A study of the adjustment of patients in relation to their prosthodontic problems.

Patients' self-concept and personality have been shown to deteriorate with an increase in prosthodontic problems. This study determined whether patients with prosthodontic problems varying in severity showed different levels of adjustment. Four groups of patients, including a control group, were studied. The three experimental groups differed from the control group and varied in their adjustment in relation to their prosthodontic problems.

Quantitative imaging of free and total intracellular calcium in cultured cells.

Techniques of fluorescence and ion microscopies were combined to study the free [Ca2+] and total Ca in NIH 3T3 fibroblast and L6 rat myoblast cells. Free Ca2+ measurements with the Ca2+ indicator fura-2 and digital imaging reveal an inhomogeneous distribution of free cytoplasmic Ca2+ in both cell lines. Fura-2 also reveals a difference in free Ca2+ activity between the nucleus and cytoplasm of cells. Ion microscopic observations on sister cells show that total Ca in the cytoplasm is also inhomogeneously distributed and that mean cytoplasmic levels of total Ca are higher than levels in the nuclei. In the nuclei of NIH 3T3 cells, the mean free [Ca2+] and total [Ca] were 110 +/- 30 nM and 225 +/- 43 microM, respectively, while regions in the cell cytoplasm contained up to 490 +/- 270 nM free [Ca2+] and 559 +/- 184 microM mean total [Ca]. Intracellular total Ca was greater than 3 orders of magnitude higher than intracellular free Ca2+ in either nuclear or cytoplasmic compartments. Perinuclear cytoplasmic regions in 3T3 cells contained higher free and total Ca than the cell nucleus. Loading of cells with fura-2 did not modify the subcellular distribution of total K, Na, Ca, or Mg. This combination of two powerful ion imaging techniques provides a comparison between free and total calcium in cells and introduces a different approach for examining the role of this important element in cell physiology.

Morphological and elemental integrity of freeze-fractured, freeze-dried cultured cells during ion microscopic analysis.

The effects of progressive ion beam bombardment on freeze-fractured, freeze-dried cultured cells during ion microscopic (SIMS) analysis were studied with scanning electron microscopy (SEM) and ion microscopy. The freeze-fracture, freeze-dry sample preparation method was generally found to preserve cell morphology to a level far exceeding the spatial resolution of the ion microscope, with splitting at the nuclear envelope being the most commonly observed artefact. SEM monitoring of surface topography of an NRK-49F fibroblast after various ion bombardment doses showed relatively uniform erosion of cellular material, with some apparent selective retention of small cytoplasmic granules. Prolonged bombardment produced no detectable lateral elemental translocation. 41K+/24Mg+ signal ratios from Swiss 3T3 fibroblasts and RBL rat basophilic leukaemia cells were shown to vary generally by less than 10% during the course of extended ion bombardment. GM0415 human skin fibroblasts containing engorged lysosomes characteristic of Hurler's Syndrome were used to evaluate the effects of ion bombardment during a typical analysis session, where ion images of 39K+, 23Na+, 40Ca+ and 24Mg+ are sequentially recorded. This cell line was chosen as a worst-case system, because these cells are often thinly spread and possess extreme surface topography. Thin cell edges were shown sometimes to sputter away during analysis, giving misleadingly low ion signals from these regions in some 24Mg+ micrographs. Various non-uniform sputtering phenomena occurring in the submicrometre spatial domain had little or no measurable impact on local intensities in ion micrographs, indicating that freeze-dried, freeze-fractured cells are sampled in a sufficiently uniform fashion that quantitative ion microscopic evaluations of intracellular elemental levels in the general cytoplasmic or nuclear regions are feasible.

Tricalcium phosphate ceramic as immediate root implants for the maintenance of alveolar bone in partially edentulous mandibular jaws. A clinical study.

This study was undertaken to probe the efficacy of tricalcium phosphate ceramic (TCP) as an immediate root implant in the maintenance of alveolar bone. Three patients had five TCP root implants placed in fresh extraction sockets with soft tissue closure. The control and implant areas were evaluated at the 20th and 78th week on the basis of radiographic and clinical measurements. Tricalcium phosphate ceramic root implants in extraction sockets produced a significant increase in height and width of alveolar bone compared with control sites. It is believed that this method is a more effective and efficient procedure to preserve alveolar bone for the retention of dentures than other methods.

A simple method for preparation of good quality cryoprecipitate.

Production of good quality cryoprecipitate from plasma was achieved, as assessed by quantitating Factor VIII coagulant activity (F VIII C) and Factor related antigen (F VIII R: Ag). The method used resulted in good recovery of FVIII in the cryoprecipitate containing F VIII C: Fibrinogen 0.82 +/- 0.015 IU/mg. The method therefore appears suitable for indigenous preparation of F VIII in standard blood banks as a replacement therapy, which is not expensive.

Detection of haemophilia A traits in carriers.

Obligate carriers of haemophilia A and normal women matched for age were studied in order to find a suitable parameter for detection of carriers of haemophilia A. The ratio of factor VIII coagulant activities (F VIIIC) to factor VIII related antigen (F VIII R : Ag) was 0.55 in obligate carriers and 1.0 in non-carrier females.

Formocresol and glutaraldehyde pulpotomies in primary teeth.

The purpose of the present study was to compare clinically and radiologically the effects of formocresol and glutaraldehyde as a medicament in pulpotomized carious exposed vital human primary molars. In formocresol group 90% clinical and radiological success rate and in glutaraldehyde group 100% clinical and radiological success rate was observed. Thus it was concluded that glutaraldehyde is better fixative and less toxic agent than formocresol.

Studies on 2,3,N,N'-substituted 4,4'-diaminodiphenylsulfones as potential antimalarial agents.

A series of new 4,4'-diaminodiphenylsulfones substituted at 2 and 3 position and also at primary amino group of the phenyl rings have been synthesized and evaluated for their antimalarial activity against Plasmodium berghei infection in mice. Some of these compounds were active and showed complete inhibition of parasitaemia which included 7a1-7a4, 7b3, 7b4 and 16a at 1 mg/kg i.p. for 4 days and 16a, at 0.3 mg/kg for 4 days. Some compounds tested for their synthetase inhibitory action in cell-free system isolated from P. berghei (7b1, 7b2 and 8b2) were found to be more active than diaminodiphenylsulphone. The difference in order of activity between these in vivo and in vitro tests may be due to differences in their pharmacokinetic properties.

The antioxidants as protectors of host stress organ injury in mice infected with Plasmodium berghei.

A study has been made of counteracting the stress organ injury in Plasmodium infection by means of antioxidants on the premise that free radicals are responsible for causing the injury to stress organs. This was evidenced by drastically altered biochemical parameters in liver and spleen of the host in terms of elevated levels of lipid peroxides and xanthine oxidase (XO) activity, and a fall in superoxide dismutase activity coupled with other drastic biochemical changes. The cardinal factor responsible for the above was considered to be XO which engenders free radicals purportedly responsible for the stress organ (biochemical) injury. Results demonstrate a lowering of lipid peroxide levels, xanthine oxidase activity, liver weight and modulation of protein level in liver of the host (mouse) in Plasmodium infection when treated with catechin, glutathione and propylgallate.

Distribution of Mallophaga on the body of Acridotheres tristis (Aves).

4 species of Mallophaga (2 Amblycera, Menacanthus eurysternus and Myrsidea sp., and 2 Ischnocera, Brueelia chayanh and Sturnidoecus affinis) coexist on the body of Common Myna, Acridotheres tristis. The distribution of eggs, nymphs and adults of these species on the body of host is discussed. The patterns of distribution coincides with their morphological adaptations.

Seasonal distribution of Aeromonas hydrophila in river water and isolation from river fish.

The seasonal distribution of Aeromonas hydrophila in water and recovery rate from live river fish was investigated. The highest isolation rates of A. hydrophila occurred in water during the late winter followed by a progressive decline in density during the summer and monsoon seasons. The organism was recovered from fish throughout the period from which it was concluded that they form a reservoir which is unrelated to their density in water. The enterotoxigenicity of some environmental strains was tested in suckling mice and rabbit ileal loop.

Findings in child deaths registered as sudden infant death syndrome (SIDS) in Madison, Wisconsin.

A review of a sequential series has been carried out on 60 Madison infants whose deaths had been registered as SIDS. In 18% it was possible to identify lesions that could have given a diagnosis other than SIDS on these same infants. When the cases were assessed on a multifactorial basis, 67% of the infants had a variety of lesions present, which in combination would probably have made their deaths explicable. It was only in children dying before the age of 18 weeks that more than half of the deaths were not explicable. The generally accepted concept of SIDS as a completely unexplained death would appear to apply only if the term is confined to a single aspect of pathology--the terminal anatomic state. If the total pathologic situation of the child is taken into consideration, the diagnosis of a completely unexplained death applies to only about a third of the cases examined in Madison. The current autopsy approach to sudden and unexpected deaths from a causal viewpoint requires critical reappraisal.
