Hormone levels in women after hysterectomy.

The serum levels of FSH, LH and estradiol-17 beta (E2) were determined in 110 women aged between 38-48 years who had been hysterectomized 2-10 years previously and were compared with a control group (n = 112). In hysterectomized women both FSH and LH levels were higher than in controls during the whole 12 year period. These differences were significant up to 43 years of age. The hypergonadotropism in hysterectomized women correlates with the higher incidence of climacteric symptoms reported in the literature.

Human immunodeficiency virus infection: affinity-mediated immunoblot detects intrathecal synthesis of oligoclonal IgG specific for individual viral proteins.

An affinity-mediated immunoblot (AMI) is introduced, able to identify virus-specific, oligoclonal IgG in unconcentrated cerebrospinal fluid (CSF) of patients infected by HIV. CSF specimens are isoelectrically focused in agarose gels and blotted to nitrocellulose (NC) strips coated with E. coli recombinant HIV-specific proteins. Virus-specific antibody clones transferred to the filter are detected by solid-phase enzyme immunoassay (EIA). Only 0.5 micrograms IgG is sufficient to demonstrate the clonal distribution of antibodies specific for an individual HIV protein. Intrathecal synthesis of oligoclonal antibodies is seen by AMI, comparing equal quantities of serum- and CSF-derived IgG. Because of its excellent sensitivity and specificity, AMI detects autochthonous HIV-specific IgG synthesis even when comparative EIA fails. Because multiple specimens can be processed within a few hours by AMI, this technique is recommendable for routine diagnosis.

DNA diagnostics--molecular techniques and automation.

Molecular biology has revolutionized the understanding of many aspects of human disease. Ongoing developments in DNA diagnostics--the analysis of disease at the nucleic acid level--will soon provide automated, rapid, and inexpensive analyses for DNA or RNA sequences associated with genetic, malignant, and infectious diseases. DNA diagnostics will also facilitate the identification of disease-associated genes at birth, thus creating new opportunities for preventive medicine.

A ligase-mediated gene detection technique.

An assay for the presence of given DNA sequences has been developed, based on the ability of two oligonucleotides to anneal immediately adjacent to each other on a complementary target DNA molecule. The two oligonucleotides are then joined covalently by the action of a DNA ligase, provided that the nucleotides at the junction are correctly base-paired. Thus single nucleotide substitutions can be distinguished. This strategy permits the rapid and standardized identification of single-copy gene sequences in genomic DNA.

Class III human liver alcohol dehydrogenase: a novel structural type equidistantly related to the class I and class II enzymes.

The primary structure of class III alcohol dehydrogenase (dimeric with chi subunits) from human liver has been determined by peptide analyses. The protein chain is a clearly distinct type of subunit distantly related to those of both human class I and class II alcohol dehydrogenases (with alpha, beta, gamma, and pi subunits, respectively). Disregarding a few gaps, residue differences in the chi protein chain with respect to beta 1 and pi occur at 139 and 140 positions, respectively. Compared to class I, the 373-residue chi structure has an extra residue, Cys after position 60, and two missing ones, the first two residues relative to class I, although the N-terminus is acetylated like that for those enzymes. The chi subunit contains two more tryptophan residues than the class I subunits, accounting for the increased absorbance at 280 nm. There are also four additional acidic and two fewer basic side chains than in the class I beta structure, compatible with the markedly different electrophoretic mobility of the class III enzyme. Residue differences between class III and the other classes occur with nearly equal frequency in the coenzyme-binding and catalytic domains. The similarity in the number of exchanges relative to that of the enzymes of the other two classes supports conclusions that the three classes of alcohol dehydrogenase reflect stages in the development of separate enzymes with distinct functional roles. In spite of the many exchanges, the residues critical to basic functional properties are either completely unchanged--all zinc ligands and space-restricted Gly residues--or partly unchanged--residues at the coenzyme-binding pocket.(ABSTRACT TRUNCATED AT 250 WORDS)

Cystic fibrosis: typing 89 German families with linked DNA probes.

Three hundred and ninety-two subjects from 89 German families were typed for restriction fragment length polymorphisms (RFLPs) detected by the probes pmetH, pmetD, pJ3.11, KM19, and XV2c known to be tightly linked to the cystic fibrosis (CF) gene. The analysis of the predictive value of this typing in individual CF families indicates that the combined use of these probes provides a powerful diagnostic system for both carrier detection and prenatal diagnosis. In 45 families the complete haplotype including all RFLPs was available. Of them 41 (91.1%) were fully informative and 4 were partly informative.

[Receptor-binding capacity for estrogens and progesterone in the uterine cervix during the menstrual cycle and in the postmenopausal period].

In biochemical studies it was found that the binding capacity of cytoplasmatic estrogen and progesterone receptors in the uterine cervix undergo characteristic changes during the menstrual cycle. The highest estrogen binding values were observed by the eighth day, after which there was a significant drop. The highest progesterone binding values were around the twelfth day, followed by a significant drop after ovulation. In sexually mature subjects the estrogen receptors predominated in the mucosa and the progesterone receptors in the myometrium. Therefore, there is obviously a parallel to the receptor content in the corpus uteri. The maximum estrogen and progesterone binding capacities were in each case ascertained some days before the maximum E2 and Pr values were attained in the serum. Hence, the estrogen receptors have already built up in the uterine cervix before optimal functional activity, measured by the cervix score, has been attained. Anti-estrogens given at the beginning of the cycle can disturb this sequence. The high estrogen receptor content in the endometrium at the beginning of the cycle creates the necessary conditions for the explosive, proliferative gland growth around this time, even though the serum estrogen concentration is still relatively low. In the postmenopause the estrogen binding capacity in the uterine cervix was higher than during sexual maturity, while the reverse was the case with the progesterone binding capacity.

[Informing angiography patients. Results of a survey and critical comments].

100 patients subjected to angiography have been interviewed about their knowledge of the previous diagnostic procedure. The group having cerebral angiography keeps more details and risks in mind than a group having an angiography of the limbs. No patient regards the informed consent as an unnecessary affair and the majority expects to be examined by the informing physician. The radiologist therefore has the opportunity to establish a trustful relationship to the patient and to master his roll as a physician.

Creation of a strain of genetically obese-hypertensive rats.

A strain of genetically obese-hypertensive rats (SHR-fa/fa) was created by transferring the fatty/fa gene of hyperlipaemic obese non-inbred rats into the genome of an SHR inbred strain by five successive crossings of SHR-fa+ brother-sister matings. SHR-fa/fa rats were heavier than their SHR littermates. They showed a severe hypertension, their systolic arterial blood pressure being higher than that previously found in genetically hypertensive rats. Their blood glucose content was not significantly different from that of their SHR littermates but their plasma triglycerides were increased by more than 500 per cent. While obesity and hypertension occurred from the 5th week following the rat's birth, the increase in blood triglycerides was manifest earlier.

Acetylated N-terminal structures of class III alcohol dehydrogenases. Differences among the three enzyme classes.

The protein chains of mammalian alcohol dehydrogenases typically lack free alpha-amino groups. The blocked N-terminal regions of the class III type of the rat (ADH-2), human (chi chi) and horse enzymes were isolated by digestions with proteases, and characterized by mass-spectrometry supplemented with chemical analysis of the peptides and their redigestion fragments. Results were confirmed by synthesis of the corresponding peptides, followed by chromatographic comparisons of the native and synthetic products. The N-terminal regions of the three class III alcohol dehydrogenase subunits are homologous but differ from the class I and II enzymes in both the exact start position and the amino acid sequence, which suggests that different N-terminal structures are typical for each of the three classes.

Microdissection and microcloning of the long arm of human chromosome 7.

DNA-fragments from the region of the long arm of human chromosome 7 to which the CF-locus has been mapped recently were isolated by microdissection and microcloning. We developed a new fixation procedure resulting in inserts of 1.0-7.0 kb in length with a mean value of 2.9 kb. Regional mapping of three clones on 7q was carried out by the use of different hybrid cell lines containing fragments of human chromosome 7.

[The menstrual cycle after hormonal growth inhibition in girls].

Up to 1979 growth was inhibited with hormones in 43 girls in puberty with constitutional gigantism. The average reduction in size as compared to the calculated final body height was 7.5 cm, treatment having been started at a skeletal age of 12.3 years and with a treatment duration of 1.4 years. In over 90% of the cases, the first spontaneous menstruation after termination of therapy occurred within three months. During the first two years, cycles were likewise regular in 90% of the cases, with a duration of 4 +/- 1 weeks. Ovarian function in adolescence was thus found not to be impaired, but rather to be more stable as compared to the total population. It may be concluded from this that the preconditions for future fertility can be regarded as favorable.
