Quantitative analysis and comparison of the physical properties of O-alkyl and S-alkyl monoethers of glycerol.

A homologous series (C(10), C(12), C(14), C(16), C(18)) of synthetic O-alkyl and S-alkyl ethers of glycerol was analyzed by gas-liquid chromatography (GLC) and thin-layer chromatography (TLC), and examined by IR and n.m.r. spectroscopy; the physical properties of the O-alkyl and S-alkyl ethers were compared. Isopropylidene derivatives of the glycerol ethers and thioethers were quantitatively analyzed by GLC on polar and nonpolar liquid phases. On a medium polar liquid phase (ethylene glycol succinate), mixtures of the O-alkyl and S-alkyl ethers were completely resolved. Isopropylidene derivatives of glycerol ethers and of thioethers could be separated as classes (though not into individual homologues) by TLC. O-hexadecyl and S-hexadecyl ethers of glycerol are easily distinguished by IR and n.m.r. spectroscopy.

Gas-liquid chromatographic analysis of free long-chain aldehydes.

Quantitative results are reported for gas-liquid chromatography of mixtures of free tetra-, hexa-, and octadecanals and cis-9-octadecenal on polar and nonpolar liquid phases. Stability of the fatty aldehydes during chromatographic analysis and up to 1 year of storage appears to be related to the solvent (carbon disulfide) used. Analysis of the free aldehydes by gas-liquid chromatography eliminates the disadvantages associated with the preparation and analysis of derivatives for this lipid class.

Gas-liquid chromatographic analyses of 1,2-ethanediol monoethers and monoesters.

Synthetic mixtures of saturated and unsaturated monoethers and monoesters of 1,2-ethanediol, ranging in chain length from 12 to 20, were analyzed as acetates, trifluoroacetates (TFA), and trimethylsilyl (TMS) ethers by gas chromatography on polar and nonpolar liquid phases. Acetates, TFA derivatives, and TMS derivatives of the glycol ethers were eluted ahead of the corresponding glycol ester derivatives on both liquid phases. The elution order of derivatives of the same compound was found to be TMS derivative before TFA derivative before acetate on the polar liquid phase, and TFA derivative before TMS derivative before acetate on the nonpolar liquid phase. Elution orders relative to methyl stearate were also determined. With one exception, all of the derivatives, and both liquid phases, were found suitable for the quantitative analysis of diol monoethers and monoesters.

Characterization and identification of glyceryl ether diesters present in tumor cells.

The previously unidentified neutral lipid present in tumor tissues has been isolated from Ehrlich ascites cells and unequivocally identified as a lipid class of glyceryl ether diesters containing various degrees of unsaturation, and ranging in approximate molecular weight from 760 to 990. The glyceryl ether diester fraction was shown to be free from neutral plasmalogens (glyceryl diacyl alk-1'-enyl ethers). The tumor lipid was subjected to saponification, transesterification, and lithium aluminum hydride reduction. The glyceryl monoethers that resulted from deacylation were the 1-isomers ranging in hydrocarbon chain length from C(12) to C(24). The predominant glyceryl ethers were the hexadecyl (49%), octadecyl (21%), and octadecenyl (14%) derivatives. Saturated and mono- and polyunsaturated fatty acids ranging in chain length from C(12) to C(24) carbon atoms were esterified to the glyceryl monoether. Gas-liquid chromatography, thin-layer chromatography, and nuclear magnetic resonance and infrared spectroscopy were used to characterize and identify the intact tumor lipid and its derived products.

Fluorescent treponemal antibody tests. A summary and comparison.

A comparison of current serologic tests for syphilis shows that treponemal tests are preferable to reagin tests in detecting specific antibodies, but that reagin tests are best for determining the response to treatment. The newly developed fta-absorption technique is suggested as a reliable, inexpensive test for treponemal antibodies.
