Cytogenetics as a tool for triatomine species distinction (Hemiptera-Reduviidae).

Several cytogenetic traits were tested as species diagnostic characters on five triatomine species: Rhodnius pictipes, R. nasutus, R. robustus, Triatoma matogrossensis and T. pseudomaculata. Four of them are described for the first time. The detailed analysis of the meiotic process and the application of C-banding allowed us to identify seven cytogenetic characters which result useful to characterize and differentiate triatomine species.

A prospective randomised European multicentre study of medium-long run mortality and morbidity comparing acetate-free biofiltration and bicarbonate dialysis.

In recent years, the progressive increase in the mean age of the population entering chronic dialysis treatment has been responsible, on the one hand, for the growing number of patients undergoing regular dialysis, and on the other, for the high number of "critical" patients, both as a result of their age and the presence of concomitant morbidity. Thus, dialysis treatment today is not only aimed at waste removal and water-electrolyte homeostasis, but also at a reduction in morbidity and mortality, and at improving the patients' quality of life, thanks to the use of biocompatible materials and the achievement of good cardiovascular tolerance to treatment. Consequently, diffusive-convective dialysis procedures have been on the increase, since they combine better depuration with the use of biocompatible high-flux membranes. Acetate-free biofiltration (AFB) is a diffusive-convective dialysis procedure which utilises a high-flux membrane, AN69, post-dilution infusion of a sodium bicarbonate solution (NaHCO3), and a dialysate which is completely free of any buffer, and thus also free of acetate, which may have various negative effects on the patient. A number of studies have already shown the better hemodynamic stability and the reduction of intradialytic side-effects during AFB. All these, however, were short-term studies. To verify the beneficial effects of AFB in the long run, a three year multicentre randomised European trial has been proposed to compare bicarbonate hemodialysis (BD), a technique used in nearly 80% of the world's dialysis population, and AFB. The specific aim of the investigation is to verify, in a large number of patients, the results of hemodialysis treatment in terms of morbidity, mortality and quality of life. The study involves 80 hemodialysis units across Italy, France, Germany, Spain, Slovenia and Croatia, with enrollment of about 400 patients considered "critical" for at least one of the following reasons: age, diabetes, dialysis cardiovascular instability. Fifty percent of the patients are to undergo AFB with the AN69 membrane and bicarbonate solution infusion (NaHCO3 145 or 167 mEq/lt), and the other fifty percent are to be treated by BD, with any membrane except the nonmodified cellulosic one. Biochemical, cardiological, and nutritional parameters will be considered throughout the study. Mortality, morbidity both in terms of intra- and interdialysis symptoms - and hospitalisation rate, as well as the patients' quality of life, evaluated by the SF36 questionnaire, will be analysed.

[Prevalence of antibodies to hepatitis C (anti HCV) in blood donors in Rio de Janeiro, Brazil. Its relation to ALT and anti HBC].

UNLABELLED: We have studied 933 volunteer blood donors from May to July, 1990. After a interview and screening tests for syphilis, Chagas disease, malaria and HIV, they underwent an enzyme immunoassay for HBsAg, anti HBc and anti HCV antibodies. Alanine aminotransferase (ALT) serum levels were determined by auto analyser. Most blood donors were male with mean age of 33 years (19-65). Anti HCV prevalence was 3.1% (29 from 933 blood donors). Among anti HCV+, blood donors, 44.8% (13/29) had ALT 40 UI/L, 31% (9/29) were anti HBc+ and 17.2% (5/29) had both surrogate markers simultaneously. From 109 donors with ALT 40 UI/L, 13 (11.9%) were anti HCV+, while among 153 anti HBc+ donors, the anti HCV was 5.8%. CONCLUSIONS: 1) we found a higher anti HCV prevalence among our blood donors than previous published reports from other countries; 2) our data show that surrogate assays do not adequately identify anti HCV blood donors, 41.4% of them would not have been excluded by anti HBc and ALT tests alone; 3) there were a correlation between anti HCV positivity with a sample to cutoff optical density ratio equal or greater than 4 and elevated ALT serum levels.

Heterogenous expression of the EGF-receptor in human breast carcinoma.

The epidermal growth factor receptor was analyzed in the membrane fraction from a series of human breast carcinomas using a radioligand assay. The results were compared to immunostaining with monoclonal antibodies on tissue sections. All tumors with detectable receptor levels as measured by ligand binding showed a positive staining with monoclonal antibodies. In such tumors more than 50 percent of the cells were receptor positive with varying intensity. Tumors classified as receptor negative with the radioligand assay could be divided into two major groups based on their immunostaining. The largest group showed no reactivity with monoclonal antibodies. In a second smaller group, a positivity was observed in small clusters of tumor cells while the majority of cells were negative. The staining intensity in such clusters was usually low. It seems possible that this heterogenous expression of the epidermal growth factor receptor is of significance with respect to clonal selection in tumor spreading.

Synergistic inhibitory effect of gamma-interferon and EGF on A431 cells.

Addition of high concentrations (100ng/ml) of epidermal growth factor (EGF) to cultures of the epidermoid-carcinoma cell-line A431, which expresses high levels of the EGF receptor, resulted in 50 percent reduction in growth rate. At a lower level (10 ng/ml) of EGF no such inhibitory effect was observed. Addition of gamma-interferon at a concentration of 100 u/ml, but not alpha-interferon at the same concentration. inhibited the growth rate of A431 cells by 50 percent. Incubation of the cells in medium containing both 10 ng/ml of EGF and 1000 u/ml of gamma-interferon led to a complete growth arrest. After 48 hours this inhibition was not reversible and resulted in excessive cell-death after approximately seven days. The human lung cancer cell-line U-1752, which expresses low levels of the EGF receptor, was not inhibited by EGF even at high concentrations. Gamma-interferon did not increase the sensitivity of these cells to EGF. The results show that the modulating effect of gamma-interferon on EGF activity is variable and probably depends on cell line, receptor concentration as well as the primary effect of EGF on different cells.

Transforming growth factor alpha in human mammary carcinomas and their metastases.

A binding competition assay was used to measure the content of transforming growth factor alpha in human mammary carcinomas and their metastases. Thirty one percent of the primary and 50 percent of the metastases expressed the transforming growth factor alpha with values ranging between 1.6 to 278 EGF equivalent ng/mg tumour extract protein. There was no correlation between the transforming growth factor alpha content and the estrogen receptor value.

Receptors for epidermal growth factor and sex steroid hormones in human colorectal carcinomas.

Iodinated epidermal growth factor was used to identify its specific receptor in 32 human colorectal carcinomas. The receptor was found in 25% of the tumors with values between 6.9 - 104.0 fmoles/mg DNA. Receptors for estrogen and progesterone were detected in 45 and 25% of the tumors respectively. No correlation was observed between short term survival and content of receptors for sex steroid hormones. Four of the 8 patients with detectable levels of epidermal growth factor receptor had experienced relapse.

Receptors for epidermal growth factor in human mammary carcinomas and their metastases.

The tumor cell content of the receptor for epidermal growth factor (EGF-R) and estrogen receptor (ER) was analyzed in 36 primary breast carcinomas and their metastatic deposits. Twenty-five percent of the primary tumors and 48 percent of the metastases exhibited measurable levels of the receptor. The frequency of EGF-R positivity was the same for cutaneous and lymph node recurrences. No discrepancy was observed with respect to ER content between primary tumors and their metastases.

Identification of transforming growth factor alpha in human primary breast carcinomas.

Primary human mammary carcinomas were found to contain an acid stable polypeptide which bound to the receptor for epidermal growth factor (EGF). This polypeptide which was antigenetically different from EGF and thus identified as Tumor Growth Factor (TGF) alpha, could be identified in 25% of the tumors. Among these tumors 40% contained measurable levels of the EGF receptor. Thus these tumors exhibit the molecular prerequisite for autocrine growth stimulation.

[Tomodensitometry: localizing value in hypertension of adrenal origin (author's transl)].

Computarized axial tomography has shown the localization of adrenal lesions in four cases of hypertension of adrenal origin: two phaeochromocytomas, one primary hyperaldosteronism, one Cushing's syndrome. This method has the advantage of being neither invasive, nor time-consuming. Unfortunately, it cannot localize tumors smaller than two cm in diameter. It seems to be highly beneficial in phaeochromocytoma, where it can advantageously take the place of arteriography. It is less fruitful in primary hyperaldosteronism and in Cushing's syndrome because lesions are smaller.
