Effect of levamisole on metabolism of phagocytic cells.

The effect of levamisole (LMS) on glucose metabolism was studied using a protozoan phagocytic model and human leukocytes. At concentrations of greater than 10 microgram/ml, LMS inhibited glucose metabolism in the protozoan phagocytic model. Glucose metabolism in both the phagocytic model and normal peripheral blood leukocytes was, however, minimally altered when exposed to levels of LMS of less than 10 microgram/ml. In contrast, myeloblasts from a child with leukemia seemed to have increased metabolic activity and 5 microgram/ml of LMS markedly reduced the glucose metabolism. These preliminary studies suggest that LMS can alter glucose metabolism of certain cells and that some malignant cells may be directly inhibited metabolically by LMS.

Effect of alternating magnetic fields (60--100 gauss, 60 Hz) on Tetrahymena pyriformis.

Tetrahymena pyriformis and neuroblastoma cells were studied following exposure to low intensity low frequency alternating magnetic fields. Tetrahymena showed cytomorphologic changes, with delayed and reduced cell division concurrent with increased oxygen uptake. The resulting dead cells appeared intact, as compared with dissolution characteristic of the control group. In contrast, magnetically exposed actively growing neuroblastoma cells showed no growth alterations in vitro, but were affected when exposed in vivo.

Effects of hypolipidemic agents on lipid synthesis in subcellular fractions from Tetrahymena pyriformis.

Lipid synthesizing systems have been prepared from subcellular fractions of Tetrahymena pyriformis, GL. These fractions, the mitochondrial fraction, the microsomal fraction and the soluble cell fraction, have been characterized as to cofactors and cations required for optimal lipid synthesis. The effects of hypolipidemic agents on lipid synthesis by all fractions are presented.

Effect of levamisole on the synthesis of macromolecules in log phase and synchronized cultures of Tetrahymena pyriformis, GL.

The effects of levamisole on log phase and synchronized cultures of Tetrahymena pyriformis, strain GL, have been examined. At a dose of 100 mug/ml, DNA, RNA and protein syntheses were inhibited by 38%, 27% and 19%, respectively, in log phase cells, and by 21%, 50% and 47%, respectively, in synchronized cells. Lipid synthesis was only slightly inhibited (15%) at this same dose in synchronized cells. Results suggest that levamisole may act by directly inhibiting nucleic acid syntheses and the inhibition of protein synthesis may be secondary to this effect.

New approach to antifolate treatment of certain cancers as demonstrated in tissue culture.

The selective toxicity of antifolates for a variety of cancers can be improved, as illustrated by the combined administration of N5-methyltetrahydrofolate and methotrexate in tissue culture. When a variety of neoplastic cell types characterized by a deficiency of vitamin B12-dependent N5-methyltetrahydrofolate methyltransferase (5-methyltetrahydropteroyl-L-glutamate:L-homocysteine S-methyltransferase, EC 2.1.1.13) and normal adult cells are grown in media containing methotrexate and either N5-methyltetrahydrofolate or N5-formyltetrahydrofolate, not only is the selective toxicity of methotrexate demonstrated, but the advantage of using N5-methyltetrahydrofolate in place of N5-formyltetrahydrofolate is also revealed. The implications and applications of this particular combination in the treatment of human cancer are discussed.
