Adoptive transfer of experimental allergic encephalomyelitis with lectin-activated spleen cells. Part 2. Studies on T cell subsets and interleukin 2 production.

Experimental allergic encephalomyelitis (EAE) can be transferred by spleen cells stimulated in vitro with D-mannose-binding lectins but not with N-acetyl-D-galactosamine (GalNAc)-binding lectins. EAE could also be passively transferred by spleen cells following incubation with wheat germ agglutinin (WGA), in which case the disease transfer was abolished by the specific hapten inhibitor, N-acetyl-D-glucosamine. In the presence of rat T cell monoclonal antibody, either W3/25 or OX-8, both concanavalin A and Wisteria floribunda agglutinin stimulated helper and suppressor T subpopulations. On the other hand, GalNAc-binding lectins were less effective than D-mannose-binding lectins in generating interleukin 2 (IL2) in the culture supernatant, whereas WGA-stimulated spleen cells did not produce IL2. Furthermore, spleen cells cultured with pure IL2 could not transfer EAE to the recipients. These data suggest that some factors distinct from IL2 are required for the differentiation of EAE-effector precursors into the final effector cells in this transfer system.

Ability of monoclonal antibody to herpes simplex virus glycoprotein gB to promote healing of herpetic skin lesions in nude mice.

The effect of monoclonal antibody (MCA) to glycoprotein gB of herpes simplex virus (HSV) was studied in athymic nude mice inoculated with HSV intracutaneously in the midflank. HS1, the MCA used in the study, had a high neutralizing titer (1:2048) and had antibody-dependent cell-mediated cytotoxicity. HS1 was injected intraperitoneally at various intervals after HSV infection. HS1 injected 3 h after infection inhibited the development of skin lesions and most mice survived. Administration of HS1 at the time the local skin erosions appeared at the inoculated site (4-7 days after infection) was also effective, and in four of eight mice skin lesions completely healed. Furthermore, in three of four mice that survived, latent infections in the ganglia were also prevented as evidenced by the failure to detect HSV by co-cultivation with Vero cells. Administration of HS1 after the development of zosteriform skin lesions (5-9 days after infection) reduced virus in the ganglia and prolonged the survival time, though the disease was not completely arrested and all the mice died eventually.

[Variations in ultrastructures and negative charges in experimental hydrosalpinx].

It is not so difficult to operate on the hydrosalpinx with macrosurgical or microsurgical methods, but difficult to initiate pregnancy after the operation because little is known about the post-operative condition of the hydrosalpinx. In this study time sequential observations of the growing experimental hydrosalpinx with the two hemoclip method was performed during morphological change using the microscope, sequential electron microscope and transmission electron microscope with cationized ferritin as an ultrastructural marker. The following results were obtained: Size of hydrosalpinx: Until 15 weeks after the operation, the hemo-clipping hydrosalpinx became longer but thereafter there was no change. The maximum diameter of the hydrosalinx became wider until 20 weeks after treatment. Peristaltic of hydrosalpinx: Tubal peristaltics were observed until 7 weeks after treatment, but not thereafter. SEM findings: Decreases in the amount of cilia started 1 week after treatment, and 7 weeks after treatment partial excoriations of epithelium were observed. 20 weeks after treatment excoriations of epithelium were observed widely. A decrease in the amount of cilia was observed but no loss was observed. The negative charges on the tubal endometrium due to TEM using cationized ferritin: The negative charges on the tubal endometrium were decreasing both on the secretory cells and ciliary cells.

Reinduction of experimental allergic encephalomyelitis in convalescent Lewis rats with cyclophosphamide.

Experimental allergic encephalomyelitis (EAE) in Lewis rats is an acute, monophasic autoimmune disease, and rats recovering from EAE are generally resistant to active reinduction. However, following immunosuppression with a high dose (100-200 mg/kg) of cyclophosphamide (CY), EAE was reinduced in convalescent rats, irrespective of whether or not they were rechallenged with the encephalitogenic inoculum. Reinduction of EAE was not observed in rats treated with low doses (20 mg/kg or less) of CY. A selective inhibition of suppressor cells during the convalescent stage may be responsible for the reinduction of EAE in Lewis rats.

Effects of combined use of acyclovir and antibody in athymic nude mice inoculated intracutaneously with herpes simplex virus.

Antiviral effects of acyclovir (ACV) and antibody were studied in athymic nude mice inoculated intracutaneously in the midflank with herpes simplex virus type 1. Three hours after virus inoculation, treatment was initiated. In ACV-treated mice, the development of skin lesions was inhibited and the mean survival time was prolonged as compared with controls. Treatment with ACV markedly reduced the viral titers both at the inoculation site and in the neural tissues (dorsal root ganglia, spinal cord and brain). Transfer of antibody was also effective in controlling infection as described previously. The use of ACV plus antibody was more effective than the use of ACV alone and in 5 of 17 mice the disease was completely inhibited. Furthermore, in the mice that survived, latent infections in the ganglia were also prevented. These results indicate that treatment with ACV plus antibody is highly effective against herpes simplex virus infection in the immunocompromised host.

Passive immunization of mice with monoclonal antibodies to glycoprotein gB of herpes simplex virus.

To investigate the protective ability of monoclonal antibodies (MCAs) to viral glycoprotein in herpes simplex virus (HSV) infection, athymic nude mice were inoculated intracutaneously with HSV type-1 (HSV-1) in the midflank. Three hours after inoculation, one group of mice was passively immunized with one of a series of MCAs to glycoprotein gB of HSV-1, and a control group of mice was given phosphate buffered saline alone. The control mice died within 16 days after infection, whereas the mice passively immunized with any of the MCA showed suppressed development of skin lesions. Three of six mice given MCA failed to develop any visible lesions and no HSV could be isolated from the lumbar dorsal root ganglia of these mice 60 days after the challenge. BALB/c mice were also protected from infection with HSV type 2 by passive immunization with MCA to HSV-1 gB.

[The evaluation of serum estradiol, progesterone, testosterone concentrations and urinary estrogen excretion level for the monitoring indices of follicular maturation].

We have evaluated serum estradiol, progesterone, testosterone, and urinary estrogen excretion in 24 hour urine samples to monitor indices of follicular maturation. The serum steroid levels were determined with the direct radioimmunoassay kit. The urinary estrogen level was measured with the estrogen micrometering kit using hemagglutination inhibition reaction. Moreover, relationships between these steroid levels and the follicular size measured with ultrasound were analyzed. The serum estradiol concentration and the estrogen excretion in 24 hour urine samples mostly showed a continuous increase during the late follicular phase, and had a positive correlation to the maximum follicular diameter of the leading follicle (MxFD) and to the total of the maximum diameter of the follicles (TFD). The serum progesterone concentration showed a remarkable increase especially on the day of the LH surge onset in many cases, and had a significant (p less than 0.01) correlation to MxFD but not to TFD. The serum testosterone concentration, however, showed neither a specific tendency on its daily change nor a correlation with the follicular size. These results indicated that the serum estradiol and progesterone, and the urinary estrogen excretion can be utilized as indices of follicular maturation.

Herpes simplex virus type 1 restriction fragment polymorphism determined using southern hybridization.

Regions of herpes simplex virus type 1 (HSV-1) DNA with variation in the size of restriction endonuclease fragments were identified by comparison of the BamHI, KpnI or SalI restriction endonuclease digestion patterns among 15 HSV-1 isolates after hybridization with specific 32P-labeled cloned HSV-1 DNA fragments. Of the types of restriction fragment polymorphism identified, one was a strain with a distinctly different restriction fragment than the prototype (loss or gain of restriction sites). Another type, the specific fragment varied only in size among strains. Thirteen distinct variations were identified. Ten were mapped to the unique sequence of the L component; two to the inverted repeat of the L component and one to the inverted repeat of the S component. The presence of a common ancestor from which some isolates of HSV-1 might derive was deduced from an analysis of the distribution of the thirteen variations among the 15 HSV-1 isolates.

Calcium antagonistic action involved in vasodilation by brovincamine.

Possible mode of vasodilative action of brovincamine was assessed in isolated cardiovascular preparations in comparison with verapamil and papaverine. Brovincamine (IC50: 1.2 x 10(-5) M), verapamil (IC50: 3.5 x 10(-7) M) or papaverine (IC50: 2.5 x 10(-5) M) caused a dose-dependent relaxation of potassium (30 mM)-contracture in the rabbit pulmonary arterial segment. This relaxation by verapamil or brovincamine, but not by papaverine, was antagonized by increasing external Ca2+ concentration to 12.4mM. Duration of slow action potentials of partially depolarized guinea-pig papillary muscle was reduced by 14 min exposure to brovincamine (5 x 10(-5) M) or verapamil (10(-5) M. These results suggest that brovincamine produces a vasodilation via a slow Ca2+-channel blockade.

Isolation of human DNAs homologous to the BamHI-Z fragment of herpes simplex virus type 1 DNA.

Human DNA hybridizes with the BamHI-Z fragment (map coordinates 0.936 to 0.949) of Herpes simplex virus type 1 (HSV-1) DNA. To characterize the BamHI-Z homologue of human DNA, we isolated six independent hybrid phages with a sequence homologous to the BamHI-Z fragment from a human genomic DNA library. Three of the six had a common 1.2-kb BamHI-EcoRI fragment homologous to the BamHI-Z, and this fragment existed as 10-60 copies per human haploid genome. A 0.29-kb MboII segment of the BamHI-Z fragment was found to be responsible for the homology with the 1.2-kb BamHI-EcoRI fragment.
