[The serum lipid and apolipoprotein levels of middle-aged male hyperlipidemics in Chengdu district].

It has been evidenced that cardiovascular diseases (CVD) relate with many risk factors and serum lipids play and essential role in the development of CVD. In order to further study hyperlipidemia (HL) in the middle-aged males, we analysed the body mass index (BMI), fast blood sugar (FBS), serum lipids and apolipoproteins A I, A II, B100, C II, C III and E in 223 male HL patients aged 41-60 and 349 normal male subjects who matched the HL patients in age. The result showed that the increase of serum triglycerids (TG), was as might be expected, the major characteristic of the middle-aged male HL patients in Chengdu District. Besides the serum lipid and apolipoprotein B100, C II, C III and E levels, the BMI and FBS levels in the IIb, IV and V types of HL patients were significantly higher than those in the normal subjects. It also showed that in TG increased patients the increased percentages of serum lipids were significantly higher than those of apolipoproteins. The age distribution in HL patients revealed that only 25% of the HL patients were between 41 to 50 years old, while 50% in the 56-60 year-old group. The relationship between apolipoprotein levels and serum lipid metabolism is also discussed. The results suggest that the 51-60 year-old males should pay attention to diet and increase physical activities to reduce incidence of HL which is directly associated with CVD.

[Relation of serum apolipoprotein B100 levels to lipids and apolipoproteins A I, A II, C II, C III and E levels in the middle and old age normal males in Chengdu area].

This study was conducted in 438 normal men aged 40-70. The age groups were: 40-44, 45-49, 50-54, 55-59, 60-64, and 65-70. The results showed that the serum apolipoproteins levels (mean+/- s, mg/L) were: B100 levels 742.5 +/- 165.8, A I 1301.2 +/- 219.9, A II 299.4 +/- 49.0, C II 42.7 +/- 21.4, C III 113.3 +/- 34.0 and E 38.7 +/- 8.2. The levels of serum apoB100 increased with age. The serum apoB100 levels in the 5th and 6th age groups were significantly higher as compared with the groups 1, 2, 3 and 4 (P < 0.05 and P < 0.05). The fasting serum mean TG (1.39 +/- 0.45 mmol/L), TC(4.79 +/- 0.82 mmol/L) and LDC-C(1.62 +/- 0.40 mmol/L levels in the groups 5 and 6 were significantly higher as compared with the groups 1, 2, 3 and 4 (P < 0.01). Serum apoC II, C III and E levels were significantly increased in the groups 5, 6 (P < 0.05). The correlation analysis indicated that there was a positive correlation of apoB100 with serum TG, TC, LDL-C, apoC II, C III and E respectively (P < 0.01); and a negative correlation with HDL-C levels (r = 0.1312); and apoB100 correlated negatively with apoA I (r = -0.0706). The results suggest that serum TG, TC, LDL-C, apoC II, C III and E are the main factors related with the serum apoB100 levels.

[A study on monoclonal antibodies against epithelial cell of endolymphatic sac in guinea pigs].

Using a microdissected endolymphatic sac of the guinea pig (n = 35) as an initial antigen preparation, serials of monoclonal antibodies were established which were used to label the epithelial cells of endolymphatic sac. The antibodies showed strong immunoreactivity with kidney, but not with other organs. It also showed isotopes IgG1, IgG2b and IgM. The results of Western blotting and SDS-PAGE indicated that the epitopes of monoclonal antibodies were proteins or glyoprotein with a molecular weight of approximately 7,400. The locations of the epitopes in epithelial cells suggest that it may play some roles in construction and function of the endolymphatic sac.

[Intrathoracic gastric function after esophagectomy for patients with esophageal cancer].

OBJECTIVE: To reveal the physiological changes of intrathoracic stomach after esophagectomy and find ways to improve patients' digestive function. METHODS: In twenty preoperative and 41 postoperative patients, the physiologic functions of stomach were studied and compared with radioisotope technique, pressure measurement, 24-hour pH monitoring and gastroscopy. RESULTS: The emptying of semisolid food from the intrathoracic stomach was much delayed (P < 0.01); its acid secretion was decreased in the early postoperative period (P < 0.01) but it restored to normal one year later. Fasting serum gastrin level was higher (P < 0.01) in the early postoperative period but showed a partial restoration one year later. The high pressure zone at anastomosis helped prevent gastroesophageal reflux. High incidence of gastritis was found in intrathoracic stomach and this may be related to gastric stasis. CONCLUSION: Performing scarf telescoping anastomosis and pyloraplasty, giving a certain amount of pepsin, diluted hydrochloric acid and anti-gastrin drug to early postoperative patients should be beneficial to reducing the symptoms and normalizing patients' digestive function.

[Separation and identification of the compounds from Achyranthes bidentata Bl].

Eight compounds were separated from the roots of Achyrathes bidentata by repeated chromatography. On the basis of physicochemical properties and spectral analysis their structures were elucidated as alpha-spinasterol (1), beta-sitosterol (2), chrysophanol (3), dibutyl phthalate (4), palmitic acid (5), alpha-spinasterol-3-O-beta-D-glucoside (6), daucosterol (7) and ecdysterone (8). Compounds 1-7 were isolated from the plant for the first time.

[Preliminary studies on outer membrane protein (OMP) patterns of Escherichia coli O1, O2 and O78 isolates of chicken origin].

Omp patterns of twenty-five isolates of Escherichia coli serotype O1, O2 and O78 of chicken origin from Jiangsu area were determined. The outer membrane proteins of these isolates were isolated with the improved N-lauroylsarcosine method and analyzed by SDS-PAGE. Major outer membrane proteins bands contributed for OMP patterns were visualized with Coomassie brilliant blue staining. Two OMP patterns were divided in eight of O1 isolates and nine O2 isolates respectively. Among these OMP patterns, 1 OMP pattern was shared by these 2 different O serotypes isolates, and eight of O78 isolates also shared this OMP pattern. These results indicated that the OMP patterns of avian pathogenic Escherichia coli isolates of O1, O2 and O78 from this area were heterogeneous, and the same OMP patterns was presented in these 3 serotypes.

[The significance of integrated function states in the field of space medical research].

The integrated functional changes of astronauts are the important aspects in the research field of space medicine, but they remain to be studied. The purpose of this paper is to discuss the significance of research on integrated function state and the ways to study it. Based on the functional changes of the nervous, the endocrine and the immune systems under adverse environmental factors, and the interrelationship among the various changes, the author put forward a proposal of three types of integrated function states, that may appear in astronauts during space flight, namely: depression type, stress type and blood stasis type. The author also recommended some aspects of research of the integrated function states of astronauts.

[Rapid determination of aspartame in compound sweetening by reversed-phase high performance liquid chromatography].

A method for rapid determination of Aspartame in compound sweetening by reversed-phase high performance liquid chromatography is presented. Aspartame in compound sweetening was separated in a short column (Ultrasphere XL-ODS, 3 microm, 4.6 mm x 70 mm) by using CH3OH-0.02 mol/L NH4Ac as mobile phase. The flow rate was 0.8 mL/min. Detection was performed with UV detector at 220 nm. The injection volume was 20 microL. It was qualitatively analysed by UV scanning at a wavelength range of 200-350 nm under no-stop flow according to their retention time. Quantitative analysis was carried out by measuring peak height and comparing it with external standard. The minimum detectable amount was 5 microg/L. The linear range of the calibration curve was 40-200 mg/L. The average recovery of Aspartame was 92%. The relative standard deviation was 2.9%. This method is simple, rapid and sensitive.

[Deviation of size-exclusion chromatographic behavior of tryptophan on TOYOPEARL HW-40S column].

A deviation of size-exclusion chromatographic behavior of amino acids on TOYOPEARL HW-40S column is reported. On the column 18 amino acids were not eluted in the same fraction in the total volume, but separated into 5 fractions with tryptophan as the last one far behind the total volume. Interaction between tryptophan and the matrices was investigated under different conditions in order to elucidate the chemical nature of the deviation. It was found that addition of trifluoroacetic acid and sodium chloride of high concentration in mobile phase, dramatically increased the retention time of trypophan, while addition of acetonitrile, sodium chloride of lower concentration and the hydrogen bond breaker urea reduced the interaction. It is concluded that adsorption of tryptophan on TOYOPEARL HW-40S is basically due to the hydrophobic nature with limited influence of hydrogen bond and ionic interaction. The former factor can be eliminated by keeping acentonitrile concentration over 50% (V/V), while the latter two can be reduced to the minimum by adding 0.17mol/L sodium chloride and 3.51mol/L urea. The results not only lead to better understanding of size-exclusion chromatography of this matrices, but also help to apply hydrophobic chromatography of this matrices to the isolation of amino acids and peptides.

[X-ray photoelectron spectroscopy (XPS) and mass spectrum (MS) study of organic tin compounds II].

Seven new synthetical (Z)-1-[2-(Triarylstannyl) vinyl]-1-cycloheptanols organic tin compounds were studied by X-ray photoelectron spectroscopy and mass spectrum method. The influence of different replacement radical in compounds on the d orbit of tin atom was discussed. The coordinate substance had an effect on the chemical displacement of Ols binding energy. The O-->Sn bond was affected by the different replacement radical. The results of X-ray photoelectron spectroscopy and mass spectrum show that Cl, Br and I had the important action.

Nitric oxide synthase inhibitors attenuate transforming-growth-factor-beta 1-stimulated capillary organization in vitro.

Angiogenesis is a complex process involving endothelial cell (EC) proliferation, migration, differentiation, and organization into patent capillary networks. Nitric oxide (NO), an EC mediator, has been reported to be antigenic as well as proangiogenic in different models of in vivo angiogenesis. Our aim was to investigate the role of NO in capillary organization using rat microvascular ECs (RFCs) grown in three-dimensional (3D) collagen gels. RFCs placed in 3D cultures exhibited extensive tube formation in the presence of transforming growth factor-beta 1. Addition of the NO synthase (NOS) inhibitors L-nitro-arginine methylester (L-NAME, 1 mmol/L) or L-monomethyl-nitro-l-arginine (1 mmol/L) inhibited tube formation and the accumulation of nitrite in the media by approximately 50%. Incubation of the 3D cultures with excess L-arginine reversed the inhibitory effect of L-NAME on tube formation. In contrast to the results obtained in 3D cultures, inhibition of NO synthesis by L-NAME did not influence RFC proliferation in two-dimensional (2D) cultures or antagonize the ability of transforming growth factor-beta 1 to suppress EC proliferation in 2D cultures. Reverse transcriptase-polymerase chain reaction revealed the constitutive expression of all three NOS isoforms, neuronal, inducible, and endothelial NOSs, in 2D and 3D cultures. Moreover, Western blot analysis demonstrated the presence of immunoreactive protein for all NOS isoforms in 3D cultures of RFCs. In addition, in the face of NOS blockade, co-treatment with the NO donor sodium nitroprusside or the stable analog of cGMP, 8-bromo-cGMP, restored capillary tube formation. Thus, the autocrine production of NO and the activation of soluble guanylate cyclase are necessary events in the process of differentiation and in vitro capillary tube organization of RFCs.

Familial occurrence of hemangioblastoma of central nervous system.

OBJECTIVE: To study the familial occurrence of hemangioblastoma of the central nervous system and to search for an effective way to diagnose and manage the disease. METHODS: From 1974 to 1993, six patients with hemangioblastoma with family history underwent surgical treatment at our hospital. Computerized tomography (CT) or magnetic resonance imaging (MRI) technique was used for the diagnoses before operation. RESULTS: Each patient had more than two relatives with hemangioblastoma. Multiple lesions were found in two of the six familial hemangioblastoma cases. All the patients recovered well after operation. So far, no victims have been discovered in the third generation. CONCLUSIONS: The high occurrence rate of familial hemangioblastoma cases suggests an involvement of hereditary factors. We need a long follow-up study on the descendants of familial hemangioblastoma patients. MRI is of diagnostic value in determining the location and nature of hemangioblastoma in both brain and spinal cord. A thorough examination is necessary for hemangioblastoma patients, and microsurgery is important in removing the tumor.

[China-made paclitaxel in the treatment of advanced ovarian cancer: a clinical analysis of 32 cases].

OBJECTIVE: To study the effect and toxicities of China-made paclitaxel in the treatment of advanced epithelial ovarian carcinoma. METHODS: Thirty-two patients were divided into two groups. single paclitaxel group (SPG): paclitaxel was only given to twelve untreated patients or those who had been given only one course of chemotherapy at a dose of 175 mg/m2 in 5% glucose 500 ml, i.v. for 3 hours; and combined paclitaxel group (CPG): paclitaxel 135 mg/m2 in 5% glucose 55 ml, i.v. for 3 hours on day 1 plus cisplatin 70-80 mg/m2 or carboplatin 300 mg/m2, i.v. on day 2, was administered to 20 patients with persistent or recurrent tumor after platinum based chemotherapy. Four weeks were defined as one course in both groups. At least two courses were given except those with tumor progression after one course. RESULTS: The overall response rate was 39%. It was 27% and 45% in the SPG and CPG. The chief side effects were moderate myelosuppression, peripheral neuropathy (myalgia, arthralgia, peripheral neuritis), severe alopecia, etc., but no hypersensitivity. One patient died from intestinal perforation. CONCLUSIONS: China-made paclitaxel is effective as in treating ovarian carcinoma especially for platinum-resistant patients. The toxic effects are tolerable to the patients.

[Kaposi's sarcoma associated with HIV in otorhinolaryngology: report of 21 cases].

Twenty one cases of kaposi's sarcoma (KS) with manifestation in the field of otolaryngology are reported. The incidence of KS is increasing following epidemic of AIDS all over the world. Epistaxis, nasal obstruction, sore-throat, dry of foreign body filling in throat and nodal mass are most frequent manifestation. The relationship of KS with HIV, it's pathology and diagnosis are discussed.
