Production of human recombinant bone morphogenetic protein-2A by high density culture of Escherichia coli with stationary dissolved oxygen fed-batch condition.

The optimization of cultivation condition in 500 ml shake flasks was carried out to produce recombinant human bone morphogenetic protein-2A (BMP-2A) in recombinant Escherichia coli YK537/pDH-B2m, followed by a 5L fermenter batch and condition-controlled fed-batch culture to obtain BMP-2A. The comparison of these two methods indicated that cultivation by keeping dissolved oxygen at 30%-40% and limiting glucose concentration could obtain BMP-2A 2.78 g/L broth, the final cell density was OD600 53 (dry cell weight 21.2 g/L), and expressed BMP-2A was 25% of the total amount of protein in E.coli. The critical fermentation conditions included: (1) keeping appropriate dissolved oxygen concentration in the process; (2) limiting glucose concentration; (3) taking heat induction at the middle-log phase and maintaining 42 degrees C for 4 hours; (4) controlling specific growth rate around 0.3 h-1 in the duration of growth.

[Detection of human cytomegalovirus by PCR in patients with gastrointestinal disease].

Human cytomegalovirus (HCMV) genomes was detected by PCR in 51 patients' biopsy or operation specimens with gastrointestinal disease. The positive rate was 9.80%. At the same time, 41 patients' serum HCMV-IgM was detected by indirect immunofluorescence assay (IFA); the positive rate was 46.34%. Patients with cancer and colitis exhibited a higher HCMV infection rate than those without cancer and colitis. The results suggest that PCR is sensitive and specific in detection of gastrointestinal HCMV infection, and the examination of biopsy or surgical specimen is superior than that of IFA of serum HMCV-IgG.

[Protective effects of shenmai cardioplegia on intracardiac operative patients with cardiopulmonary bypass].

Forty intracardiac-operative patients were divided into two groups randomly. Twenty patients were received by the improved St. Thomas Crystal Blood (1:4) Cardioplegia(Group A); 20 by the Shenmai liquid and improved St. Thomas Crystal Blood(1:4) Cardioplegia(Group B). During perioperation, the blood samples and myocardial tissue were collected, and clinical observations were recorded. The results showed that in Group B, contents of myocardial specific isoenzyme of creatine kinase(CK-MB), lactate dehydrogenase(LDH), malonyldialdehyde(MDA), and myocardial Ca2+ were significantly lower, the contents of superoxide dismutase(SOD) were, significantly better than that in Group A; and the histological findings showed less damage in Group B than in Group A. We conclude that Shenmai Cardioplegia can increase the myocardial protection of the intracardiac-operative patients. The possible mechanisms are that Shenmai liquids inhibit the production of oxygen free radicals and calcium paradox.

[The protective effects of cardiac ischemic preconditioning on lung in cardiac operation with cardiopulmonary bypass].

UNLABELLED: Twenty direct vision intracardial operation patients were divided into two groups randomly. After cardiopulmonary bypass, ten patients were treated with myocardial ischemic preconditioning. The aorta were clamped for 3 minutes and released for 3 minutes (Group IP). Another ten patients were not treated with ischemic preconditioning (Group C), only underwent 6 minutes cardiopulmonary bypass. Then the aorta were clamped and intracardial operation were done. The left atrium blood and lung tissue were collected just after thoractomy and half an hour after cardiac reperfusion in both groups. RESULTS: (1) The numbers of polymorphonuclear (PMN) of the two groups were increased significantly after cardiopulmonary bypass (P < 0.01). (2) The number of PMN and SOD, PaO2 contents were significantly higher in Group IP than in Group C (P < 0.05). (3) The numbers of PMN in lung interstitum under microscopy were less in Group IP than in Group C. (4) MDA contents were less in Group IP than in Group C (P < 0.05). (5) Histological finding showed less damage in Group IP than in Group C. It is evident that cardiac ischemic preconditioning could protect lung against ischemia reperfusion injury. The possible mechanisms are that ischemic preconditioning inhibites the accumulation and activation of PMN in lung tissue and reduces the production of oxygen free radicals.

[Study on multidrug resistant gene (MDR1) expression between neoplastic cells and peripheral blood lymphocytes in ovarian carcinoma].

OBJECTIVE: To explore the correlation of MDR1 (P-glycoprotein, P170) gene expression P170 contents among neoplastic tissues, peripheral blood lymphocytes (PBL) and ascites cancerous cells (ACC) during chemotherapy. METHODS: Content of P170 in cancer tissue, ACC and PBL was quantiated dynamicly by using flow cytometric-immunologic method in 48 cases suffered from ovarian carcinoma. RESULTS: Cancer tissue showed positive P170 in 25% of patients. During chemotherapy, P170 content in PBL and ACC were statistically increased. A significant correlation of P170 was found between PBL and ACC (0.01 < P < 0.05). CONCLUSION: The results indicated that multidrug resistance was produced gradually during chemotherapy. Content of P170 in PBL could probably be used as an indirect index of multidrug resistance for recurrent carcinoma or residual tumor.

[Effects of plasma endothelin-1 and aldosterone on sodium retention in children with nephrotic syndrome].

The effects of plasma endothelin-1 (ET-1) and aldosterone (Ald) on sodium retention in children with nephrotic syndrome were investigated. 14 cases were divided into the edematous stage and non-edematous stage. Plasma ET-1, Ald, and serum osmolarity, albumin (Alb), and Na+ were measured in each stage. The results showed that the plasma ET-1 in edemaous stage increased significantly, compared with that in no-edematous stage (P < 0.01). There was no significant difference in plasma Ald between the edematous stage and the non-edematous stage. Plasma ET-1 was positively correlated with the serum sodium ionic concentration and negatively correlated with the serum Alb(r = 0.486, P < 0.01; r = 0.490, P < 0.01, respectively). In conclusion, the sodium retention with nephrotic syndrome might be correlated with reduction of secreted sodium in the kidney, suggesting that ET-1 plays an important role in pathogenesis.

Effects of Huangdan capsule on plasma cGMP and ANP in rats with chronic renal failure.

The model of chronic renal failure (CRF) was made in 5/6 nephrectomized rats and Huangdan capsule was used to treat these rats. The levels of cyclic guanosine monophosphate (cGMP) and atrial natriuretic peptide (ANP) in plasma were examined. The results showed that Huangdan capsule could postpone the increase in the levels of cGMP and ANP, suggesting that by regulating the water and sodium metabolism, Huangdan capsule could ameliorate the glomerular filtration rate, and that Huangdan capsule could lower the levels of cGMP and ANP in plasma via body regulation.

[Surgical treatment of perplexing renal cell carcinoma].

OBJECTIVE: To summarize the experience of surgical treatment of perplexing renal cell carcinoma in order to increase survival rate. METHODS: From 1987 through 1996, 20 patients with perplexing renal cell carcinoma were surgically treated. There were 3 patients in stage I, 9 in stage II, and 8 in stage III. The operative proceduce was described and the follow-up results were analysed. RESULTS: Among 20 patients, 18 were evaluable. The 1-, 3-, 5-year survival rate was 100%, 91.7% and 75% for stage I-II; 87.5%, 62.5% and 37.5% for stage III, respectively. CONCLUSION: Radical nephrectomy is still effective for the patients with perplexing renal cell carcinoma.

[Plasma neuropeptide Y (NPY) levels in patients with gastric and colorectal carcinomas].

OBJECTIVE: To investigate the plasma NPY concentration in patients with gastric and colorectal carcinomas in relation to tumor progression. METHODS: Blood samples were obtained from patients with gastric cancer (n = 18) and colorectal cancer (n = 20). Plasma NPY concentration was determined by radioimmunoassay. RESULTS: The average plasma NPY level in the 38 patients studied (125.3 +/- 31.5 pg/ml) was significantly(P < 0.001) lower than that in 28 healthy control individuals (145.1 +/- 44.1 pg/ml). Decreased NPY levels were correlated with loss (> 3 kg) of body weight (P < 0.01) and tumor size (> 5 cm) (P < 0.05). The stage of gastric cancer, but not that of colorectal cancer, was negatively correlated with plasma NPY level. Plasma NPY level did not correlate with sex, age, blood pressure, depth of tumor invasion, degree of differentiation, lymph node metastasis, hemoglobin and albumin concentrations. CONCLUSION: Plasma NPY level is decreased in patients with gastric and colorectal carcinomas, but it reflects only in part the progression of gastric cancer.

[A novel rotavirus causing large scale of adult diarrhea in Shi Jiazhuang].

An outbreak of adult diarrhea causing more than 1,000 hospitalized cases among graduates in a university in Shi Jiazhuang city (Hebei province), occurred in 10-28, April, 1997. The non-bacterial pathogenic agent is confirmed as Rotavirus first by PAGE. Forteen samples have been demonstrated sharing the same nucleic acid electrophoresis pattern of 4-2-1-1-1-1-1. The positive rate was 47% (thirty samples have been checked) with no other RV pattern. Dada showed that RV with this kind of nucleic acid electrophoresis pattern were responsible for the outbreaks acute diarrhea in adults. The novel RV and known ADRV was RT-PCR with end-primers 5, 9 of typical ADRV, the positive control typical ADRV's results are positive, the novel RV's are negative. Experimental results showed that the novel RV did not belong to group B RV, totally different from all the known ARDV. This case was the first reported one in Hebei province with an outbreak of adult diarrhea not by any known ADRV.

[Polymorphism at the LDL receptor gene locus in patients with cholesterol gallstone disease].

OBJECTIVE: To investigate the association between the dinucleotide repeat polymorphism at the 3' end of the LDL receptor gene and cholesterol gallstone disease. METHODS: Polymorphism of the (dTA)n was amplified by using polymerase chain reaction, and the alleles identification was performed with denaturing polyacrylamide gel electrophoresis and silver stain technique. RESULTS: Analysis of allele frequencies in 131 unrelated Chinese gallstone patients and 79 controls indicated that the C allele was more frequent in the patients with cholesterol gallstones than that in the controls (0.40 vs 0.27, P < 0.01), and the frequency of the BB genotype of the LDL receptor gene was markedly lower (P < 0.005), while the CC and BC genotypes were significantly higher in the patients with cholesterol gallstones than in the controls respectively (P < 0.05, P < 0.05). CONCLUSION: The present study suggests that the LDL receptor gene polymorphism may be associated with cholesterol gallstone disease.

[Myocardioprotective effects of preconditioning in patients undergoing open heart operation].

OBJECTIVE: To investigate the myocardial protective effects of ischemic preconditioning (IP) on adult patients who undergoing prolonged open heart operation for complex heart diseases. METHODS: 30 patients were evenly divided into two groups at random. IP was elicited twice for 2 minutes ischemia from occlusion of vena cava and aorta followed by 3 minutes of reperfusion under cardiopulmonary bypass. Dynamic changes of ATP and purine metabolites in myocardium were measured. Electrocardiogram, leakage of myocardial enzymes, activities of SOD and LPO in serum and myocardial mechanics were investigated. RESULTS: IP reduced ATP repletion and maintained a higher ATP content in ischemic myocardium (P < 0.05 vs control group at 30, 60, and 90 minutes of ischemia). Reduced inosine content was found in IP group (P > 0.05). The inosine/adenosine ratio at 30, 60 and 90 minutes of ischemia was 4.2 vs 14.1, 17.6 vs 28.2, 29.2 vs 71.3 for the IP and control groups, respectively, P < 0.05). There were also significant differences between the two groups in changes of ST-segment shifting, myocardial contractile and early postoperative recovery. CONCLUSIONS: IP can reduce ischemia-reperfusion injury on prolonged ischemic myocardium in humans even when combined with cold cardioplegia. Decreased inosine production and inosine/adenosine ratio in ischemic myocardium may be an important mechanism of IP, by which the production of oxygen free radicals are decreased during early reperfusion.

[Ineffective platelet transfusion in patients with hematology malignancy].

OBJECTIVE: To explore the relationship between the effects of platelet transfusion and histocompatibility antigen (HLA). METHODS: The relationship between the corrected count increment (CCI) of platelet with lymphocytotoxicity test (LCT) and human histocompatibity antigen system-I subtype (HLA-I) in 126 hospitalized patients who accepted platelet transfusion. RESULTS: Ineffective platelet transfusion was strongly related to LCT positive and LCT intensity (P < 0.01). The HLA A2 gene frequency was 0.91. The frequency was much higher than that of health human being (0.54). CONCLUSION: Positive LCT was mainly immune factor for ineffective platelet transfusion. CCI and LCT should be the routine test for platelet patients. It may improve the effects of platelet transfusion.

The purification and identification of heme oxygenase isoforms from spleen tissue of rat and the expression of heme oxygenase-1 cDNA in COS-1 cells.

OBJECTIVE: To demonstrate that heme oxygenase (HO) isoforms exist in rat spleen treated with hematin and phenylhydrazine and to confirm that the isolated cDNA actually encodes HO-1 by expressing cDNA in monkey kidney cells (COS-1 cells) in order to prepare HO-1 mutant for inhibiting the natural enzyme. METHODS: The rat spleen microsomal fractions were first purified by diethylaminoethyl (DEAE)-Sephacel and hydroxylapatite. The activity of two isoforms (HO-1 and HO-2) of enzyme and their apparent molecular weight on sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (SDS-PAGE) were measured. Secondly, using the isolated HO-1 cDNA clone, the expression plasmid pcDNA3HO1 was constructed and transfected into cultured COS-1 cells. The transfected cells were collected and disrupted by sonication, and the microsomes were prepared by ultracentrifugation. The activity of HO-1 was measured. RESULTS: Two isoforms were purified and identified in treated rat spleen and HO-1 was the predominant form. The ratio of HO-1 to HO-2 was 3.2:1. The apparent molecular weights of HO-1 and HO-2 were about 30 kD and 36 kD under reducing conditions, respectively. The HO-1 was highly expressed in endoplasmic reticulum of transfected cells. The specific band was located in molecular weight of 30 kD. The specific activity was five times higher than that of the control. CONCLUSION: The activity of expressed HO-1 in COS-1 cells is higher than that of purified enzyme from rat spleen tissue. It is suggested that this clone having an insert of 1030 base-pairs encodes HO-1 and that we can prepare HO-1 mutant by site-directed mutagenesis of HO-1 cDNA to prevent and treat hyperbilirubinemia.

Ischemic preconditioning enhances donor lung preservation in canine lung transplantation.

OBJECTIVE: To prove the protective effect of lung ischemic preconditioning on enhancing the canine lung preservation and reducing allograft lung dysfunction after transplantation. METHODS: Ten pairs of adult canines underwent left lung allotransplantation. Five donors were treated with ischemic preconditioning [their left hilus was clamped for 10 minutes and released for 15 minutes (Group IP)], and five donors were not treated with ischemic preconditioning (Group C). The donor lungs were flushed with 4 degrees C Euro-Collin's solution (ECS) and stored in the same solution for two and a half hour, then transplanted to the recipient canines, who were observed for one to two hours after transplantation. The lung venous blood of the recipient and the donor lung tissue were collected just after thoracotomy and one hour after reperfusion of the transplanted lung in both groups. RESULTS: The number of polymorphonuclear (PMN) was significantly higher in Group IP than in Group C (P < 0.05). However, the number of PMN in lung interstitium under microscope was less in Group IP than in Group C. The thromboxane (TXB2), malondialdehyde (MDA) and mean pulmonary artery pressure (MPAP) contents were significantly lower in Group IP than in Group C (P < 0.05). The superoxide dismutase (SOD) and the lung venous blood oxygen tension (PvO2) contents were significantly higher in Group IP than in Group C (P < 0.05). Histological findings showed less damages in Group IP than in Group C. CONCLUSIONS: The protective effect of ischemic preconditioning together with ECS flush and storage is superior to using ECS alone. The possible mechanisms may be that ischemic preconditioning inhibits the accumulation and activation of PMN in lung tissue and reduces the production of oxygen free radicals.
