Naturally occurring R.ColBM plasmids belonging to the IncFIII incompatibility group.

Two Escherichia coli strains isolated from urinary tract infections were resistant to streptomycin, kanamycin, neomycin, tetracycline and sulphonamides. The strains also produced colicins B and M. The resistance to streptomycin, kanamycin and neomycin and the ability to produce colicins B and M could be transferred to an E. coli K12 recipient. Resistance and colicinogeny markers were transferred together by conjugation, and did not segregate even after interrupted mating or phage P1-mediated transduction. Hence, the drug-resistance and colicinogeny markers were carried by the same plasmid, designated as R.ColBM plasmid. The two R.ColBM plasmids were Fi+ and produced an F-like pilus. They belonged to the IncFIII incompatibility group, being thus the first R plasmids identified in this group. The two plasmids were isolated and their molecular sizes were determined by electrophoresis in agarose gels and by contour length measurements. Both methods showed that both plasmids were about 52 megadaltons.

Interference of Neisseria gonorrhoeae growth by aerobic bacterial representatives of the urogenital flora.

Aerobic bacterial isolates obtained from endocervical, vaginal and urethral swabbings were tested for interference of neisseria gonorrhoeae growth on solid medium. Simultaneous antagonism was studied using the lawn spotting method, and delayed antagonism by the basal spot/lawn method. From 58 swabbings we recuperated a total of 181 isolates, 71 of those were found interfering with at least one out of four gonococcal strains (G-1, G-2, G-3 and G-4). Similar percentages of interfering isolates were obtained from each of the isolation sites. The identification of the interfering isolates has revealed that similar numbers of coagulase negative staphylococci and identical numbers of group D streptococci were found for each of those sites. The majority of the interfering isolates and also of the inhibitory coagulase negative staphylococci showed only simultaneous antagonism. To complete the interference spectrum, we have tested all the active urogenital isolates against four other gonococcal strains (G-7, G-9, G-10 and G-11). This spectrum showed clearly that interference is not an all or none phenomenon. While the gonococcal interference spectrum of most of the Gram positive cocci and the Acinetobacter sp. strains is broad, that of all the other isolates is relatively narrow. Gonococcal strains G-7 and G-9 were the most susceptible to inhibition by the interfering urogenital isolates while strain G-3 was the most resistant one.

An etiologic survey of clinical factors in cervical intraepithelial neoplasia: a transverse retrospective study.

In this retrospective survey 235 patients were studied: 123 patients with cervical intraepithelial neoplasia (CIN) and 112 controls. Fourteen epidemiologic variables were reviewed. The stepwise logistic multiple regression analysis demonstrated that age at first sexual contact was the variable most related to CIN appearance, followed by the occupation of the husband. School attendance and socioeconomic indices of the patients' fathers, husbands or partners were lower in the CIN group, as shown in the descriptive analysis. Age at first pregnancy was also found to be lower among CIN patients. Promiscuity and contraception did not seem to play a significant role in the incidence of neoplastic disease. A carcinogenic model is proposed.

Detection of Epstein-Barr virus antigens and antibodies by peroxidase-labeled specific immunoglobulins.

Detection of the Epstein-Barr (EBV) antigens, early antigen (EA), viral capsid antigen (VCA), and nuclear antigen (EBNA) by the indirect immunoperoxidase technique was highly sensitive. Antibody titers to EBNA, EA, and VCA were determined in more than 25 sera of patients with Burkitt lymphoma (BL), nasopharyngeal carcinoma (NPC), or normal persons. A good correlation between the titers of these antigens was obtained by the immunoperoxidase and immunofluorescence methods. The indirect (anti-IgG) immunoperoxidase technique for the detection of EBNA is, in contrast to the indirect immunofluorescence method, highly sensitive. EBNA was associated with the chromosomes in cells arrested in the metaphase with colchicine.

[Effect of open heart surgery on the defense mechanisms against bacterial infections].

Extracorporeal circulation during open-heart surgery may damage cellular and humoral factors in the blood. Phagocytic functions of circulating polymorphonuclear leukocytes were investigated in 10 patients 1 day before, then 1 hour, 2, 4 and 10 days after open-heart surgery. Transient diminutions in the phagocytic capacity of polymorphs was found for Staphylococcus aureus and Escherichia coli in one patient, and E. coli only in two patients up to the 4th postsurgical day. A positive correlation was found between the duration of extracorporeal circulation and the transient diminutions in phagocytic capacity for these three patients. However, the bactericidal capacity of polymorphs and their capacity to reduce nitroblue tetrazolium were normal in all patients. The serum concentrations of total hemolytic complement, C3, IgG and IgM were generally diminished up to the 4th day after surgery, but the opsonic power of the serum was almost normal in all the patients.

[Selective deficiencies of phagocytosis and opsonization towards Staphylococcus aureus. 1. Preliminary study in normal humans and patients with debilitating diseases].

We studied simultaneously the bactericidal activity (BA) and the serum opsonic capacity (OC) towards S. aureus and Escherichia coli during 2 hours of phagocytosis in vitro. The PMNs and the sera from 17 seriously ill patients and from 10 normal individuals were tested. Both BA and OC towards E. coli were the same for all patients and controls. The BA for one patient and the OC for three other patients were found to be diminished significantly. The last three patients had infectious complications due to S. aureus only. The selective deficiency of OC towards S. aureus in these three patients may be connected with the pathogenesis of their infectious complications.

Toxoplasmosis and the compromised host.

A high percentage of population shows antibodies to Toxoplasma gondii yet clinical manifestations of primary infestations are rare. Congenital and secondary forms (chorioretinitis) are serious medical conditions. An outburst in a compromised host is always serious and may be fatal. Therefore, patients subjected to immunosuppressive therapy or with pathologic immune suppression should be clinically and serologically screened and, when toxoplasmosis is diagnosed, treated for the entire period of immunosuppression.
