Structural requirements for FokI-DNA interaction and oligodeoxyribonucleotide-instructed cleavage.

The FokI restriction endonuclease recognizes the double-stranded (ds) 5'-GGATG-3' site and cuts at the 9th and 13th nucleotides downstream from the 5'-3' and 3'-5' strands, respectively. To elucidate the interaction between FokI and DNA, and the effect of Mg2+ on this interaction, we used FokI with various combinations of dsDNA, single-stranded (ss) DNA and oligodeoxyribonucleotides (oligos) containing a double-stranded hairpin carrying the FokI recognition site. Oligo- and dsDNA-FokI interactions showed that for fully effective recognition, two or more base-pairs were required outside the 5'-GGATG-3' site. When using FokI with ssDNA and oligos, precise cutting with no observable byproducts was observed at the 9th or 13th nucleotide. This was independent of whether the region between the recognition and cut sites was perfectly complementary or whether there were up to four mismatches in this region, or a single mismatch within the cut site. Moreover, FokI cleavage, when followed by step-wise filling-in of FokI cohesive ends in the dsDNA, allowed FokI to recleave such sites when two or more nucleotides were added, releasing 2-mer, 3-mer, or 4-mer single-stranded chains. Electrophoretic mobility shift assays showed that the DNA helix was bent when complexed with FokI (without Mg2+. Such a complex, when formed in the absence of Mg2+, did not accept the subsequently added Mg2+ for several minutes. This suggests a tight, diffusion-resistant contact between the enzyme and the cognate DNA sequence. In the presence of Mg2+, the half-life of the complex FokI and dsDNA was 12 minutes at 22 degrees C. In the absence of Mg2+, such a complex, possessing a terminally located 5'-GGATG-3' site, had a half-life of 1.5 to 2 minutes. However, if magnesium ions were present, this complex had a stability similar to that of a complex formed with dsDNA containing a centrally located 5'-GGATG-3' site.

A novel antimicrobial peptide from Bufo bufo gargarizans.

A potent and structurally novel antimicrobial peptide was isolated and characterized from the stomach tissue of Bufo bufo gargarizans, an Asian toad. The 39-amino acid peptide, named buforin I, was purified to homogeneity by heparin-affinity column and reverse-phase HPLC. The amino acid sequence of buforin I was identical in 37 of 39 amino-terminal residues of Xenopus histone H2A. The buforin I showed strong antimicrobial activities in vitro against a broad-spectrum of microorganisms and was found to be more potent than magainin 2. In addition, a 21-amino acid peptide, named buforin II, which was derived from buforin I, showed more potent antimicrobial activities than buforin I.

Generalized multinucleate cell angiohistiocytoma.

Multinucleate cell angiohistiocytoma is a rare benign fibrohistiocytic vascular condition that mainly affects middle-aged women. Grouped reddish-brown papules are found in the extremities. We describe a case of multinucleate cell angiohistiocytoma occurring in a 24-year-old man who presented with multiple papules on the trunk and extremities. This is the first report of generalized multinucleate cell angiohistiocytoma.

Sequential amplification of cloned DNA as tandem multimers using class-IIS restriction enzymes.

In order to make high-copy-number multimers of DNA fragments in a tandem unit, two different gene amplification vectors (pSK9 and pBBS1) were developed. Two identical class-IIS restriction enzyme sites (BspMI for pSK9 and BbsI for pBBSI) were inversely oriented in each vector with the same cut site, creating asymmetric and complementary cohesive ends (5'-CCCC and 5'-GGGG). Multimers were made by: (i) cloning a target DNA into the class-IIS restriction enzyme cut site of each vector; (ii) excision of the monomeric insert by digestion with the class-IIS restriction enzyme; (iii) isolation of the fragments; (iv) self-ligation of the fragments; (v) cloning into the original vector digested with the class-IIS restriction enzyme; and (vi) repeating steps (i) through (v) to generate higher-order multimers. Various-sized multimers of a 93-bp DNA fragment encoding magainin, an antimicrobial peptide, were obtained with the gene amplification vector, pBBS1. Larger multimers, up to about 108 copies, were constructed from the monomer by the sequential amplification procedure. Of six different Escherichia coli hosts examined for the stability of multimers, the multimers were the most stable in E. coli D1210. The gene amplification vector system described here is very efficient and can be applied in the construction of tandem multimers of any kind of DNA, as long as the cloned DNA does not contain the cut site of the class-IIS restriction enzyme to be utilized.

Treatment of posttraumatic chronic high-flow priapisms by superselective embolization of cavernous artery with autologous clot.

Two cases of high-flow priapism that occurred after blunt perineal trauma and lasted for 35 days (case 1) and 40 days (case 2) were successfully treated by embolization of the lacerated cavernous arteries with autologous clots. Cavernous arterial blushes were demonstrated on selective internal pudendal arteriograms in both cases. Case 1 with left cavernous arterial laceration restored 80% of premorbid erectile function with successful intercourse 4 months after superselective embolization. In case 2 with bilateral cavernous arterial lacerations, premorbid erectile function was fully resumed 2 months after bilateral embolization of the cavernous arteries at intervals of 15 days. None developed local or systemic complications.

Effect of ginsenoside Rb1 on rat liver phosphoproteins induced by carbon tetrachloride.

We investigated the effects of ginsenoside Rb1 (G-Rb1), a major saponin from Panax ginseng C. A. MEYER, on rat liver protein phosphorylation after intraperitoneal administration of CCl4 alone or together with G-Rb1. We found that 118, 63, and 34kDa proteins were prominently phosphorylated in liver homogenates prepared from CCl4-administered rats, while these protein-phosphorylations were inhibited in the homogenate prepared from the G-Rb1 plus CCl4-administration group. When inhibitors of protein kinases were exogenously added to the homogenates from either the CCl4-administered group or the G-Rb1 plus CCl4-administered group, their phosphorylations were inhibited much more by W-7, an inhibitor of Ca2+/calmodulin-dependent protein kinase (CaM-PK), than by H-7, an inhibitor of protein kinase C (C-kinase). Interestingly, only 34kDa was phosphorylated in homogenates prepared from the corn oil-, G-Rb1-, and G-Rb1 plus CCl4-administered groups by the exogenous addition of sodium fluoride (NaF), an inhibitor of glycogen synthase. Additionally, G-Rb1 inhibited the Ca(2+)-accumulation induced by CCl4 both in liver homogenates and microsomes. The above results imply that G-Rb1 inhibits the CCl4-induced protein phosphorylations by modulating CaM-PK rather than C-kinase, and that 34kDa protein may play a different biological role in cellular environment from 118 and 63kDa proteins. Therefore, a study in which G-Rb1 is employed as a modulator of critical CCl4-induced phenomena ranging from the disturbance of Ca2+ concentration to protein phosphorylation may be successfully applicable to investigate the diverse physiological functions of liver.

[Population density of chigger mites, the vector of tsutsugamushi disease in Chollanam-do, Korea].

The geographical distribution and population density of rodents and chigger mites at six localities of Chollanam-do were investigated from October to December in 1993. Among total 142 field rodents collected by the modified Chemla wooden traps, 131 rodents were Apodemus agrarius (92.3%) and 11 were Crocidura lasiura (7.7%). Out of 142 field rodents, 92 were parasitized by chiggers, showing 69.0% of the infestation rate and 74.2 of the chigger index. Infestation rate and chigger index of A. agrarius and C. lasiura were 73.3%, 80.4 and 18.2%, 0.5, respectively. From the trapped field rodents, 10,532 chiggers were collected and identified with 11 species of 4 genera. Leptotrombidium pallidum, the vector species of tsutsugamushi disease, was the dominant species, showing 8,038 chiggers (76.3%). L. scutellare was the second dominant species showing 1,359 chiggers (12.9%). The distribution of chigger mites was clearly localized by the species, showing the different dominant species according to localities. The predominant species was L. scutellare (100%) in Changhung-gun, L. scutellare (41.5%) in Posong-gun, L. pallidum (88.8%) in Hwasun-gun, L. pallidum (59.2%) in Koksong-gun, L. zetum (77.3%) in Hampyong-gun, and L. palpale (63.4%) in Tamyang-gun. Regarding to the geographical distribution of chigger population density, the infestation rate and chigger index was most high in Hwasun-gun as 62.4% and 216.2 respectively, and next high in Koksong-gun as 22.4% and 77.7% respectively.

Minocycline treatment for confluent and reticulated papillomatosis.

We report six cases of confluent and reticulated papillomatosis in which the skin lesions cleared almost completely after treatment with minocycline. Patients with confluent and reticulated papillomatosis often do not respond well to a variety of therapeutic agents. The response of confluent and reticulated papillomatosis to minocycline was first described in 1965. Although the pharmacologic mechanisms of minocycline in confluent and reticulated papillomatosis are unknown, we suggest that it should be considered as a first-choice therapeutic agent because of its marked effectiveness.
