMC DNA synthesis was mimicked by dibutyryl-cAMP but not by dibutyryl-cGMP. These results suggest that PTHrP, as an autocrine and/or paracrine regulator, may inhibit VSMC DNA synthesis mediated at least partly by cAMP, a second messenger which has also been proposed to mediate vascular relaxation.

[Expression of endothelin-1 mRNA, endothelin receptor-A and nitric oxide synthase mRNA in pulmonary artery and right ventriculus cordis of rats exposed to hypoxia].

Endothelins (ETs) are a family of novel regulatory peptides which can constrict the vascular and promote the proliferation of vascular smooth muscle cells. Serum ET-1 was elevated, and nitric oxide (NO) levels were reduced in hypoxic pulmonary hypertension. However, the exact mechanism remains unclear, and no study has elucidated if hypoxia could stimulate directly overgrowth of right ventricle in patients with chronic cor pulmonale. To investigate the role of ET-1 and NO in hypoxia-induced pulmonary hypertension and right ventricular hypertrophy, we measured the levels of ET-1 mRNA, ET receptor-A (ETR-A) mRNA and nitric oxide synthase (NOS) mRNA in the pulmonary artery and right ventricle of rats exposed to hypoxia (FiO = 0.1, 8 hours daily for 1, 2 and 3 weeks) by reverse transcription-polymerase chain reaction (RT-PCR). ET-1 mRNA level of pulmonary artery raised after 1 week's hypoxia (P < 0.05), and after 2 weeks' hypoxia, it returned to near normal, but elevated significantly again after 3 weeks' hypoxia. Pulmonary artery ETR-A mRNA in 1 and 2 weeks hypoxic groups showed no significant change, but it raised significantly in 3 weeks' hypoxic group. After exposure to hypoxia for 1, 2 and 3 weeks, NOS mRNA in the pulmonary artery all reduced significantly (P < 0.05). The right ventriculus cordis showed a significant increase in weight after 3 and 2 weeks' hypoxia. ET-1 mRNA showed no significant change but ETR-A mRNA increased significantly after 2 weeks' hypoxia; both ET-1 mRNA and ETR-A mRNA showed significant increase in weight after 3 and 2 weeks' hypoxia. In the ventriculus cordis of rats exposed to hypoxia for 2 and 3 weeks, NOS mRNA had no significant change.

[Morphometric study on the cerebral cortex of partial status epilepticus rats induced by coriaria lactone].

Twenty adult male Wistar rats, divided into two groups were studied. Experimental rats were embedded with a particle containing Coriaria Lactone into the fore limb's representative area of cerebral motor cortex to induce epilepsy. Control rats were embedded with a particle containing distilled water. After 6 h motor cortex was cut and the structure was compared morphologically with control rats. Some pale neurons, dark shrunken neurons and severe sponginess of neuropil were observed in layers I-V of the epileptic rat under LM. Under EM, the mitochondria became swollen and other organelles migrated and close associated with the nucleus in pale neurons, and in dark neurons both cytoplasm and karyoplasm showed highly electron dense. The spongiform neuropil was mainly occupied by swollen glial processes. The number of neuron and neuroglia were counted from photos under LM. The number of synaptic terminal was counted directly and the areal fraction of the constituents in the neuropil was measured stereologically in the EM photos. The number of neuron and neueroglia decreased significantly. Both the number of synaptic terminal in neuropil and the areal fraction of synaptic terminal decreased significantly. These changes were most prominent in the focus area of epileptic rats and in the parafocus area, too. This may indicate that the decrease of areal fraction of synaptic terminal results from the decrease of the number of synaptic terminal.(ABSTRACT TRUNCATED AT 250 WORDS)

Catalytic properties of NAD(P)H:quinone acceptor oxidoreductase: study involving mouse, rat, human, and mouse-rat chimeric enzymes.

NAD(P):quinone acceptor oxidoreductase (quinone reductase) (DT-diaphorase, EC 1.6.99.2) is involved in the process of reductive activation of cytotoxic antitumor quinones and nitrobenzenes. In this study, we initially examined the relative abilities of mouse, rat, and human quinone reductases to reduce two prodrugs, CB 1954 [5-(aziridin-1-yl)-2,4-dinitrobenzamide] and EO9 [5-(1-aziridinyl)-3-(hydroxymethyl)-2-(3-hydroxy-1-propenyl)-1- methyl-1H-indole-4,7-dione]. By using Escherichia coli-expressed quinone reductases and evaluating them under identical conditions, we confirmed previous finding showing that the human enzyme is not as effective as the rat enzyme in reducing CB 1954 and EO9, although the two enzymes have similar NAD(P)H-menadione reductase activities. Interestingly, although the amino acid sequence of mouse quinone reductase is more homologous to that of the rat enzyme, we found that the mouse enzyme behaves similarly to the human enzyme in its ability to reduce these compounds and to generate drug-induced DNA damage. To determine the region of quinone reductase that is responsible for the catalytic differences, two mouse-rat chimeric enzymes were generated. MR-P, a chimeric enzyme that has mouse amino-terminal and rat carboxy-terminal segments of quinone reductase, was shown to have catalytic properties resembling those of rat quinone reductase, and RM-P, a chimeric enzyme that has rat amino-terminal and mouse carboxyl-terminal segments of quinone reductase, was shown to have catalytic properties resembling those of mouse quinone reductase. In addition, MR-P and RM-P were found to be inhibited by flavones with Ki values similar to those for rat and mouse quinone reductases, respectively. Based on these results, we propose that the carboxyl-terminal portion of the enzyme plays an important role in the reduction of cytotoxic drugs and the binding of flavones.

V-J junctional sequences of T cell receptor gamma gene in acute lymphocytic leukemia.

T cell receptor TCR gamma gene rearrangement in a series of acute lymphocytic leukemia (ALL) patients was studied using PCR technique. 18 V-J junctional sequences (designated by N sequence) of TCR gamma gene were amplified with the V gamma and J gamma primers, using an unsymmetrical PCR and analysed by direct sequencing, demonstrating that in Chinese ALLs, the N sequences of TCR gamma gene are indeed clone-specific. Based on the known N sequences, several oligo-nucleotides were synthesized as probes, which were specific for the leukemic clone, and were used to detect the minimal residual disease (MRD) in 4 ALL cases. The sensitivity of this method was 0.1%-0.01%.

Age-related changes of male mouse submandibular gland: a morphometric and radioautographic study.

The DNA synthesis and the changes of cell population kinetic of aging male mouse submandibular glands were studied by light and electron microscopic radioautography from embryonic day 19 to 21 months postnatal. At the embryonic day 19, the gland consisted of developing acinar cells (49%), intercalated duct (ICD) (37%), juxta-acinar (JA) (3%) and striated duct (SD) cells (11%). At birth, JA cells increased rapidly to 32%, thereafter decreased gradually. At 1 month of age, granular convoluted tubule cells appeared and increased rapidly in number with age. They reached a maximum at 6 month after birth. Then they appeared to decrease gradually from 6 to 21 months. The most conspicuous feature of the aged submandibular glands was the regressive changes of the granular ducts. The extent and height of the granular ducts as well as the content of mature secretory granules decreased. These changes may play an important role during the development of mice, because the granular duct synthesizes many biological factors. The 3H-thymidine labeling index of each cell type of male mouse submandibular glands reached a peak 1 day after birth and decreased with aging. The labeling index of intercalated duct cells was higher than the others and persisted for a long time. On the other hand, the labeling indices of the other cells were significantly depressed at one week after birth. From these observations on morphology, it was also shown that ICD cells were concerned with the generation of the other cell types.

[Endodermal sinus tumor of the testis in children: report of 9 cases].

Nine children with endodermal sinus tumor of testis have been treated since 1982. Their age ranged from 7 months to 5 years. Radical inguinal orchiectomy was carried out in all the patients. Four patients have received irregular chemotherapy. The follow-up showed that 4 died of the metastasis within 5 years and 5 (stage I clinically) survived without evidence of the recurrence for 1.5-7 years after operation. Serum AFP determination may give important information for the classification of the tumor and the evaluation of the follow-up results. For the cases of clinical stage I, radical orchiectomy, short course of chemotherapy, and frequent observations of AFP level are advocated.

Identification and classification of Neisseria gonorrhoeae by RAPD fingerprinting.

Using the RAPD method, we identified and classified Neisseria gonorrhoeae on the basis of the DNA level. Except OPG20 the other 19 primers of OPG arbitrary primer kit showed informative arrays of amplified prominent segments. Most of the segments were common to all three Neisseria gonorrhoeae groups; some segments were amplified from two groups or only one group so that the fingerprint maps of different Neisseria gonorrhoeae groups were distinctive. Based on these findings, i.e. on the genomic level, we can classify the three Neisseria gonorrhoeae groups. Using OPG10 and OPG11 to amplify the five Neisseria species, we found marked diversity among them, which made it easy for us to identify Neisseria gonorrhoeae from four other Neisseria species. The use of the RAPD method needs some special reaction conditions, e.g. annealing temperature that should not be too low or too high and should be consistently stable. The quality of Taq polymerase is also very important. The reproducibility of the results under these conditions was very good.

The potassium channel subunit KV3.1b is localized to somatic and axonal membranes of specific populations of CNS neurons.

Potassium channels play major roles in the regulation of many aspects of neuronal excitability. These channels are particularly well suited for such multiplicity of roles since there is a large diversity of channel types. This diversity contributes to the ability of specific neurons (and possibly different regions of the same neuron) to respond uniquely to a given input. Neuronal integration depends on the local response of spatially segregated inputs to the cell and the communication of these integration centers with the axon. Therefore, the functional implications of a given set of K+ channels varies depending on their precise location on the neuronal surface. Site-specific antibodies were utilized to characterize the distribution of KV3.1b, a subunit of voltage-gated K+ channels in CNS neurons. KV3.1b subunits are expressed in specific neuronal populations of the rat brain, such as cerebellar granule cells, projecting neurons of deep cerebellar nuclei, the substantia nigra pars-reticulata, the globus pallidus, and the ventral thalamus (reticular thalamic nucleus, ventral lateral geniculate and zona incerta). The KV3.1b protein is also present in various neuronal populations involved in the processing of auditory signals, including the inferior colliculus, the nuclei of the lateral lemniscus, the superior olive, and some parts of the cochlear nuclei; as well as in several other neuronal groups in the brainstem (e.g., in the oculomotor nucleus, the pontine nuclei, the reticulotegmental nucleus of the pons, trigeminal and vestibular nuclei, and the reticular formation) and subsets of neurons in the neocortex, the hippocampus and the caudate-putamen shown by double staining to correspond to neurons containing parvalbumin. KV3.1b subunits are localized predominantly in somatic and axonal membranes (particularly in axonal terminal fields) but are much less prominent in dendritic arborizations. This distribution is different than that of other subunits of voltage gated K+ channels and is consistent with a role in the modulation of action potentials. KV3.1b proteins have a cellular and subcellular distribution different than the related KV3.2 subunits which express in Xenopus oocytes currents similar to those expressed by KV3.1b.

In situ hybridization analysis of hepatic cytochrome P450 2E1 messenger ribonucleic acid in mice. Modulation of expression by acetone.

BACKGROUND: Cytochrome P450 2E1 is a distinct enzyme induced by diverse agents and notably by solvents including acetone. The P450 2E1 apoprotein is predominantly localized in the centrilobular region of the hepatic lobule in the constitutive state and after induction with acetone. In this study, we investigated the level and pattern of distribution of the 2E1 mRNA within the hepatic lobule in control mice and in mice exposed to acetone under acute and chronic conditions. EXPERIMENTAL DESIGN: Male CD-1 mice were administered acetone acutely (5 ml/kg) or chronically (1% in drinking water for 8 days). Cytochrome P450 2E1 mRNA was detected in liver tissues, using in situ hybridization with a 3H-labeled RNA probe coupled with quantitative image analysis. RESULTS: Acetone exposure did not alter the distance spanning a central vein and an adjacent portal vein. In control mice, 2E1 mRNA transcripts were localized most abundantly in the centrilobular zone of the hepatic lobule; intermediate levels were found in the midzonal region, and minimal levels were found in the periportal region. In acetone-treated mice, 2E1 mRNA expression within the hepatic lobule was significantly reduced and was manifested, firstly, in a decreased hepatocyte population with high expression, as assessed by reduction of total acinar areas containing 2E1 mRNA transcripts, and, secondly, in reduced 2E1 mRNA content within the hepatocytes. These alterations were observed to a significantly greater extent in mice after chronic versus acute acetone exposure. CONCLUSIONS: Our results showed that acetone exposure in mice results in both a reduction and a more restricted distribution of 2E1 mRNA within the hepatic lobule. Furthermore, these findings suggest that 2E1 enzyme induction is associated with post-transcriptional events that are more marked after chronic than after acute exposure.

Interaction of flavones and their bromoacetyl derivatives with NAD(P)H:quinone acceptor oxidoreductase.

Flavones are a new type of inhibitor of NAD(P)H:quinone acceptor oxidoreductase (DT-diaphorase, EC 1.6.99.2). To further characterize the flavone binding site, three bromoacetyl derivatives of flavones, i.e., 7-bromoacetylflavone, 5-hydroxyl-7-bromoacetylflavone, and 7,8-dibromoacetylflavone, have been synthesized. These compounds have been found to be potent inhibitors that inactivate the rat quinone reductase in a time-dependent manner, suggesting that they can be used as affinity labels for the enzyme. Among the three bromoacetyl derivatives, 7,8-dibromoacetylflavone is the most potent inhibitor; however, its labeling of the quinone reductase is the least stable, so that the enzyme regains activity after a short incubation. In contrast, the inactivation of the quinone reductase by 5-hydroxyl-7-bromoacetylflavone is stable. Accordingly, this flavone derivative is the most suitable compound for labeling the flavone binding site of the enzyme. Electrospray mass spectrometry has been applied to demonstrate that 5-hydroxyl-7-bromoacetylflavone labels this enzyme in a stoichiometric manner.

[National notification of incidence of chronic occupational poisoning during 1984-1993].

Incidence and distribution of chronic occupational poisoning caused by toxic chemicals during 1984 to 1993 throughout the country were summarized and analyzed. Totally, 28,901 cases of chronic occupational poisoning were notified during this period. Lead, benzene and trinitrotoluene were three major toxic chemicals which caused poisoning, with chronic lead poisoning listed the first place, accounting for 47.5% of the total cases. Most of the cases distributed in Laioning, Hunan and Jiangsu Provinces, and 75.4% of the cases occurred in machinery, chemical, metallurgical, non-ferrous, light industries, etc. Incidence of poisoning presented a decreased trend during the past 10 years and the reason of it was described in the paper. The authors indicated that quality assurance in notification of occupational diseases depended on regular implementation of the work in prevention and treatment of occupational diseases.
