Purification and biophysical properties of acute haemorrhagic conjunctivitis virus.

The buoyant density of acute haemorrhagic conjunctivitis virions labeled with either [(3)H]uridine or [(3)H]leucine was 1.34 g/ml in CsCl and 1.25 g/ml in sucrose. RNA extracted from the virions gave a sedimentation coefficient of approximately 34S in sucrose, and was found to be sensitive to RNase. Molecular weight of RNA was calculated to be 2.5 x 10(6) using poliovirus RNA for reference.

Enterovirus type 70: the etiologic agent of pandemic acute haemorrhagic conjunctivitis.

A new enterovirus, now classified as enterovirus type 70, was isolated from the conjunctiva of patients with acute haemorrhagic conjunctivitis during the 1971 epidemics that occurred in Japan, Singapore, and Morocco. These epidemics were parts of a pandemic involving Africa (Algeria, Ghana, Morocco, Nigeria, and Tunisia), Asia (Cambodia, China (Province of Taiwan), Hong Kong, India, Indonesia, Japan, Malaysia, the Philippines, Singapore, and Thailand), and England during 1969-71. A representative strain from each of the three epidemic areas was studied cooperatively. The strains exhibited the physicochemical characteristics of enteroviruses. Cross-neutralization tests showed that these viruses were distinct from all known human enterovirus immunotypes, but that they were antigenically closely related. The human origin of the viruses was demonstrated by the appearance of homologous neutralizing antibodies during convalescence in patients with acute haemorrhagic conjunctivitis.

Effect of a modified kaolin treatment on serum immunoglobulins.

After kaolin treatment of fetal rabbit serum, 7S antibody titers were reduced more than 19S titers. This reduction was less when the kaolin treatment was performed at pH 9.0 than when it was performed at pH 7.3. A modification of the kaolin treatment of sera for use in the hemagglutination-inhibiting antibody titration, in which the hemagglutination reaction is performed at a neutral pH, is recommended. The advantage of the modified method is that adsorption of immunoglobulins to kaolin is minimized when serum is treated at a lower dilution with pH 9.0 kaolin, followed by reduction of the pH of the supernatant fluid to neutrality with a "serum adjusting diluent." When the serum was diluted with physiological saline before kaolin treatment, a great decrease in serum immunoglobulin concentrations occurred. This decrease was found to be less in the modified kaolin treatment than in the conventional pH 7.3 kaolin treatment.
