Characterisation of cell-wall-derived polypeptide antigens from different species of Mycobacterium.

Cell walls from different species of Mycobacterium were purified on a sucrose step gradient. The components derived from these preparations were characterised by sodium dodecyl sulphate-polyacrylamide gel electrophoresis, followed by staining or by Western blotting. Surface-exposed polypeptide molecules were also identified by biotinylation. Many protein and glycoprotein molecules were identified in the cell walls. Some of these molecules were immunogenic in man and experimental animals and showed wide variability from species to species. The data suggest that these molecules could be of significance in the diagnosis and pathophysiology of mycobacterial diseases.

Characterization of a monoclonal antibody that selectively recognizes a subset of Entamoeba histolytica isolates.

Monoclonal antibody 2D7.10 recognized an antigen present in seven of nine isolates of axenically cultured Entamoeba histolytica and absent in all other Entamoeba isolates studied. The antigen was absent in two isolates: 200:NIH and Rahman. All nine isolates belonged to pathogenic zymodeme II. Western blot (immunoblot) analysis and treatment with periodate and the proteolytic enzyme trypsin suggest that the antigen recognized by 2D7.10 is a carbohydrate moiety.

Impairment of the regulation of gonadal function in Channa punctatus by metacid-50 and carbaryl under laboratory and field conditions.

Regulation of gonadal function by gonadotropic hormone (GtH) and gonadotropin-releasing hormone (GnRH) in Channa punctatus was significantly affected by nonlethal levels of Metacid-50 and Carbaryl. Under laboratory conditions, the time-dependent decrease in serum GtH level was higher in Carbaryl-treated fish than in Metacid-50-treated fish. The situation was reversed in the field, with a higher inhibitory effect of Metacid-50 being recorded. On the other hand, pituitary GtH content and GnRH activity were inhibited to a greater extent by Metacid-50 than by Carbaryl under both field and laboratory conditions. The present findings highlight that even low doses of Metacid-50 and Carbaryl are effective enough to cause reproductive damage, as evidenced by homeostatic unbalance of the reproductive regulatory system.

Lovastatin-acetaminophen subchronic toxicity in mice.

A subchronic, toxicity study of lovastatin-acetaminophen combination in s/w mice is reported. Oral administration of lovastatin in corn oil 30 mg/kg three times a week and acetaminophen 0.75% w/v in drinking water for 13 weeks resulted in significant increase in SGPT (ALT) in both male and female mice. Histopathology of the liver revealed centrilobular hepatocyte swelling. These results provide evidence of hepatic injury in s/w mice when concomitantly exposed to lovastatin and acetaminophen.

Localization of the microsatellite probe DXS426 between DXS7 and DXS255 on Xp and linkage to X-linked retinitis pigmentosa.

The microsatellite marker DXS426 maps to the interval Xp21.1-Xp11.21, the chromosomal region which contains two loci for X-linked retinitis pigmentosa (XLRP; RP2 and RP3). We have refined the localization of DXS426 both physically, by mapping it to a deletion which spans the interval Xp21.3-Xp11.23, and genetically, by studying multiply informative crossovers which indicate that DXS426 lies between DXS7 and DXS255 (i.e., Xp11.4-Xp11.22). As this is the region which contains the RP2 gene, RP2 families could be identified on the basis of linkage of XLRP to DXS426. Multiply informative crossovers in two RP2 families indicate that the most likely location of the RP2 gene is between DXS426 and DXS7. DXS426 is therefore an important highly informative marker for the purposes of carrier detection and early diagnosis of RP2 and for the localization of the disease gene.

Biological significance of [14C]phenol accumulation in different organs of a murrel, Channa punctatus, and the common carp, Cyprinus carpio.

Phenol, a ubiquitous component of industrial effluents, is a common pollutant of water resources and a serious threat to fish. The present work demonstrates that a significant amount of phenol is retained by various tissues of the common carp, Cyprinus carpio, and the snake-headed murrel, Channa punctatus. The rate of [14C]phenol accumulation was higher carp than in the murrel. It is suggested that retention of phenol in the brain and ovary may seriously affect the reproductive potential of the fish.

C-reactive protein (CRP) in haemolymph of a mollusc, Achatina fulica Bowdich.

C-reactive protein (CRP) was found to be one of the major components of the haemolymph of A. fulica with a normal level of approximately 2 mg CRP/ml haemolymph. Immunological cross-reactivity was found between CRP from A. fulica, a mollusc and Limulus polyphemus, an arthropod. The present finding demonstrates the evolutionary significance of C-reactive protein.

Possible role of C-reactive protein in detoxication of mercury.

In mercury-treated rats, C-reactive protein (CRP) has been found to be synthesized in the liver which, in turn, sequesters mercury resulting in the denaturation of the protein into subunits. The subunits retain the mercury and are released into the serum from where it gets excreted. Native CRP and CRP-subunits have been found to have different antigenicity. Thus one of the physiological roles of CRP seems to be detoxication of heavy metals.

Purification of a metallothionein-like protein from serum of mercury-treated rats using phosphorylcholine-Sepharose affinity column.

A metallothionein-like protein (MLP) from the serum of mercury-treated rats was isolated while purifying an acute phase plasma protein, C-reactive protein, by its Ca2(+)-dependent affinity to phosphorylcholine (PC)-Sepharose column. The MLP was further purified by a single DEAE-Sephacel ion-exchange chromatography. The MLP is similar to mammalian hepatic Zn-metallothionein on the basis of its low molecular weight of approximately 7000, 7 g Zn atoms/molecule of MLP and an absorption maximum at 220 nm. The purity of the protein was confirmed by double immunodiffusion test against anti-MLP antiserum raised in a rabbit. Immunologically MLP was detected not only in the hepatic cytosol, serum and urine from Hg-treated rats but also in the serum and hepatic cytosol of untreated rats. Using PC-Sepharose column, MLP could not be purified from normal serum.

Pattern of acute myocardial infarction in a district hospital in Brunei Darussalam--a pilot study.

The objective of the authors was to study the pattern of presentation, risk factors, and natural course of acute myocardial infarction in the general population of Belait District in Brunei Darussalam. A prospective study was done of 100 consecutive cases of acute myocardial infarction admitted to the coronary care beds of a District General Hospital. The patients were followed up to 12 weeks after admission to hospital. There were three times more males than females (75 males, 25 females). Nine cases out of 75 males were below the age of 40 years, 3 being below 30 years. The mean age of the male denominator was 57.4 years while that of the female counterpart was 67.48 years showing a mean difference of 10.08 years. There were significant association with hypertension (31%), smoking (30%) and diabetes mellitus (27%). The majority of the patients had prodromal symptoms, the most common presenting symptoms were chest pain (63%) and shortness of breath (27%). Only 4% of the patients had silent infarction. Acute myocardial infarction is common in Brunei forming 2.6% of all the patients admitted to the medical wards and the relative rate is 3 times higher in males than in females. There is significant association of IHD with 3 main risk factors namely hypertension, smoking and diabetes mellitus.
