Evaluation of (D-Pro2, D-Trp7,9)-substance P as an antagonist of substance P responses in the rat central nervous system.

The (D-Pro2, D-Trp7,9) analogue of substance P has been tested for substance P antagonist activity in the caudal trigeminal nucleus in vivo, and in the isolated spinal cord in vitro. In neither case was the analogue found to be a specific antagonist of substance P, although the analogue did have weak antagonist actions in the isolated guinea-pig ileum preparation. It is concluded that the analogue is not a suitable tool for the identification of putative substance P systems in the spinal cord or the caudal trigeminal nucleus of the rat.

Autogenous vaccine therapy for condyloma acuminatum. A double-blind controlled study.

In an attempt to substantiate claims that autogenous vaccine is an effective form of immunotherapy for condyloma acuminatum, a double-blind cross-over study was carried out on 34 patients, in which autogenous wart vaccine was compared with a placebo vaccine identically prepared from each patient's own normal skin. It was found that the duration of disease was an important determinant of curability in patients with condylomata acuminata (P less than 0.001) and that when this factor was taken into account autogenous wart vaccine was not significantly more effective than the placebo (P = 0.43).

The epidemiology of indoor nitrogen dioxide in the U.K.

The results of three studies on the effect of gas cooking on the lung function of children are reported. The studies which have been carried out in England and Scotland with children aged 6-11, 6-7 and 5-6 years, respectively, show that there is only a weak association between indoor levels of NO2 and respiratory illness. Nevertheless, the consistency of the difference in the rate of respiratory diseases in children from homes with electric and gas cookers suggest there is some hazard to the use of unflued gas appliances which may warrant further epidemiological investigation.

Neuropsychology and cluster analysis: potentials and problems.

This report presents a selective overview of the cluster analysis literature and its potential uses in neuropsychology. In addition, an actual problem involving data from the Florida Longitudinal Project is presented to provide a practical example of many of the processes and problems involved in cluster analytic techniques. It is hoped that the reader will gain a theoretical and practical understanding of such methods and their potential usefulness in neuropsychology and other related areas.

The uptake and transmission of protein by neonatal rat enterocytes.

1. Proximal enterocytes transmitted intact immunoglobulin G (IgG) preferentially in the order rat, human, sheep and bovine; the removal from the vascular compartment of these transmitted molecules occurred at about the same rate. 2. Heterologous IgGs are processed similarly to rat IgG: they are either transmitted intact or broken down to less than 1000 mol. wt. fragments. 3. All of the human transferrin removed from the intestine was broken down to less than 1000 mol. wt. fragments, but a small amount of bovine serum albumin (BSA) was transmitted intact. 4. The IgGs and BSA are relatively indigestible molecules whereas human transferrin is relatively digestible. 5. These observations are discussed in the context of receptor-mediated transmission.

Effect of adrenalectomy on the transmission of IgG in young rats.

Antibody transmission in rats aged 17 days was unaffected by adrenalectomy performed at 13 days. The normal 'closure' of the jejunum to antibody is delayed by about 2 days in adrenalectomised rats. Adrenalectomy at 13 days has no apparent effect on plasma volume or haematocrit. Plasma corticosterone levels increase fivefold between 14 and 20 days, but in early weaned animals with 'closed' guts, corticosterone levels were not raised.

Radioimmunoassay of oestriol in the urine of non-pregnant women.

A radioimmunoassay for oestriol in the urine of non-pregnant women is described. Urine is hydrolysed rapidly at room temperature with an E. coli glucuronidase preparation and extracted prior to a simple radioimmunoassay procedure. An antiserum raised against oestriol-6-(O-carboxymethyl) oxime bovine serum albumin is used with (3H)-oestriol as tracer. Free and antibody-bound fractions are separated by partition between ammonium sulphate solution and scintillant without centrifugation. The accuracy and precision of the assay are satisfactory. It gave values approximately 80% of those obtained for total oestrogens by a fluorimetric method.

The anatomy and blood system of the kidney in the river lamprey, lampetra fluviatilis.

The anatomy and blood system of the kidney in the river lamprey, Lampetra fluviatilis, was studied after injecting microfil into nephrons or the arterial system. Renal arteries arose at irregular intervals from the dorsal aorta and gave rise to a regular arrangement of afferent arterioles supplying the network of glomerular capillaries. Nephron units were arranged in two longitudinal series on each side of the glomerular capillaries, with the capsule of each nephron closely related to the capillary network. A short neck segment lead into the convoluted proximal segment, which accounted for over half the length of each nephron and was surrounded by a network of capillaries and sinusoids supplied by efferent glomerular arterioles. The end of each proximal segment formed the descending limb of a nephron loop, which lay parallel to the ascending distal limb and the end of each collecting duct. Peritubular blood flow in this region was generally opposite the flow of tubular fluid and blood eventually drained into large thin-walled sinuses connected to the post-cardinal vein.

Actions of microiontophoretically applied oxytocin, and immunohistochemical localization of oxytocin, vasopressin and neurophysin in the rat caudal medulla.

Oxytocin-, vasopressin- and neurophysin-containing axons were visualized within the rat caudal medulla using the immunoperoxidase technique. The highest densities of axons and terminals were found in the nucleus tractus solitarius, nucleus dorsalis vagus, nucleus commissuralis, nucleus reticularis lateralis and within the marginal layer of the nucleus trigeminalis. In these areas, oxytocin fibres predominated markedly over vasopressin fibres. In a series of electrophysiological experiments, neurones in these and surrounding areas were predominantly depressed following the iontophoretic application of oxytocin. This depression was seen on both spontaneous and glutamate-evoked neuronal firing.

The effect of exogenous steroids and steroid inhibitors on IgG transmissions in young rats.

The transmission of labelled IgG by the proximal small intestine was assessed in 16- and 18-day-old rats. Closure had commenced by 18 days and in some animals was well advanced by this age. The injection of large doses of cortisone acetate, hydrocortisone sodium succinate, deoxycorticosterone acetate and corticosterone induced precocious closure of the small intestine by 16 days; and all but the latter steroid also induced precocious cell replacement in the ileum. Maternal deprivation for about 18 h daily led to closure of the proximal small intestine but did not induce precocious replacement in the ileum. The injection of metopirone, aminoglutethimide and ACTH resulted in partial closure of the proximal small intestine but did not induce precocious cell replacement in the ileum. Maternal deprivation for about 8 h daily produced a similar result.

A simple procedure for the radioimmunoassay of 17 alpha-hydroxyprogesterone in serum: comparison with an immunological purification technique.

A simple radioimmunoassay for the measurement of 17 alpha-hydroxyprogesterone in 0.1-0.25 ml serum is described. An antibody prepared against 17 alpha-hydroxyprogesterone 3(O-carboxymethyl)-oxime-BSA as immunogen was used. A correlation of the 17 alpha-hydroxyprogesterone concentration found in the serum of normal subjects and of patients with that obtained by a more complex immunological purification method was good (r=0.924, P less than 0.001). Accuracy, precision, and specificity were acceptable with the simple method. Normal values for infants, children, and adults have been established, and the individual concentrations of 17 alpha-hydroxyprogesterone in the serum of patients withcongenital adrenal hyperplasia were well separated from normal values.

A micropuncture study of kidney function in the river lamprey Lampetra fluviatilis adapted to sea water.

Micropuncture techniques have been used to investigate kidney function in lampreys adapted to hyperosmotic media. Plasma electrolyte concentrations were maintained well below corresponding concentrations in the external environment. Urine composition was variable, but generally showed high concentrations of magnesium, sulphate and chloride ions. Lampreys in 50% sea water produced urine which was hypo or iso-osmotic to plasma, whereas those in 100% sea water produced hyperosmotic urine. Urine flow rate in 50% sea water was one tenth of that in fresh water, due to a reduction in filtration rate and an increase in water reabsorption by the kidney. As in fresh water, little if any filtered water was reabsorbed by the proximal segment. Almost 90% of filtered water was reabsorbed by the kidney of 100% sea water lampreys and most of this must have occurred in the distal and collecting segments.
