[Physiologic studies of maturity diagnosis in the calf and its relationship to the postnatal condition].

The parameters of kidney, lung, and skin maturity, known from human medicine, were for the first time determined from amniotic fluid of cattle. Given the difference in placentation, it is not known whether the conditions of the human fetus can be fully applied to the bovine fetus, the detectability of such maturity criteria may be indicative of comparable morphokinetic conditions in the bovine fetus. All physiological parameters investigated exhibited difference to the benefit of surviving calves. Positive correlations were found to exist between the lecithin/sphingomyelin quotient and body weight gain in the first week of age as well as drinking behaviour of the calves. Hence, insufficient concentration of surface-active phospholipids in calf breeding has measurable effects on quantitatively definable characteristics.

Histopathologic evaluation of application of labeled neoglycoproteins in primary bronchus carcinoma.

Neoglycoproteins are readily available conjugates of a histochemically inert carrier protein and histochemically crucial carbohydrate moieties which are covalently attached to the carrier protein by chemical synthesis. Biotinylation renders these conjugates detectable in formalin-fixed, paraffin-embedded tissue sections of human lung cancer by standard staining protocols, thereby localizing endogenous receptors for carbohydrate moieties. Examination of 30 cases of main types of human lung cancer revealed the presence of alpha-fucosyl-, alpha-mannosyl-, and alpha-glucosyl-specific receptors in adenocarcinomas or epidermoid carcinomas with high positivity rates. The extent of the expression of receptors for alpha- and beta-galactosides appeared to be comparatively lower. Within the standard protocol, using a concentration of the biotinylated probes of 10 micrograms/mL, this panel of probes consistently failed to detect endogenous sugar receptors in ten cases of small cell anaplastic carcinoma of the lung. Whereas none of the sections from the tumor cases bound the sulfated fucan fucoidan, the accompanying inflammatory cells, especially the granulocytes, expressed receptors for the sulfated fucan. Pronounced labeling for macrophages was observed for the alpha-galactoside-specific probe, whereas no binding to inflammatory cells and pneumocytes was detectable for the beta-galactoside-specific probe. The results indicate that expression of endogenous receptors for neoglycoproteins may be useful in discriminating between small cell and non-small cell lung carcinoma and carcinomatous cells from accompanying inflammatory cells.

DNA-content, inflammatory tissue response and tumour size in human lung carcinoma.

Tumour imprints of 75 resection specimens with human lung carcinoma (lobe and lungs) were fixed with alcohol and Feulgen-stained. Resection specimens were cut into serial sections 6 mm thick and tumour mass, tumour involvement into intrapulmonary and extrapulmonary lymph nodes, detailed pTN-stage were determined. DNA-content, DNA-index, percentage of diploid/hyperploid tumour cells, and morphometric nuclear features were measured using an automated image analyzing system (VISIAC). Only 10% of the measured carcinomas were diploid. The DNA-index showed the broadest variance in large cell anaplastic carcinoma (1.2-3.3). Carcinomas growing predominantly within the alveolar space, i.e. without destroying the interstitial tissue showed a lower DNA-content above 3c and above 5c compared to carcinomas destroying the interstitial tissue. Carcinomas with severe stroma reaction were found to have 33%-48% of DNA above the 3c value whereas carcinomas without stromal reaction had a percentage above 3c ranging 46%-64% (confidence limits, p less than 0.05). DNA-index increases with increasing tumour volume and decreases for large tumours (greater than 100 ccm). Hyperploid and polyploid tumours were found more frequently in case of pT2 and pT3-stages compared to pT1-stages. No relation of DNA-content or ploidy was found to lymph node involvement and inflammatory response of host tissue.

Perspectives on self and object.

The normal adult is able both to experience himself and to know himself as an object in the world; he is capable of both subjective and objective self-awareness. Both types of awareness are developmental achievements, and their integration is related to the development of self-constancy and the sense of reality and identify. They are perspectives depending on where the line is drawn between the self observing and the self observed, and they vary in altered states of consciousness. Psychological theories also vary along these dimensions and although psychoanalysis is distinctive in attempting to integrate both viewpoints, the language of the theory and certain technical parameters change as the emphasis changes from defects in objective self-awareness to defects in subjective awareness.

[Experimental use of prostaglandin F2 alpha to synchronize estrus in cattle].

Experiments were conducted for the use of prostaglandin to synchronise oestrus in dairy cows. One single dose of 5 mg F2-Alpha prostaglandin was administered via the intrauterine route. The reference values of the control groups were matched by first insemination following induced oestrus among the best of the test animals which were inseminated twice. These values reached 72 and 96 hours following application accounted for 46.7 and 50.0 per cent. The yellow body phase, a prerequisite for oestrus production by prostaglandins, had been induced in the test animals by cycle blocking, using gestagen. Possible use of the method in a bio-engineering context for large-scale oestrus synchronisation in cows is discussed.

On the nature of the presumed receptor for IgE on mast cells. V. Enhanced binding of 125I-labeled IgE to cell-free particulate fractions in the presence of protease inhibitors.

Cell-free particulate preparations derived from sonicated, purified rat peritoneal mast cells can be radiolabeled with IgE. The IgE concentration-dependence and the kinetics of the binding reaction at pH 6.6 were very similar to those observed with intact mast cells. The number of IgE molecules bound per mast cell equivalent at saturation varied somewhat from one particulate preparation to the next and, on average, was about 50% of the binding capacity of the intact cells. Incubation of intact mast cells or of particulate fractions with low concentrations of IgE in serum-free media resulted in a decrease in the amount of bindable IgE which remained in the supernatants in excess over the amount of IgE which was bound to the cells. Specific binding of IgE to both particles and to intact cells at limiting IgE concentrations was stimulated up to approximately twofold by a variety of antibiotic, synthetic or high molecular weight (protein) protease inhibitors of which soybean trypsin inhibitor and p-nitrophenyl-p'-guanidinobenzoate were the most active. These substances also markedly protected the IgE, leaving more of it bindable to rat basophil leukemic cells in a second incubation. Binding to particulate fractions also occurred at pH 4.8 when bovine serum albumin was added to the incubations. The results are consistent with the view that normal peritoneal mast cells have a membrane-bound protease which has high selectivity for IgE.

[Planned control of reproduction processes in industrialized cattle production].

Reference is made to the authors' own experience and to evaluated literature in an account of possibilities which so far have been explored for planned control of reproduction processes in industrialised cattle production. Synchronisation of oestrus in heifer has been found to be suitable for reproduction control even under conditions of industrialised production. The method has been in use on several farms. Synchronisation of oestrus in cow is an approach which still is in the process of translation into practice. Obtainable advantages, like those in the context of heifer, will include better job conditions. Systematic puerperal monitoring, too, will yield positive effects in controlling post-partum readiness of cow for reproduction. Results so far achieved include progress towards the introduction of a complex reproduction technology in cattle production. Before such complete system of reproduction control can be used, further studies are required into some fundamental aspects of the problem, including calving control, pregnancy examination, and gravidity hagiene.

[Studies on the use of megestrol acetate for estrus synchronization in heifers].

Megestrol acetate has been tested for its applicability to synchronisation of oestrus in 463 mature heifers in the framework of preclinical and clinical experiments. Clinical tests and studies in the context of laboratory diagnostics have shown that 40 mg/die megestrol acetate, administered in two applications with twelve hours in-between and over 15 days, are necessary for maximum synchronisation effect. The resulting oestrus was found to be prolonged, with ovulations taking place within two or three days. Technological requirements in this context are discussed. Megestrol acetate is found to be suitable for oestrus synchronisation in heifer. Tests conducted under industrialised conditions have shown conception rates to be identical with those obtained by administration of chloromadinone acetate.
