Dependence of renal (Na+ + k+)-adenosine triphosphatase activity on thyroid status.

In thyroidectomized rats, a single injection of L-2,,5,2'-triiodothyronine (T3) (50mug/100 g body weight) elicited at 45% increase in (Na+ + k+)-dependent adenosine triphosphatase (NaK-ATPase) activity of the membrane-rich fraction of renal cortex at the optimal time of response, 48 h after injection. Three successive doses of T3 (50 mug/100 g body weight), given on alternate days, increased NaK-ATPase by 67% in the renal cortex but had no significant effect on the outer medulla or the papilla. Moreover, T3 had no effect on Mg2+-dependent adenosine trisphatase (MgATPase) in cortex, cedulla, or papilla. Three doses of T3 (50 mug/100 g body weight) given on alternate days to thyroidectomized rats elecited a 134, 79, and 46% increase in Vmax for ATP, Na4, and K+, respectively. There were no changes in the Km for ATP or the K1/2 values for Na+ and K+. Two methods were used to estimate the effect of T3 on the number of NaK-ATPase units (assumed to represent the number of Na+ pump sites); rat renal plasma membrane fractions were incubated with [gamma-32P]ATP, Mg2+, and Na+; the 32P-labeled membrane protein yeild was quantitatively dependent on Na+ and was hydrolyzed on addition of K+. There was a linear correlation between the specific activity of NaK-ATPase (Vmax) and the amount of phosphorylated intermediate formed, in renal cortical membrane fractions from thyroidectomized rats given T3 or the diluent. There was also a linear correlation between the specific activity of NaK-ATPase (Vmax) and the amount of [3H]ouabain specifically bound (Na+-, Mg2+-, APT-dependent) to the NaK-ATPase preparation. Injection of T3 resulted in a 70% increase in NaK-ATPase activity, a 79% increase in formation of the phosphorylated intermediate, and a 65% increase in the [H]ouabain specifically bound to the NaK-ATPase system. The T3-dependent increases in Vmax for ATP, Na+, and K+ and the proportionate increases in the phosphorylated intermediate and in the amount of [3H]ouabain bound indicate that T3 increases the number of NaK-ATPase units and that this increase accounts for the increase in NaK-ATPase activity.

Performance characteristics of a laser scanner and laser printer system for radiological imaging.

This paper describes the performance characteristics of a laser scanner and laser printer system for radiological imaging. The laser scanner can digitize a 14'' X 17'' X-ray film to 2000 X 2400 X 10 bit in about 100 s. The laser printer can format 16 images on a 8'' X 10'' or 14'' X 17'' film with up to 2300 X 3050 X 8 bit resolution in about 50 s. Our experiments show that the laser scanner and the laser printer have a spatial resolution of 3 cycles/mm with excellent spatial linearity and flat field response. This system is a major component of a picture archiving and communication system in a digital-based radiology department. As a stand-alone system, it has certain clinical applications such as a contrast enhancement of radiographs and potential dose reduction to the patient.

Projection domain compensation of missing angles for fan-beam CT reconstruction.

An improved method is proposed for fan-beam computed tomographic (CT) reconstruction from data with limited views. Compensation for the missing projections for fan-beam CT can be partially accomplished by using the coincident ray or by an interpolation technique using circular sample theory. In this article, the authors propose a more accurate compensation method for the missing projections whether the coincident ray pairs exist or not. The fan-beam reprojection algorithm, which is the inverse operator of the convolution filter, was extended from the projection space iteration reconstruction-reprojection (PSIRR) in parallel beam geometry. In addition, this algorithm was validated by applying the Shepp-Logan phantom for a computer simulation in the equi-angular fan-beam CT geometry.

Genetic and serologic relatedness between Mycoplasma fermentans strains and a mycoplasma recently identified in tissues of AIDS and non-AIDS patients.

A mycoplasma previously identified in the tissues of both AIDS and non-AIDS patients dying of an acute fatal disease was earlier shown to share some biologic and genetic properties with a strain of Mycoplasma fermentans, an organism occurring infrequently in the human lower urogenital tract. More extensive genetic and serologic comparisons using DNA/DNA hybridization, DNA base composition (guanine + cytosine), restriction endonuclease DNA analysis, cellular protein patterns and metabolism inhibition serologic procedures confirm that the organism previously designated as "Mycoplasma incognitus" (Mi) is indeed very closely related to strains of M. fermentans. While the genetic and serologic features observed among the newly isolated mycoplasma and two M. fermentans strains suggest a species relationship, it now seems useful to re-examine the biological activities of other freshly isolated M. fermentans strains from man.

Herpes simplex type II and Mycoplasma genitalium as risk factors for heterosexual HIV transmission: report from the heterosexual HIV transmission study.

OBJECTIVES: Two hundred twenty-four human immunodeficiency virus (HIV) discordant couples (one HIV negative, one HIV positive) were compared with 78 seroconcordant heterosexually infected couples with HIV with regard to sexually transmitted diseases. METHODS: Serologic testing and cultures were used to determine exposure of participants to sexually transmitted pathogens. These data were compared with HIV concordance of partners to investigate possible risk factors for HIV transmission. RESULTS: Syphilis, chlamydia, and hepatitis B virus (HBV) serologies did not distinguish between concordant and discordant couples nor did cultures for Neisseria gonorrhoeae and Trichomonas or Chlamydia enzyme immunoassay (EIA). Risk of transmission increased with positive serologies for herpes simplex virus (HSV)-2 (P = 0.002), cytomegalovirus (CMV) (P = 0.04), and Mycoplasma genitalium (P = 0.01), but not with Mycoplasma fermentans or Mycoplasma penetrans. Cytomegalovirus was not a significant risk factor when controlled for HSV-2 status. Examination by partner status showed increased risk of concordance with: HSV-2 positive serology in both partners (odds ratio [OR] = 3.14; confidence interval [CI] = 1.62-6.09; P = 0.007); HSV-2 in female secondary partner (OR = 2.10; CI = 1.12-3.93; P = 0.02) or the male primary partner (OR = 2.15; CI = 1.15-4.02; P = 0.017); M. genitalium antibody in both partners (OR = 3.44; CI = 1.68-7.04; P < 0.001); M. genitalium antibody in the primary male partner (OR = 2.51, CI = 1. 27-4.91; P = 0.008) and M. genitalium antibody in the secondary female partner (OR = 2.52; CI = 1.21-5.23; P = 0.01). CONCLUSIONS: These data support the role of HSV-2 in transmission of HIV and, for the first time, suggest a role for M. genitalium as an independent risk factor.

A method for splitting digital value in radiological image compression.

A new decomposition method using image splitting and gray-level remapping has been proposed for image compression, particularly for images with high contrast resolution. The effects of this method are especially evident in this radiological image compression study. In these experiments, the impact of this decomposition method was tested on image compression by employing it with two coding techniques on a set of clinically used CT images and several laser film digitized chest radiographs. One of the compression techniques used as zonal full-frame bit-allocation in the discrete cosine transform (DCT) domain, which is an enhanced full-frame DCT technique that has been proven to be an effective technique for radiological image compression. The other compression technique used was vector quantization with pruned tree-structured encoding, which through recent research has also been found to produce a low mean-square error and a high compression ratio. The parameters used in this study were mean-square error and the bit rate required for the compressed file. In addition to these parameters, the differences between the original and reconstructed images were presented so that the specific artifacts generated by both techniques could be discerned through visual perception.
