Natural radionuclide distribution in soils of Gudalore, India.

The concentration of primordial radionuclides in soil samples of Gudalore Taluk in the Udagamandalam district has been measured from the gamma ray spectrum of the soil. The mean activities of 232Th, 238U and 40K are 75.3 +/- 44.1, 37.7 +/- 10.1 and 195.2 +/- 85.1 Bq kg-1 dry weight, respectively. The average outdoor absorbed dose rate in air at a height of 1 m above ground is 74.3 +/- 27.8 nGy h-1, corresponding to an annual effective dose equivalent of 455.6 microSv. The dose equivalent ranges from 168.3 to 1250.5 microSv. The results have been compared with other global radioactivity measurements and evaluations.

Growth hormone directly stimulates testosterone and oestradiol secretion by rat Leydig cells in vitro and modulates the effects of LH and T3.

The modulatory effect of GH on basal, LH and T3 mediated secretion of testosterone and oestradiol by purified rat (60 day old) Leydig cells was studied in vitro. Percoll gradient purified Leydig cells (1 x 10(3)) were cultured for 48 hours at 34 degrees C in a medium containing different concentrations of rat GH (5-400 ng/mL), after an initial culture for 24 hours at 37 degrees C. GH increased testosterone and oestradiol secretions in a dose dependent manner. While testosterone secretion reached the saturation point with 50 ng GH, oestradiol secretion reached the saturation point with 150 ng GH, followed by diminished secretions. Co-administration of minimum (10 ng) effective does of GH with minimum (25 ng) or maximum (100 ng) effective doses of oLH significantly decreased the testosterone secretion. However, an increased secretion of testosterone was observed when maximum effective doses of rGH (50 ng) and oLH (100 ng) were co-administered. Minimum effective (25 ng) or maximum effective (50 ng) doses of T3 inhibited GH mediated secretion of testosterone in vitro. Oestradiol concentration in the culture medium increased when either dose of rGH was co-administered with the minimum or maximum effective doses of oLH. T3 50 ng augmented the secretion of oestradiol by Leydig cells in the presence of GH. These results indicate that GH acts as a gonadotrophin to stimulate testosterone and oestradiol secretions by Leydig cells, and that it modulates LH or T3 induced secretion of these steroids, depending on the intensity of their stimulation.

Successful replacement of fetal calf serum with human urine for in vitro culture of Leishmania donovani.

Leishmania donovani causes visceral leishmaniasis claiming several thousand lives every year in Indian sub-continent. The etiological agent is grown in cell free media supplemented with fetal calf serum (FCS). Urine from human beings and other mammalian species has also been reported to stimulate growth of Leishmania species No study has been carried out Leishmania donovani. Therefore, we studied the feasibility of culturing Leishmania donovani promastigotes in M199 medium supplemented with 10% human urine and compared the phenotypic and genotypic characters under supplementation with urine vis-a-vis FCS. The growth curve showed no significant difference in the promastigote counts in urine vs. FCS supplementation. The best growth was observed in cultures supplemented with the post-menopausal urine. No difference in antigenic bands and RFLP pattern was seen indicating that no alteration occurred in strain specific characters of the parasites cultured with human urine.

Tensile set behavior of Foley catheter balloons.

The removal of indwelling urinary balloon catheters from patients is usually associated with many problems. The problems such as balloon deflation failure; encrustations on balloons, eyes, and lumen; and catheter associated infections are widely discussed in the literature. The tensile set exhibited by the catheter balloon material could also play a role and further complicate the removal process. This article addresses this issue by comparing the tensile set behavior of the balloon material from three commercially available Foley catheters. The balloon materials were subjected to aging in synthetic urine at 37 degrees C for 28 days to simulate clinical conditions. The deflation time of catheter balloons aged in similar conditions were also measured. It was found that different brands of catheters exhibited statistically significant differences in their properties. The tensile set data of the aged samples could be correlated with the deflation time of the balloons. The clinical significance of the tensile set is also highlighted.

Morphological & ultrastructural changes of platelet concentrates stored in PVC bags.

To assess the effect of storage bags on platelets, we studied the morphological and ultrastructural changes of samples drawn from platelet concentrates (PC) prepared and stored in triple, poly vinyl chloride (PVC) bags, manufactured in India. Using the scanning electron microscopy, we demonstrate formation of long pseudopods, and interaction through these to form aggregates. When platelets were stored at 23 +/- 2 degrees C, morphological changes were severe compared to the deleterious effects when kept at 22 +/- 0.5 degrees C. Ultrastructural analysis also showed that maintenance of discoid shape and prevention of granule secretion could be improved by storing the platelets at 22 +/- 0.5 degrees C. Significant degree of platelet fragmentation took place when the storage temperature was high. The morphology score done for platelets stored at both 22 +/- 0.5 degrees C and 23 +/- 2 degrees C showed that preservation of discoid shape was better with the former.

Biochemical lesions of platelets stored as concentrates in PVC bags.

Acid-base status of platelet suspension during storage is a measure of the gas permeability of the bag material. To assess the efficacy of the bags available in our market to store platelets, we compared biochemical lesions of platelets stored in an Indian polyvinyl chloride (PVC) triple bag against a Japanese PVC bag standardized for 5 days platelet storage. Platelet concentrates prepared in both control and test PVC bags were stored for 72 h. Two ml samples were drawn 1 h after preparation, and then at 24 h intervals, for analysis. Our data show that the mean pH value in the test bags was maintained above 6.5. However, the CO2 tension was high and O2 tension was low. We also analyzed malondialdehyde (MDA) formation which is a measure of arachidonic acid metabolism, and seemed to be unaffected in stored platelets. Lactate dehydrogenase (LDH) was not released into the plasma excessively and hence significant platelet lysis was absent during storage. Hypotonic shock response (HSR) of platelets stored in both test and control bags was comparable, indicating the possibility of satisfactory post-transfusion recovery.

Water vaporization from heated tissue: an in vitro study by differential scanning calorimetry.

BACKGROUND AND OBJECTIVE: The heat balance equation is central to modeling laser vaporization of tissue. Because about 70% of tissue is water, energy loss due to water vaporization becomes an important consideration in modeling. In this study, the rate and pattern of water vaporization from various animal tissues studied by differential scanning calorimetry (DSC) are reported. STUDY DESIGN/MATERIALS AND METHODS: DSC thermograms of fresh muscle, liver, and kidney tissues were made at heating rates of 2 degrees C/min, 5 degrees C/min, and 10 degrees C/min in the temperature range of 30-150 degrees C. Thermograms of vacuum-dried muscle sample were also made to establish that the endotherms obtained were essentially due to the water in the tissue. RESULTS AND CONCLUSION: Two broad peaks-one below and the other above 100 degrees C-are seen in all the endotherms. The peaks are attributed to the structural and compositional features of tissue and tissue water. The two-stage vaporization pattern has importance in modeling tissue heating by a continuous-wave Nd:YAG laser.

Structural studies on bovine bioprosthetic tissues and their in vivo calcification: prevention via drug delivery.

Cardiovascular calcification, the formation of calcium phosphate deposits in cardiovascular tissue, is a common end-stage phenomenon affecting a wide variety of bioprostheses. To study the process of calcification in tissue prosthetics, glutaraldehyde-treated bovine pericardium, dura mater and fascialata were implanted subcutaneously in rats and retrieved 21 days later and thereby morphological findings were correlated with biochemically determined levels of calcium. Transmission electron microscopy showed that calcification primarily involved the surface of collagen fibrils and the interfibrillar spaces. The deposition of calcium was higher with dura and fascia prostheses compared to pericardium. However, the release of Fe3+ ions from chitosan matrix had substantially inhibited the deposits of calcium in all implanted tissues. It seems that the structural and anatomical features of the tissue is one of the important factors for tissue-associated calcification. It is also confirmed that glutaraldehyde-preserved pericardium is the most suitable material for the development of cardiac prosthesis, with an appropriate drug combination therapy for prevention of pathological calcification.

Structural and stereochemical studies of potent inhibitors of glucosylceramide synthase and tumor cell growth.

Analogs and homologs of PDMP were synthesized, based on its structure (D-threo-1-phenyl-2-decanoylamino-3-morpholino-1-propanol). This compound had previously been found to block the synthesis of GlcCer (glucosylceramide). Increasing the acyl chain length from 10 to 16 carbon atoms greatly enhanced the efficacy of the enzyme inhibitor, as did the use of a less polar cyclic amine, especially a pyrrolidine instead of a morpholine ring. Replacement of the phenyl ring by a chain corresponding to sphingosine also yielded a strongly inhibitory material. By using a chiral synthetic route, we showed that the isomers active against GlcCer synthase had the R,R-(D-threo)-configuration. However, strong inhibition of the growth of human cancer cells in plastico was produced by both the threo and erythro racemic compounds, showing involvement of an additional factor (beyond simple depletion of cell glycosphingolipids by blockage of GlcCer synthesis). The growth arresting effects could be correlated with increases in cellular ceramide and diglyceride levels. The aliphatic pyrrolidino compound was strongly inhibitory toward the glucosyltransferase and produced almost complete depletion of glycolipids, but did not inhibit growth or cause an accumulation of ceramide. Attempts were made to see whether the differences in growth effects could be attributed to the influence of the inhibitors on related enzymes (ceramide and sphingomyelin synthase and ceramidase and sphingomyelinase). While some stimulation of enzyme activity was noted, particularly at high inhibitor concentrations (50 microM), these findings did not explain the differing effects of the different inhibitors. The best inhibitors of GlcCer synthase compared favorably in efficacy with some cancer chemotherapeutic drugs in current use when tested with a battery of human cancer cells.

Preparation of omega trideuterated sulfidopeptide leukotrienes: analysis by FAB/MS.

C-20 Trideuterated leukotriene A4 methyl ester was prepared by Wittig condensation of a deuterated C9-phosphonium salt with a C11-epoxydienal. It was then treated separately with glutathione, cysteinylglycine and cysteine. The resulting esters were saponified to give C-20 trideuterated leukotrienes C4, D4 and E4 respectively. Fast atom bombardment mass spectrometry showed that the synthetic compounds contained from 0.5 to 1.0% protium impurity. Introduction of deuterium at C-20 ensures that it is stable to a wide variety of reagents and reaction conditions. The deuterated leukotrienes will serve as internal standards for the quantitative analysis by mass spectrometry of endogenous sulfidopeptide leukotrienes present in biological fluids.

Duration-dependent effect of transient neonatal hypothyroidism on sertoli and germ cell number, and plasma and testicular interstitial fluid androgen binding protein concentration.

The impact of transient neonatal hypothyroidism on growth and function of puberal testis during different milestones of postnatal testicular development was studied in Wistar rats. Rat pups were made hypothyroid for 10, 15, 30, 40 and 60 days of postnatal age from birth by providing 0.05% (W/V) methimazole (MMI) in the drinking water of the mother, from day 1 postpartum till weaning (25 days postpartum) and thereafter in the drinking water. Control rats were raised without MMI treatment. Sertoli cell number and its function was assessed on day 60 postpartum. Sertoli cell number increased consistently in 10, 15, 30 and 40 days transient hypothyroid rats but decreased in rats subjected to continuous hypothyroidism from birth to 60 days postpartum. Rats subjected to continuous hypothyroidism from birth showed spermatogenic arrest at puberty and had only a single layer of spermatogonia. Transient neonatal hypothyroidism for 10 (or) 15 days from birth increased spermatocytes (pachytene and zygotene), spermatids (elongated and round) whereas, that of 30 and 40 days decreases the number of germ cells. Plasma androgen binding protein (ABP) concentration decreased in puberal rats belonging to all groups, whereas the testicular interstitial fluid (TIF) concentration of ABP increased significantly in 10 and 15 days hypothyroid rats while it decreased in all other groups. These findings indicate that the mitogenic activity of Sertoli cell is increased irrespective of the duration of transient neonatal hypothyroidism. However, the functional activity of Sertoli cells (ABP production) in these puberal rats varies depending upon the postnatal period at which the animals were in hypothyroid state.

Recent advances in the diagnosis of leishmaniasis.

Leishmaniasis is a parasitic disease caused by a haemoflagellate Leishmania. There are more than 21 species causing human infection. The infection is transmitted to humans through the bites of female sandflies belonging to 30 species. The disease manifests mainly in 3 forms: the visceral, the cutaneous and the mucocutaneous leishmaniasis. The diagnosis of visceral form is conventionally made by the demonstration of amastigotes of the parasite in the aspirated fluid from the bone marrow, the spleen, and rarely from the lymph nodes, or the liver. The parasite demonstration and isolation rates are rather poor from cutaneous and mucocutaneous lesions due to low parasite load and high rate of culture contamination. Recently several recombinant proteins have been developed to accomplish accurate diagnosis. Recombinant kinesin protein of 39 kDa called rK 39 is the most promising of these molecules. The antigen used in various test formats has been proved highly sensitive and specific for visceral leishmaniasis. It is useful in the diagnosis of HIV-Leishmania co-infection and as a prognostic marker. Molecular techniques targeting various genes of the parasite have also been reported, the PCR being the most common molecular technique successfully used for diagnosis and for differentiation of species.

Synthesis and CNS activities of 2-methyl/5-chloro-8-(3-substituted propoxy)quinolines.

In the present study, a series of 2-methyl/5-choloro-8-(3-substituted propoxy) quinolines were synthesized. The compounds were characterized by IR, 1H-NMR and elemental analysis. The compounds were investigated for anticonvulsant and decrease in the elevated motor activity by introceptive chemical stimuli (amphetamine antagonistic activity) at the dose level of 80 and 160 mg/kg. Compounds 2, 10, 11, 12 and 13 exhibited significant anticonvulsant and amphetamine antagonistic activity. Compounds 2 and 11 exhibited highest anticonvulsant and amphetamine activity respectively.
