Flash photolysis of the carbon monoxide compounds of wild-type and mutant variants of cytochrome bo from Escherichia coli.

The carbon monoxide compounds of the fully reduced and mixed valence forms of cytochrome bo from Escherichia coli were laser photolysed under anaerobic conditions at room temperature. The carbon monoxide recombined with characteristic rate constants of 50 s-1 or 35 s-1 in the fully reduced and mixed valence forms, respectively. Rates of CO recombination with the fully reduced enzyme were examined in a variety of mutant forms of cytochrome bo, produced by site-directed mutagenesis. A method was developed to deconvolute cytochromes bo and bd, leading to some reassessment of histidine ligands to the metals. Significant changes in the rate constant of recombination of carbon monoxide occurred in many of these mutants and these results could be rationalised generally in terms of our current working model of the folding structure of subunit I. In the mixed valence form of the enzyme the transient photolysis spectra in the visible region are consistent with a rapid electron redistribution from the binuclear centre to the low-spin haem. This electron transfer is biphasic, with rate constants of around 10(5) and 8000 s-1. The process was also examined in the His-333-Leu mutant, in which a putative histidine ligand to CuB is replaced by leucine, and which results in the loss of the CuB. It appeared that rapid haem-haem electron transfer could still occur. The observation that CuB is apparently not required for rapid haem-haem electron transfer is consistent with the recently proposed model in which the two haems are positioned on opposite sides of transmembrane helix X in subunit I of the oxidase.

Expression of PNA-binding sites on specific glycoproteins by human melanoma cells is associated with a high metastatic potential.

Lectin-binding patterns of seven human melanoma clones and variants selected from the same parental cell line and differing in their spontaneous metastatic potential in an animal model were compared by flow cytometry and Scatchard analysis. Human melanoma clones and variants with high and low metastatic potential could be distinguished by their peanut agglutinin (PNA)-binding patterns, but not by their wheat germ agglutinin (WGA)-, Ulex europaeus agglutinin I (UEA I)-, and soybean agglutinin (SBA)-binding patterns. Low metastatic clones and variants proved to be made up of single poorly peanut agglutinin-binding cell population (2.20-3.52 x 10(6) sites/cell, Ka = 2.48-2.75 x 10(6) M-1). By contrast, highly metastatic variants were found to be constituted by two cellular subpopulations, exhibiting respectively a moderate 2.62-3.72 x 10(6) sites/cell) and a high peanut agglutinin staining (17.68-18.76 x 10(6) sites/cell). One highly metastatic clone was found to be homogeneously constituted by a single population of cells strongly binding this lectin (18.86 x 10(6) sites/cell) with an association constant of 4.06 +/- 10(6) M-1. Using an EPICS V cytometer, these two subpopulations were sorted from a highly metastatic variant and tested for their metastatic abilities: cells with high PNA binding generated a higher frequency of metastases than did moderately PNA-binding cells. Following treatment with Vibrio cholerae neuraminidase, all cells from all variants and clones were brightly labeled by PNA, collecting in a single peak with similar fluorescence intensities. Electrophoresis of total cellular proteins and subsequent detection with labeled PNA om Western blots show two major PNA-reactive glycoproteins with apparent molecular weights of 140 and 110 kDa (MAGP1 and MAGP2), expressed only in highly metastatic cells, but which can be strongly labeled by PNA in slightly metastatic cells following a treatment with neuraminidase. These results provide evidence that the expression of terminal galactose (beta 1-3)N-acetyl galactosamine structure, positioned on MAGP1 and MAGP2 glycoproteins, is associated with the metastatic potential of human melanoma cells.

Expression and visualization during cell cycle progression of alkaline phosphatase in B lymphocytes from C3H/HeJ mice.

The expression of alkaline phosphatase (APase) activity by purified B cells from lipopolysaccharide (LPS)-hyporesponsive C3H/HeJ mice was determined. Optimal APase activity was expressed after costimulation with interleukin-5 and dextran sulfate (DXS), whereas LPS, which is highly effective on B lymphocytes from normal mice, was unable to induce enzyme expression, even in the presence of DXS. The simultaneous determination by flow cytometry of both cellular APase, by using a fluorescent azo dye technique, and DNA content showed that APase was highly expressed by about one-tenth of cells in G1 phase, whereas it was present in more than 50% of cells in S and G2/M phases. The enzyme, as visualized by confocal microscopy after cell sorting on the basis of DNA content, was found to be localized mainly in vesicular structures distributed throughout the cytoplasm in G1 cells. It was distributed in patches and essentially localized at the cell periphery in S cells, whereas clear capping of activity was observed in G2/M cells.

Comparison of virulence of Mycobacterium avium complex (MAC) strains isolated from AIDS and non-AIDS patients.

Mycobacterium avium complex (MAC) strains from AIDS and non-AIDS patients and from the environment were studied for their colony morphology and virulence in beige mice. The majority of the MAC isolates from AIDS patients, in contrast to those from non-AIDS patients and the environment, showed increased virulence. Similarly, the majority of the MAC isolates from AIDS patients formed smooth transparent (ST) colonies, whereas most of the non-AIDS isolates formed smooth opaque (SO) or intermediate (IM) type of colonies. MAC isolates from the same AIDS patient obtained at different times were found to be heterogenic with respect to serotype, RFLP and glycolipid patterns, suggesting that these patients might be infected with more than one strain of MAC.

Effect of cadmium on bone calcium and 45Ca in mouse dams on a calcium-deficient diet: evidence of Itai-Itai-like syndrome.

To test whether Cd exposure would increase Ca release from bone during pregnancy and lactation in relation to the etiological mechanism of Itai-Itai disease, virgin female mice with 45Ca prelabeled skeletons (15 microCi/mouse) were subjected to one round of pregnancy/lactation and were exposed to a Ca-deficient diet containing 0, 5, or 25 ppm Cd or 25 ppm Pb for 32 days, from conception until Lactation Day 14. A striking loss of 45Ca was found in the dam's total skeleton (-40%), right femur (-47%), and lumbar vertebrae (L1-L5) (-75%) due only to pregnancy/lactation in conjunction with Ca deficiency. At both 5 and 25 ppm, Cd administered through food induced an additional significant 45Ca loss from the total skeleton (-25% at 5 ppm Cd, -30% at 25 ppm Cd) and right femur (39% at 5 ppm Cd, -32% at 25 ppm Cd) compared to 0 ppm animals. Almost all of the 45Ca lost from the dam's skeleton appeared in the pups, with 80% transferred via the dam's milk during lactation and only 20% transferred during gestation; a very small fraction of the dam's skeletal 45Ca was excreted. Considering stable Ca values, Cd exposure nearly doubled the loss of Ca from the dam's skeleton (-78 mg Ca/mouse at 0 ppm; -146 mg Ca/mouse at 5 and 25 ppm Cd). Paralleling 45Ca losses, a Ca-deficient diet in combination with pregnancy/lactation alone caused significant decreases in weight and mineral content of the right femur and lumbar vertebrae (dry weight, ash weight, ash/dry, Ca content, Ca/dry, and Ca/ash) (-8 to -52%). Cd at both 5 and 25 ppm showed additional decreases (-15 to -32%, Cd groups compared to 0 ppm animals). Responses were specific to Cd in that no significant effect occurred due to 32 days of Pb exposure (25 ppm). This experiment supports the view that Cd exposure in conjunction with Ca deficiency and pregnancy/lactation are key etiological factors of Itai-Itai disease and that Cd at both 5 and 25 ppm in conjunction with one round of gestation/lactation and Ca deficiency can induce an extreme demineralization characteristic of Itai-Itai-like syndrome.

Inguinal herniotomy in young infants, with emphasis on premature neonates.

Over a period of 6 years, 251 infants under 6 months of age underwent repair of inguinal hernias (IH; n = 311). There were 241 males and 10 females. Incarceration occurred in 59 infants (24%), one of whom had bilateral incarceration. As a result of the authors' policy to operate on infantile IH within 7 days of diagnosis, only 6% of the incarcerations occurred in already diagnosed cases. Sedation and taxis did not reduce the hernia in 22 cases (38%); transperitoneal closure of the internal ring was performed in 14 of these. Eighty-nine infants (36%) were born premature; thirty-nine (41%) of these had been ventilated before, a possible cause of the hernia. Bilateral presentation was more common in the premature infants (35% v 17%); surprisingly, incarceration was less common (13% v 24%). Hence, the policy of delaying herniotomy until discharge from the neonatal unit was justified. During follow-up, six recurrences were noted and two cases of testicular atrophy.

Epilepsy, pseudoseizures and perceived family characteristics: a controlled study.

Previous research in the area of pseudoseizures has focused upon their phenomenology and the characteristics of the individual with pseudoseizures. This study set out to examine the role of pseudoseizure behaviour in fulfilling a function within the family context. Pseudoseizure patients, patients with epilepsy and healthy controls completed questionnaires measuring the following variables--anxiety and depression, locus of control, self-esteem, family characteristics and perceived seizure severity. People with pseudoseizures perceived their families as displaying less commitment and support to each other (family Cohesion scale) and less emphasis on ethical issues and values (family Moral-religious scale) than both the epilepsy and the control groups. People with pseudoseizures reported levels of family interest in political, social and recreational activities (family Intellectual-cultural scale) similar to people with epilepsy, both these group scores being lower than the control group. The two patient groups also reported higher depression scores than controls, yet only the epilepsy group had lower self-esteem than the controls. These initial findings support a role of family involvement in therapy for people with pseudoseizures and may lead to a better understanding of the aetiology of pseudoseizures, as well as clarifying characteristics which may well aid the differentiation of pseudoseizures from epilepsy.

Effects of different antihypertensive treatments on morphologic progression of diabetic nephropathy in uninephrectomized dogs.

We previously reported the renal hemodynamic effects of different antihypertensive regimens in uninephrectomized, alloxan-induced, diabetic (DM) beagle dogs following one year of treatment. Dogs were prospectively randomized to one of five groups (N = 26): nondiabetic controls, Group I; dogs with DM on no antihypertensive drugs, Group II; dogs on a converting enzyme inhibitor, lisinopril (L), Group III; dogs on a calcium antagonist, TA3090 (diltiazem-like), Group IV; and dogs on a combination of each drug, in reduced doses, Group V. The current paper extends our previous studies by describing the morphologic changes that occurred within each group of dogs studied. More than 100 glomeruli from the renal cortex of each dog were evaluated for increases in mesangial volume fraction (Vv), glomerulosclerosis (GS) and ateriolar hyalinosis. The interstitium was also evaluated for associated changes. Increases in Vv were attenuated in all treated groups (0.28 +/- 0.04, DM alone versus 0.16 +/- 0.05 L; 0.21 +/- 0.07, TA-3090; 0.19 +/- 0.06 micron 2/micron 2, L+TA 3090; P < 0.05) compared to untreated DM. An attenuated increase in Vv also correlated with a blunted rise in proteinuria in Groups III (r = 0.79) and V (r = 0.81) but not Group IV (r = 0.29). Development of focal GS was blunted in all treated groups; however, global GS was fourfold greater in Group IV compared to untreated DM. The degree of interstitial fibrosis also correlated with the degree of global GS. These data support the concept that both a converting enzyme inhibitor and heart rate lowering calcium antagonist attenuate morphologic progression of diabetic renal disease.(ABSTRACT TRUNCATED AT 250 WORDS)

Selection of cDNAs using chromosome-specific genomic clones: application to human chromosome 13.

We have developed a general method for en masse isolation of cDNAs present in a normalized library by hybridization to arrayed chromosome-specific phage lambda clones; we have used this approach to initiate exon-mapping of human chromosome 13. An advantage of the simultaneous isolation of cDNA/lambda pairs is that it allows cytogenetic assignment of a bona fide genomic clone by in situ hybridization, which also verifies that the corresponding cDNA or a homologous expressed sequence resides on chromosome 13. This information is enriched by partial sequencing of a selected cDNA from both ends. The sequence of the 3' noncoding region provides an 'identifier' that is used to develop STSs, while the sequence from the 5' end, often corresponding to a coding region, is used for homology searches in databases that occasionally reveal gene functions.

Waterloo Vision and Mobility Study: gait adaptations to altered surfaces in individuals with age-related maculopathy.

Walking is an extremely complex task that can become seriously challenged if one of the sensory systems which provides input to the motor system is compromised. The present study evaluated gait adaptations to altered surface characteristics and high and low ambient light conditions by subjects with age-related maculopathy (ARM). Twenty subjects with ARM and 20 control subjects walked along a 6 m path, along which they met 1 of 3 altered surfaces (compliant, uneven, or shiny). Kinematic data and ground reaction forces information were analyzed to discern gait adaptation strategies used by the ARM group. Ten trials on each surface were collected under both high and low ambient light levels. The ARM subjects were found to be generally more cautious when walking on the altered surfaces. For example, they walked more slowly, with a longer swing time. However, gait adaptations in the ARM group were not merely scaled versions of normal gait but were adjustments to adapt to environmental changes. Gait was modified to avoid tripping over a surface edge, to prevent slipping at heel contact, and to balance during stance. These adaptations enabled subjects to maintain safe mobility when walking in a challenging environment.

Satisfaction with care in labor and birth: a survey of 790 Australian women.

Data on satisfaction with care in labor and birth were gathered in a survey conducted in conjunction with a review of maternity services in Victoria, Australia. All women who gave birth in one week in 1989 (> 1000) were mailed questionnaires eight to nine months after the birth, with a response rate of 790 (71.4%). When adjusted for parity in a logistic regression model, the following factors were highly related to dissatisfaction with intrapartum care: lack of involvement in decision making (p < 0.001), insufficient information (p < 0.001), a higher score for obstetric intervention (p = 0.015), and perception that caregivers were unhelpful (p = 0.04). No association was found between satisfaction and maternal age, marital status, total family income, country of birth, or health insurance status. The survey results were influential in shaping final recommendations of the Ministerial Review of Birthing Services by countering stereotypes about women who become dissatisfied with their care, providing evidence of far greater dissatisfaction with intrapartum than antenatal care, and demonstrating the importance of information, participation in decision making, and relationships with caregivers to women's overall satisfaction with intrapartum care.

Comparison of IgG subclasses in foetal serum, maternal serum at delivery and milk in IgA-deficient and control women.

Immunoglobulin G subclass concentrations were measured in paired foetal (cord) and maternal serum specimens at delivery from 27 IgA-deficient (serum IgA < 0.01 g/l) and 15 control women. IgA-deficient women had significantly higher serum IgG1 and IgG3 concentrations than control women but 2 of the group had concomitant IgG2/IgG4 deficiency and a further 12 had low IgG4 concentrations (serum IgG4 < 0.025 g/l). Foetal serum also had significantly higher IgG1 concentrations than control foetal serum but lower IgG2 and IgG4 levels. Concentrations of IgG subclasses and IgM were measured in breast milk collected on the fifth day postpartum from 19 of these IgA-deficient and 18 control women. Between-group differences in IgG subclass levels resembled those in serum. Compared with serum, proportionally less IgG3 was present in milk in both groups although the contribution of IgG3 to total IgG was not less than that of IgG4. Slightly higher IgM was found in milk from the IgA-deficient mothers.

Birth events, birth experiences and social differences in postnatal depression.

A postal survey of all women who gave birth in Victoria in one week in 1989 was used to assess the contribution of birth events, satisfaction with care and social differences to depression after birth. The questionnaire was mailed to women eight to nine months after birth. The survey response was 71.4 per cent (790/1107) with underrepresentation of young women, single women and women of non-English-speaking background. Assessment of depression was made using the Edinburgh Postnatal Depression Scale, with a score of 13 or more indicating probable clinical depression. The point prevalence of depression was 15.4 per cent (95 per cent confidence interval 12.8 to 18.0). In the final logistic model two factors were associated with lower odds of depression: nonmetropolitan residence and maternal age over 34. Factors associated with increased odds of depression in the model were having a first child over the age of 34, assisted delivery (caesarean, forceps, vacuum extraction), bottle feeding, dissatisfaction with antenatal care, having unwanted people present at the birth and lacking confidence to look after the baby at the time of leaving hospital. When those factors were taken into account, being unmarried, being born overseas of non-English-speaking background, being dissatisfied with care in labour and after birth, and not having an active say in decision making, all of which were significantly associated with depression in univariate analyses, did not make any additional contribution to the model.

Antimicrobial susceptibility of pathogenic Yersinia enterocolitica isolated in Canada from 1972 to 1990.

Yersinia enterocolitica has emerged as an enteropathogen associated with several types of human infections that often require antimicrobial therapy, but little is known about the antimicrobial susceptibilities of pathogenic strains isolated from humans in Canada. To determine the present patterns of antimicrobial susceptibility, to identify changes in these patterns that occurred during the past two decades, and to investigate the relationships between O serotypes and patterns of susceptibility, we tested a total of 1,105 pathogenic Y. enterocolitica strains isolated during 1972 to 1976, 1980, 1985, and 1990 for their susceptibilities to 22 antimicrobial agents. Susceptibility testing was performed by using a single breakpoint concentration in agar procedure. The results showed that all strains were susceptible to ciprofloxacin and piperacillin, and 98% or more of the strains from each period were susceptible to trimethoprim, sulfamethoxazole, cotrimoxazole, tetracycline, chloramphenicol, cefamandole, cefotaxime, aztreonam, and four aminoglycosides. In contrast, all strains were nonsusceptible to erythromycin, furazolidone, and clindamycin and 90% or more of the strains from each period were nonsusceptible to ampicillin, carbenicillin, ticarcillin, and cephalothin. Strains belonging to serotypes O:3, O:5,27, and O:8 had different patterns of susceptibility to ampicillin, carbenicillin, ticarcillin, and amoxicillin-clavulanic acid. No major difference in susceptibility was observed between any of the groups of human or animal strains included in the study, but nonsusceptibility to tetracycline increased from 0.4% in 1985 to 2% in 1990 in human strains isolated in those years. Our results indicate that between 1972 and 1990 there was no marked decrease in susceptibility to agents commonly used for therapy among pathogenic Y. enterocolitica strains isolated in Canada.

Effect of 4.5S RNA depletion on Escherichia coli protein synthesis and secretion.

We examined the synthesis of individual proteins following depletion of 4.5S RNA by using a strain deficient in the induction of heat shock proteins. We found that initially the synthesis of all proteins was equally affected, and the peptide elongation rate was reduced by approximately 10%. For up to 1 generation time after the onset of inhibition of total protein synthesis, the processing of secreted proteins was unaffected. After further depletion of 4.5S RNA, accumulation of precursors of secreted proteins was observed under some growth conditions.
