Studies of the endoplasmic reticulum and plasma membrane-bound ribosomes in erythropoietic cells.

Erythropoietic cells of 5 species, including man, contain endoplasmic reticulum present as individual cisternae or tubules scattered throughout the cytoplasm of all stages except mature RBCs. The endoplasmic reticulum is mainly agranular but occurs frequently as a variant of granular ER which is characterized by an asymmetrical and irregular distribution of ribosomes along one cytoplasmic face. In most cells, the endoplasmic reticulum occurs in close proximity to mitochondria or the plasma membrane , suggesting that the organelle may be involved in functions related to these structures, e.g. haem biosynthesis. Endoplasmic reticulum is more abundant in early than in late erythroid cells. Its exact role in RBC development is unclear. Since endoplasmic reticulum could account for 'plasma membrane-bound ribosomes' reported in lysed reticulocytes, studies were performed which ruled out this possibility and which suggested that such ribosomes were an artifact of the lysing conditions. Hypotonic lysis in less than 20 vol. of magnesium-containing buffers yielded ghosts variably contaminated by ribosomes and other structures. Lysis of reticulocytes in 20-30 vol. of magnesium-free buffer or homogenization of whole cells or crude membrane fractions in hypotonic buffer removed virtually all contaminating ribosomes from the purified membrane fraction.

Percutaneous ethanol injection as a possible curative treatment for malignant portal vein thrombosis in hepatocellular carcinoma.

Postoperative residual hepatocellular carcinoma (HCC) with malignant portal vein thrombosis in a 48-year-old man was cured with transarterial chemoembolization (TACE) for the parenchymal portion and percutaneous ethanol injection (PEI) for the malignant portal vein thrombosis. No evidence of tumor recurrence was noted after 18 months of follow-up. The only severe complication in our patient was biliary stricture which was treated with an internal stent via endoscopic retrograde pancreatico-cholangiography (ERCP).

Improved sonographic visualization by fluid challenge method of renal lithiasis in the nondilated collecting system. Experience in seven cases.

Ultrasound is an effective means of detecting renal stones in patients with hydronephrosis. The filled urinary bladder in a normal person under hydration frequently results in the sonographic appearance of minimal or moderate hydronephrosis. For optimal visualization of stones and associated acoustic shadows, six patients with radiographically opaque renal stones were evaluated with fluid-loaded renal ultrasonography, which was compared to conventional renal sonogram in basal state. Although ultrasound in basal state can be used for detection of intrarenal lithiasis, fluid-challenged renal sonogram appears more accurate for the determination of contour, location, size, and number of small stones. In one case, this fluid-loaded ultrasound well demonstrated the tramline appearance of an isolated renal artery, which mimics stone before fluid challenge.

Urinary excretion of furazabol metabolite.

Furazabol, a newer anabolic steroid, is metabolized into 16-hydroxyfurazabol and excreted in urine. The presence of this compound in urine can be monitored with a GC/MS procedure. It can be incorporated into the general dope testing protocol for anabolic steroids.

Analysis of bumetanide in human urine by high-performance liquid chromatography with fluorescence detection and gas chromatography/mass spectrometry.

Bumetanide is a potent diuretic. Its concentration in urine after therapeutic doses is in the low ng/mL range. A procedure using reversed-phase HPLC with fluorescence detection is described. Using this procedure, the authors have been able to measure bumetanide in a urine sample at 10 ng/mL with a signal-to-noise ratio of 7. Therefore, the detection limit is lower than this concentration. Confirmation is by a GC/MS procedure after methylating the diuretic. Derivatization is by extractive alkylation with tetrabutylammonium hydrogen sulfate as the phase transfer reagent and iodomethane as the methylating agent.

Screening for acidic and neutral drugs by high performance liquid chromatography in post-mortem blood.

A procedure for screening acidic and neutral drugs in post-mortem blood is presented. The procedure, which is sensitive and specific, employs high performance liquid chromatography with a reversed-phase column and an ultraviolet detector. The volume of specimen used is 1 mL. Over 40 drugs are identifiable by this technique, including analgesics, anti-inflammatory agents, barbiturates and other hypnotic-sedatives, and anti-epileptic drugs. To date, over 500 post-mortem blood specimens have been analyzed by the authors.

Enhancement of the cytolytic effect of anti-bacterial cecropin by the microvilli of cancer cells.

A comparison between IC50s of cecropin B on tumor cells such as KG-1 leukemia and Ags stomach carcinoma and non-tumor cells like fibroblasts and red blood cells was conducted. The IC50s of cecropin B for KG-1 leukemia and Ags carcinoma cells were 20.8 +/- 2.3 microM (MTT) and 18.9 +/- 3.3 microM (trypan blue) and 16.0 +/- 3.5 microM (MTT) & 15.3 +/- 3.7 microM (trypan blue), respectively. The IC50 of cecropin B for 3T6 fibroblast cells was 92.0 +/- 9.1 microM by MTT assay and the HE50 of cecropin B for human red blood cells was 180.0 +/- 20.1 microM at OD414nm. The cytolysis induced by cecropin peptides was more effective for the cancer cells than for the normal cells. Based on the observations from scanning electron microscopy, this may mainly due to the cancer cells having a high population of the irregular microvilli on the cell surface. Since peptides bound to the cell membrane are non-specific, the attraction of peptides by microvilli may be one of the main driving forces before the lysis in membrane bilayers can be efficiently initiated.
