Subsets of T lymphocytes in peripheral blood of patients with oral lichen planus.

The T lymphocyte subsets in the peripheral blood were studied in 26 patients with oral lichen planus and in 11 normal controls. Lymphocyte subsets were identified with monoclonal antibodies (Ortho Diagnostic System Inc. Raritan, New Jersey, USA) by an indirect immunofluorescent antibody method. This study shows no significant difference between patients with oral lichen planus and healthy controls. The conclusion is that local immunity, due to the breakdown of skin-associated lymphoid tissue, not a generalized immunologic disorder, is considered to be involved in the pathogenesis of oral lichen planus.

Peripheral ameloblastoma with metastasis.

Peripheral ameloblastoma is an uncommon clinical entity, while ameloblastoma with distant metastasis is still more unusual. A case of peripheral ameloblastoma with left supraclavicular lymph node metastasis is presented here.

Serum constituents and liver photomicrographs of wild Suncus murinus in the south of Taiwan.

Serum constituents and liver of the wild male suncus as well as of those bred and fed in our laboratory were examined serologically and histologically. The following results were obtained: The serum glutamic-oxaloacetic transaminase, gamma-glutamyl transaminase, inorganic phosphate, blood urea nitrogen, creatinine, uric acid, and zinc turbidity test levels of wild suncus were higher than those of 4 and 8 week old fed suncus. The total cholesterol level of wild suncus was lower than those of 4 and 8 week old fed suncus. In wild suncus, the total bilirubin and direct bilirubin levels were higher, and the Ca and albumin levels were lower than those of 4 week old fed suncus. However, no significant differences were observed between the wild and the 8 week old fed suncus. Although the alkaline phosphatese and thymol turbidity test levels of wild suncus were larger than those of 8 week old fed suncus, no significant differences were observed between the wild and the 4 week old fed suncus. The histological study revealed the presence of fatty droplets in only 2 of the 67 wild suncus examined while fatty droplets (grade +) were observed in almost all the fed suncus. In one case of the wild suncus, moderate round cell infiltration in interlobular connective tissue was found.

Studies on muscarinic binding sites in human brain identified with [3H]pirenzepine.

Pirenzepine, a potent antimuscarinic agent with apparent selectivity for a subtype (M1) of muscarinic receptors, was used in tritiated form to characterize its binding to human brain tissue. Specific [3H]pirenzepine binding showed rapid association and dissociation. From kinetic and competitive binding experiments, its KD was 5.5 nM and 9 nM, respectively. Regional distribution of [3H]pirenzepine binding determined in parallel with [3H]quinuclidinyl benzilate binding, a nonselective muscarinic antagonist, indicated a significant correlation for the maximum number of binding sites for the two radioligands in 13 brain regions, with the highest amount of binding for each in the putamen and the least in the cerebellum. Binding for [3H]pirenzepine averaged 57% of that for [3H]quinuclidinyl benzilate, with a range of 20% (cerebellum) to 77% (frontal cortex). Most antidepressants and neuroleptics tested had affinities for [3H]pirenzepine binding sites that were not significantly different from their previously reported values obtained with the use of [3H]quinuclidinyl benzilate.

Studies on the diurnal pattern of serum 3,5,3'-triiodothyronine.

Seventeen subjects, including 12 mildly obese women, were investigated to determine the presence of a diurnal variation in serum T3 levels. A subset of 8 subjects was studied after fasting for 6 days, while another subset of 5 subjects was studied 2 days after receiving 3 mg T4, orally, to suppress TSH secretion. To negate the influence of hemoconcentration produced by ambulation, serum T3 to T4 ratios (nanograms per microgram) rather than total T3 values were used for analysis. A synchronous diurnal rhythm for mean serum T3 to T4 ratios and TSH values was found, with mean nighttime increases of 7.8% and 49.5%, respectively. The timing of the T3 to T4 and TSH nocturnal peaks for individual subjects, however, were not correlated, suggesting that T3 to T4 ratios changes were not caused by TSH stimulation of thyroid T3 release. During fasting, the diurnal rhythm of serum T3 to T4 was obliterated within 24 h of beginning the fast, although TSH rhythmicity persisted. After the 3-mg oral T4 dose, serum TSH became undetectable, while the diurnal serum T3 to T4 ratio changes persisted. In contrast, there was no detectable diurnal rhythmicity of serum T3 to T4 ratios during either the control or fasting period. We conclude that the diurnal rhythm of serum T3 is not TSH dependent, but, rather, is influenced by some as yet unidentified dietary signal, which alters the efficiency of the peripheral tissue T4 to T3 conversion.

Effects of morphine and B-endorphin on Ca2+-ATPase activity of synaptic plasma membranes.

Morphine and B-endorphin were found to inhibit the high calcium -affinity Ca2+-ATPase of synaptic plasma membranes which is believed to be involved in the regulation of intracellular Ca2+ that triggers the release of neurotransmitters. The inhibition was blocked by naloxone and reversed by calmodulin. Kinetic analyses revealed that opioids decreased the affinity of Ca2+-ATPase for Ca2+ and calcium bound to the enzyme decreased the affinity of the enzyme for opioids.

Plasma levels of cortisol, corticotropin, and beta-endorphin in patients with major depression.

Efforts to elucidate the abnormal mechanism of corticotropin and beta-endorphin in major depression have yielded conflicting findings. The relationship of plasma levels of cortisol, corticotropin, and beta-endorphin in 42 patients with a Research Diagnostic Criteria diagnosis of major depression, endogenous subtype was examined. Following the DST, 32 patients were nonsuppressors and 10 were suppressors. The differences between the median values for plasma corticotropin and beta-endorphin immunoreactivity were not significant at any time of measurement after the DST.

Serum levels of beta-2 microglobulin in oral tumor patients.

Beta-2 microglobulin (beta-MG) is a low molecular weight protein of 100 amino-acids. It is the light chain of HLA-A, -B or -C antigens which on the surface of nearly all nucleated cells. Clinically, the serum levels of beta-MG are increased in patients with renal failure, immune disorders, and various malignant diseases. In this study, the serum levels of beta-MG were measured clinically for a total of 114 cases. Diverse conditions were ensured by biopsy except for 10 control cases, and the following eight groups were diversified: (1) control group, 10 cases; (2) oral carcinoma, 36 cases; (3) mixed tumor, 7 cases; (4) pyogenic granuloma, 12 cases; (5) leukoplakia, 18 cases; (6) mucocele, 15 cases; (7) ameloblastoma, 7 cases; (8) fibroma, 9 cases. The serum levels of beta-MG in oral cancer and leukoplakia patients were found to be significantly higher compared with that of the control group and patients with benign oral tumors. Further study to unravel the correlations between the clinical stages, the treatment of oral cancer, and beta-MG is now in progress.

Serum constituents of Suncus murinus.

Blood samples were collected by cardiac puncture from nine-week-old suncus, mice and rats those had been fasted for 16 hours, and the serum was assayed for the levels of corticosterone, cortisol, and other constituents. The following results were obtained: The serum levels of corticosterone and cortisol and corticosterone/cortisol ratio of suncus were about the same as those of human. The SGPT, gamma-GT, ALP, LDH, Ca and K values in suncus were all within the normal range of the respective values in human. SGOT, amylase, BUN, Na, Cl, Fe and inorganic phosphate values were higher, and total cholesterol, triglyceride, phospholipid, creatinine, urea, total protein, albumin and bilirubin levels were lower in suncus than the respective normal values in human. The values of SGOT, amylase, ZTT, and K were higher in female than in male suncus, while the values of gamma-GT and ALP were higher in the male. Feeding the animals individually in the individual cages for a week increased the values of direct bilirubin, total cholesterol, ZTT, Na, K and Cl and lowered the value of urea. The values of SGPT, ALP and triglyceride of suncus obtained in 1984 were higher, the value of albumin was lower, than the respective values obtained in 1983. The value of cholinesterase in suncus was very small.

Species difference in the fibrinogenolytic effects of alpha- and beta-fibrinogenases from Trimeresurus mucrosquamatus snake venom.

Alpha- and beta-fibrinogenases prepared from Trimeresurus mucrosquamatus venom digested specifically the alpha(A) and beta(B) chains of the fibrinogen molecule, respectively. alpha-Fibrinogenase digested bovine fibrinogen more markedly than human fibrinogen, while beta-fibrinogenase digested human fibrinogen more markedly than bovine fibrinogen. Human fibrin was also digested by both enzymes. Plasma fibrinogens of 4 animal species were digested by alpha-fibrinogenase to the same degree, while those by beta-fibrinogenase in the following order: human greater than dog greater than guinea-pig greater than rabbit. The fibrinogenolytic effects of alpha-fibrinogenase on human fibrinogen were strongly inhibited by sera of the 4 animal species, while those of beta-fibrinogenase were inhibited in the following order: rabbit greater than guinea-pig greater than dog greater than human. It was concluded that the different activities of the protease inhibitors in the plasma of animal species are mainly responsible for the sensitivity differences.
