Quantitative suspension tests performed with Staphylococcus aureus ATCC 6538 and different preparations of povidone-iodine (PVP-I) show that the germicidal activity of a preparation depends on the content of free, non-complex-bound iodine. A comparison between the results obtained with different PVP-I preparations showed that other substances present influenced microbicidal activity.

The number of chlorine-impaired E. coli organisms obtainable with the aid of the membrane-filtration-Endo-agar-method is so reduced that this method cannot be considered equivalent to liquid enrichment. Notwithstanding the well documented difficulties that arise from differences between batches of the same filters e. g. from problems to assure their standardization, the main obstacles do not stem from the questionable suitability of membrane filters but from the toxicity of the Endo-agar. The use of lactose-pepton (L.P.)-broth (German Standard Methods 1971) equally does not lead to satisfactory results if compared to the use of casein-soya (C.S.)-broth.
A description of a casein-soya-lactose broth (CSLB) for the cultivation of chlorine-impaired E. coli bacteria is given. In this liquid medium the recovery rates of chlorine impaired E. coli are superior or at least equal to recovery rates observed when casein-soya-broth (CSB) is used. Differences are regularly seen when lactose-pepton-broth (LPB) according the German Standards (DEV.K6) is used between direct inoculation into a liquid enrichment medium and inoculation into the same medium following membrane filtration are no longer found when CSSL-broth is used instead of LPB.

The particular problems that arise in connection to prophylaxis to infections, food hygiene, sanitation, refuse disposal and the maintenance lavoratory facilities are emphasized. Relevant rules and recommendations are quoted.

The preparation of suspensions of bacterial cells impaired by the action of chlorine require a very accurate standardization of all the procedural detail of controlled experimental impairment by chlorine. All measures suitable to spread the kinetics of reduction of bacterial numbers in time will facilitate the reproducibility of results. The exact amount of hypochlorite--determined experimentally prior to the actual test--is transformed to chloramine at 0 degrees C then a defined quantity of bacteria is added. After different periods of contact samples are taken and stabilized by adding thiosulphate of sodium. When the bactericidal action is not abrupt but sufficiently drawn out a relatively high proportion of bacteria impaired by chlorine but still viable is obtained (50-90%). In suspension the proportion remains relatively stable during storage in the refrigerator (4 degrees -6 degrees C). Resuscitation experiments carried out with suspensions of organisms damaged as explained above showed resuscitation times of 3 h in casein soy broth not to grow as well as those after 24 h of resuscitation. Obviously the process of reparation after impairment by chlorine must be assumed to be relatively slow under these conditions.
