Complete nucleotide sequence of immunogenic protein MPB70 from Mycobacterium bovis BCG.

The extracellular protein MPB70 is a heat-stable immunogenic protein which was found in the culture filtrate of Mycobacterium bovis BCG Japanese. We determined the complete nt and aa sequences of MPB70 and correlated with the previously reported data. The N-terminal sequence revealed that the signal peptide (SP) consisted of 30 aa and that the mature protein had 163 aa with a molecular weight of 16,305. The SP displayed a characteristic feature of an Ala-rich property which would be efficient in a SP function.

A case of bullous pemphigoid induced by tiobutarit (D-penicillamine analogue).

A 61-year-old woman with severe rheumatoid arthritis developed bullous lesions on the trunk, conjunctiva, and oral mucosa during treatment with tiobutarit, a new antirheumatic drug analogus to D-penicillamine. Histology showed a subepidermal blister, and circulating antibodies to the basement membrane zone were detected. The bullous lesions improved rapidly after discontinuation of tiobutarit. However, 3 years later, she was re-exposed to tiobutarit and the bullous lesions recurred within a week. This is the first confirmed case of tiobutarit-induced BP.

Antigenic variation among Sendai virus strains detected by monoclonal antibodies.

Thirteen strains of Sendai virus isolated from various sources in the 1950's and after 1976 were compared for their reactivities with monoclonal antibodies prepared against the prototype strain MN of Sendai virus. Results revealed that while the 5 strains isolated in the 1950's reacted with all the monoclonal antibodies as the prototype strain did, the 2 strains isolated in 1976 and 1978 did not react with an F-specific monoclonal antibody, and the other 6 strains isolated after 1978 lacked reactivity with an HN-specific monoclonal antibody.

Distribution of mass transfer rate and wall shear stress behind simple rectangular stenosis in pulsating flow.

The distributions of mass transfer rate and wall shear stress in sinusoidal laminar pulsating flow through a two-dimensional asymmetric stenosed channel have been studied experimentally and numerically. The distributions are measured by the electrochemical method. The measurement is conducted at a Reynolds number of about 150, a Schmidt number of about 1000, a nondimensional pulsating frequency of 3.40, and a nondimensional flow amplitude of 0.3. It is suggested that the deterioration of an arterial wall distal to stenosis may be greatly enhanced by fluid dynamic effects.

Cloning and characterization of the gene for immunogenic protein MPB64 of Mycobacterium bovis BCG.

The gene for immunogenic protein MPB64 found in culture filtrates of only Mycobacterium tuberculosis and some strains of Mycobacterium bovis BCG was cloned by using a single-probe method and was sequenced. The gene analysis revealed that the structural gene for MPB64 consisted of 618 base pairs, and its deduced molecular weight was 22,400. Twenty-two amino acids for a putative signal peptide and 205 amino acids for the MPB64 protein were observed. In the coding region, the third letter of the codon showed a biased codon and a high G+C content (78.5%). The gene was expressed in Escherichia coli by using an E. coli expression vector. The product showed migration similar to that of the authentic MPB64 protein by electrophoresis and reacted with the polyclonal and the monoclonal antibodies raised against the MPB64 protein. The strict specificity of MPB64 could be applied to immunodiagnosis of tuberculosis.

Thermal expansion of visible-light-cured composite resins.

Linear coefficients of thermal expansion were measured on seven visible-light-cured composite resins from 0 to 60 degrees C. The values ranged from 29.0 to 83.5 x 10(-6)/degrees C at 37 degrees C. Statistical analysis revealed significant differences among temperature ranges and products and between runs. Significant correlation was not found between the coefficient of thermal expansion and volume fraction of filler particles.

Immunogenic protein MPB57 from Mycobacterium bovis BCG: molecular cloning, nucleotide sequence and expression.

Using a single-probe method, we have cloned the gene for an immunogenic MPB57 protein of Mycobacterium bovis BCG. The nucleotide sequence includes an ORF of 300 base pairs encoding a protein of 99 amino acids with an Mr of 10,818. This cloned gene was expressed in an Escherichia coli expression vector to give a mature protein which reacted with a polyclonal antibody raised against MPB57.

Variation of virulence and other properties among Sendai virus strains.

The virulence of five Sendai virus strains (MN, Z, KN, Mol, and Hm) isolated from laboratory rodents was compared, using 3-week-old female Jcl-ICR mice. The virulence of the strains was Mol, MN, KN, Z, and Hm in decreasing order. The 50% lethal dose and 50% lung consolidation inducing dose of the highest virulent strain differed by the order of more than 10(3) and 10(6), respectively, from those of the lowest virulent one. Other properties such as the growth rate in LLC-MK2 cells, neuraminidase activities, and molecular weights of structural proteins also differed among the virus strains. These results indicate that Sendai virus prevailing in laboratory rodents is not homogenous with respect to virulence and some other properties.

Cloning and expression of the Mycobacterium bovis BCG gene for extracellular alpha antigen.

The gene for the extracellular alpha antigen of Mycobacterium bovis BCG was cloned by using a single probe restricted to G or C in the third position. This technique should have great potential for the isolation of mycobacterial antigen genes. The gene analysis revealed that the alpha antigen gene encoded 323 amino acid residues, including 40 amino acids for signal peptide followed by 283 amino acids for mature protein. This is the first report on the structure of the mycobacterial signal peptide. The promoter-like sequence and ribosome-binding site were observed upstream of the open reading frame. In the coding region, the third position of the codon showed high G + C content (86%). The gene was expressed as an unfused protein in Escherichia coli by using an E. coli expression vector. This protein, which reacted with polyclonal antibody raised against alpha antigen from Mycobacterium tuberculosis, would be applicable to the immunodiagnosis of tuberculosis.

Parameters that affect in vitro bonding of glass-ionomer liners to dentin.

The purpose of this study was to evaluate the effects of two concentrations of poly (acrylic acid) (10 and 25%), three treatments (untreated, passive conditioning, and active conditioning), and two storage conditions (24 hours in 37 degrees C water and thermal cycling) on the in vitro tensile bond strength of three commercial glass-ionomer liners to human dentin. Bond strengths to untreated dentin after storage for 24 hours ranged from 19.0 to 21.7 kg/cm2 for Glasionomer Base Cement, Cement/Liner, and Ketac-Bond, but dropped to a range of 4.9 to 9.7 kg/cm2 after thermal cycling. Active conditioning with 10% acid resulted in bond strengths after 24-hour storage that ranged from 23.5 to 44.0 kg/cm2, compared with values from 21.7 to 38.0 kg/cm2 with active conditioning using 25% acid. Active conditioning with 10% acid resulted in bond strengths after thermal cycling that were in the range of 15.8 to 27.4 kg/cm2 and were 80 to 320 percent higher than values resulting from passive conditioning under these conditions. Active conditioning with 10% acid for 30 seconds produced a bond strength for Glasionomer Base Cement of 44.0 kg/cm2, compared with a bond strength of 28.7 kg/cm2 for a 10-second active conditioning. Qualitative analysis of scanning electron photomicrographs showed that dentin tubules were opened to a greater extent by active conditioning with 25% acid than by passive conditioning with 10% acid.
