Distribution and molecular forms of C-type natriuretic peptide in plasma and tissue of a dogfish, Triakis scyllia.

A radioimmunoassay (RIA) to measure C-type natriuretic peptide (CNP) of a dogfish, Triakis scyllia, was established, and plasma and tissue levels of CNP were measured. Molecular forms of CNP in plasma and tissues were also examined using a combination of the RIA and cation-exchange high-performance liquid chromatography (HPLC). The antibody used in the assay cross-reacted with all forms of Triakis CNP, as well as eel CNP, against which the antibody was raised. The antibody exhibited no cross-reaction with any atrial, brain (B-type), and ventricular natriuretic peptides examined and showed a weak cross-reaction with porcine CNP. The detection limit of this assay was 0.8 fmol/tube of Triakis CNP-22 which was used as standard. The CNP level in the Triakis plasma was 1.97 +/- 0.38 pmol/ml (n = 5) which far exceeded the physiological levels of any natriuretic peptides in other species. Among various tissues examined, the highest concentration of CNP was measured in the atrium, followed by the ventricle, brain, and pituitary. Low levels were detected in the kidney, liver, and digestive tracts. HPLC analyses revealed that the major form of CNP in the brain was CNP-22, while it was proCNP (CNP-115) in the heart. In contrast to other species from teleosts to mammals thus far examined, the majority of CNP in dogfish plasma was prohormone instead of processed, mature forms.

Coculture of mouse embryos with cryopreserved human oviduct epithelial cells.

PURPOSE: The effect of coculturing mouse embryos with cryopreserved human oviduct epithelial cells was investigated. The cryopreserved cells in Cellbanker were thawed and cultured in Richard D. Goldsby culture medium to establish monolayers. Two-cell-stage mouse embryos were cultured alone (control group) or cocultured with monolayers established from cryopreserved cells (cryopreserved coculture group) or from fresh cells (fresh coculture group). The rates of embryo development and the qualities of the blastocysts in the three groups were compared. RESULTS: The two coculture groups had significantly higher blastocyst development rates (cryopreserved coculture group, 81.6%; fresh coculture group, 82.2%) than the control group (63.1%). The two coculture groups had significantly more blastomeres (cryopreserved coculture group, 108.3 +/- 25.9; fresh coculture group, 108.4 +/- 25.1) than the control group (87.7 +/- 31.9). CONCLUSION: The method of cryopreservation of human oviduct epithelial cells using Cellbanker is simpler than conventional cryopreservation methods. These cryopreserved human oviduct epithelial cells may provide a constant supply of cells for coculture for in vitro fertilization and embryo transfer.

Modelling of movement of the lamprey: control of oscillators.

This article discusses the control methods of the central pattern generator (CPG). First a control model of the CPG is presented using 2 oscillators, and we suggest that phasic modulation to the CPG by means of phasic information is effective for controlling the phase difference between oscillators. Next, two models for controlling the CPG of a lamprey are proposed. One model describes a control system from the brain stem, in which the reticulospinal neurons control the CPG by receiving feedback signals and sending control signals to the neck region of the CPG. The other is a model for learning an localized control system to generate a desired motor pattern. By means of these models, a role of the efference copy is suggested.

[Survival of Vibrio cholerae O139 Synonym Bengal in water from a river].

Survival of five strains of Vibrio cholerae, including serotypes O139 Synonym Bengal, O1 El Tor, and non-O1 were compared in water from a river. These bacteria were mixed with water from a river and the water filtered through 0.45 micron millipore filters, respectively, to yield a concentration of 10(6) CFU/ml and incubated at 5 degrees C and 20 degrees C for 21 days. The survival curve of V. cholerae O139 was almost the same with V. cholerae O1 and non-O1. The number of these bacilli decreased to less than 10(2) CFU/ml at 7 days of the incubation at 20 degrees C. When incubated at 5 degrees, however, these bacilli survived much longer and the number decreased to the same value after 14 days. Therefore, these results indicated that the temperature during the incubation greatly affects the survival of V. cholerae. On the other hand, when the water was filtered and used for the experiment, V. cholerae survived longer than in the polluted water. From these observations, if the river is polluted with V. cholerae O139 in the future, it is suggested that the distribution of V. cholerae O139 in the river may be the same as the present condition with V. cholerae O1 and non-O1.

Aftereffects of high-intensity DC stimulation on the electromechanical performance of ventricular muscle.

To clarify the mechanisms underlying cardiac dysfunction after electrical defibrillation, we investigated the effects of direct current field stimulation (10 ms, 1-80 V/cm) on isolated guinea pig papillary muscles. Shocks (S2) > 15 V/cm lowered the plateau height of the S2-induced action potential and inhibited its terminal repolarization. Subsequent responses to basic stimuli (S1, 1.0 Hz) for 1-3 min were characterized by a decrease in the maximum diastolic potential, a shortening of action potential duration, and an increase of the developed tension. With S2 > 30 V/cm, a marked delay in repolarization of the S2-induced action potential was followed by oscillation of membrane potential, resulting in repetitive spontaneous activity and often refractoriness to S1 stimulation. The aftereffects were independent of the phase of S2 application. Most of the aftereffects were preserved in the presence of nifedipine (1 microM) or ryanodine (1 microM). Only sodium channel blockade by tetrodotoxin (10 microM) modified the aftereffects by depressing the generation of spontaneous activity. These findings suggest that strong shocks (> 15 V/cm) will produce abnormal arrhythmogenic responses probably through a transient rupture of sarcolemmal membrane (electroporation) leading to a disturbance of the ionic equilibrium of the myocyte.

Against-the-rule astigmatism judged at one week after phacoemulsification and aspiration procedures.

To determine the characteristics of astigmatism 1 week postoperatively or whether the final astigmatism is predictable from that present 1 week postoperatively, we analyzed 544 postcataract against-the-rule (AR) cases of astigmatism among 1,649 standard phacoemulsification cataract operations, all utilizing 10-0 Mersilene in double-running fashion. Postoperative AR astigmatism < or = 1.0 dpt increased markedly 1 week postoperatively (2.72 +/- 0.09 dpt) but decreased 6 months postoperatively. However, in postoperative AR astigmatism > 2.0 dpt, the increase 1 week postoperatively was not as great (2.16 +/- 0.13 dpt) but tended to continue to increase even 6 months postoperatively, with a flatter corneal curvature being the responsible factor. Eyes with less astigmatism 1 week postoperatively showed exacerbation of final astigmatism 6 months postoperatively. This course was different from that of small incision cataract surgery.

Reduction of intraocular pressure after phacoemulsification and aspiration with intraocular lens implantation.

We prospectively evaluated the pre- and postoperative intraocular pressures (IOP) of 1,118 cataract patients. All surgical procedures were performed by the same skilled surgeon using the same method of phacoemulsification and aspiration with intraocular lens implantation. The mean +/- SE preoperative IOP was 13.29 +/- 0.12 mm Hg, while the mean IOP values were 10.87 +/- 0.12, 9.94 +/- 0.09, and 10.43 +/- 0.14 mm Hg 1, 3, and 6 months, respectively, after surgery. When the preoperative IOP was in the low 20 s, a decrease of approximately 40% was achieved. The present study shows that the IOP decreased postoperatively to a greater extent than previously considered, and the rate of decrease was dependent on the initial measurement.

Alterations in plasma concentrations of natriuretic peptides and antidiuretic hormone after subarachnoid hemorrhage.

BACKGROUND AND PURPOSE: Hyponatremia is a common complication after subarachnoid hemorrhage. In this study we investigated the relations among hyponatremia, plasma natriuretic peptides, and antidiuretic hormone concentrations after subarachnoid hemorrhage. METHODS: Blood samples for radioimmunoassay measurement of plasma brain natriuretic peptide-like immunoreactivity, atrial natriuretic peptide-like immunoreactivity, and antidiuretic hormone were obtained every 2 to 4 days until day 14 after subarachnoid hemorrhage. RESULTS: Eleven of 20 patients with verified subarachnoid hemorrhage demonstrated mild hyponatremia (126 mEq/L < serum sodium < 135 mEq/L) during their clinical course. Atrial natriuretic peptide and antidiuretic hormone concentrations were significantly elevated on days 0 to 2 after onset of subarachnoid hemorrhage. Atrial natriuretic peptide concentrations remained high in patients who developed mild hyponatremia on days 6 to 14 after onset of subarachnoid hemorrhage. In contrast, antidiuretic hormone concentrations became significantly lower during the second week in these patients. CONCLUSIONS: Mild hyponatremia after subarachnoid hemorrhage may be attributable not to the syndrome of inappropriate secretion of antidiuretic hormone but to cerebral salt-wasting syndrome. Atrial natriuretic peptide may be a causal natriuretic factor in cerebral salt-wasting syndrome.

The role of superoxide anions in the pathogenesis of cerebral vasospasm.

BACKGROUND AND PURPOSE: To determine the role of superoxide anions in the pathogenesis of cerebral vasospasm after aneurysmal subarachnoid hemorrhage, we studied the preventive effect of human recombinant copper-zinc superoxide dismutase (h-r SOD) in a rabbit subarachnoid hemorrhage (SAH) model. METHODS: Forty-five rabbits receiving intracisternal injection of 3 mL autologous nonheparinized blood or 3 mL saline were divided into four groups as follows: (1) saline injected and no treatment (control group, n = 6); (2) blood injected and no treatment (SAH group, n = 20); (3) blood injected and treated by multiple intracisternal injections of 30,000 U of h-r SOD in 0.5 mL saline (SOD group, n = 9); and (4) blood injected and treated by multiple intracisternal injections of 0.5 mL saline (saline group, n = 10). Serial angiograms were performed after the blood injection, and the diameter of the basilar artery was measured. Three animals from the control group and five animals from the SAH and SOD groups each were killed 2 days after SAH, and their basilar arteries were processed for transmission electron microscopic observations. RESULTS: In the SAH and saline groups, the diameter of the basilar arteries was significantly reduced (28 +/- 14% and 27 +/- 9%, respectively) at 2 days after the blood injection, then recovered to pre-SAH levels until 11 days. In the SOD group, the diameter of the basilar artery was only minimally changed during the follow-up period. Transmission electron microscopy revealed endothelial injury in all basilar arteries in the SAH group, whereas endothelial injury was minimal in the SOD group. CONCLUSIONS: We determined that h-r SOD prevents the occurrence of vasospasm, possibly as a result of preventing endothelial injury initiated by superoxide anions.

Surgically induced angiogenesis to compensate for hemodynamic cerebral ischemia.

BACKGROUND AND PURPOSE: The ischemic brain may stimulate angiogenesis to compensate for impaired circulation. We examined the conditions promoting such angiogenesis to provide the basis for surgical treatment. METHODS: The degree of cerebral hemodynamic stress was studied in patients with moyamoya disease using the stable xenon-enhanced computed tomographic acetazolamide tolerance test and positron emission tomography. Patients were subjected to surgery in which scalp arteries were placed on the cerebral cortex without vessel-to-vessel anastomosis. Formation of the newly vascularized collateral network connecting the implanted artery to cortical arteries was assessed angiographically 12 to 17 months after surgery. RESULTS: Preoperative average resting cerebral blood flow for cortex that developed revascularization of cortical arteries was not significantly different from that for cortex that did not. However, cortex that developed revascularization had an average preoperative increase of blood flow by acetazolamide treatment of -3.29 +/- 4.6 mL/min per 100 cm3 (n = 20), which was significantly less (P = .0034) than that of cortex that did not show revascularization (20.7 +/- 4.3 mL/min per 100 cm3; n = 9). Good revascularization developed when the cortex showed increase of blood flow by acetazolamide treatment of less than 0 (steal phenomenon). Preoperative positron emission tomography data indicated that revascularization developed when the cortex was under "misery perfusion." Postoperative hemodynamics were ameliorated by revascularization. CONCLUSIONS: Angiogenesis to connect the implanted scalp arteries to the cerebral cortical arteries was selectively initiated when ischemia of hemodynamic origin existed.

[Enhancement of rectal absorption of rifampicin by sodium para-aminosalicylate dihydrate in human subjects].

The suppositories of rifampicin (RFP) containing sodium para-aminosalicylate dihydrate (PAS-Na) were prepared in order to enhance the rectal absorption of RFP. By the addition of PAS-Na, the in vitro release of RFP from the suppositories was enhanced and the hardness of the suppositories decreased. The rectal absorption of RFP from the suppositories containing no PAS-Na (control suppositories) was significantly lower compared to oral administration of it (26%) in human subjects. When PAS-Na was added to the suppository (300 mg), both the area under the plasma concentration-time curve (AUC) and the maximum plasma concentration (Cmax) increased significantly compared to those of the control suppositories. The rectal absorption of PAS-Na itself from the suppositories seemed to be fast. PAS-Na might increase the absorption of RFP dissolved in the rectal fluid from the suppositories, but not affect the undissolved RFP.

Modulation of production of matrix metalloproteinases from ovine endometrial cells by ovine trophoblast interferon.

Ovine trophoblast interferon modulates the secretion of a number of proteins by ovine endometrium, but only one of these proteins has so far been identified. We examined the effects of trophoblast interferon on the secretion of matrix metalloproteinase-1, -2 and -3 by cultured ovine endometrial cells and determined whether they are mediated via effects on prostaglandin synthesis. Both ovine trophoblast interferon (30 ng ml-1) and human recombinant interferon alpha (50 U ml-1) inhibited the production of latent matrix metalloproteinase-1 and -3 (P < 0.05), as measured by enzyme assays, but had no effect on the secretion of latent matrix metalloproteinase-2. These inhibitory effects were not overcome by PGE2 or PGF2 alpha (each 10 mumol l-1) either alone or in combination. Indomethacin (12 mumol l-1) similarly inhibited the production of latent matrix metalloproteinase-1 and -3, but production was partially restored by adding the prostaglandins either singly or in combination. PGE2 and PGF2 alpha together had no effect on enzyme production. These data were confirmed by gelatin and casein zymography. Northern analysis showed a 4.5-fold increase in the abundance of specific mRNA for latent matrix metalloproteinase-1 following treatment of cells with phorbol myristate acetate, but a marked decrease following interferon treatment. Thus, ovine trophoblast interferon inhibits the production of the latent forms of matrix metalloproteinase-1 and -3 by ovine endometrial cells, and this is independent of its effect on prostaglandin production.

Trans-suppression of gene expression by hepatitis C viral core protein.

We have demonstrated that the truncated hepatitis C (HCV) core protein with its C-terminal hydrophobic domains deleted is translocated to the nucleus of transfected cells (22). In this study, intact and truncated core proteins of HCV were transiently expressed in a human hepatoblastoma cell line, HepG2, and their effects on the expression of the chloramphenycol acethyl transferase (CAT) gene driven by viral and cellular promoters were examined. The intact core protein of 22 kDa which is localized in the cytoplasm of the transfected cells suppressed the expression in all of the promoters tested. They were promoters of the SV40 early region, the c-fos oncogene, the retinoblastoma susceptibility gene, the beta-interferon gene and the beta-actin gene. In contrast, the truncated HCV core protein located in the nucleus did not show such a suppressive activity. The HCV core protein appears to function not only as a viral structural protein but as a regulator of gene expression and it might act as a suppressive factor for the cellular gene expression.

[Mass screening for prostate cancer at a local town--five-year results and screening system].

The results of a mass screening examination for prostate cancer conducted from 1989 to 1993 at a local town, Kawagoe-cho, in Mie Prefecture were evaluated. Among the 216 examinees, 4 were found to have prostate cancer. The most accurate examination was the prostate specific antigen (PSA), which was followed by digital examination and transrectal ultrasound. The applicants for the prostate cancer screening accounted for only 8% of the Kawagoe-cho male residents over 40 years old. An educational campaign of prostate disease in the area must be started to increase the number of applicants. We concluded that the most effective modality for the screening program was a combination of PSA and digital examination in the field study, and transrectal ultrasound accompanied by systemic biopsy results in the second tool for screening.

Reduction of the severity of bronchial hyperresponsiveness by the novel leukotriene antagonist 4-oxo-8-[4-(4-phenyl-butoxy)benzoylamino]-2- (tetrazol-5-yl)-4H-1-benzopyran hemihydrate.

There is much evidence that the cysteinyl-leukotrienes (C-LTs) are important in the pathogenesis of asthma. The clinical effect of a new leukotriene antagonist, ONO 1078 (4-oxo-8-[4-phenylbutoxy)benzoylamino]-2-(tetrazol-5-yl)-4H- 1-benzopyran hemihydrate, CAS 103177-37-3) on symptoms, pulmonary lung function and bronchial hyperresponsiveness was evaluated in patients with bronchial asthma. Eleven patients were treated for 24 weeks with 450 mg of ONO 1078 twice daily. The score of asthma symptom severity and the number of inhaled procaterol were significantly reduced after 2 weeks of ONO 1078 treatment and remained decreased for another 22 weeks. Their FEV1 and PC20 to histamine significantly improved 12 and 24 weeks after ONO 1078 treatment. The effectiveness of ONO 1078 suggests that the C-LTs play an important role in the pathogenesis of asthma. ONO 1078 might help to favorably modify the pathophysiologic condition in patients with bronchial asthma.

Remarkable elevation of interleukin 6 and interleukin 8 levels in the bone marrow serum of patients with rheumatoid arthritis.

OBJECTIVE: Characteristic cellular changes have previously been reported in the bone marrow of patients with rheumatoid arthritis (RA). We investigated the levels of various cytokines in RA bone marrow. METHODS: We studied 25 patients with RA (22 women and 3 men) and 10 trauma patients (7 women and 3 men) as non-RA controls. Twelve kinds of cytokines [interleukin (IL)-1 alpha, IL-1 beta, IL-2, IL-3, IL-4, IL-6, IL-7, IL-8, granulocyte colony stimulating factor, granulocyte/macrophage colony stimulating factor, tumor necrosis factor (TNF)-alpha, and TNF-beta] were assayed by ELISA in iliac bone marrow serum (BMS), tibial BMS, and peripheral blood serum. RESULTS: Markedly elevated levels of IL-6 and IL-8 were detected in iliac BMS, and much lower levels were found in tibial bone marrow and peripheral blood serum. The levels of IL-6 and IL-8 in iliac BMS showed a close relationship to the extent of synovial proliferation. CONCLUSION: Iliac bone marrow may be an important site for the production or accumulation of IL-6 and IL-8 in RA, and these cytokines may influence synovial proliferation in patients with polyarthritis.

[Prevalence of specific IgE antibodies for Dermatophagoides farinae and six inhalants in the general population].

A seroepidemiologic study of house dust allergy and pollinosis was performed among a general population aged 17 to 69 in Aichi Prefecture, Japan. The prevalence of blood containing IgE antibodies to four perennial antigens and three seasonal antigens was examined among 198 persons whose serum total IgE antibodies were above 40 IU/ml. The prevalence of blood containing IgE antibodies was from 9 to 18 percent among perennial antigens and from 6 to 30 percent among seasonal antigens. These results show that Dermatophagoides farinae, Cryptomeria japonica pollen and Anthoxanthumodoratum pollen are important antigens in this population.

[Secondary leukemia after etoposide treatment involved MLL gene rearrangement].

The 11q23 chromosomal abnormality is frequently observed in infantile leukemia and secondary leukemia, and the translocation associated gene in infantile leukemia is called mixed-lineage leukemia (MLL) gene. A 50-year-old man was admitted because of left axillary lymphadenopathy and IBL like T cell lymphoma was diagnosed by lymph node biopsy. The patient responded to the LSG-9 protocol with complete remission. After 10 months he was readmitted because of fever and was diagnosed acute myeloblastic leukemia by bone marrow aspiration. Chromosome analysis revealed 11q23 abnormality, suggesting that the leukemia was induced by etoposide treatment. Southern blot analysis demonstrated DNA rearrangement in the MLL gene at 11q23. It was suggested that the breakpoint region of the MLL gene in secondary leukemia is the same as that of infantile leukemia.
