Role of the polyadenylate segment in the translation of globin messenger RNA in Xenopus oocytes.

The translations of native messenger RNA for rabbit globin and that of poly(A)-free globin messenger RNA have been compared after injection into Xenopus oocytes. The initial rate of translation of poly(A)-free mRNA is close to that found with intact mRNA. However, at longer incubation periods, the rate of globin synthesis with poly(A)-free mRNA is considerably lower than with native mRNA. Similar differences in the template activity of the two mRNA preparations were found with a cell-free extract of Krebs II ascites tumor. It is concluded that the presence of the 3' poly(A)-rich sequence in mRNA is required to ensure high functional stability.

Glycosylation of VIP receptors: a molecular basis for receptor heterogeneity.

Apparent molecular weights of VIP-binding proteins differ greatly according to species and to tissue. In this study, we used plasma membranes from various species (human, rat, pig) and tissues (melanoma, intestine, liver), which display major 125I-VIP-labeled components with molecular weights ranging from M(r) = 51,800 to 66,800. With the exception of porcine receptor, the various VIP receptors had similar apparent molecular weights after removal of their N-linked carbohydrates. In addition to differences in the amount of asparagine-linked glycans, our results also revealed differences in the composition of the oligosaccharide chains, which can also account for the heterogeneity in the molecular weights of the VIP receptor.

Endothelin-3 gene mutations in isolated and syndromic Hirschsprung disease.

Hirschsprung disease (HSCR, aganglionic megacolon) is a frequent congenital malformation regarded as a multigenic neurocristopathy. Four susceptibility genes have recently been identified in HSCR, namely the RET proto-oncogene, the glial cell line-derived neurotrophic factor (GDNF), the endothelin B receptor (EDNRB) and the endothelin-3 genes (EDN3). Homozygosity for EDN3 mutations has been previously shown to cause the Shah-Waardenburg syndrome, a combination of HSCR with features of the Waardenburg syndrome. Here, we report on heterozygous EDN3 missense mutations in isolatec HSCR. The present data give further support to the role of the endothelin signaling pathway in the development of neural crest-derived enteric neurons. They also suggest the possibility that either recessive or weakly penetrant dominant alleles could occur at the EDN3 locus, depending on the nature of the mutation.
