Naevus superficialis lipomatosus (NSL) is a rare malformation consisting of multiple or solitary papules usually occurring on the lower trunk or gluteal regions. In this study the clinical features of twenty new cases are described with a detailed account of the histopathology. Notable were the marked variability in the amount of ectopic fat found in the lesions, and the presence of associated abnormalities of the connective tissue, the vessels, and the skin appendages. In early lesions, mononuclear cells differentiating into lipoblasts could be identified around proliferating capillaries. Two clinical types were distinguished. First, the classical type (7 cases) consisting of multiple lesions occurring in a zonal distribution, and secondly the solitary papule or nodule (13 cases). The latter type can be difficult to distinguish from a skin tag with fatty herniation.
Allotype-reactive polystyrene tubes were made by first coating their inner surface with immunglobulin (Ig) solutions containing the allotype of interest; this was followed by the application of anti-allotype antibody. The latter was covalently cross-linked to the adsorbed allotype antigen by means of glutaraldehyde to prevent the dissociation of antibody from the surface of the tube in subsequent steps (glutaraldehyde treatment did not affect the specificity of these tubes). Under conditions of antibody excess, allotype-reactive tubes were treated next with 125I-labeled 7S Ig from standard solutions in order to estimate the concentration of Ig allotypes in these solutions. Then, to determine serum allotype concentrations, we used the standard solutions and the serum unknown to carry out competition experiments between labeled and unlabeled allotype antigen. Quantitation of serum Ig allotypes in this way featured a high signal to noise ratio (30 to 100), an error estimate of 3 to 7%, and sensitivity in the nanogram range.

Quantitative histochemical techniques demand constancy in sectioning tissue and preferably knowledge of the true thickness of a section. In this investigation a simple, rapid, accurate, and reproducible technique is described for measuring section thickness using an instrument known as a Surfometer. It has been shown that: (a) cryostat microtome settings bear little relationship to the true section thickness of the sections produced; (b) that different instruments differ in their inaccuracies; (c) that this inaccuracy varies slightly with different tissues.If densitometric methods are used and either the absolute amount of a tissue component is to be measured or the amount of a tissue component is to be compared in two tissues then it is mandatory to determine the actual thickness of the section. The method described here seems the most appropriate one for these investigations.
