Identification of Aspergillus fumigatus and related species by nested PCR targeting ribosomal DNA internal transcribed spacer regions.

Aspergillus fumigatus is the most common species that causes invasive aspergillosis. In order to identify A. fumigatus, partial ribosomal DNA (rDNA) from two to six strains of five different Aspergillus species was sequenced. By comparing sequence data from GenBank, we designed specific primer pairs targeting rDNA internal transcribed spacer (ITS) regions of A. fumigatus. A nested PCR method for identification of other A. fumigatus-related species was established by using the primers. To evaluate the specificities and sensitivities of those primers, 24 isolates of A. fumigatus and variants, 8 isolates of Aspergillus nidulans, 7 isolates of Aspergillus flavus and variants, 8 isolates of Aspergillus terreus, 9 isolates of Aspergillus niger, 1 isolate each of Aspergillus parasiticus, Aspergillus penicilloides, Aspergillus versicolor, Aspergillus wangduanlii, Aspergillus qizutongii, Aspergillus beijingensis, and Exophiala dermatitidis, 4 isolates of Candida, 4 isolates of bacteria, and human DNA were used. The nested PCR method specifically identified the A. fumigatus isolates and closely related species and showed a high degree of sensitivity. Additionally, four A. fumigatus strains that were recently isolated from our clinic were correctly identified by this method. Our results demonstrate that these primers are useful for the identification of A. fumigatus and closely related species in culture and suggest further studies for the identification of Aspergillus fumigatus species in clinical specimens.

Comparison of three mechanical thrombus removal devices in thrombosed canine iliac arteries.

PURPOSE: To assess and compare intimal and medial vascular damage caused by three mechanical wall-contact thrombectomy devices: Fogarty embolectomy catheter, Arrow-Trerotola peripheral thrombectomy device, and MTI-Castañeda over-the-wire brush. MATERIALS AND METHODS: Bilateral external iliac arteries of 15 canines were thrombosed before mechanical thrombolysis. Ten thrombosed arteries were randomly assigned to receive each device. Animals were sacrificed immediately, and histologic assessment of endothelial and medial damage in the vessels was performed. RESULTS: The vascular damage found with all devices extended into the tunica media. The Fogarty embolectomy catheter and the Arrow-Trerotola device caused significantly more damage than the Castañeda brush. CONCLUSION: All devices caused lesions extending into the media. Previous research has shown that the extent and depth of the vascular lesion may be contributing factors in promoting early atherosclerotic and accelerated hyperplastic intimal and medial changes. These findings warrant further study of these devices in an atherosclerotic model with longer follow-up.

The Tem1 small GTPase controls actomyosin and septin dynamics during cytokinesis.

Cytokinesis in budding yeast involves an actomyosin-based ring which assembles in a multistepped fashion during the cell cycle and constricts during cytokinesis. In this report, we have investigated the structural and regulatory events that occur at the onset of cytokinesis. The septins, which form an hour-glass like structure during early stages of the cell cycle, undergo dynamic rearrangements prior to cell division: the hourglass structure splits into two separate rings. The contractile ring, localized between the septin double rings, immediately undergoes contraction. Septin ring splitting is independent of actomyosin ring contraction as it still occurs in mutants where contraction fails. We hypothesize that septin ring splitting may remove a structural barrier for actomyosin ring to contract. Because the Tem1 small GTPase (Tem1p) is required for the completion of mitosis, we investigated its role in regulating septin and actomyosin ring dynamics in the background of the net1-1 mutation, which bypasses the anaphase cell cycle arrest in Tem1-deficient cells. We show that Tem1p plays a specific role in cytokinesis in addition to its function in cell cycle progression. Tem1p is not required for the assembly of the actomyosin ring but controls actomyosin and septin dynamics during cytokinesis.

Determination of glycerol in plasma by an automated enzymatic spectrophotometric procedure.

The plasma concentration of glycerol, the backbone of triglycerides and the end product of triacylglycerol breakdown, is considered to reflect lipolysis in adipose tissue. We evaluated an automated enzymatic procedure for the measurement of glycerol in plasma. The assay was linear up to 250 micromol/l. The detection limit was 8 micromol/l. Recovery averaged 94% from spiked plasma and 104% from diluted plasma. An extensive precision study performed according to the guidelines of the National Committee for Clinical Laboratory Standards showed within-run coefficients of variation between 14.8% and 2.6% for concentrations ranging from 28 micromol/l to 164 micromol/l. The reference range for fasting healthy adult men was 14-69 micromol/l. Glycerol levels were significantly correlated with free fatty acid levels. This automated enzymatic method is rapid and reliable, and provides greater sensitivity or convenience than previously described procedures.

Cloning of rat interleukin 11 and interleukin 11 receptor alpha chain and analysis of their expression in rat uterus in the peri-implantation period.

Studies in mice have shown that interleukin 11 (IL-11) signalling is required for female fertility. In the absence of IL-11, decidualization is markedly retarded and implantation fails. IL-11 acts via a heterodimeric receptor composed of a ligand-specific receptor alpha chain (IL-11R alpha) and the signalling moiety gp130. This study reports the cloning of genes encoding rat IL-11 and IL-11R alpha. RNase protection was used to demonstrate that expression of IL-11 is upregulated in the rat uterus at the initiation of implantation at 5.5 days after mating. Expression of the genes encoding the two receptor components, IL11Ra and gp130, did not change throughout the peri-implantation period. In situ hybridization studies revealed that, as in mice, expression of IL-11 was high in the primary decidual zone at the time of the attachment reaction, whereas IL11Ra was expressed throughout primary and secondary decidua. Conservation of the temporal and spatial expression of IL-11 and IL-11R alpha in the uterus of the mouse and rat during the peri-implantation period will facilitate future studies investigating the role of IL-11 in fertility.

Reverse transcriptase template switching: a SMART approach for full-length cDNA library construction.

Here, we describe a fast, simple method for constructing full-length cDNA libraries using SMART technology. This novel procedure uses the template-switching activity of Moloney murine leukemia virus (MMLV) reverse transcriptase to synthesize and anchor first-strand cDNA in one step. Following reverse transcription, three cycles of PCR are performed using a modified oligo(dT) primer and an anchor primer to enrich the cDNA population for full-length sequences. Starting with 1 microgram human skeletal muscle poly(A)+ RNA, a cDNA library was constructed that contained 3 x 10(6) independent clones with an average insert size of 2 kb. Sequence analysis of 172 randomly selected clones showed that 77% of cDNA clones corresponding to known genes contained intact open reading frames. The average length of complete open reading frames was 2.4 kb. Furthermore, 86% of the full-length clones retained longer 5' UTR sequences than the longest 5' end deposited in the GenBank database. cDNA libraries generated using this method will be useful for accelerating the collection of mRNA 5' end sequence information, which is currently very limited in GenBank.

Qualitative and quantitative analyses of resin-bound organic compounds.

Methods for qualitative and quantitative analyses of resin-bound organic compounds are essential tools for chemistry development in solid-phase combinatorial and parallel syntheses. Here we discuss the use of gel-phase 19F NMR, the fluoride ion-selective electrode method, and spectrophotometry for monitoring solid-phase reactions. Our results indicate that the application of these diverse methods for analyzing the outcome of solid-phase combinatorial synthesis are sensitive and conclusive.

Change of hypervariable region proteins of hepatitis C virus E2 in two infants.

Two infants vertically infected with hepatitis C virus (HCV) were followed-up from 3 or 4 months to 2.5 years of age. We analyzed five complementary DNA (cDNA) clones from each patient and compared the genetic drift of the HCV E2 gene hypervariable region (HVR) between the two infants and between the infants and their mothers within the two families. The HCV strains initially detected in infant 1 were identical to those found in her mother, while the HCV strains initially detected in infant 2 were very different from those of her mother. The mutation rate of HVR-1 proteins was higher in mother 1 than in mother 2, but was 1.6 to 2-fold higher in infant 2 than in infant 1 during the follow-up period. Serum ALT levels or serum HCV-core protein activity did not correlate with the mutation rates of HVR-1 proteins in either infant. However, the mutation rate of HVR-1 proteins significantly increased from 6 months of age in both infants, with concomitantly increased serum HCV antibody (anti-HCV) levels.

[Identification of mutation of the X-linked juvenile retinoschisis gene].

OBJECTIVE: To elucidate the pathogeny of X-linked retinoschisis(XLRS) and evaluate its value in direct gene diagnosis. METHODS: Polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP) assay was performed to examine exons of XLRS1 gene in six unrelated retinoschisis cases (3 families and 3 sporadic cases), and fragments with a mobility shift were sequenced to identify the mutation. The deletion mutations were further identified by Southern blotting analysis. RESULTS: Three deletions that eliminate exon 1, exon 2, and exon 3 were found in 4 patients of one family. There was severe effect of the mutation in the coding region. Three kinds of mutations were found in exon 4: Glu72Lys, Glu72Gln, Gly70Ser. CONCLUSION: XLRS is caused by mutation of the XLRS1 gene. The finding helps establish a fast and effective direct diagnosis.

[Cloning the differentially expressed genes in the retina of rds mouse during the development of retinitis pigmentosa].

OBJECTIVE: To clone the differentially expressed genes in the retina of rds mouse (the animal model of congenital retinitis pigmentosa) during the disease development. METHODS: The retinal mRNA of rds mouse during the development of retinitis pigmentosa was analyzed by the mRNA differential display. The differentially expressed mRNA fragments were cloned and sequenced. RESULTS: There was obvious difference of gene expression between rds mouse and the control during the development of retinitis pigmentosa. Five differentially expressed bands were cloned and sequenced. One of those had 86% identity (132/154) with the sequence of the human cDNA DKFZp434D1227 from adult testis in GenBank, which was submitted lately (15-Oct-1999) and without much information. The other had lower identity with the sequences in GenBank. A highly expressed clone in the rds mouse on postnatal day 25 had the same length as another clone in the normal on postnatal day 37, which was not expressed in the rds mouse on day 37. The sequences of the two clones were identical in all but two base pairs. CONCLUSION: These results indicate that there are a lot of novel differentially expressed genes in the chronic processing diseases, such as retinitis pigmentosa.

[Establishment of suc2 signal sequence trap system].

The secretion of proteins depends on the signal peptide located to the N-terminal of the protein precursor. We established a genetic system in yeast to screen cDNA library for the signal peptide encoding sequences. To do it, we mutated genomic suc2 gene (encoding yeast invertase) of EGY48 by one-step gene disruption method, and got yeast cell lines without invertase expression (EGY48-delta suc). To get vector for library screening, we inserted suc2 gene encoding mature peptide of invertase downstream to yeast promoter P-ADH1, and multiple cloning sites for insertion of library is between suc2 and P-ADH1. EGY48-delta suc transformed with the vector can grow on the medium with glucose as carbon source, but not on the medium with raffinose. Signal peptide of suc2 and alpha chain of human interleukin-2 was fused in frame to suc2 gene, then the two resulting vectors were transformed into EGY48-delta suc, all the transformants can grow in the medium with either raffinose or glucose as carbon source. Hence, the system established here can discern cDNA encoding signal peptide from the one not encoding signal peptide.

The principle and technique of using Chinese drugs in the treatment of hypertension.

Differential use of Chinese and western drugs, and combination of traditional theory of materia medica with the results of modern pharmacological research can be regarded as a general principle in the treatment of a disease, while combination of lowering blood pressure with preventing and treating complications is a specific method in the treatment of hypertension. As to the use of drugs based on differentiation of diseases and/or symptoms, drugs which have the action of relieving symptoms but also reducing blood pressure should be used as the first choice. For those drugs which have the action of relieving symptoms but can not lower blood pressure should not be used or should be used as less as possible. Drugs with action of raising blood pressure are contraindicated. When patients have the symptoms of hyperactivity of yang resulting from yin deficiency, Huang Bo ([symbol: see text] Cortex Phellodendri) and Zhi Mu ([symbol: see text] Rhizoma Anemarrhenae) are used together to enhance the effects of both nourishing yin to reduce pathogenic fire, and reducing blood pressure. This practice conforms to the TCM theory and also to the modern pharmacological research.

[Effect of shenfukang on nephropathy rats induced by adriamycin].

UNLABELLED: Adriamycin-induced nephropathy (AIN) model in rats (mainlining 7.5 mg/kg avoirdupois adiramycine) was adopted to explore the effect of Shenfukang (SFK). The results showed that SFK could remarkably reduce the content of urine protein, cholesterol, creatinine and urinary nitrogen in serum, MDA in serum and renal cortex of AIN model rats. And SFK also could increase the content of album and golbulin in serum, the activities of SOD in serum and renal cortex of AIN model rats. CONCLUSION: SFK played an important role on AIN.

[Experimental and mechanism analyses on HCl emission control during PVC combustion in fixed beds].

Incineration of chloridated plastic (PVC) in solid wastes contributes much to HCl pollutant in air environment. The characteristics of HCl emissions and reduction during PVC combustion were conducted in a nearly isothermal, externally heated, horizontal quartz-tube reactor. With increasing temperature from 700 to 900 degrees C, the conversion of HCl increased leniently from 82.5% to 88.3%. High excess air factor enhanced a small amount of HCl existing in form of Cl2. The calcium-based sorbents including CaCO3, Ca(OH)2, Ca(CH3COO)2 exhibited high HCl capture efficiencies from 68% to 79%, while magnesium-based sorbent less than that of 3%. Influences of operational variables, i.e., size of sorbent, Ca/Cl molar ratio and combustion fume compositions, on the HCl reduction were discussed. From a point of view of equilibrium constant of dechloridization reaction the experimental results were explained in detail.

[Effect of rare earth elements on the seedling ratio of crops].

The effects of rare earth elements(REEs) on the relative seedling ratio of three crops(rice, rape and soybean) in three soil(red soil, yellow fluvo-aquic soil and yellow cinnamon soil) were studied according to OECD method, and the LC50 were obtained. Toxicity effect of REEs on rice was minimum among the crops tested. The toxicity on crops in yellow cinnamon soil was lower, whereas on soybean in yellow fluvo-aquic soil and on rape in red earth were higher.

[Effects of metformin on hyperinsulinemic anovulation].

OBJECTIVE: To investigate the effects of metformin on serum testosterone (T) levels and ovulation in hyperinsulinemic women. METHODS: Twenty-nine patients with hyperinsulinemic anovulation were treated with metformin for 12 weeks. Among them 19 had history of clomiphene(CC) treatment with 11 cases failure. CC was given again after one month of metformin treatment. Serum T, luteinizing hormone, follicle-stimulating hormone, insulin(Ins) levels were measured after 4 weeks of treatment. Basic body temperature was also observed. RESULTS: Serum T and Ins levels declined significantly after 4 weeks of metformin treatment [(2.7 +/- 1.6) nmol/L Vs (1.9 +/- 1.0) nmol/L, (49.2 +/- 34.1) mU/L Vs (25.3 +/- 23.9) mU/L, P < 0.05, P < 0.05 respectively]. Eight cases ovulated spontaneously and 8 cases with history of CC failure became ovulatory during CC treatment. CONCLUSIONS: Metformin is effective for suppressing serum T levels and improving the outcome of ovulation.

[Imaging diagnosis of intestinal tuberculosis].

OBJECTIVE: To assess significance and limitation of CT and radiological gastrointestinal examination in the diagnosis of the intestinal tuberculosis. METHODS: 22 cases of intestinal tuberculosis proven pathologically were analyzed. CT scans and radiological examination were performed (barium meal examination in 18 cases, intestinal or colonic double contrast examination in 10 cases and hypotonic duodenography in 3 cases). RESULTS: Sensitivity and specificity of diagnosing the intestinal tuberculosis with CT were inferior than that with radiological examination. Lesions in small intestine (not including ileocecal region) were not easily identified by CT scan. However, CT scan was more preferable in identifying abdominal tuberculosis other than intestinal tuberculosis, especially typical tuberculous lymphadenopathy. Intestinal radiological examination was of great significance for the assessment of the status of intestinal tuberculosis by demonstrating mucosal alterations, morphologic appearance of ulcer, bowel deformation, and the involvement of lesion and fissures. CONCLUSIONS: Gastrointestinal radiological examination and CT scans possess their diagnostic value, but have limitation in diagnosis of intestinal tuberculosis. CT scans combined with gastrointestinal radiological examination further improve the accuracy of diagnosis.

[Distinguishing apoptotic cells from DNA strand-broken cells by comet assay].

Apoptosis of immature thymocyte of mice induced by dexamethasone and DNA strand-broken cells induced by K2Cr2O7 were tested by comet assay. The results showed that the morphology of apoptotic cells assayed by comet was special. Under the same condition, the time of apoptotic cells running on electrophoresis was shorter than DNA-broken cells. The length of comet tail of DNA strand-broken cells increased in a dose-response relation with the increase of K2Cr2O7, whereas, the apoptotic index increased with the dosage of dexamethasone. While DNA damage was repairing, apoptosis continued to process. The DNA strand-broken cells could be obviously seen while DNA damage was repairing. It was suggested that comet assay could be used to detect DNA strand break sensitively, but also could distinguish different grades of DNA damage, which is helpful for studies on the etiology, progressing and outcome of cancer.
