A rapid, objective method for the detection of lymphocytotoxic antibodies using flow cytometry.
Whilst several centres have reported lymphocytotoxic antibody detection using single and dual fluorescent stains with analysis of the fluorescence emitted from the cell population present in a well of a multiwell plate, problems are encountered with cell concentration and light emission overlap. A method we have developed using flow cytometry produces similar values of percentage cell death using single or double staining techniques (correlation coefficient = 0.9896). This method is not influenced by slight variation in cell number or light emission overlap. The effect of introducing red cell impurities into the normal lymphocyte preparation is described.

Enzyme replacement in nervous tissue after allogeneic bone-marrow transplantation for fucosidosis in dogs.
Bone-marrow transplantation after total lymphoid irradiation in an alpha-L-fucosidase-deficient dog raised the enzyme activity in both visceral and neural tissues with consequent reduction in the severity of storage lesions. These results offer hope that early marrow transplantation may prevent the development of disease in neurovisceral storage disorders.

Nonlever action of the mandible.
This study considers the current concept of the mandible as a lever of the third order. The concept requires a fulcrum, and this function has been ascribed to the condyle region, but it tends to be overlooked that the fulcrum of a third-order lever in this case would sometimes have to bear a considerable stress. Certain changes, attributed to stress, have been observed in anatomical components of the articulation, but they cannot be explained in terms of the lever concept. They are accounted for by the changing anatomical relations in the working and contralateral sides during mandibular function. They arise from minor stress, especially when dental conditions indicate a period of abnormal function.
Aspirate cellularity as a rapid indicator of renal allograft rejection.
No rapid and unequivocal test exists to diagnose renal allograft rejection. Despite advances in immunosuppression over the last 20 years, rejection is still the major problem in clinical organ transplantation. Most transplant centres report a renal graft survival rate of about 60 per cent at 5 years and the majority of graft losses are due to rejection. An influx into the graft of various cell types has been implicated in the diagnosis of rejection but it takes up to 24 h to obtain such information histologically. We have carried out fine needle aspiration biopsy of the allografts and estimated the number of white cells in the aspirate using a fluorescence activated cell sorter. Studies were performed in 34 cases of normal allograft function, 8 of rejection, 8 of acute tubular necrosis and 8 of cyclosporin nephrotoxicity. There was a statistically significantly higher cell number in the group undergoing rejection (P less than 0.01; Wilcoxon unpaired test) when compared to each of the other three groups. We believe that this technique, which takes only a few minutes to perform, could yield much valuable information. It deserves further study.
Effect of perioperative blood transfusion on recurrence of colorectal cancer.
The aim of the present study was to examine the outcome of 517 patients undergoing curative surgery for colonic and rectal cancer, and to compare the recurrence and mortality rates in transfused and non-transfused groups of patients. The two groups were evenly matched for age, sex, Dukes' stage and histological differentiation. There were significantly more rectal tumours in the transfused group (P less than 0.01), but the distribution of colonic lesions did not differ. Life table analysis revealed that the transfused patients had a 20 per cent greater probability of recurrence at 5 years (P less than 0.005) and there were 16 per cent more cancer related deaths (P less than 0.01). Even when all rectal cancers were excluded, a similar trend was seen for the colonic lesions: a 24 per cent greater probability of recurrence at 5 years (P less than 0.025) and 15 per cent more cancer related deaths (P less than 0.02). We conclude that blood transfusion may be associated with increased mortality and recurrence in patients undergoing curative surgery for colorectal cancer.
Gastric hyalinization presenting in life and mimicking gastric cancer.
Gastric hyalinization is an unusual abnormality that has in the past been regarded by some authorities as a postmortem artifact. We describe a 69-year-old patient who presented with symptoms due to the condition, and who was treated by surgical resection of the diseased stomach. To our knowledge, no other case presenting in life has been identified in the literature, which we reviewed in the light of the present case. The cause of the condition remains unknown.
