The WHI1+ gene of Saccharomyces cerevisiae tethers cell division to cell size and is a cyclin homolog.

WHI1-1 is a dominant mutation that reduces cell volume by allowing cells to commit to division at abnormally small sizes, shortening the G1 phase of the cell cycle. The gene was cloned, and dosage studies indicated that the normal gene activated commitment to division in a dose-dependent manner, and that the mutant gene had a hyperactive but qualitatively similar function. Mild over-expression of the mutant gene eliminated G1 phase, apparently entirely relaxing the normal G1 size control, but revealing hitherto cryptic controls. Sequence analysis showed that the hyperactivity of the mutant was caused by the loss of the C-terminal third of the wild-type protein. This portion of the protein contained PEST regions, which may be signals for protein degradation. The WHI1 protein had sequence similarity to clam cyclin A, to sea urchin cyclin and to Schizosaccharomyces pombe cdc13, a cyclin homolog. Since cyclins are inducers of mitosis, WHI1 may be a direct regulator of commitment to division. A probable accessory function of the WHI1 activator is to assist recovery from alpha factor arrest; WHI1-1 mutant cells could not be permanently arrested by pheromone, consistent with a hyperactivation of division.

Status of calcium influx in cell cycle of S. cerevisiae.

The transport of calcium was assayed in exponentially growing and G1 arrested temperature sensitive cdc mutants of Saccharomyces cerevisiae. There was no statistically significant difference in the rate of Ca2+ influx in cdc 28, cdc 37 and cdc 4 arrested cells, as well as in wild type cells arrested in G1 phase in comparison to exponentially growing cells. There was however a significant increase in Ca2+ uptake in cdc 7 and cdc 24 arrested cells. The former is known to arrest before bud emergence and initiation of DNA synthesis; arrest of the latter affects bud formation while DNA synthesis continues. The results suggest that Ca2+ may have a role in bud formation and growth.

Histopathological changes induced in rat tissues by oral intake of lead acetate.

In the present study an attempt has been made to observe the pathological alterations brought about in the intestine, liver, and kidney of lead-intoxicated rats. A short-term exposure to a sublethal dose of lead (44 mg/kg body wt/day) is seen to cause conspicuous degenerative changes in the three tissues. The intestinal mucosal epithelium is affected which leads to malabsorption, while in the kidney proximal tubular cells degenerate causing secretion of essential materials such as glucose, amino acids, etc., in the urine.

Atracurium infusion in the intensive care unit.

An open study of 38 infusions of atracurium in 33 patients requiring neuromuscular blockade for ventilation was carried out. Consecutive patients requiring ventilation were studied and there were no exclusions. The initial rate of infusion was 0.4 mg kg-1 h-1 and this was adjusted according to clinical observation and the results of neuromuscular stimulation. The average infusion rate in the series was 0.59 mg kg-1 h-1. Patients were sedated using a morphine infusion at a rate of 2-4 mg kg-1 h-1. Satisfactory blockade was achieved in all cases. Time for spontaneous recovery following termination of infusion was recorded, the average recovery time being 39.2 min. Emphasis was placed on recording any possible serious toxic effects attributable to long-term atracurium infusions, possible toxic effects were considered and a satisfactory explanation of such effects was found in all cases.

Contraception in male monkeys by intra-vas deferens injection of a pH lowering polymer.

A new male method of contraception by injecting a polymer, styrene-maleic anhydride (SMA), into the lumen of the vas deferens has been studied on rhesus monkeys. The polymer has the dual feature that it can occlude the vas deferens lumen and also it can inhibit the fertilising ability of spermatozoa by virtue of the pH lowering effect. Matings with females were carried out when the lumen was completely occluded giving azoospermia as well as with partial block and spermatozoa present in the semen. All matings were infertile. Data up to one year is presented. All indications are that the contraceptive effect will last for a considerably long period.

Prenatal and neonatal exposure to cimetidine results in gonadal and sexual dysfunction in adult males.

Exposure of rats to cimetidine during intrauterine life and the immediate neonatal period results in hypoandrogenization in adult life with decreased weights of androgen-dependent tissues and decreased concentrations of testosterone. Moreover, sexual behavior patterns in adult life are disturbed as shown by a lack of sexual motivation and decreased performance.

Comparison of electrical field plethysmography with electrical impedance plethysmography.

As a means for assessing cardiac function, electrical field plethysmography (EFP) has been shown to have some features quite different from electrical impedance plethysmography (EIP). Here the two techniques are compared by using the two systems simultaneously on a subject and also with independent use in different electrode configurations. The results conform with the view that EIP is related primarily to volumetric changes of the aorta, whereas EFP is affected predominantly by changes in cardiac dimensions and orientation. Because of this difference, the standard time differential formula used for EIP is not applicable for the computation of cardiac output from the EFP waveforms. An alternative method of computation based on the amplitude of the EFP waveform is suggested.

Dynamics of the ampullary-isthmic junction in rabbit oviduct.

A delay in the ovum transport at the ampullary-isthmic junction (AIJ) has been confirmed by various investigators, but the mechanism of retention and release at this junction is not yet well understood. Using a technique for monitoring unobstructed Fallopian tube motility with an impedance plethysmograph, an increase in luminal diameter was observed at the time when the ova are released from the AIJ. This enlargement persists till all ova emerge from the AIJ. Hence, it seems likely that the AU relaxation is a dominant factor contributing to ova release from the AIJ.

Copper status of Indian pregnant women of low socio-economic group and its effect on the outcome of pregnancy.

Copper status of 109 women, (36 nonpregnant, 17 in the first trimester, 17 in the second trimester of pregnancy and 39 in the postpartum period) and its effect on the outcome of pregnancy was evaluated by a cross-sectional study. The results documented a low intake of copper--1.2 mg-1.5 mg, by the women and a statistically significant correlation (P less than 0.01) between the intake of copper and the intake Calories and protein. Serum copper levels rose with the progress of pregnancy and the increase was statistically significant (P less than 0.005) in the second trimester. Thereafter, a slight decline was noted. No correlation was found between serum copper and serum total proteins, albumin or globulin in either the pregnant or the nonpregnant women. The correlation between the dietary intake of copper and serum copper concentration was also not significant. Birth weight, length and head circumference of the neonate exhibited no correlation with material dietary or serum copper levels.
