Development of organometallic ruthenium-arene anticancer drugs that resist hydrolysis.

With a view to develop drugs that could resist hydrolysis in aqueous media, organometallic arene-capped ruthenium(II) 1,3,5-triaza-7-phosphatricyclo[3.3.1.1]decane (RAPTA) complexes bearing chelating carboxylate ligands have been prepared and studied. The new complexes, Ru(eta6-cymene)(PTA)(C2O4) (1) and Ru(eta6-cymene)(PTA)(C6H6O4) (2), were found to be highly soluble and kinetically more stable than their RAPTA precursor that contains two chloride ligands in place of the carboxylate ligands. They were also able to resist hydrolysis in water and exhibited significantly lower pKa values. Importantly, they showed a similar order of activity in inhibiting cancer cell-growth proliferation (as determined by in vitro assays) and exhibited oligonucleotide binding characteristics (as evidenced by matrix-assisted laser desorption ionization mass spectrometry) similar to those of the RAPTA precursor, hence realizing a strategy for developing a new generation of stable and highly water-soluble RAPTA adducts.

Lanthanide triple-stranded helicates: controlling the yield of the heterobimetallic species.

Two unsymmetrical ditopic hexadentate ligands designed for the simultaneous recognition of two different trivalent lanthanide ions have been synthesized, L(AB2) and L(AB3), where A represents a tridentate benzimidazole-pyridine-benzimidazole coordination unit, B2 a diethylamine-substituted benzimidazole-pyridine-carboxamide one, and B3 a chlorine-substituted benzimidazole-pyridine-carboxamide moiety. Under stoichiometric 2:3 (Ln/L) conditions, these ligands self-assemble with lanthanide ions to yield triple-stranded bimetallic helicates. The crystal structures of four helicates with L(AB3) of composition [LnLn'(L(AB3))3](ClO4)6.solv (CeCe, PrPr, PrLu, NdLu) show the metal ions embedded into a helical structure with a pitch of about 13.2-13.4 A. The metal ions lie at a distance of 9.1-9.2 A and are nine-coordinated by the three ligand strands, which are oriented in a HHH (head-head-head) fashion, where all ligand strands are oriented in the same direction. In the presence of a pair of different lanthanide ions in acetonitrile solution, the ligand L(AB3) shows selectivity and gives high yields of heterobimetallic complexes. L(AB2) displays less selectivity, and this is shown to be directly related to the tendency of this ligand to form high yields of HHT (head-head-tail) isomer. A fine-tuning of the HHH left arrow over right arrow HHT equilibrium and of the selectivity for heteropairs of Ln(III) ions is therefore at hand.

Synthesis, characterization, and in vitro evaluation of novel ruthenium(II) eta6-arene imidazole complexes.

Ten complexes of general formula [Ru(eta6-arene)Cl2(L)], [Ru(eta6-arene)Cl(L)2][X], and [Ru(eta6-arene)(L)3][X]2 (eta6-arene = benzene, p-cymene; L = imidazole, benzimidazole, N-methylimidazole, N-butylimidazole, N-vinylimidazole, N-benzoylimidazole; X = Cl, BF4, BPh4) have been prepared and characterized by spectroscopy. The structures of five representative compounds have been established in the solid state by single-crystal X-ray diffraction. All the new compounds were assessed by the same in vitro screening assays applied to [imidazole-H][trans-RuCl4(DMSO)(imidazole)] (NAMI-A) and [Ru(eta6-arene)Cl2(1,3,5-triaza-7-phosphaadamantane)] (RAPTA) compounds. It was found that the new compounds show essentially the same order of cytotoxicity as the RAPTA compounds toward cancer cells. Several of the compounds were selective toward cancer cells in that they were less (or not) cytotoxic toward nontumorigenic cells that are used to model healthy human cells. Thus, two of the compounds, [Ru(eta6-p-cymene)Cl(vinylimid)2][Cl] (vinylimid = N-vinylimidazole) and [Ru(eta6-benzene)(mimid)3][BF4]2 (mimid = N-methylimidazole), have been selected for a more detailed in vivo evaluation.

Use of dipicolinate-based complexes for producing ion-imprinted polystyrene resins for the extraction of yttrium-90 and heavy lanthanide cations.

Highly selective separation of yttrium (and lanthanides) is of interest for the design of radiopharmaceuticals, and an efficient method based on the ion-imprinting concept is proposed here. The synthesis and structural, thermodynamic and photophysical characterization of complexes of trivalent yttrium and lanthanides with two new vinyl derivatives of dipicolinic acid, HL1 and L2, are described. The feasibility of using ion-imprinted resins for yttrium and lanthanide separation is demonstrated. The resins were obtained by copolymerization with styrene and divinylbenzene and subsequent acid treatment to remove the metal ion. High-resolution Eu luminescence experiments revealed that the geometry of the complexation sites is well preserved in the imprinted polymers. The ion-imprinted polymer based on HL1 proved to be particularly well adapted for yttrium extraction, having a sizeable capacity (8.9+/-0.2 mg g(-1) resin) and a fast rate of extraction (t(1/2)=1.7 min). In addition, lighter and heavier lanthanide ions are separated. Finally, the resin displays high selectivity for yttrium and lanthanide cations against alkali and alkaline earth metals. For instance, in a typical experiment, 10 mg of yttrium was extracted from 5 g of milk ash sample by 2 g of the resin. The good separation properties displayed by the resin based on HL1 open interesting perspectives for the production of highly pure (90)Y and radiolanthanides for medical applications, and for trace analysis of these radiochemicals in food and in the environment.

Enhanced hydrolytic activity of Cu(II) and Zn(II) complexes in highly cross-linked polymers.

The chelate ligand tris[(1-vinylimidazol-2-yl)methyl]amine (5) was synthesized in five steps from commercially available starting materials. Upon reaction with ZnCl2 or CuCl2 in the presence of NH4PF6, the complexes [Zn5Cl]PF6 (6) and [Cu5Cl]PF6 (7) were obtained. The structure of both complexes was determined by single-crystal X-ray crystallography. Immobilization of 6 and 7 was achieved by co-polymerization with ethylene glycol dimethacrylate. The supported complexes P6-Zn and P7-Cu were found to be efficient catalysts for the hydrolysis of bis(p-nitrophenyl)phosphate (BNPP) at 50 degrees C. At pH 9.5, the heterogeneous catalyst P7-Cu was 56 times more active than the homogeneous catalyst 7. Partitioning effects, which increase the local concentration of BNPP in the polymer, are shown to contribute to the enhanced activity of the immobilized catalyst.

NIR lanthanide luminescence by energy transfer from appended terpyridine-boradiazaindacene dyes.

Mononuclear trivalent lanthanide complexes with formula [Ln(L)(NO(3))(3)] [in which L=4,4-difluoro-8-(2':2'';6'':2'''-terpyridin-4''-yl)-1,3,5,7-tetramethyl-2,6-diethyl-4-bora-3a,4a-diaza-s-indacene (Boditerpy)] are reported for Ln=Yb, Nd, Er, La and Gd. According to the crystal structure of the Yb complex, the lanthanide ion is bound to the terdentate terpyridine and the inner coordination sphere of the nine-coordinate lanthanide ion is completed by three bidentate nitrate anions. The coordination polyhedron can be described as a distorted tricapped antiprism. The terpyridine chelate is almost planar and tilted by nearly 60 degrees from the indacene subunit. FT-IR spectra confirm the bidentate binding mode of the nitrate anions for the other complexes. NMR and ES-MS spectra (through characteristic isotopic patterns) confirm the chemical formulation. The complexes have high molar absorption coefficients in the visible spectral region (65,000 M(-1) cm(-1) at 529 nm) and display sizeable NIR luminescence (900 to 1600 nm, for Ln=Yb, Nd and Er), upon irradiation through the electronic state of the indacene moiety at 514 nm. Crystal-field splitting was analysed at low temperature. The quantum yield of the Yb solution (10(-4) M) in dichloromethane amounts to 0.31 %, corresponding to a sensitisation efficacy of the ligand of ca. 63 %.

Ionic solid-state dimers and polymers derived from imidazolium dicarboxylic acids.

A series of imidazolium dicarboxylic acids have been prepared from the reaction of the 1,3-bis(carboxymethyl)imidazolium zwitterion with the Brønsted acids HX (X = F, Br, Cl, ClO4). The structures of these acids have been established in the solid state by single-crystal X-ray diffraction, which revealed that the cations and anions form strong hydrogen bonds through O-H...X interactions, leading to the formation of dimeric and polymeric networks. These acids react with elemental zinc and cobalt to form stable polymeric coordination complexes, some of which have also been characterised by X-ray diffraction.

Dual emission from luminescent nonalanthanide clusters.

Clusters containing nine lanthanide and six sodium ions are crystallized with the help of a tridentate ligand bearing a phosphonic acid group; the structure of the Eu(III) compound reveals a host-guest assembly Eu [symbol: see text] Na6Eu8L16 with a substantial quantum yield (32%). A similar Eu(III)-Yb(III) structure displays dual luminescence in the visible (Eu(III)) and NIR (Yb(III)) ranges.

Utilization of self-sorting processes to generate dynamic combinatorial libraries with new network topologies.

The synthesis of water-soluble, organometallic macrocycles is described. They were obtained by self-assembly in reactions of the half-sandwich complexes [[Ru(C6H5Me)Cl2]2], [[Ru(p-cymene)Cl2]2], [[Rh(Cp)Cl2]2], and [[Ir(Cp*)Cl2]2] with the ligand 5-dimethylaminomethyl-3-hydroxy-2-methyl-4-(1H)-pyridone in buffered aqueous solution at pH 8. The structure of the Ru-(p-cymene) complex was determined by single-crystal X-ray crystallography. Upon mixing, these complexes undergo scrambling reactions to give dynamic combinatorial libraries. In combination with structurally related complexes based on amino-methylated 3-hydroxy-2-(1H)-pyridone ligands, an exchange of metal fragments but no mixing of ligands was observed. This self-sorting behavior was used to construct dynamic combinatorial libraries of macrocycles, in which two four-component sub-libraries are connected by two common building blocks. This type of network topology influences the adaptive behavior of the library as demonstrated in selection experiments with lithium ions as the target.

Bioactive diterpenes from the fruits of Detarium microcarpum.

A fruit pulp extract of Detarium microcarpum showed inhibition of the growth of the plant pathogenic fungus Cladosporium cucumerinum and of the enzyme acetylcholinesterase, implicated in Alzheimer's disease. Fractionation of this extract led to the isolation of four new clerodane diterpenes, 3,4-epoxyclerodan-13E-en-15-oic acid (1), 5alpha,8alpha(2-oxokolavenic acid) (2), 3,4-dihydroxyclerodan-13E-en-15-oic acid (4), and 3,4-dihydroxyclerodan-13Z-en-15-oic acid (5). Also isolated were 2-oxokolavenic acid (3) and copalic acid (6). Structure elucidation of the compounds was carried out by spectroscopic data interpretation and by the X-ray crystallography of 2 and 4. Three of the new clerodane diterpenes (1, 2, and 5) showed both antifungal activity and inhibition of acetylcholinesterase, and 3 showed a slight inhibition of this enzyme.

Synthesis and characterization of platinum(IV) anticancer drugs with functionalized aromatic carboxylate ligands: influence of the ligands on drug efficacies and uptake.

A series of trans-platinum(IV) complexes with functionalized aromatic carboxylate ligands, cis,cis,trans-Pt(NH3)2Cl2(CO2C6H4R)2 (R = H (3), p-vinyl (4), p-methoxy (5), p-iodo (6), p-cyano (7), or o-carboxyl (8)) was synthesized and characterized by spectroscopic methods. Crystal structures of 3, 4, 7, and 8 were obtained, which revealed that their structural conformations were influenced by intramolecular H-bonding interactions. The complexes were evaluated for cellular uptake and inhibition of cell proliferation against a panel of lung, colon, and breast carcinoma cell lines. The functionalization of the aromatic carboxylate ligand was found to have a profound influence on the uptake, and hence, efficacy, of this class of complex.

Highly cross-linked polymers containing N,N',N' '-chelate ligands for the Cu(II)-mediated hydrolysis of phosphoesters.

Three immobilized Cu(II) complexes were generated by the following: (a) homopolymerization of the N,N',N' '-chelate ligand tris[2-(1-vinylimidazolyl)]phosphine (1) and subsequent metalation with CuCl2; (b) copolymerization of 1 with ethyleneglycol dimethacrylate (EGDMA) und subsequent metalation with CuCl2; or (c) molecular imprinting with the organometallic Mo-complex [Mo(eta3-C4H7)(CO)2(1)](TsO) (5) and EGDMA and subsequent replacement of Mo(II) by Cu(II). All three polymeric Cu complexes were found to efficiently promote the hydrolysis of activated phosphoesters with the relative activity being dependent on the nature of the polymer and the substrate.

A nearly planar water sheet sandwiched between strontium-imidazolium carboxylate coordination polymers.

The crystal structure of a strontium-imidazolium polymeric material obtained from the reaction of zwitterionic 1,3-bis(carboxymethyl)imidazolium with SrCO(3) (2:1) in water is reported. The compound crystallized in the orthorhombic space group Pcc2 with a = 14.545(3) A, b = 8.514(5) A, c = 8.5619(14) A, V = 1060.2(6) A(3), and Z = 2. The macromolecular structure is constructed from 8-coordinate strontium, which binds to six different imidazolium carboxylate molecules. The two-dimensional polymer is separated by water sheets in which the water molecules form near-planar hexagons.
