High-performance liquid chromatography of glycosides on a new type of hydroxyapatite column.

High-performance liquid chromatography on a column of newly developed hard spherical hydroxyapatite was applied to the separation of a variety of glycosides of tri- and diterpenes including Ginseng saponins and Stevia sweet glycosides, affording satisfactory results by using a simple solvent system, aqueous acetonitrile. This normal phase chromatography is superior to chromatography on a silica gel column for the separation of water-soluble glycosides.

Effect of 1 alpha-hydroxyvitamin D3 on osteoporosis induced by immobilization combined with ovariectomy in rats.

The effect of low-dose 1 alpha-hydroxyvitamin D3 [1 alpha(OH)D3] on experimental osteoporosis in rats induced by a combination of hemicordotomy and ovariectomy was evaluated. 1 alpha(OH)D3 (0.1 microgram/kg 1 alpha(OH)D3 three times a week) increased the femur score and photo-density. The administration of 1 alpha(OH)D3 also significantly increased the dry weight and ash weight of the bone. Histomorphometry revealed a significant decrease of bone resorption by the administration of 1 alpha(OH)D3. Significant decreases in 45Ca accretion rate and 14C-proline uptake were also observed. Bone formation in the 1 alpha(OH)D3 treated group was thought to decrease following the decrease of bone resorption.

Scintigraphy in nontraumatic femoral head necrosis.

We performed serial 99mTc-MDP bone scintigraphy of nontraumatic femoral head necrosis in 14 hips of 10 patients. The uptake of isotope was decreased in the very early stage, and either variable or increased at the stage when symptoms appeared. A variable uptake changed to increased uptake in 3 of 10 patients, but there was no hip in which an increased uptake changed to a variable uptake. No relationship was demonstrated between the scintigraphic appearance of the femoral head and the pain and function of the hip.

[Bone modeling in experimentally induced osteoporosis in rats].

Osteoporosis was experimentally induced in rats by ovariectomy (OVX), surgical excision of the sciatic nerve (NX), unilateral cordotomy (CX) and their combined operations. Each experimental group was sacrificed 12 or 24 weeks after the operation. Microdensitometrical findings, bone weight and the amount of tetracycline absorbed into the bone were examined and were compared with the control (sham op.) in order to study the bone modeling dynamics. The rats which underwent CX combined with OVX were given 1 alpha (OH)D3 and the effects of the drug were examined in a similar manner. It was concluded that bone formation decreased in OVX, that bone formation decreased and bone resorption increased simultaneously in NX, and that both bone formation and bone resorption increased in CX. 1 alpha (OH)D3 was considered to inhibit the progression of osteoporosis.

Age-related changes in bone mass in the senescence-accelerated mouse (SAM). SAM-R/3 and SAM-P/6 as new murine models for senile osteoporosis.

Age-related changes of the femoral bone mass in several strains of the senescence-accelerated mouse (SAM) were investigated. Microdensitometrically, all strains exhibited essentially the same patterns of age changes, that is, bone mass corrected by the diameter of the shaft reached the peak value when the mice were 4 or 5 months of age and then fell linearly with age up to over 20 months of age. Two strains, SAM-R/3 and SAM-P/6, which originated from the same ancestry on pedigree, had a significantly lower peak bone mass than other strains (SAM-R/1, SAM-R/2, SAM-P/1, and SAM-P/2). On the other hand, the strains with a low peak bone mass had the same rate of decrease as other strains. Mineral and collagen contents per dry weight of bone showed little difference among the strains. Histologic studies of tibia, femur, and lumbar spine revealed that the osteopenia was not due to osteomalacia but, rather, to osteoporosis. The elderly mice in these two strains were prone to fracture, thus should be important models for study of senile osteoporosis seen clinically.

Probable assignment of soluble isocitrate dehydrogenase (IDH1) to 2q33.3.

Gene dosage effects for soluble isocitrate dehydrogenase (IDH1) were investigated in four unrelated cases with abnormalities involving the long arm of chromosome 2. Case 1 was trisomic for 2q33.3----qter, Case 2 monosomic for 2q33.3----q35, Case 3 trisomic for 2q11.2----q24.2, and Case 4 monosomic for 2q23----q24.2. These abnormalities were de novo except in Case 1, where trisomy 2q resulted from a maternal translocation. The red cell IDH1 levels were significantly reduced in Cases 1 (41.4% of normal value) and 2 (51.9%), while they were normal in Cases 3 and 4. The low IDH1 level also in the father of Case 1 (43.6%), together with the common electrophoretic phenotype of IDH1 in red cells as well as leukocytes, led us to suppose that Case 1 was really heterozygous for common and probable null alleles, and that the IDH1 gene locus could be excluded from 2q33.3----qter. On the other hand, normal IDH1 values in the parents of Case 2 were consistent with the hemizygosity for this locus in Case 2. The results suggested that the IDH1 locus could be assigned to the 2q33.3 band, especially the proximal portion of it.
