A study of human growth hormone and insulin gene regions in relation to metabolic control of non-insulin-dependent diabetes mellitus.

The possible association of human growth hormone (hGH) and insulin (INS) gene regions with metabolic control in diabetes was investigated in 98 subjects with non-insulin-dependent diabetes mellitus (NIDDM); 54 control subjects from the same population were also studied. Two polymorphic restriction sites in the region of the hGH cluster (BGLIIA and BGLIIB) show significant association with both glycemic and hemoglobin A1c (HbA1c) levels. Mean values for plasma glucose and HbA1c show a maximum in the BGLIIA *1/*1 genotype and a minimum in the BGLIIA *2/*2 genotype. Mean values for plasma glucose and HbA1c show a maximum in the BGLIIB *1/*2 genotype. The BGLIIA*2/BGLIIB*1 haplotype shows a negative correlation with plasma glucose and HbA1c levels. Since the two markers are located in the area surrounding the hGH-V locus, the expression of this gene in NIDDM warrants further investigation.

ACP1 and human adaptability. 2. Association with season of conception.

We have studied the pattern of association between the season of conception and cytosolic low molecular weight phosphotyrosine phosphatase (ACP1) genetic polymorphism in 329 consecutively newborn infants from the population of Penne and 361 consecutively newborn infants from the population of Rome. In addition, 329 mothers were studied in the population of Penne. A concordant, highly significant association was observed in the two populations between ACP1 parameters and the season of conception of newborn infants. The total activity of ACP1 shows a minimum in infants conceived in January-February and a maximum in those conceived at the end of the solar year. Analysis of the joint mother-newborn ACP1 distribution in relation to the season of fertilisation has shown that among mothers carrying ACP1*A (the allele showing the lowest activity), the proportion of newborns carrying this allele is higher in those conceived in the first months of the year than in those conceived subsequently. Since ACP1 probably functions as a phosphotyrosine phosphatase and as a flavin mononucleotide phosphatase, low activity could enhance the metabolic rate and would be advantageous in a cold environment. The cycle of variation of ACP1 in infants follows the cycle of solar illumination. It is possible that individuals who have a genetic background allowing them to adapt easily and readily to seasonal demand are more successful in reproducing themselves. The population of zygotes conceived in a given season would therefore reproduce the pattern of gene combination more fit for that season.

Is there a genetic basis for gestational diabetes?

Rh E-PGM1 (phosphoglucomutase locus 1) joint genotype frequencies (chromosome 1) have been determined in 90 women with gestational diabetes mellitus (GDM) and in 140 pregnant women with preexisting insulin-dependent diabetes mellitus (IDDM) and non-insulin-dependent diabetes mellitus (NIDDM). The pattern of Rh E-PGM1 association differs among types of diabetes. The distortion of the pattern tends to assume opposite values in GDM and NIDDM and this is particularly evident in PGM1*2,2 subjects. The proportion of PGM1*2,2 subjects homozygous for the Rh e allele is higher in GDM than in IDDM and NIDDM. IDDM women show an intermediate proportion and NIDDM women show the lowest proportion of this genotype. The opposite pattern is observed among PGM1*2,2 subjects carrying the Rh E allele. Genotype frequencies of the hypervariable region flanking the insulin gene (chromosome 11) have been determined in 77 women with GDM, in 52 pregnant women with preexisting diabetes (IDDM and NIDDM), and in 62 normal adults. In GDM and NIDDM subjects the frequencies are similar. In IDDM women the frequency of homozygotes for the class 1 allele is higher than the frequency found in GDM and NIDDM women. The data suggest a possible genetic basis for the differentiation of a subclass of GDM from both IDDM and NIDDM.

DNA restriction fragment analysis of the somatostatin gene in familial isolated growth hormone deficiency type I.

The somatostatin (SST) gene was analyzed to detect possible molecular variations in subjects with familial isolated growth hormone deficiency type I (IGHD I). No gross alterations in restriction fragments were observed with 18 used enzymes. The association with two RFLPs closely linked to the SST gene was also negative, adding weight to the evidence that the SST gene is not involved in the etiology of IGHD I. The nucleotide variability of a 23-kb DNA segment containing the SST gene and its flanking sequences was studied by restriction analysis of a sample of 19 Italians. The data suggest that approximately 1 in 400 bp is variant in this region.

Restriction fragment length polymorphism of the D5S4 locus in Italy.

Five Italian samples were examined for an EcoRI restriction fragment length polymorphism associated with a DNA sequence of unknown function, located on chromosome 5. No significant difference was observed between the samples. The allele frequencies in Italy were D5S4ES = 0.697, D5S4EF = 0.303.

Genetic variations of insulin-like growth factor I in Italy.

Four Italian populations were examined for a HindIII RFLP associated with the human insulin-like growth factor I gene. No differences were observed among the four samples. The allele frequencies in Italy were: IGF-I HF = 0.834; IGF-I HS = 0.166. The polymorphism appears to be due to a 400-bp sequence insertion-deletion mechanism.

Polymorphism of S-adenosylhomocysteine hydrolase in Italy.

S-adenosylhomocysteine hydrolase (SAHH) polymorphism has been investigated in the Italian population. Three common alleles, SAHH*1, SAHH*2 and SAHH*3, have been observed and the estimated gene frequencies are 0.968, 0.023 and 0.009, respectively. SAHH activity has been assayed in 50 healthy individuals and the mean activity was 0.043 +/- 0.017 mumol uric acid/min/g Hb at 37 degrees C. Five heterozygotes for adenosine deaminase deficiency and three heterozygotes for purine nucleoside phosphorylase deficiency showed SAHH within the range of the normal distribution. The effects of some thiol reagents on red blood cell SAHH electrophoretic pattern have been investigated.

Human placental glucose dehydrogenase: IEF polymorphism in two Italian populations and enzyme activity in the six common phenotypes.

Glucose dehydrogenase (hexose-6-phosphate dehydrogenase) has been assayed qualitatively and quantitatively in more than 600 human placentae collected in two Italian populations. The gene frequencies for GDH1, GDH2 and GDH3 were, respectively, 0.66, 0.21 and 0.12 in Continental Italy and 0.65, 0.23 and 0.12 in Sardinia. Among the six common phenotypes there was no difference in catalytic activity.

Relationship between human serum placental alkaline phosphatase and the ABo and Rh compatibility status in pregnancy.

458 unrelated healthy women at various gestational ages were examined for serum heat-stable alkaline phosphatase (HSALP) activity. The sample was subdivided into four groups according to the compatibility mating type in the ABO and Rh systems: double compatible, ABO incompatible, Rh incompatible and double incompatible. The results confirm the exponential growth of serum placental isoenzyme as a function of gestational age and show that the moment of appearance of the placental isoenzyme is six weeks earlier in double incompatible matings.

Human phosphoglucomutase locus 1: red cell enzymatic activities associated with common isoelectric focusing phenotypes.

Human phosphoglucomutase activity has been determined in red blood cells obtained from 348 unrelated subjects. The mean activities attributed to the four common PGM1 alleles, expressed as micromoles of G6P produced per gram of Hb per hour were 53 for PGMa31, 60 for PGMa11, 61 for PGMa41 and 72 for PGMa21. The relative amount of variation associated with the electrophoretic polymorphism was estimated as 24%.

Human placental alkaline phosphatase electrophoretic alleles: quantitative studies.

Human placental alkaline phosphatase (ALP) activity has been determined in specimens obtained from 562 Italian subjects. The mean activities of the three common homozygotes (Pl 2 = 4.70 +/- 0.24, Pl 1 = 4.09 +/- 0.08, and Pl 3 = 2.15 +/- 0.71 mumol of p-nitrophenol produced) were significantly different. The differences among the various allelic forms account for 10% of the total quantitative variation of the human placental alkaline phosphatase.

A survey of serum protein and enzyme polymorphisms in the district of L'Aquila (Italy).

A random sample of more than 500 subjects collected in the district of l'Aquila (Italy) has been studied for the following genetic markers: ADA, EsD, G-6-PD, 6-PGD, SOD-A, Hp, C3. The gene frequencies of all the systems examined were similar to those observed in Rome. Rare variants have been found for 6-PGD, C3 and SOD-A. The last one showed an electrophoretic pattern different from the only other variant so far observed in the Italian population.
