Locations and nucleotide sequences of three major class III promoters for bacteriophage T3 RNA polymerase on T3 DNA.

The DNA sequences of three major class III T3 RNA polymerase promoters located at 45.0, 55.0, and 64.8% on the standard T3 genetic map have been determined. The precise RNA initiation sites were also determined by 5'-terminal RNA sequence analysis of the transcripts synthesized from the promoter-containing DNA fragments. Alignment of these three class III promoters and a previously determined T3 RNA polymerase promoter at 1.05% on T3 genetic map, with start points of transcription (+1) in register, indicates a high degree of sequence conservation among the four T3 RNA polymerase promoters. The sequences are identical between positions -12 and +4 and are uniformly A-T between -12 and -17. The conserved portion of the (-) strand sequence is 5' (sequence in text) Upstream from -17 and downstream from +4 the sequences diverge. Comparison of this sequence with a prototype 23-base pair promoter sequence for T7 RNA polymerase shows overall homology between positions -17 and +4 with conserved divergence at residues -2 and between -10 and -12. Furthermore, careful examination of the nucleotide sequences around 45.0 and 64.8 T3 map units shows that the putative RNA sequences arising from these regions by overlapping transcription from upstream promoters can be arranged into stable stemloop structures thought to be required for RNase III cleavage. This pattern is similar to those reported for the corresponding T7 RNA polymerase promoters on T7 DNA (Dunn. J. J., and Studier, F. W. (1983) J. Mol. Biol. 166, 477-535).

Biosynthesis in vitro of gangliosides containing Gg- and Lc-cores.

On the basis of our previous and present studies with embryonic chicken brain system, we have proposed stepwise biosynthesis of GD1a (Gg-series) and LD1 (Lc-series) gangliosides, starting from ceramide (Fig. 4). At least three different galactosyltransferases GalT-2 (UDP-Gal:Glc-Cer), GalT-3(UDP-Gal:GM2) and GalT-4(UDP-Gal:LcOse3-Cer) and three different sialyltransferases SAT-1(CMP-NeuAc:Lac-Cer), SAT-2(CMP-NeuAc:GM3) and SAT-3(CMP-NeuAc:nLcOse4 Cer) are involved in the biosynthesis in vitro of these gangliosides. All six of these glycosyltransferases have been solubilized using nonionic detergents. Two forms of glycolipid:galactosyltransferases (GalT-3 and GalT-4) have been separated by DEAE sepharose CL-6B chromatography from solubilized supernatant of 11- to 13-day-old embryonic chicken brain. Using microisoelectric focusing (pH gradient 3 to 8) the galactosyltransferases (GalT-3 and GalT-4) have been separated from SAT-3. Two beta-N-acetylglucosaminyltransferases (GlcNAcT-2(UDP-GlcNAc:nLcOse4Cer(beta 1-3] and GlcNAcT-3(UDP-GlcNAc:nLcOse4Cer(beta 1-6] have also been solubilized from mouse T-lymphoma, P-1798, using Triton CF-54. These enzymes are involved in the synthesis of Ii-core gangliosides and 3H-products have been characterized by methylation studies. Further separation of these two GlcNAcT's are in progress.

Biological characteristics of Salmonella weltevreden typing phages.

The important biological characteristics of Salmonella weltevreden (3, 10 : r :Z6) typing phages were studied. On the basis of these, the phages could be classified into three groups: phages phi I and phi II, phages phi III, phi IV and phi VI, and phage phi V.

Renal excretion of prostaglandin metabolites, arginine vasopressin, and sodium during endotoxin and endogenous pyrogen induced fever in the goat.

Responses to intravenous injections of an endotoxin (E. coli-lipopolysaccharide, 1 microgram/kg b.wt.) and endogenous pyrogen were studied in euhydrated and hyperhydrated goats. The biphasic febrile response to the endotoxin was associated with a pronounced increase in the renal excretion of measured prostaglandin (PG) metabolites (11-ketotetranor PGF metabolites). This increase was time-correlated with the elevation of the rectal temperature, and (in hyperhydrated animals) with an inhibition of the water diuresis and an increase in renal excretion of arginine vasopressin (AVP). Other effects of the endotoxin were an immediate depression of renal Na and K excretion followed by the development of pronounced natriuresis, and a reduction of plasma Fe and Zn concentrations. The appearance of the febrile reactions (peripheral vasoconstriction and shivering) was accompanied by miosis. The maximum elevation of the rectal temperature was significantly greater during euhydration than during hyperhydration. Also endogenous pyrogen elicited miosis concomitant with febrile reactions, and an elevation of the renal excretion of PG metabolites which was closely correlated in time with the monophasic febrile response, and (during hyperhydration) with temporary inhibition of the water diuresis and an increase in the renal AVP excretion. However, the responses were much weaker than the corresponding endotoxin effects. No appreciable changes in renal excretion of Na and K were observed in response to the endogenous pyrogen. It is concluded that the observed effects on renal cation excretion were manifestations of direct endotoxin influences on kidney function.(ABSTRACT TRUNCATED AT 250 WORDS)

Suicide reconstruction by glue-lift of gunshot residue.

Based upon the recently developed glue-lift collection of gunshot residue particles for examination in the scanning electron microscope with energy dispersive X-rays, this laboratory has undertaken a research program to determine if reconstruction of gunshot deaths is feasible. Because undisturbed conditions of suicide victims may help in securing unambiguous results and high success rates, the program has been carried out to reconstruct suicides only. Data obtained from 13 firearms suicides and their laboratory reconstruction which involved primarily shotguns and handguns, indicate that reconstruction can be immensely useful to interpretation of the gunshot residue distributions on a suspect's or victim's hands. This reports outlines the basic experiments performed to relate the residue emission from the gun to the deposits found on the firing hand(s) and a description of the reconstruction technique that uses a target to simulate a human body. The technique can determine the specifics of a victim's hand position at the time of firing and which hand was used to pull the trigger. Because the technique is sensitive to the nature of the grasp of the firing hand and of the supporting hand, in the case of a shotgun, it has been possible in all cases to date, to determine which one of the potential eight hand positions existed at the time of shooting.

Immunochemical studies on Shigella dysenteriae type 9 bacterial polysaccharide.

On graded hydrolysis and Smith degradation, the O-somatic polysaccharide isolated from Shigella dysenteriae type 9 bacteria, strain NCTC 7919, yielded five oligosaccharides which were characterized. The positions of the O-acetyl and pyruvic acetal groups in the repeating unit were identified. Immunochemical studies indicated that D-galactose is the immunodominant sugar in the polysaccharide, and one of the oligomers, having the structure Gal-(1 leads to 3)-GlcNAc-(1 leads to 3)-Gal-(1 leads to 4)-Man, showed maximum inhibition of the homologous precipitation.

Plaque morphology and antigenic relationship of the Salmonella weltevreden typing phages.

The plaque morphology and antigenic relationship of the six typing phages of Salmonella weltevreden were studied. Under identical conditions of plating, the phages could be classified into three groups based on plaque morphology. Neutralization tests with anti-phage sera showed that typing phages phi I and phi II were antigenically similar. Phages phi III, phi IV and phi VI also showed antigenic similarity. Typing phage phi V was antigenically distinct from all other phages. Thus the phages could be classified into three groups on the basis of both plaque morphology on their respective indicator strains and velocities of neutralization by homologous/heterologous anti-sera.

Current trends in the diagnosis and treatment of carcinoma of the pancreas.

Causes of diagnostic delay and survival figures were compared in three groups of patients (total 191 patients) with ductal adenocarcinoma of the pancreas between the years 1951 and 1960 and 1971 and 1980. The main causes of delay between onset of symptoms and definitive treatment were patient delay in seeking medical advice and delay by the initial doctor in referring the patient. Recent direct imaging techniques of ultrasound, computerized axial tomographic scanning, endoscopic retrograde cholangiopancreatography, and percutaneous transhepatic cholangiography did not significantly decrease this diagnostic delay. Operative mortality in the groups treated by pancreatoduodenectomy between 1951 and 1960 and 1971 and 1980 were 13.9 and 3.4 percent, respectively. The mean survival rates for these groups were 14 and 20.4 months, respectively. Patients with unresectable carcinoma of the pancreas between 1971 and 1980 had an operative mortality rate of 10 percent and a mean survival of 6.7 months. Various aspects of symptomatology and operative findings in patients with carcinoma of the pancreas are emphasized to aid early diagnosis.

Immunochemical studies on a polysaccharide from Shigella dysenteriae type 2.

The polysaccharide isolated from Shigella dysenteriae type 2, strain NCTC 566, on Smith degradation and graded hydrolysis yielded three oligosaccharides which were characterised using methylation studies. Using homologous rabbit antiserum and the monosaccharides that constituted the polysaccharide and the oligosaccharides isolated from it and the cross-reactions in some type-specific pneumococcal antisera, immunochemical specificities of different sugar groupings in the polysaccharide molecule were determined. These results indicated that N-acetylglucosamine was the immunodominant sugar in the polysaccharide and the oligosaccharide isolated from the Smith-degraded product and having the structure (formula; see text) gave maximum inhibition of the specific precipitation.

Surgical treatment of bronchobiliary fistulas.

We have reviewed our experience with thoracobiliary fistulas due to biliary tract disease. Early diagnosis and treatment of subphrenic abscesses and bile duct obstruction will decrease the incidence of these fistulas. Once established, relief of biliary obstruction, subphrenic drainage, excision of the fistulous tract and appropriate supportive measures will decrease the morbidity rate and mortality of this rare but distressing condition. In our series, all of the patients ultimately had relief of the symptoms. There was no mortality.

Biosynthesis in vitro of sialyl(alpha 2-3)neolactotetraosylceramide by a sialyltransferase from embryonic chicken brain.

A sialyltransferase activity present in 7- to 12-day-old embryonic chicken brain catalyzes the transfer of sialic acid from CMP-sialic acid to the terminal galactose residue of [3H]nLcOse4Cer ([3H]Gal(beta 1-4).GlcNAc(beta 1-3)Gal(beta 1-4)Glc-Cer) to form NeuAc(alpha 2-3)-[3H]nLcOse4Cer (LM1 ganglioside). The product is sialidase-labile (96%), and the NeuAc group is linked to O-3 of the terminal galactose residue. The (alpha 2-3) linkage between sialic acid and the terminal galactose was determined on the basis of identification of 2,4,6-tri-O-methyl[3H]galactose obtained after hydrolysis of the permethylated enzymatic product. The CMP-sialic acid:nLcOse4Cer (alpha 2-3)sialyltransferase activity sediments (90%) at the junction of 1.2 M and 1.5 M on a discontinuous sucrose density gradient when still membrane bound (insoluble in 0.2% Triton X-100). The enzyme preparation also catalyzes the transfer of sialic acid from CMP-sialic acid to O-3 of GgOse4Cer (Gal(beta 1-3)GalNAc(beta 1-4)Gal(beta 1-4)Glc-Cer) to form NeuAc (alpha 2-3)GgOse4Cer (GM1b). Substrate inhibition studies indicate that these two reactions are probably catalyzed by the same enzyme.

Biosynthesis in vitro of a blood group B-active fucose-containing hexaglycosylceramide from neolactopentaosylceramide in bovine spleen.

A solubilized alpha-fucosyltransferase activity has been isolated from a bovine spleen Golgi-rich membrane fraction. The enzyme transfers fucose from GDP-beta-L-fucose to a blood group B-active pentaglycosylceramide acceptor (Gal(alpha 1-3)Gal(beta 1-4)GlcNAc(beta 1-3)Gal(beta 1-4)Glc-ceramide) isolated from rabbit erythrocytes. Treatment of the membranes with 0.2% (final concentration) sodium taurodeoxycholate detergent produced maximal recovery (90%) of activity. A cationic detergent, G-3634-A, is required for optimal activity and the enzyme does not require addition of exogenous metal ion for activation. The purified 14C-labeled product of the reaction migrated with human blood group B-active hexaglycosylceramide on Silica Gel G thin layer plates. After treatment with fig alpha-galactosidase, the radioactive pentaglycosylceramide migrated with human H-active glycosphingolipid. The 14C-labeled product inhibited the hemagglutination reaction of B-type erythrocytes with Bandeiraea simplicifolia lectin and anti-B serum and formed a precipitin line with Euonymus europeus lectin. Treatment of the 14C-labeled product with alpha-fucosidase (Venus mercenaria) or weak acid at 100 degrees C for 2 h released 80-90% of the bound radioactive fucose.

Host specificity of Salmonella weltevreden typing phages.

The host range of the six S. weltevreden typing phages was studied on 1469 strains belonging to 37 different Salmonella serotypes. In addition to S. weltevreden, only S. nchanga, S. give, S. lexington and S. anatum, all belonging to O group E, showed varying degrees of susceptibility to the action of some of the typing phages. Typing phage VI lysed only one strain other than S. weltevreden. All serotypes tested other than S. weltevreden were resistant to phages III and IV even at 1000 times the routine test dilution. Thus, typing phages III and IV were specific for S. weltevreden. The sensitivity patterns of S. weltevreden typing phages were not found to bear much correlation with either somatic of flagellar antigens of Salmonellae.

Probable involvement of a glycoconjugate in IMR-32 DNA synthesis: decrease of DNA polymerase alpha 2 activity after tunicamycin treatment.

Multiple forms of DNA polymerase alpha activity (alpha 1, alpha 2, and alpha 3) from human neuroblastoma IMR-32 cells untreated or treated with tunicamycin (3 microgram/ml) were separated by DEAE-cellulose column chromatography. Loss of 40--60% of DNA polymerase alpha 2 activity was observed in tunicamycin-treated cells. Ricin 1B, a subunit of intact ricin (Mr, 64,000), was found to be a specific inhibitor of DNA polymerase alpha 2 isolated from control IMR-32 cells. However, DNA polymerase alpha 2 isolated from tunicamycin-treated cells was insensitive to ricin 1B. Heat treatment studies at 50 degrees C showed two completely different inactivation profiles for the DNA polymerase alpha 2 enzymes isolated from the tunicamycin-treated and untreated cells. A probable involvement of a beta-linked galactose-containing carbohydrate chain in the catalytic subunit of DNA polymerase alpha 2 is suggested.
