[Permanent interstitial implantation of 125iodine in case of carcinomas of the oral cavity and the oropharynx. Preliminary report].

The authors present their first experiences gained during the last year with the permanent implantation of 125iodine seeds in thirty patients suffering from carcinomas of the oral cavity and the oropharynx. The properties and advantages of 125iodine, the method of implantation, the dosimetry and the problems of radioprotection are particularly underlined. In every case, we apply pre- or postoperative external radiotherapy, i.e. we expose the primary tumor to a dose of 50 Gy fractionated in the usual manner over four or five weeks. This combined treatment is intended to overcome the resistance to therapy of ORL malignomas and to obtain a high local rate of tumor control and a low rate of functional and cosmetic troubles.

[Prevention of accidents related to alcohol drinking].

In its first part the article questions the notion of alcohol-related accidents. It is stressed that the rates of alcohol-related accidents are influenced by changes of the public consciousness of problems. In the second part the share of alcohol involvement in accidents of various nature is shortly indicated. Finally, in the third part, the effect of primary and secondary prevention is discussed, based on foreign studies. It is concluded that there is a high probability that only a combination of various measures will bring success.

Enzymatic synthesis of double-stranded DNA containing radioactively labeled O(6)-ethylguanine as the only modified base.

This paper describes the enzyme-catalyzed in vitro synthesis of double-stranded DNA (ds-DNA) containing [3H]-labeled O(6)-ethylguanine (O(6)-EtGua), an alkylation product strongly implicated in mutagenesis and carcinogenesis by ethylating N-nitroso carcinogens. Single-stranded DNA (ss-DNA) containing O(6)ethyl-[8-3H]-2'-deoxyguanosine was synthesized using terminal deoxynucleotidyl transferase. The parameters determining yield of reaction, base ratios, and DNA chain length, were investigated. The O(6)-EtGua-containing ss-DNA could be replicated by DNA polymerase I, as indicated by the incorporation of [8,5'-3H]-2'-deoxyguanosine-5'-monophosphate and by the resistance of the replication product to nuclease S1 digestion. ds-DNA's with chain lengths between approximately 200 and 1000 base-pairs and O(6)-EtGua/guanine molar ratios of approximately 10(-2)--10(-3) were synthesized. Their use in the analysis of enzymatic mechanisms involved in the elimination of O(6)-alkylguanine from the DNA of mammalian cells is discussed. The procedure of synthesis described for (O(6)-EtGua)-containing ds-DNA may also be applicable for the production of ds-DNA containing structurally modified bases other than O(6)-EtGua.

Detection of hepatitis B virus-specific DNA in the genomes of human hepatocellular carcinoma and liver cirrhosis tissues.

Hepatitis B virus-related DNA was detected in the chromosomal DNA of three out of seven hepatocellular carcinomas and two out of five cirrhosis samples examined, by means of the blot-hybridization technique, described by Southern (1975). The integration patterns were not identical but some similarities raise the question of whether there are some preferred sites of viral integration.

Coexisting suppressor cells for myeloid colony formation in the bone marrow of patients with aplastic anaemia.

Suppressor cells for colony forming cells (CFUC) were found in the bone marrow of 1 of 6 patients with aplastic anaemia. The assay applied was based on the fact that coexisting suppressor cells are more sensitive to dilution than CFUC, as long as adequate stimulation is provided by a source of colony stimulating activity. In the patient with suppressor cells, a 4 d trial of 1000 mg methylprednisolone per d failed to improve the blood cell production and had to be discontinued due to gastric irritation.

Haemorheology and cerebrovascular disease: multifunctional approach with pentoxifylline.

New aspects in the pathophysiology of chronic cerebral ischaemia as the cause of the multiform syndromes of cerebrovascular disorders are reviewed and therapeutic approaches are discussed. It is suggested that the fatal vicious circles of the disturbances of nutritive blood flow within the decompensated cerebral microcirculation can by interrupted by pentoxifylline. By amelioration of the disturbed flow properties of blood in the affected capillary regions through inhibition of red cell and platelet aggregation, improvement of red cell deformability and reduction in blood viscosity, oxygen supply is enhanced and more glucose is made available. Membrane permeability and function of the cell are normalized by the favourable inhibitory action of pentoxifylline on oedematous changes in the brain tissue and mechanical obstacles in the micro-circulation are removed. The disturbed cerebral metabolism is directly stimulated by compensation of the impaired energetic balance. The combined rheological, antioedematous and metabolic effects of pentoxifylline offer a multifunctional therapy of cerebrovascular circulatory disorders.

Hemorheology and peripheral vascular diseases: a new therapeutic approach.

New findings on the physiology of flow properties of blood are reviewed. Particular attention is paid to hemorheologic occurrences in the decompensated microcirculation in peripheral vascular disease. The therapeutic possibilities for obtaining functional compensation are discussed. Trental (pentoxifylline) largely satisfies the complex requirements of a hemorheologically active therapeutic agent.
