Study of thymidylate synthetase-function by laser Raman spectroscopy.

The Laser-Raman spectra of thymidylate synthetase have been obtained with 488 nm excitation from an argon ion laser. Raman bands observed in the range 600-800 cm-minus-1 have been assigned to functional groups of constituent amino acids. The band positions and intensities in the Amide I (1600-1700 cm-minus-1) and Amide III (1200-1300 cm-minus-1) regions, suggest that the enzyme is a mixture of alpha-helical and unordered conformations. Low levels of beta-structure cannot be excluded. The spectra of the ternary complex formed by reacting thymidylate synthetase with (+)-L-methylenetetrahydrofolate and fluorodeoxyuridylate reveals a new band at 1618 cm-minus-1 assigned to the C=N stretching vibration. This band may be due to formation of dihydrofolate or an iminium ion. The overall secondary structure of thymidylate synthetase does not change on formation of the ternary complex. However, the spectrum of the complex indicates local changes in groups such as ionized carboxyl (1400 cm-minus-1), tryptophan (1003 cm-minus-1) and CH-3, CH-2 deformation modes (1440-1470 cm-minus-1).

Cyclin nucleotide binding sites of the smooth endoplasmic reticulum from normal and neoplastic liver in the rat.

The studies are presented which demonstrate that smooth endoplasmic membrane of normal liver has a single apparent binding site for cAMP with a KD of 0.6 X 10(-8) M. In contrast to this, however, cyclic AMP binding to the intracellular membrane of hepatoma 7800 exhibit two binding sites; the binding constant of one site on the tumor membrane is comparable to that of the normal liver whereas the value of the second intrinsic association constant differ by a factor of 10. It is suggested that there may be an association between abnormal cyclic nucleotide metabolism and the intracellular membrane modulation of the expression of genetic information in normal and neoplastic cells.

Growth hormone and liver mitochondrial protein synthesis effect of cortisol and estradiol.

The effect of cortisol and estradiol on the stimulation by growth hormone of liver mitochondrial protein synthesis in hypophysectomized rats has been studied by measuring radioactive leucine incorporation in vivo and in vitro. Estradiol inhibited the stimulatory effect of growth hormone on incorporation into mitochondrial and microsomal proteins. Cortisol blocked the stimulatory effect of growth hormone on mitochondrial protein synthesis. The stimulatory effects of cortisol and growth hormone on microsomal incorporation were not additive. Gain in body weights corresponded with their effects on mitochondrial and microsomal protein synthesis. These results show that steroid hormones may act as regulators of the growth hormone effect.

Tryptophane metabolism in bronchial asthma.

Seventeen out of 21 children with severe asthma had an abnormal increase in urinary Kynurenic (KA) and Xanthurenic (XA) acid following oral tryptophane administration. In a second study 11 asthmatic children had significantly greater increases in XA and KA than nine normal healthy children. A micro-assay of KA and XA by column chromatography was performed. The data suggest the presence of a partial block in the metabolism of tryptophane in some children with bronchial asthma.
