[Synthesis and antiinflammatory activity of 2-(E)-benzylidene-5-(N-substituted aminomethyl) cyclopentanones].

Nineteen kinds of 2-(E)-benzylidene-5-(N-substituted aminomethyl) cyclopentanones were synthesized via Mannich reaction or amine exchange reaction and identified spectrometrically. One compound exhibited significant antiinflammatory activity, showing obvious inhibitory effect on xylene-induced mice ear swelling, carrageenin-induced rats paw edema and increased capillary permeability induced with acetic acid in mice. Its ED50 values in these inflammatory models were calculated to be 67.8 mg.kg-1, 25.3 mg.kg-1 and 41.8 mg.kg-1 respectively, nearly equal to those of ibuprofen and aspirin.

[Researches on immunogenicity identification of recombinant Sj22.6(rSj22.6) kDa antigen gene of schistosoma japonicum].

AIM: To identify the immunogenicity and the potentiality of rSj22.6 as a candidate vaccine antigen for schistosomiasis japonica. METHODS: rSj22.6 was tested by Western blot with specific antibodies to identify its immunogenicity. The rSj22.6 protein was isolated and purified by SDS-PAGE and injected twice into rabbits to raise antirSj22.6 serum that was tested by Western blot to recognize the native protein of adult S. japonicum and SEA. C57 mice were immunized with rSj22.6 and challenged with S. japonicum cercariae to identify the protective immunity raised by rSj22.6. RESULTS: rSj22.6 could be recognized by specific antibodies and could stimulate the rabbits to produce high level of anti-rSj22.6 antibodies. Anti-rSj22.6 serum could recognize an adult native protein with the same molecular weight as rSj22.6. The preliminary immune-challenge test resulted in significant reduction in adult worm burden. CONCLUSION: rSj22.6 possesses immunogenicity in stimulating rabbits to produce high level of specific antibody and has protective role as a candidate vaccine antigen against S. japonicum infection.

[Researches on the nucleotide sequencing of the recombinant Sj 22.6 kDa antigen gene of Schistosoma japonicum].

AIM: To sequence the cloned gene in pGSj24 and to identify the encoded protein. METHODS: The cloned gene in pGSj24 was digested from the recombinant plasmid by EcoRI, ligated into the M13mp19 vector and sequenced by automatic sequencer. The sequence was analyzed by Goldkey DNA and Protein Analytical Program and DNASIS Program. RESULTS: The pGSj24 cloned gene was demonstrated to be 840 bp containing one opened reading frame (ORF) with an initiation codon ATG at position 23 nt and a termination codon TAA at position 596 nt, encoding a protein with a molecular weight of 22.6 kDa. At the upstream and downstream of the ORF there were termination codons, so the encoded protein was unable to be larger. However, there was a termination codon TAA at position 11 nt, suggesting why the 22.6 kDa protein expressed separately. The nucleotide sequence of the pGSj24 cloned gene shared 95% identity with that of the corresponding part of S. japonicum 22.6 kDa antigen gene, and 99.7% identity in the encoding part. The deduced amino acid sequence analysis showed a sequence motif known as EF-Hand calcium binding domain, several endoplasmic reticulum targeting sequences and microbodies C-terminal targeting signals. The possible antigen determinants were predicted within the amino acid fragments of 29-32aa, 63-68aa and 87-101aa. CONCLUSIONS: The cloned gene in pGSj24 is the gene that encodes Sj22.6 kDa antigen.

[Screening and characterization of Schistosoma japonicum reinfection-related cDNA clones].

AIM: To obtain cDNA clones coding for S. japonicum (Sj) reinfection-related specific immunologic molecules for vaccination or diagnosis. METHODS: An adult S. japonicum cDNA library, of which the cloning efficiency is 3.13 x 10(6), with directional inserts in the vector lambda gt11, was screened with pooled serum from 43 reinfected human cases in a schistosomiasis-endemic area with high level of Sj specific-lgG4 antibodies. Primers located several bases away from the insert position on the lambda gt11 arm were used to amplify the insert fragments by standard PCR. SILVER SEQUENCE (PCR-silver) DNA sequencing system was then used to sequence selected positive clones with relatively large inserts and homology comparison with GenBank database was also carried out. RESULTS AND CONCLUSION: Eleven phage plaques were defined as positive out of the 8.6 x 10(4) phage plaques immunoscreened by human specific lgG4 antibody. GenBank database retrieval showed that 4 new Sj gene clones including Sj reinfection-related specific protein-encoding gene and Sj mitochondrial protein-coding gene have been obtained.

[Segmentation of osteomas X-ray image using mathematical morphology].

In this paper, we introduce a method for the segmentation of osteomas X-ray images. This method is mainly based on mathematical morphology. Structuring elements are selected according to the shape of osteomas. After gray scale morphology operations, osteomas boundary is extracted by histogram threshold. Further processing of resulting boundary with binary morphological operation produces a closed osteomas contour. So osteomas is isolated from background.

[Primary studies on the establishment of cytomegalovirus murine model].

Intraperitoneal inoculation of Murine cytomegalovirus(MCMV) into Balb/C mice at age of 4 weeks resulted in acute MCMV infection of the mice. The infected mice had body weight reduction, growth retardation, salivary gland swelling and death. A lethal dose was 5 x 10(6) PFU/ml of MCMV. Virus could be recovered from salivary gland and the infectivity titer of MCMV reached to a high titer of 2 x 10(5) PFU/ml. Plaque formation of MCMV in 3T3/Swiss albino cells was identified. Histopathological examination showed the appearance of MCMV-specific intracytoplasmic inclusion bodies in neuron cells of the mouse brain.

[Studies on fermentation conditions of cytokinin produced by rhizobia strain 4012a].

The conditions of cytokinin fermentation of the rhizobia strain 4012a were detected by the ELISA. The results indicated that the optimal medium for cytokinin production by strain 4012a was composed of glucose 10 g/L, (NH4)2SO4 1.0, K2HPO4.3H2O 0.6, MgSO4.7H2O 0.1, CaCl2.2H2O 0.4, FeCl3.6H2O 0.04, Na2MoO4.2H2O 0.1 mg/L, calcium pantothenate 100 micrograms/L, adenine 200 mg/L. When strain 4012a was grown in 250 ml flask containing 50 ml of the medium on the rotary shaker (150 r/min) at 27 degrees C for 96 h, the yield of CTK 908 micrograms/L culture solution was obtained. It displayed bioactivity kinetin equivalents (KE) 1 mg/L medium with the radish cotyledon expansion test.

[Hyaluronic acid and laminin in patients with ischemic necrosis of femoral head].

To study the course and clinical significance of hyaluronic acid (HA) and laminin(LN), the main components of extracellular matrix in ischemic necrosis of femoral head(INFH), the quantities of HA and LN were measured by radioimmunoassay in 45 patients suffering from INFH with different pathogenesis, and in 30 normal subjects(NS). The results showed that the level of HA in patients with INFH was markedly higher than that of NS group(t = 3.29, P < 0.01). The patients with hormone type INFH had the highest level of HA(t = 3.62, P < 0.01). The level of LN in patient with INFH was significantly higher than that of NS group(t = 2.84, P < 0.01). The increased levels of HA and LN were positively correlated with the development of the disease. These suggest that quantity analysis of HA and LN may be used as a good indicator in early diagnosis and prognosis of INFH.

[Determination of Y, Ce, Ba and Cu in Y(1-x) Ce(x),Ba(2)Cu(3)O(y), using x-ray fluorescence fundamental parameter method].

The determination of Y, Ce, Ba and Cu in a series of Y(1-x)Ce(x)Ba(2)Cu(3)O(y) samples has been performed using X-ray fluorescence fundamental parameter method. The composition is given in atomic percentage. The results agree well with those obtained by ICP. It is shown that this method is appropriate for the determination of compositions of the high temperature superconductors. In addition, the influence of oxygen on analytical elements has been discussed in this paper. The fraction of the matrix total mass absorption that is associated with oxygen for each analytical line can not be larger than 3.44%. The determined composition of the samples show different from their nominal composition, which may affect the properties of high temperature superconductor.

[Studies on complex chromosomal translocation and their relevance to clinical prognosis in acute promyelocytic leukemia].

OBJECTIVE: To study the relationship between the complex chromosome translocation in acute promyelocytic leukemia(APL, M3) and clinical therapy and prognosis. METHODS: Chromosome translocation and PML-RAR fusion transcript in three APL patients were studied by using karyotypic analysis, fluorescence in situ hybridization and reverse transcriptase/polymerase chain reaction. RESULTS: The findings revealed that all these cases had PML/RAR A gene rearrangement. Apart from the chromosomes 15 and 17 involved in the translocation, other multiple chromosomes including 5, 11, 16, 22 were also implicated in complex translocations, which to some extent seemed to be related with clinical prognosis. CONCLUSION: This study provides additional information for monitoring clinical therapy and prognostic evaluation.

Bone marrow repopulation by human marrow stem cells after long-term expansion culture on a porcine endothelial cell line.

In vitro exposure of murine hematopoietic stem cells (HSCs) to cell cycle-inducing cytokines has been shown to result in a defect in the ability of these cells to engraft. We used a porcine microvascular endothelial cell (PMVEC) line in conjunction with exogenous interleukin (IL)-3, IL-6, granulocyte-macrophage colony-stimulating factor (GM-CSF), and stem cell factor (SCF) to expand human HSCs that express the CD34 and Thy-1 antigens but lack lineage-associated markers (CD34+Thy-1+Lin- cells). Ex vivo expansion of hematopoietic cells was evaluated in comparison to stromal cell-free, cytokine-supplemented cultures. Cells expressing the CD34+Thy-1+Lin- phenotype were detectable in both culture systems for up to 3 weeks. These cells were reisolated from the cultures and their ability to engraft human fetal bones implanted into SCID mice (SCID-hu bone) was tested. HSCs expanded in PMVEC coculture were consistently capable of competitive marrow repopulation with multilineage (CD19+ B lymphoid, CD33+ myeloid, and CD34+ cells) progeny present 8 weeks postengraftment. In contrast, grafts composed of cells expanded in stroma-free cultures did not lead to multilineage SCID-hu bone repopulation. Proliferation analysis revealed that by 1 week of culture more than 80% of the cells in the PMVEC cocultures expressing the primitive CD34+CD38- phenotype had undergone cell division. Fewer than 1% of the cells that proliferated in the absence of stromal cells remained CD34+CD38-. These data suggest that the proliferation of HSCs in the presence of IL-3, IL-6, GM-CSF, and SCF without stromal cell support may result in impairment of engraftment capacity, which may be overcome by coculture with PMVECs.

A more flexible regression-to-the-mean model with possible stratification.

We consider a regression-to-the-mean model that includes both additive and multiplicative treatment effects. We allow either or both of these treatment effects to be stratified by ranges of the first measurement. We focus on the situation where there is a very large sample on the first measurement and a relatively small subsample for the second measurement is selected, which often occurs in screening trials. We propose some asymptotically efficient estimators for the parameters of the model that are very simple to compute. We begin with a discussion of the full model, and more on tests and estimation for reduced models follows. An example from a large screening trial is discussed.

Enhanced aggregability of human red blood cells by diving.

In vitro studies have shown that mild pressure increases red blood cell (RBC) aggregation. Enhanced RBC aggregation in pathologic states can drive the circulation into stasis. This investigation examined the effects of pressure on RBC aggregation in human volunteers. The hypothesis tested is that RBC aggregation is increased during hyperbaric exposure. Eleven subjects participated in dives to 300 feet of seawater (fsw) in a man-rated chamber complex. Blood samples were taken at the surface, at 66 fsw, and at 300 fsw. Data were analyzed with a repeated measures one-way analysis of variance for a complete randomized design. Statistical significance was achieved at P < 0.05. Data are expressed as mean +/- SEM. The median aggregate size (number of RBC/aggregate) of RBCs was significantly increased at depth. At a shear stress of 0.1 dyne/cm2, median aggregate size was 12.0 +/- 2.1, 33.0 +/- 7.3, and 48.8 +/- 10.8 at the surface, at 66 fsw, and at 300 fsw, respectively. These results show that mild pressure increases RBC aggregation in the human circulation.
