Incorporation of (5-3H) uridine into ribonucleotide pools and RNA during thyroxine-induced metamorphosis of Xenopus laevis tadpoles.

Incorporation of [3H] uridine into the ribonucleoside triphosphates UTP and CTP, total RNA, and nuclear and cytoplasmic RNA was followed in Xenopus laevis tadpole liver during thyroxine (T4)-induced metamorphosis. Pool sizes of UTP and CTP were found to remain unchanged, although turnover the ribonucleoside triphosphates was found to be greatly stimulated after 4 days of hormone treatment. The time course of labeling of the 40-S pre-rRNA was very similar to that of UTP in both thyreostatic and T4-treated tadpoles, thus reflecting a direct relationship between turnover of the immediate precursors and labeling of RNA. Although a faster depletion of labeled UTP and pre-rRNA (precursor ribosomal RNA) was noted in T4-treated tadpoles, labeled cytoplasmic rRNA continued to accumulate almost linearly for 25 h. In thyreostatic larvae no further increase in labeled cytoplasmic rRNA occurred beyond 4 h of labeling. From these results we conclude that both enhanced transcription and more effective utilization of pre-rRNA are responsible for the net accumulation of rRNA observed on the 4th day of T4-induced metamorphosis.

Tobramycin sulfate: a summary of worldwide experience from clinical trials.

Tobramycin sulfate, a new bactericidal aminoglycoside antibiotic, has been evaluated in 3,506 patients. Its in vitro activity is similar to that of other aminoglycoside antibiotics, except that it is more active against Pseudomonas aeruginosa. Clincal studies have shown that tobramycin sulfate is well tolerated and effective in the treatment of the infections listed below when they are caused by susceptible gram-negative organisms and by staphylococci: infections of the respiratory tract (including cystic fibrosis), central nervous system, skin, soft tissue, and bone (including burns); urinary tract and intraabdominal infections; and septicemia. The overall response was satisfactory in greater than 86% of the patients. Drug-related adverse effects were reported in 3.9% of the patients, and included reactions in the nervous system in 0.6% and in the kidney in 1.5%.

[Ageing changes in collagen -- in vitro ageing changes in collagen (part III) (author's transl)].

In the present three parts of the survey ageing-changes in collagen physical and chemical qualities are discussed at first. Further a summary on questions of the biosynthesis of collagen, changes of the metabolism in the ageing organism and changes of the ripening collagen in the living organism is given. Following in vitro-ageing-changes of collagen and causes of the changes of the ageing organism are described. A summarizing appreciation of the ageing-changes in the collagen finishes the survey.

[The role of radiotherapy in the treatment of malignant bone tumors (author's transl)].

The role of radiotherapy and adjuvant chemotherapy in the primary treatment of osteogenic sarcomas and of Ewing's sarcoma is reviewed. In osteosarcoma radiotherapy can take the form of prophylactic total irradiation of the lung, but preoperative irradiation of the primary tumor has not proved successful. On the other hand, in Ewing's sarcoma primary and local irradiation is the therapy of choice, and is followed by adjuvant polychemotherapy over a long period.

[Effect of generalized edema on blood flow distribution in the brain].

The correlation between increase in brain tissue pressure and cerebral blood flow distribution was studied in generalized brain edema induced by water intoxication in cats. Brain tissue pressure increased as the brain edema progressed. The intracranial vascular component was suggested to play a certain role in the volume buffering capacity at increased intracranial pressure. Transtentorial redirection of carotid blood flow observed in the early stages of water intoxication, is related to tissue pressure gradients. The meaning of tissue pressure gradients for blood flow distribution is discussed.
