Reflexes of the external urethral sphincter in children.

The electromyographic responses in the external urethral sphincter to electrical stimulation of the pudendal nerve and stretch of the external sphincter were analyzed in a pediatric population; the population included a group with recurrent urinary tract infections and a group with complete or almost complete upper spinal cord lesions. Electrical stimulation produced a response of reflex nature mediated in the sacral spinal cord, frequently beginning with two components that could be individually characterized and that behaved similarly to a flexor reflex. The response produced by stretch appeared to be similar to that produced by electrical stimulation.

Asymptomatic scrotal masses in children.

The ultrasound scans of 16 boys with asymptomatic scrotal masses were retrospectively studied. Ultrasound localized seven of the masses as primary testicular lesions and nine as extratesticular lesions, six of which were hydroceles. Ultrasonography also characterized the scrotal masses as predominantly cystic or solid. Ultrasound could not differentiate benign from malignant disease when a solid mass was found. When a hydrocele was identified, ultrasound was able to image the underlying testis and accurately evaluate it for primary disease. These ultrasound observations can help the surgeon decide when to explore the scrotum and whether to approach the testes via an inguinal or scrotal incision. Conversely, ultrasonographic confirmation of a normal testis may prevent needless surgery and the removal of a normal testis.

[Thermitase, a thermostable serine protease of Thermoactinomyces vulgaris: interaction of the active center and the SH-group of the enzyme].

Modification of the serine and histidine residue in the active centre of thermitase with diisopropylfluorophosphate (DFP) or L-1-tosylamide-2-phenylethyl chloromethylketon (TPCK), and of the only SH-group of the enzyme, with Hg-compounds causes an activity loss against hydrolysis of 4-nitrophenylacetate. While the modification of cysteine prevents reaction of serine and histidine in the active centre of the enzyme with DFP and TPCK, respectively, the Hg2+- and CF3Hg+-binding to the SH-group after modification of essential amino acid residues in the active centre is retained. To elucidate the interaction of the SH-group with the active centre, the modified products of thermitase were investigated for their thermostability. Ca2+-ions were found to have a stabilizing effect on all the modified products of thermitase, as well as on the native enzyme. Simultaneous modification of the cysteine and serine leads to an increase in thermostability of thermitase, whilst double modification at the cysteine and histidine causes destabilization of the enzyme.

Preparation of mutants of Trichoderma viride with increased production of cellulase.

Four mutant strains exhibiting increased production of cullulases were prepared by UV irradiation of conidia of Trichoderma viride QM 9414. Selected mutants were tested for production of cellulases in submerged cultivations in shake flasks and in a 30-L fermentor in a synthetic medium containing 1 % microcrystaline cellulose as the carbon source. Some mutants showed considerable morphological differences when compared to the parent strain, the most noticeable being a higher degree of branching of the mutant hyphae. The branched mutants produced 2 to 3 times higher levels of beta-glucosidase than the parent strain QM 9414.

Growth of Aureobasidium pullulans on waste water hemicelluloses.

Strain Aureobasidium pullulans capable of utilizing hemicelluloses and xylan was cultivated on processed waste dialysis liquor from the production of viscose fibres, containing about 1.5% hemocelluloses. Basic conditions of biomass production were tested on a laboratory scale. The dialysis waste liquor adjusted with mineral acids to pH 4--5 and supplemented with 0.05% yeast autolyzate and 0.2% ammonium sulphate affords protein yields of about 0.8 g/l, corresponding to 4.0--4.5 g dry biomass. Biomass is isolated together with residual water-insoluble hemicelluloses which are not utilized by the microorganism. The total utilization of hemicelluloses attains about 70%.

Xylan-degrading activity in yeasts: growth on xylose, xylan and hemicelluloses.

The ability to grow in liquid media with D-xylose, xylan from decidous trees, and hemicelluloses from conifers was tested in 95 strains of 35 genera of yeasts and yeast-like organisms. Of 54 strains thriving on xylose, only 13 (genera Aureobasidium, Cryptococcus and Trichosporon) utilized xylan and hemicelluloses as growth substrates. The árowth media of these strains were found to contain xylan-degrading enzymes splitting the substrate to xylose and a mixture of xylose oligosaccharides. The ability of these yeasts to utilize the wood components (hitherto unknown in the genus Crytococcus) makes them potential producers of microbial proteins from industrial wood wastes containing xylose oligosaccharides, xylan, and hemicelluloses as the major saccharide components without previous saccharification.

Arterial hypoxemia following the administration of sublingual nitroglycerin.

The effect of nitroglycerin on arterial blood gases and cardiovascular hemodynamics were studied in patients with coronary artery disease. In 13 premedicated patients blood gases and cardiovascular hemodynamics were studied before and 10 minutes after sublingual nitroglycerin (0.6 mg). In eight unpremedicated patients only blood gases were determined before and 10 minutes after sublingual nitroglycerin 0.6 mg. All studies were performed before induction of anesthesia with the patients in the supine position breathing room air. In both groups, arterial PO2 decreased significantly (p less than 0.001); pH and pCO2 did not change. In the 13 patients on which hemodynamic studies were performed, the mean arterial pressure (p less than 0.001), cardiac index (p less than 0.001), central venous pressure (p less than 0.001), pulmonary artery (p less than 0.001) and pulmonary artery wedge pressure (p less than 0.001) decreased. Calculated values for systemic and pulmonary vascular resistance were not significantly altered (p greater than 0.4). This study gives conclusive evidence that nitroglycerin reduces arterial PO2 in most patients with coronary artery disease breathing room air in the supine position. The possible mechanisms and clinical implications are discussed.

[Analysis of the biological effect of city smog extract. III. Comparative investigations on the effect of city smog extracts on cell replication and DNA-synthesis of kidney cells in vitro from the primate Cercopithecus aethiops (author's transl)].

We analysed the cytotoxic effect of city smog extracts from a heavy industrialized area using kidney cell cultures from the primate Cercopithecus aethiops.--Using logarithmically growing cell cultures we determined cell replication and the rate of DNA-synthesis after incorporation of 3H-thymidine.--In presence of city smog extracts we found a dose dependent reduction of cell replication and of DNA-synthesis. In presence of high concentrations (BP-aquivalent 0.25-0.5 microgram/ml) of city smog extract we found no increase in cell number over a period of 72 h. Under the same conditions hardly any DNA-synthesis was detected.--In presence of middle and low concentrations of city smog extract a dose- and time-dependent increase in cell number and rate of DNA-synthesis was detected.

Inhibition of pancreatic ribonuclease by 2'-5' and 3'-5' oligonucleotides.

Forty different oligonucleotides were investigated as possible inhibitors of the depolymerizing activity of RNase A. The strongest inhibitors among the diribonucleoside 2'-5' mono- phosphates were: G2'-5'G, C2'-5'G and U2'-5'G, and among the diribonucleoside 3'-5' monophosphates: ApU, ApC and GpU. Of the eight trinucleotides investigated, ApApUp, ApApCp and ApGpUp were the strongest inhibitors. All four dinucleotides studied (ApUp, ApCp, GpUp and GpCp) were very strong inhibitors, ApUp being the strongest one. The results show that the nature of the various bases in the oligonucleotide has an effect on the degree of inhibition, and that the 3' phosphomonoester group increases the binding of the oligonucleotide to RNase A. These inhibitors can be used in physicochemical and biochemical studies of ribonuclease.
