The effects on merino lambs of chronic infection with Trichostrongylus rugatus.

Four groups of six 5-month-old merino lambs were exposed to infection with 12,000 larvae of Trichostrongylus rugatus per week and one group was killed at each of 3, 6, 9 and 12 weeks. Numbers of adult worms increased in an approximately linear manner throughout the experiment; the percentage of larvae developing to adults remained between 35 and 25% throughout the course of the experiment. Sub-total villus atrophy was evident in the anterior small intestine of each group of infected sheep and significant reductions occurred in the plasma concentrations of albumin and phosphate. Serum globulin and calcium concentrations rose in infected animals. No clinical signs were observed in infected sheep, no alteration in bone histology was detected and no decrease in growth rate occurred.

Comparison of the effects of infection with Trichostrongylus colubriformis, T. vitrinus and T. rugatus in merino lambs.

Groups of six worm-free merino lambs were infected with 100,000 larvae of T. colubriformis, T. vitrinus, T. rugatus or a mixture of the three species and were slaughtered 21 days later. Nematodes were distributed predominantly in the first 3 m of the small intestine where they caused subtotal villus atrophy. T. vitrinus caused severe erosion of the epithelium; erosion was less severe with T. colubriformis infections and mild with T. rugatus infections. Plasma concentrations of albumin declined during the experiment and were most marked in sheep infected with T. vitrinus. Plasma phosphate concentrations declined during infection with all three species, while plasma concentrations of globulin and calcium declined only in sheep infected with T. vitrinus. Food intake was reduced in all groups, but was most marked with sheep infected with T. vitrinus. T. vitrinus appeared to be more pathogenic than the other species.

Effects of temperature and relative humidity on development and survival of the free-living stages of Trichostrongylus colubriformis, T. rugatus and T. vitrinus.

Temperatures of 4 and 10 degrees C reduced the rate of hatching of eggs and the development of the pre-infective stages of T. colubriformis and T. rugatus, but not T. vitrinus. Eggs of T. rugatus hatched at 4 degrees C, but did not develop to the infective stage. At 20 and 30 degrees C, no differences were detected between species in egg hatching or the development of infective larvae. Third-stage larvae of T. rugatus survived for significantly longer periods of time at 33 and 56% relative humidities at a temperature of 20 degrees C than did either T. colubriformis or T. vitrinus. At 30 degrees C, differences were less marked, but T. rugatus and T. colubriformis survived longer than T. vitrinus. The results are discussed in relationship to the known distribution of these species in eastern Australia.

Study of upper extremity growth and development using human embryos and computer-reconstructed models.

This study investigated the apical ectodermal ridge and early vasculature of the human limb bud by use of histologic techniques and a new method of computer modeling. Fourteen human embryos were studied (more than 10,000 sections) microscopically and the embryonic limbs were reconstructed three-dimensionally by use of a serialized programming method. The apical ectodermal ridge consists of an extensive band of specialized cells, located over the rim of the limb surmounting a strip of rapidly-dividing mesenchymal cells. It is consistently supplied by a large vessel that shows extensive central branching.

Identification and characterization of the potato leafroll virus putative coat protein gene.

Complementary DNA clones representing approximately 6100 nucleotides of potato leafroll virus (PLRV) were generated, restriction-mapped, and partially sequenced. Within one of the cDNA clones an open reading frame (ORF) encoding a 23K protein was identified and further characterized. Amino acid sequence comparison of this protein showed significant homology (47.1%) with the barley yellow dwarf virus (BYDV-PAV) coat protein. This and other observations suggested that this gene encodes the PLRV coat protein. Other similarities were observed between PLRV and BYDV sequences in this region of their genomes, including an ORF of 17K within the ORF encoding the 23K putative coat protein.

Double stranded RNA in natural isolates of Neurospora.

Thirty-six wild type isolates of Neurospora were surveyed for the presence of dsRNA. The survey identified seven strains which contain dsRNA molecules. These seven strains are all from different geographic locations. The sizes of the dsRNAs range from 500 bp to 18 kb and a total of seven distinct dsRNA species was identified. Cross homologies of some of the dsRNAs were apparent. There was homology between the 9.0 kb dsRNA and genomic DNA prepared from all strains in the survey, indicating a possible cellular rather than viral origin for this dsRNA species. None of the other dsRNAs hybridized with genomic DNA suggesting a viral origin for these dsRNAs.

An experimental increase in stuttering frequency.

The purpose of the present experiment was to investigate whether stuttering frequency could be increased from baseline by an experimental condition. Initially, 10 adult stutterers spoke spontaneously while alone. Next, they continued speaking, but with an adult, male conversationalist. Finally, the subjects again spoke while alone. Percent stuttering increased in the conversational situation compared with the baseline (alone) situation, and decreased in the final baseline (alone) situation compared with the conversational situation.

Measurement of alveolar gas volume by ambient pressure changes in isolated lungs.

Alveolar gas volume (AGV) may be measured in humans (Peslin et al., J. Appl. Physiol. 62: 359-363, 1987) by applying very slow sinusoidal variations of ambient pressure (delta Pam) around the body and studying the relationship between delta Pam and the resulting gas displacement at the mouth (delta Vaw): AGVapc = (PB.delta Vaw)/(delta Pam.cos phi), where AGVapc is AGV measured by ambient pressure changes, PB is barometric minus alveolar water vapor pressure, and phi is the phase angle between Pam and Vaw. The applicability of this method to excised lungs at various transpulmonary pressures was assessed in six rabbit lungs and three dog lobes by reference to AGV measurements by He dilution (AGVdil) and by a volumetric method (AGVvol). Except in one instance, AGVapc did not change significantly when the frequency of delta Pam was varied from 0.02 to 0.2 Hz. AGVapc was highly correlated (P less than 0.001) to both AGVdil and AGVvol. It did not differ significantly from AGVdil (81.4 +/- 50.6 vs. 80.2 +/- 44.2 ml) and was only marginally higher than AGVvol (64.6 +/- 26.9 vs. 62.4 +/- 24.4 ml, P less than 0.05). We conclude that the method usually provides accurate results in excised lung preparations. Its main advantages are that it does not require manipulating the lung or changing its volume and that the measurement takes less than 1 min.

Clinical, functional and pathological correspondence in early stage idiopathic pulmonary fibrosis: evidence for small airway obstruction 1-2.

We describe the clinical, physiological and pathological features of 23 subjects with early stage idiopathic pulmonary fibrosis. Thirteen subjects who had no symptoms had been fortuitously recruited by a routine chest radiograph, whereas the 10 other subjects complained of dyspnea. Twenty-one subjects showed only light to moderate extent of abnormalities on the chest radiograph. Fourteen subjects had a reduced vital capacity whereas 16 and 17 showed a reduced pulmonary compliance and an increase in lung elastic recoil, respectively. Transfer factor was significantly reduced in 18 subjects. Evidence for significant airway obstruction, mainly located at the peripheral level, was demonstrated by a reduced specific lung conductance and upstream conductance in 13 subjects. Airway obstruction was not associated with smoking habits. Bronchial hyperresponsiveness was noted in 50% of the 18 subjects studied. Although fibrosis was mild to moderate in 15 instances, it was only focal, i.e. at least one zone of normal parenchyma in the lung specimen in 17 subjects. Peribronchial fibrosis was established in 8/11 satisfactory biopsy specimens. Significant correlations were observed between rales, the radiological score, some functional indices and the characteristics of fibrosis. We conclude that small airway obstruction documented by physiological and pathological means is frequent in early stage idiopathic pulmonary fibrosis.

Bronchoconstriction due to exercise combined with cold air inhalation does not generally influence bronchial responsiveness to inhaled histamine in asthmatic subjects.

We investigated immediate and late changes in airway responsiveness to histamine, after bronchoconstriction due to exercise combined with inhalation of cold air, in ten asthmatic subjects who came on six days. On the first visit, the provocation concentration producing 20% fall in FEV1 (PC20) histamine was obtained. After functional recovery, each subject walked on a treadmill whilst breathing dry, cold air. This resulted in an immediate fall greater than 15% (mean +/- SD = 31.9 +/- 11.0%) in forced expiratory volume in one second (FEV1). Following recovery, PC20 was measured again. FEV1 was then monitored for up to 6-8 h. PC20 was measured then and on the two following days at the same time of the day. Subjects were studied on three control days using the same design except that a resting period replaced the exercise with cold air. The mean changes in PC20 at each interval after exercise combined with cold air were not statistically significant. Physiologically significant changes were obtained in only two subjects, reaching a maximum 8 h after the manoeuvre. In these subjects, changes in PC20 were reproducible during a second series of visits. It is concluded that bronchial responsiveness to histamine is not generally influenced by the bronchoconstriction due to exercise combined with cold air.

Kinetics of the recovery from bronchial obstruction due to hyperventilation of cold air in asthmatic subjects.

The timecourse of recovery from bronchial obstruction due to inhaled cold air was studied in eight adult asthmatic subjects. On the first visit, bronchial responsiveness to inhaled histamine was assessed. On the other two visits, after assessment of baseline lung resistance (RL) and spirometry, dry cold (-20 degrees C) air was inhaled for three minutes. RL was monitored continuously until its return to baseline +/- 20%. The baseline concentration of histamine causing a 20% fall in FEV1 (PC20) varied from 0.03 to 5.9 mg.ml-1. The mean maximum increase in RL was 2-fold (2.03 +/- 0.41) and was reached 2-11 min after the challenge. RL values were linearly correlated to time (r2 values greater than 0.80 in 14/16 instances). The two slopes of recovery were not significantly different. Slopes of recovery and total time of recovery (14-55 min) varied greatly between subjects. No relationship was found between baseline airway calibre, bronchial hyperresponsiveness and maximal increase in baseline RL on the one hand and the slopes of recovery on the other.

Fatal poisoning from sodium phosphate enema. Case report and experimental study.

An overdose of a common over-the-counter sodium phosphate enema solution was fatal in an infant. The marked hypernatremia, acidemia, hyperphosphatemia, and hypocalcemia observed before death were also produced in a porcine model. A study using pigs showed that the enema solution was lethal if retained in doses above 20 mL/kg, equivalent to four pediatric-sized enemas in a 2-year-old child. Even normal doses of the enema solution caused measurable changes in serum phosphorus and calcium levels.

Machete wounds to the head: report of three cases.

Three cases of machete wounds to the head are described. The resulting injuries from this unusual mode of trauma varied in severity and included transection of a previously placed ventriculoperitoneal shunt. Complications of these injuries resulted from delayed and inadequate treatment.

Within- and between-day reproducibility of isocapnic cold air challenges in subjects with asthma.

Eight adult subjects with asthma had isocapnic cold air challenges on 4 different days. Three consecutive tests were performed on each visit with functional recovery between tests. Subjects were asked to breathe dry cold air (-20 degrees C) for progressively increasing levels of minute ventilation (7.5, 15, 30, and 60 L/min and maximal voluntary ventilation) until a 20% fall in FEV1 had been reached or when maximal voluntary ventilation was done. FEV1 was assessed between each level. The doses of respiratory heat exchange and minute ventilation causing 10%, 15%, and 20% changes in FEV1 were interpolated from dose-response curves. The within- and between-day 95% confidence intervals based on a single determination on the loge scale varied from +/- 0.32 to 0.59 for the indices derived from respiratory heat exchange. Reproducibility of the between-day results was more satisfactory than for the corresponding within-day assessments. No significant within-day tachyphylaxis was demonstrated for these indices.

Lack of acute effects of ascorbic acid on spirometry and airway responsiveness to histamine in subjects with asthma.

Sixteen adult subjects with asthma in a clinical steady state were studied. On day 1, after baseline spirometry, they underwent four histamine inhalation tests with functional recovery between each test. The provocative concentration causing a 20% fall in FEV1 (PC20) was obtained after each test. On days 2, 3, and 4, after baseline spirometry, active and placebo ascorbic acid (2 gm) was administered orally, double-blind, according to a 4.3.1 two-treatment crossover study design. One hour later, spirometry was performed, and PC20 was reassessed. We found no significant changes in FEV1 and FVC after ascorbic acid as compared with placebo administration. There was no difference between PC20 on days 2, 3, and 4 and by standardizing for the four PC20 results obtained on day 1. We conclude that ascorbic acid has no acute bronchodilator effect and does not alter bronchial responsiveness to histamine in subjects with asthma.

Theophylline partially inhibits bronchoconstriction caused by inhaled histamine in subjects with asthma.

Sixteen adult subjects in a clinical steady state had four consecutive histamine inhalation tests on the same day when they were not receiving oral theophylline medication. The provocative concentration of histamine causing a 20% fall in FEV1 (PC20) was used to assess the response. They were then administered active or placebo sustained-release theophylline preparations according to a double-blind, randomized 4.4.1. two-treatment crossover design. Medication was administered for a minimum of 3 consecutive days, and PC20 was reassessed on 4 different days, 3 to 4 hours after receiving active or placebo medication (two visits for each medication). A significant but small bronchodilator and blocking effect on histamine excitability was demonstrated for the active medication. This latter effect was present even by adjusting for changes in baseline airway caliber and for the intraindividual variability of the four PC20 values obtained on day 1. We conclude that theophylline partially blocks bronchial responsiveness to inhaled histamine.
