Development of IVM-IVF produced 8-cell bovine embryos in simple, serum-free media after conditioning or co-culture with buffalo rat liver cells.

Development of 8-cell bovine embryos derived from in vitro matured/in vitro fertilized (IVM/IVF) oocytes was evaluated in two simple, serum-free media (CZB and SOM) with buffalo rat liver cells co-culture (BRLC) or after conditioning compared to a commonly used, serum-supplemented complex medium TCM-199. In a 3 x 4 factorial design, 578 eight-cell embryos were randomly assigned to 12 treatment groups. The factors were: first, type of culture medium (M199/FBS, CZBg and SOM), and second, the use of BRLC (as co-culture or to condition media for 24 hr and 48 hr) and unconditioned media. Development to morula was not affected by the type of medium, but co-culture and 48 hr conditioning within media type resulted in better development when compared to the 24-hr conditioned or unconditioned groups. Blastocyst development in SOM (38.9%) was different (P < 0.05) than in CZBg (46.6%) and M199/FBS (48.7%) and was lowest in the unconditioned group (27.8%) followed by 24 hr conditioned (33.3%), 48 hr (56.3%), and co-culture (59.6%). No blastocyst expansion was observed with unconditioned media and 24 hr conditioned SOM. Significant differences (P < 0.05) were found among all treatment groups except the co-culture and 48-hr conditioned groups. Hatching occurred only with co-culture and 48-hr conditioned groups of M199/FBS and CZBg media. These data show that CZB with glucose conditioned by BRLC monolayers for 48 hr can support the development of IVM/IVF produced bovine embryos to blastocyst compared to culture in TCM-199 with serum.

Effect of sperm exposure time on in vitro fertilization and embryo development of bovine oocytes matured in vitro.

This study was designed to investigate the effect of sperm exposure time on the fertilization rate and subsequent developmental capacity of bovine oocytes matured in vitro. Cumulus oocyte complexes (COCs) obtained from 2 to 6 mm follicles were matured for 24 h in TCM-199 supplemented with fetal bovine serum (FBS) and hormones (FSH, LH and estradiol 17-beta). In vitro fertilization (IVF) was performed by incubating 15 to 20 matured oocytes with 1 x 10(6) percoll separated frozen-thawed spermatozoa in 1 ml of IVF-TL medium for either 4, 8, 12, 16, 20, 24 or 28 h. Following sperm exposure for different periods of times, the presumptive zygotes were co-cultured with Buffalo Rat Liver cells (BRLC) monolayers in CZB medium without glucose, a simple semi-defined medium developed for mouse embryo culture, for 3 d post-insemination and then in M199/FBS (TCM-199-HEPES supplemented with 20% heat-treated FBS and 1 mM sodium pyruvate) for 5 d. The fertilization rates differed significantly among the 7 treatment groups, with higher frequencies obtained by co-incubation of gametes for 20, 24 or 28 h (67 to 76%) than for 4, 8 and 12 h (26 to 54.5%), with 16 h (57%) being intermediate. However, the length of sperm exposure time did not significantly affect subsequent embryo development, although an increasing trend was noted from 4 h to 20 h. The number of fertilized oocytes at 3 d post-insemination cleaving to 2- to 4-cell vs 8-cell stage was not different among treatment groups. Development of 8-cell embryos to morulae and blastocysts did not differ among the treatment groups. These data suggest that the optimum duration of sperm-oocyte incubation is 24 h, and periods shorter than 16 h may result in a reduced fertilization rate.

Development of in vitro matured and fertilized bovine oocytes co-cultured with Buffalo Rat Liver cells.

This study was designed to evaluate the efficacy of Buffalo Rat Liver cells (BRLC) monolayers in supporting the development of in vitro matured and fertilized (IVM/IVF) bovine oocytes through to the hatched blastocyst stage compared to the commonly used co-culture system of bovine oviduct epithelial cells (BOEC). Cumulus oocyte complexes (COCs) obtained from 2- to 6-mm ovarian follicles at slaughter were matured for 24 h in TCM-199 supplemented with FBS and hormones (FSH, LH and estradiol 17-beta). In vitro fertilization (IVF) was performed using 1 x 10(6) percoll separated frozen-thawed spermatozoa in 1 ml of IVF-TL medium containing 18 to 20 matured oocytes. After 20 to 22 h of sperm exposure, 584 presumptive zygotes in 2 separate trials were randomly assigned to 3 treatment groups (BRLC co-culture, BOEC co-culture and control, consisting of medium alone). Zygotes were cultured in CZB media, a simple semi-defined medium, without glucose for the first 2 d, transferred to M199/FBS (TCM-199-HEPES supplemented with 20% HTFBS, 1 mM Sodium pyruvate), and cultured for an additional 8 days. Cleavage and development to morula and various blastocyst stages were recorded between d 3 and 11 after the start of IVF. Overall average cleavage rate was 75% (440 584 ) and did not vary across the treatments or trials. The proportion of embryos that reached the morula stage in both co-culture systems did not differ (P > 0.05) and was significantly higher (P > 0.05) compared to the control group. However, the percentage of the number of blastocysts, expanded blastocysts and hatched blastocysts varied across the treatment groups (P < 0.05), with the highest results obtained in the BRLC co-culture system. The production of blastocysts in BOEC co-culture was inconsistent between the 2 trials where a significant difference (40.6 vs 53.0%; P > 0.05) was observed. Rate of development to the blastocyst stage was similar between the 2 co-culture systems, with most of the embryos reaching the blastocyst stage by d 8 post insemination. The results of this study show that BRLC from a commercially available established cell line offer a more reliable alternative to a BOEC co-culture system for in vitro maturation, fertilization and culture of bovine embryos.

Pathologic acetabular fractures: new concepts in surgical management.

Pathologic fracture of the pelvis and acetabulum secondary to metastases is a disabling condition for cancer patients. Management has for the most part remained nonoperative because the complexities of pelvic anatomy and reconstruction yield risks which have outweighed potential benefits. No advances in surgical reconstruction have been reported in one and a half decades. Previous reports and ideas addressing surgical reconstruction have focused on which type of total hip arthroplasty best suits the acetabular bone stock remaining following removal of the tumor. As improving medical management of metastatic cancer increases longevity, improved methods for surgical management of pathologic fractures are required. A new concept for surgically managing periacetabular fractures due to metastases is introduced whose premise is based on pelvic rather than hip pathology. Resection of tumor-infiltrated acetabular columns creates reconstructive challenges best met by the techniques and knowledge gained from trauma surgeons repairing acetabular fractures. The additional use of reinforced polymethylmethacrylate and subchondral bone cement augments fixation. These concepts have been incorporated into a new classification system and treatment strategy which is critically examined in 13 individuals. Tumor resection and pelvic-periacetabular reconstruction can satisfactorily be performed through extensile pelvic approaches without necessarily performing a hip arthroplasty. A more complete oncologic resection, superior reconstruction, and quicker rehabilitation for affected individuals can result in approximately one half of individuals.

Fibroblast cytotoxicity and blood cell integrity following exposure to dermal wound cleansers.

Cellular toxicity of dermal wound cleansers may be an important consideration in wound healing. To study the effects of dermal wound cleansers on wound closure, human fibroblasts were cultured and survival rates were established. Cellular integrity of both red and white blood cells was established by quantifying the number of intact cells using a hemocytometer. In addition, the concentration of hemoglobin in plasma was determined. All tests were conducted against saline controls. Cellular viability and function in each experiment were used to rank solutions according to the cytotoxic effects demonstrated. Significant differences were found in the toxicity profile of available cleansers. Cellular toxicity of dermal wound cleansers is an important consideration in wound healing. Initial steps in wound management should not undermine subsequent phases in wound healing.

Superovulation of cattle with equine pituitary extract and porcine ESH.

Superovulation has been practiced in cattle for more than 50 years but the results have been highly variable. Scientists at six locations compared a horse pituitary extract (HAP) with a single batch of porcine FSH (pFSH) to determine the efficacy of these hormones to induce superovulation and to test for variability in the superovulatory response. Acetone-dried equine pituitaries were suspended in 40% ethanol containing 6% ammonium acetate, and the supernatant was mixed with 2.5 volumes of cold ethanol. The resulting precipitate was washed with cold ether and dried. Total doses of 18 mg of HAP and 36 mg of pFSH were injected intramuscularly (i.m.) over 4 days, two injections per day, and prostaglandin (PGF(2)alpha; 25 mg, i.m.) was administered on Day 3. Injections were begun on Days 6 to 13 of the estrous cycle. The overall ovulation rates (mean +/- SEM) for HAP and FSH were 8.8 +/- 0.7 and 15.1 +/- 1.0, respectively (n=231; P<0.01). Location interacted (P<0.01) with the type of gonadotropin for the ovulation rate. When expressed as a proportion of the number of corpora lutea, the total number of embryos recovered was greater (P=0.03) for pFSH than for HAP, but there was no difference in the number of Quality 1 and 2 embryos. The results show that HAP can induce a satisfactory superovulatory response, but there was no evidence of reduced variability of response to HAP compared with pFSH.

Prenatal lesions in an ovine fetus with GM1 gangliosidosis.

Sheep affected with ovine GM1 gangliosidosis are normal at birth and develop clinical signs, initially ataxia, commencing at approximately 5 months of age, which progresses rapidly to recumbency. Superovulation and embryo transfer techniques were applied to a flock of carrier sheep of ovine GM1 gangliosidosis to increase the numbers of carrier and affected animals. A recipient ewe with 3 at-risk fetuses died at 4 months of gestation (normal ovine gestation is 5 months), and spectrofluorimetric assay of cerebral lysosomal beta-galactosidase of the fetuses showed that 2 were carriers and one was an affected fetus. The affected fetus had marked cytoplasmic enlargement and vacuolization of central and peripheral nervous system neuronal soma and of hepatocytes and renal epithelial cells. Lectin histochemistry indicated abnormal storage of complex carbohydrates, with terminal saccharide moieties consisting of beta-galactose, N-acetylneuraminic acid, and N-acetylgalactosamine. This case underlines the need for prenatal initiation of therapy and also demonstrates that vacuolization alone is not the cause of clinical signs in this lysosomal storage disease in that clinical signs do not commence until at least 5 months after vacuolization is histologically apparent.

Glucose metabolic pathways in the anaerobic rumen fungus Neocallimastix frontalis EB188.

Primary pathways for glucose metabolism were established in the anaerobic rumen fungus Neocallimastix frontalis EB188. This highly capable cellulolytic organism demonstrated a strict anaerobic integration of metabolic pathways. Glycolysis in N. frontalis EB188 was coupled to malate dehydrogenase, 'malic' enzyme and specified hydrogenosome reactions. Pyruvate, as in most life forms, was a pivotal compound. The major fermentation products of N. frontalis EB188 were acetate, ethanol and lactate, with the concomitant generation of H2. On the basis of its unique characteristics and streamlined fermentation pathways, it was concluded that N. frontalis EB188 should be an important contributor to programs generating energy and selected chemicals from currently intractable biomass.

Methyl viologen as a preferred electron acceptor in metabolic experiments.

Widely used artificial electron acceptors, including dichlorophenol-indophenol, methylene blue, and phenazine ethosulfate, were shown to induce the nonenzymatic decarboxylation of pyruvate. This characteristic rendered these electron acceptors unsuitable for use in metabolic experiments. In contrast, methyl viologen stimulated the pentose phosphate pathway in mouse preimplantation embryos without decarboxylating pyruvate in the process. Furthermore, methyl viologen had no effect on the Embden-Myerhoff pathway and was not overtly toxic to mouse embryos. These features made methyl viologen a preferred artificial electron acceptor for metabolic studies.

Quantity and quality of embryos collected from mice passively immunized against estradiol.

The objective of this study was to determine the effects of passive immunization against estradiol on the occurrence and timing of estrus, ovulation and fertilization rates and on early embryonic development in mice. Swiss Webster female mice were randomly assigned to one of the three treatment groups to be injected with 0.1 ml saline (control; n=15), 0.1 ml anti-estradiol antisera (high dose: n=17) or 0.1 ml antiestradiol antisera diluted 1:10 with saline (low dose; n=17) at seven weeks of age. Immediately after injection mice were placed with males and observed daily for the presence of vaginal plugs for 10 d. Three days after vaginal plugs were observed, mice were terminated and the uteri were removed and flushed to determine the number and quality of unfertilized ova and embryos. No differences were observed in the timing of vaginal plug formation, the proportion of mice with vaginal plugs, or the mean number of unfertilized ova or embryos collected from each treatment group. However, the proportion of excellent or good quality embryos was reduced in the high dosage treatment. It was concluded from this study that passive immunization of mice against estradiol did not increase the number of embryos obtained from mice and that a high dosage of antisera against estradiol reduced embryo quality. These results provide evidence that alterations in levels of estradiol may adversely affect embryonic development.

Determination of pentose phosphate and Embden-Meyerhof pathway activities in bovine embryos.

Quantitative determination was made of the activity of pentose phosphate pathway (PPP) and Embden-Meyerhof pathway (EMP) in individual bovine embryos from the six-cell to the hatched blastocyst stage. Embryos were collected from superovulated cross-bred heifers and classified into good and poor categories. A single embryo in 1 microl of medium was mixed with 2 microl of medium containing 3 to 30 nCi radiolabeled glucose previously placed on a detached lid of the 1.5-ml polypropylene microcentrifugé vial. The lid was then fitted to its vial which had been loaded in advance with 1.5 ml of 0.1 N NaOH. Vials were then incubated at 37 degrees C for 3 h. At the end of the incubation period, a 1.5-ml NaOH trap was inverted and placed into a 20-ml scintillation vial containing 10 ml of aqueous counting solution and counted in a liquid scintillation spectrophotometer. The PPP activity in good-quality embryos was greatest at the six-cell stage and decreased with increasing embryo development. The EMP activity showed the reverse trend. Poor- quality embryos had a lower glucose metabolism and higher PPP activity. Similar measurements were made on embryos following 24 h of culture, and total glucose metabolism and percentage of PPP activity were increased. In conclusion, these data suggest that in good quality bovine embryos total glucose utilization is low until 16-cell stage, with PPP being the predominant pathway. Total glucose utilization increases significantly at the morula stage; EMP activity increases with increasing embryo development; and PPP activity increases significantly in poor quality embryos and in embryos 24 h in culture.

Pattern of sex steroids secretion and their relationship with embryo yield in Jersey cows superovulated with PMSG.

The levels of progesterone and estrogen secretion were studied in relationship to the superovulatory response in Jersey cows. Progesterone and estrogen concentrations were measured in superovulated Jersey cows with the objective of correlating the patterns of steroid secretion with embryo yield and quality. Pregnant mare serum gonadotropin (PMSG) was used in combination with prostaglandin F(2) alpha analogue to induce superovulation in 18 multiparous, cyclic cows. Serum progesterone and estradiol levels from cows which exhibited estrus within 24 to 48 h after prostaglandin administration (n=13) were used to estimate the superovulatory response. Sex steroid concentrations at the day of estrus (Day 0) was a strong indicator of embryo yield. Progesterone was negatively (r=-0.56) and estrogen positively (r=0.80) correlated to the number of embryos collected. Dramatic increase in progesterone from Day 0 to Day 7 was a significant indicator of embryo yield. A higher rise of estrogen in the follicular phase was an indicator of a larger number of growing follicles and, consequently, better superovulatory response. Nonresponding animals did not show any significant change in the hormonal profile from the day of PMSG treatment to the day of embryo collection. The estimation of progesterone and estradiol concentrations, simultaneously, gave a more objective prediction of embryo yield.

Glucose utilization by sheep embryos derived in vivo and in vitro.

Embryos were collected from superovulated donors at various intervals from onset of oestrus, ranging from Day 1.5 to Day 6. In addition, blastocysts obtained from the culture of 1-cell embryos collected in vivo or of oocytes matured and fertilized in vitro were used to assess the effects of in vitro manipulation and culture on glucose utilization. Glycolytic activity was determined by the conversion of [5-3H]glucose to 3H2O, and oxidation of glucose was determined by the conversion of [U-14C]glucose to 14CO2. Glucose utilization increases significantly from the 8-cell stage and during compaction and blastulation. Glucose oxidation was at a relatively low level (5-12% of total utilization) compared with glycolysis. No difference was observed between the glycolytic activity of blastocysts derived from in vivo or in vitro sources. However, glucose oxidation was lower (P less than 0.05) in blastocysts derived from the culture of 1-cell embryos or from oocytes matured and fertilized in vitro. Exogenous tricarboxylic acid cycle substrates (i.e. pyruvate and lactate supplied in the medium) affected the level of glucose oxidation.

Evaluation of contact dermatitis using the TRUE patch test.

Patch testing is a dermatological diagnostic procedure for contact dermatitis utilizing selected allergens. It has been suggested that suspected cases of contact dermatitis cannot be definitively diagnosed without a patch test. Physicians have avoided the use of patch testing for a variety of reasons, chief of these being inconvenience and inconsistency. The TRUE patch test (Pharmacia AB, Uppsala, Sweden) offers a test modality that circumvents these problems. Background information on the TRUE test is presented along with two studies demonstrating its efficacy.

Bovine amniotic and allantoic fluids for the culture of murine embryos.

This study evaluated bovine amniotic and allantoic fluids as culture media for two-cell murine embryos to the hatched blastocyst stage. Amniotic and allantoic fluids were collected from four 70-d periods of pregnancy and pooled from at least five different animals. In Experiment 1 (n=470) the fluids were frozen twice. Treatments consisted of twice frozen amniotic or allantoic fluid from each pregnancy period, Whitten's medium and fetal calf serum. The later two media were controls. Twice-frozen amniotic fluid<70 d pregnancy period, fetal calf serum and Whitten's medium supported the development of embryos to the hatched blastocyst stage. Whitten's medium was superior to twice-frozen amniotic fluid<70 d pregnancy period or fetal calf serum (P<0.01). Biochemical analysis showed lower glucose in amniotic and allantoic fluids than in Whitten's medium. Experiment 2 (n=425) was performed to evaluate the effect of glucose supplementation to amniotic fluid. No benefit of glucose supplementation of the amniotic fluid was observed. In Experiment 3 (n=432), the fluids were transported nonfrozen on ice. Treatments consisted of nonfrozen amniotic fluid<70 d pregnancy period; nonfrozen amniotic fluid<70 d pregnancy period+glucose), nonfrozen allantoic fluid<70 d pregnancy period; and Whitten's medium. The percentages of embryos developing to hatched blastocyst stage were 66.6, 56.5, 57.4 and 63.9% respectively, for each of the four treatments. No differences were found between any two treatments (P<0.05). In Experiment 4 (n=231) the fluids were stored at -20 degrees C for 15 d. Whitten's medium was superior to amniotic or allantoic fluid<70 d pregnancy period in sustaining embryo development (P<0.05). In conclusion, these data indicate that nonfrozen bovine amniotic or allantoic fluid<70 d pregnancy period can support the development of murine embryos to the hatched blastocyst stage comparable to culture in Whitten's medium. Glucose supplementation of the amniotic fluid offered no advantage, and freezing of fluids had an adverse effect on in vitro embryo development.

Expression of beta-galactosidase in preimplantation ovine and porcine embryos.

Knowledge regarding the timing of embryonic expression of the mammalian genome is of relevance for the development of preimplantation diagnostic methods for human genetic diseases. For development of preimplantation diagnosis of lysosomal storage diseases, it will be necessary to know at which embryonic stage the genes for lysosomal enzymes are expressed. In previous studies by other investigators, it has been shown that lysosomal alpha- and beta-galactosidase and beta-glucuronidase in murine embryos increase 50- to 100-fold in activity between the two-cell and late blastocyst stage. We describe here expression of lysosomal beta-galactosidase in preimplantation ovine (two-cell through midblastocyst) and porcine (two-cell through late blastocyst) embryos. Expression of beta-galactosidase in ovine and porcine preimplantation embryos followed a similar rate of increase as that described for murine embryos. Activity of beta-galactosidase increased over 10-fold between the two- to four-cell and midblastocyst stages in ovine embryos, and 300-fold between the two- to four-cell and late blastocyst stages in porcine embryos. Activity expressed on a per cell basis was relatively constant in ovine embryos, as has been described in murine embryos, and increased approximately 5-fold on a per cell basis in porcine embryos. Activity of beta-galactosidase in ovine and porcine embryos initially was greater than 12-fold on a per cell or per embryo basis than in murine embryos evaluated. The knowledge of beta-galactosidase embryonic expression may provide the basis for preimplantation diagnosis of genetic beta-galactosidase deficiency in these species.

Deep freezing of mouse one-cell embryos and oocytes using different cryoprotectants.

The objective of this study was to compare iso-osmolar concentrations (1.5 M) of 1,2-propanediol, glycerol, dimethylsulphoxide and a combination of 1 M propanediol + 0.5M glycerol (PDGLY) as cryoprotectants for murine ovulated oocytes and one-cell embryos. A higher (P < 0.01) percentage of one-cell embryos developed to the two-cell stage when frozen-thawed with 1,2-propanediol (83%) as compared with glycerol (43%), dimethylsulfoxide (51%) or PDGLY (7%). Data recalculated on the basis of two-cell embryos/number of normal one-cell embryos after thawing indicated no differences among single cryoprotectant groups. More (P < 0.01) frozen-thawed, in-vitro fertilized oocytes developed to the two-cell stage when 1,2-propanediol (35%) was used as cryoprotectant as compared with glycerol (15%). Freezing-thawing resulted in a reduced number of two-cell embryos after oocytes were fertilized in-vitro as compared with fresh oocytes. 1,2-propanediol was a better cryoprotectant than glycerol, dimethylsulphoxide or PDGLY for deep freezing of murine oocytes or one-cell embryos.
