Both di- and triglycerides were synthesized when microsomes isolated from mammary glands of lactating mice were incubated with dihydroxyacetone phosphate (DHAP), 1(-14)C)palmitate, ATP, CoASH, GSH, KF, MgC12, and NADPH. When NADH replaced NADPH, glyceride synthesis was very low. In the absence of either NADPH or NADH, DHAP was acylated to palmityl-DHAP. Since microsomes do not have glycerol 3-phosphate NAD:oxidoreductase activity , we inferred that glycerol 3-phosphate (GP) is not an intermediate in triglyceride biosynthesis from DHAP. This reductase, present in the cytosol, was active only with NADH. With the same concentration of either GP or DHAP, microsomes yielded essentially similar amounts of di- and triglycerides. Mitochondria, while capable of synthesizing palmityl-DHAP, did not produce di- and triglycerides.

The morphology of the outflow apparatus in human eyes which had been treated topically with either 2 or 4% pilocarpine on 4 occasions at 6-hourly intervals before enucleation was compared with an untreated control series of eyes from patients in a similar age range. In the pilocarpine-treated group the scleral spur was pulled posteriorly and internally, so that the angle formed by the scleral spur and the scleral sulcus was significantly greater in the treated than the untreated series. The change in attitude of the scleral spur produced widening of the spaces between the corneoscleral trabeculae and distension of the endothelial meshwork. It was shown from counts of giant vacuoles in the endothelium of Schlemm's canal that the incidence of giant vacuoles in the pilocarpine-treated group was greater than twice that in the controls. It would appear that pilocarpine produces alterations in the configuration of the outflow apparatus which would promote the drainage of aqueous humour.

Mouse mammary epithelial (MME) cell antigens were cell type-specific and were retained, to a large extent, by MME cells after neoplastic transformation. These MME cell antigens were expressed in mammary tumors of BALB/c, C3H, GRS/A, RIII, and Is/Bi mice tested and were not expressed in tumors whose normal counterpart was other than mammary tissue. They were not dependent on cell culture conditions or on the presence of murine mammary tumor virus; therefore, they can be used in an ubiquitous cell type marker for MME cells in both normal and neoplastic tissues.
Liver slices, from mice fasted for one day and then refed for three days either a 15% corn oil diet or a 15% corn oil diet containing eicosa-5,8,11,14-tetraynoic acid (TYA), were incubated with [1-14C] acetate or [3H] H2O to determine lipogenic capacity. Dietary TYA produced a twofold stimulation in fatty acid and cholesterol synthesis. TYA also caused an increase in the relative proportion of linoleate (C18:2) and a decrease in that of arachidonate (C20:4) in liver. Thus, (a) despite high levels of C18:2, hepatic lipogenesis can be increased, and (b) even short term feeding of TYA can alter the hepatic fatty acid composition presumably by inhibition of arachidonate synthesis from linoleate.
Rabbit antibodies against components of the human milk fat globule bind specifically to normal human breast epithelial cells and cell lines derived from breast carcinomas, as well as to the outer surface of the human milk fat globule. Variation in indirect immunofluorescence staining in both intensity per cell and percentage of cells stained is observed for the different brest cell lines. Cells derived from other epithelial and other ectodermal tissues, fetal fibroblasts, cells of the blood buffy coat, and even fibroblasts of the breast itself do not bind the antibodies. This suggests that these antibodies are detecting cell-type-specific antigens. These normal breast epithelial cell antigens are on the cell surface and their expression is stable in long-term cultured cell lines, even after much chromosomal variation in a given line. By affinity chromatography, three distinct antigenic components can be isolated from the milk fat globule, one of which contains carbohydrate. These differentiation antigens of the human breast epithelial cell are not only useful as specific cell-type markers, but also can provide a tool to study the role of the cell surface in normal and neoplastic mammary development.
Female 3-month-old C3H mice were given sc injections of 5-mg pieces of mammary adenocarcinoma and fed a linoleate-containing (15% corn oil) diet in the presence or absence of eicosa-5,8,11,14-tetraynoic acid (TYA), an inhibitor of prostaglandin synthesis. After 6 weeks, the weights of tumors of mice fed the TYA-free linoleate diet were three to five times greater than those of mice fed the TYA-containing linoleate diet. Dietary TYA caused a reduction in the levels of arachidonate and an elevation in the levels of linoleate in mammary tumors and livers. Aspirin, another known inhibitor of prostaglandin synthesis, when added to the linoleate diet, did not affect the tumor size or the composition of fatty acids in the tumors and livers. Thus we concluded that a) the growth of mammary tumors was not related to prostaglandin synthesis but was related to the availability of arachidonate, and b) TYA was an effective inhibitor for the conversion of linoleate to arachidonate.
The outflow apparatus from adult baboon and rabbit eyes was stained with the inorganic dye ruthenium red. The ruthenium reaction product coated the surface of the trabecular meshwork cells and the canalicular endothelial cells. Deposits also impregnated the various connective tissue elements within the trabeculae and the extracellular spaces of the endothelial meshwork. A fine fibrillar network could also be identified with ruthenium red and this was present in the trabecular cores and the extracellular spaces of the endothelial meshwork. It was considered that the fibrillar network may represent a matrix of glycosaminoglycans and glycoproteins. The significance of these materials in relation to aqueous outflow was discussed.
